1.Effect on the expression of proto-onogene c-mpl in megakaryocytes line-HEL by interleukin-13

AIM:To investigate the relevance of the proliferation of megakaryocytic cell line-HEL stimulated by the recombinant human interleukin-13 (IL-13) to the expression of pro-oncogene c-mpl in HEL cells. METHODS: MTT colorimetric assay and reverse transcrition polymerase chain reaction (RT-PCR) are separately used in this study to observe the effect on the proliferation of HEL cells and the expression of c-mpl mRNA in HEL cells by rhIL-13. RESULTS: RhIL-13 stimulated the proliferation of HEL cells and upregulated the expression of c-mpl mRNA in HEL cells. CONCLUSION: Our results suggest that rhIL-13 stimulated the proliferation of HEL cells and provide the evidence that its mechanism is partly because of increasing the pro-oncogene c-mpl expression in HEL cells.
2.Accuracy of intraocular lens power prediction by using six different formulas

Objective To compare the accuracy of intraocular lens(IOL)power calculations by using 6 formulas(SRK-T,Haigis,Binkhorst-Ⅱ,Hoffer Q,Holladay-1,SRK-Ⅱ)in different axial lengths of the eye,in order to improve the accuracy of predicating IOL powers for patients after cataract surgery.Design Retrospective comparative clinical analysis.Participants 169 patients(169 eyes)with age-related cataract underwent regular phacoemulsification and posterior chamber IOL implantation.Methods 169 eyes with cataract were divided into five groups according to the axial lengths:AXL≤22mm,22mm28.4mm.The axial lengths and keratometry of the 169 eyes were measured with A-mode ultrasound before operation,and the actual postoperative refraction was measured with the methods of computer and retinoscopy optometry when patients could gain best corrected visual acuity one month after surgery or later.Inputting the axial lengths,keratometry and IOL powers into A-mode ultrasound,the predicted postoperative refraction was calculated with SRK-T,Haigis,Binkhorst-Ⅱ,Hoffer Q,Holladay-1,SRK-Ⅱ formulas,and the differences of predicted and actual postoperative refractions in different axial lengths were compared.Main Outcome Measures The absolute value with the differences of the predicted and actual postoperative refractions,i.e.absolute predicted errors.Results(1)Comparing the difference of predicted and actual postoperative refraction,there was no statistical significance between the five formulas in the groups of axial length(AXL)≤22mm,22mm28.4mm,but the average absolute predicted error of Haigis and SRK-T formulas is significantly smaller than SRK-Ⅱand Binkhorst-Ⅱ formulas.Conclusions In existing sample size,there was no obvious influence on postoperative refraction when using other five formulas except Binkhorst-Ⅱ.But in the extremely long AXL group,Haigis and SRK-T formulas have some ascendancy over the other formulas.
3.Application of Edaravone,Ozagrel sodium combined nursing intervention for patients with cerebral infarction in speech and limb rehabilitation

Objective To investigate the clinical effect of Edaravone,Ozagrel sodium combined nursing intervention for patients with cerebral infarction in speech and limb rehabilitation. Methods 116 patients with cerebral infarction methods met inclusion criteria were randomly divided into control group (n=58) and study group (n=58). Two groups were given treatment with Edaravone andOzagrel sodium,the control group was given routine mode nursing, the study group was given early targeted rehabilitation nursing. Compare patient language, physical recovery and daily life recovery between control group and study group after nursing. Results Contrast group and the study group after the nursing intervention, rehabilitation effect of significant efficiency were 48.28% and 75.86%, the difference between groups was statistically significant (P<0.05);nursing before the implementation of 2 groups of patients with speech and limb function showed different degrees of impairment, nursing after the implementation of the 2 groups of patients with ABC score and Fugl-Meyer score were significantly increased, the difference between groups was statistically significant (P<0.05), and the patients in the study group ABC the scores of Fugl-Meyer and enhance the rate higher, the difference between groups was statistically significant (P< 0.05); the study group ADL Barthel index was significantly better than the control group, the rank sum test (Z=3.063, P=0.002 ) that has a statistically significant difference between the two groups. Conclusion Edaravone,Ozagrel sodium combined rehabilitation nursing intervention can effectively improve the speech and limb function recovery, and improve the quality of life of patients, nursing satisfaction, is worthy of clinical application.
4.Study on Isolation of Pathogenic Dematiaceous Fungi from Wild Toads

Objective To compare the carriage of dematiaceous fungi in wild toads from both en-demic area and non-epidemic area of chromoblastomycosis.Methods A total of354and45wild toads(Bufo Chinese)were caught from two counties of Shandong Province,Zhangqiu county,an endemic area of chromoblastomycosis,and from Laiyang county,an area where the infection of dematiaceous fungi in humans has not been found,respectively.The toads were killed and the suspension of the internal organs were used to isolate dematiaceous fungi.Results Eleven species including162strains of dematiaceous fungi were iso-lated from151of354(42.7%)toads from the endemic area.The isolated strains consisted of49Phialopho-ra verrucosa,33Exsophiala,19Phialophora(nonpathogenic),19E.jenselmei,12Cladosporium carrionii,3suspected of C.carrionii,7Rhinocladiella sp.,5Fonsecaea pedrosoi,4C.trichoides,3E.spinifera,1C.mi-nourae,4unidentified.Only4nonpathogenic dematiaceous fungi were isolated from4of45(8.9%)toads obtained from non-epidemic area.Conclusions Varieties of pathogenic dematiaceous fungi,especially C.carrionii,are isolated from wild toads in endemic area of chromoblastomycosis,while none from non-endemic area.The results suggest that the colonization of pathogenic dematiaceous fungi in wild toads may be associa-ted with endemicity of chromoblastomycosis.
5.Difference of gene expression in bone marrow mononuclear cells between a chronic myeloid leukemia patient and a healthy person

BACKGROUND: Chronic myeloid leukemia (CML) is characterized by the clonal expansion of hematopoietic stem cells. Without effective treat ment, individuals in the indolent, chronic phase (CP) of CML will undergo blast crisis (BC), the prognosis for which is poor. Therefore, it is important to clarify the mechanism underlying CML from a whole-genome perspec tive. OBJECTIVE: To investigate the gene expression profile of bone marrow mononuclear cells from CML with Applied Biosystems Expression Array System.DESIGN: Observation and controlled analysis.SETTING: Institute of Gene Engineering, Southern Medical University PARTICIPANTS: Samples of two cases of bone marrow (a chronic myeloid leukemia patient and a healthy person).METHODS: This experiment was conducted at the Institute of Gene Engineering, Southern Medical University from October 2004 to September 2005.The total RNAs were extracted and purified from bone marrow mononuclear cells derived from a CML patient and a healthy person. mRNAs were purified using an oligo (dT)-cellulose mRNA purification kits and labeled using reverse transcription, in vitro transcription (RT-IVT), then hybridized with microarray. Gene expression differentiation of the bone marrow mononuclear cells were examined by ABI 1700 Chemiluminescent Microarray Analyzer. Reproducibility of microarray results was assessed by comparing data sets obtained from the same sample and analyzed by two different arrays.MAIN OUTCOME MEASURES: ①Assessment of quality of total RNA and labled cRNA. ②Reproducibility of microarray. ③ Hybridization of array.④Results of semi-quantitative reverse transcription-polymerase chain reaction RESULTS: ①Using statistical data analysis tools, we identified 6 706 genes that were up- or down-regulated in CML patient compared with the healthy person. In these genes, we found that 17 genes were up-regulated while 51 genes were down-regulated among 68 genes closely related to CML. ②most differentially expressed genes in C/EBPalpha mediated path way and CD40L signaling pathway had reduced expression. ③Good repro ducibility of microarray was confirmed by analysis of correlation and detection concordance in technical replicates. The correlation coefficient of the detectable probe in technical replicates was 0.991 for the CML patient and 0.988 for the healthy person. ④The results of semi-quantitative RT-PCR experiments supported the reliability of our microarray analysis.CONCLUSION: By comparing expression patterns of CML with those of the healthy person, we identified a large number of genes that, were up- or down-regulated in CML patients. These data should provide useful information for finding candidate genes whose products might serve as molecular targets for treatment of CML patients.
6.Inhibitory effects of lobaplatin on human ovarian cancer and its regulation of apoptosis-related genes in nude mice

Objective To explore the inhibitory effects of lobaplatin and cisplatin and their regulation of apoptosis-related genes in ovarian cancer cells in nude mice.Methods SKOV3 cells were implanted into nude mice.In monotherapy treatment study,the nude mice bearing human SKOV3 cells were randomly divided into control,lobaplatin,and cisplatin groups,with 7 mice in each group.The mice in each group were received corresponding treatment.The volume of tumor and the weight of nude mice were measured three times per week,respectively.Tumor inhibitory rate was calculated.The protein expressions of bax and bcl-2 were detected by flow cytometry.Results The growth inhibitory rate was 47.2% in lobaplatin group and 42.8% in cisplatin group,without significant difference between two groups (P > 0.05).The expression of bcl-2 was decreased but the bax was increased in lobaplatin and ciaplatin groups compared to the control group.Conclusions Lobaplatin can significantly inhibit the growth of ovarian cancer cells,induce apoptosis by down-regulation of bcl-2 and up-regulation of bax.
7.Absolute Quantification of Human Growth Hormone by Isotope Dilution-High Performance Liquid Chromatography-Mass Spectrometry

A method was developed for the quantification of human growth hormone ( hGH ) by protein purification and isotope dilution-high performance liquid chromatography mass spectrometry ( HPLC-IDMS ) . The hGH was purified and fractionated by fast protein liquid chromatography ( FPLC ) , then hGH molecular weight was accurately determined by Fourier transform ion cyclotron resonance mass spectrometer ( FTICR-MS). The purified hGH was hydrolyzed and the separation was performed on an KINETEX C18 column (150 mmí2 mm I. D. , 2. 6 μm) with water ( containing 0. 1% TFA) and acetonitrile isocratic solution as the mobile phase at a flow rate of 0 . 2 mL/min and 40℃. The electrospray source was operated in the positive ion mode, and monitored in the multiple reaction monitoring ( MRM) mode. The measured hGH molecular weight by FTICR-MS was only 0. 31 Da difference from theoretical value. Three amino ( proline, valine and phenylalanine) were clearly separated by isocratic elution within 5 min. Under the optimized conditions, the content of hGH was 186 . 80 μg/g with a RSD of 0 . 5%. The detection results of hGH in international comparison by this method were consistent with the reference value, which validated the feasibility of the established method. The developed method is simple, practical, accurate, reliable and reproducible, and can be used for the hGH quantitation of pure hGH CRM to provide reference for the routine detection of hGH.
8.Typing of Resistance Genes of ESBLs from Three Gram-negative Bacilli of Children in Urumqi

OBJECTIVE To study antibiotic-resistant phenotypes and genotypes of ESBLs-producing Escherichia coli,Klebsiella pneumoniae and Klebsiella oxytoca isolated from children with pneumonia in Urumqi,to know the distribution and difference of these three Gram-negative bacilli from 2003 to 2007.METHODS Bacterial strains were identified by VITEK32,ESBLs were detected by confirmatory test recommended by CLSI.Microarray technique was used to determine the genotypes of ESBLs.RESULTS Antibiotic-resistant phenotypes showed ESBLs-producing K.oxytoca decreasing from 84.3% to 35.3%,K.pneumoniae stabling in 50-60%,E.coli increasing from 34.4% to 72.1% during the five years;genotypes indicated there were most of ctx-m-9 and tem+ctx-m-9 in E.coli,tem and shv in K.pneumoniae,and the most of tem+ctx-m-3 in K.oxytoca.CONCLUSIONS There is high percentage of ESBLs production from children in Urumqi;resistant phenotypes and genotypes of ESBLs are different in three Gram-negative bacilli;and must further enhance the regional epidemiology surveillance about ESBLs.
9.Effects of living donor liver transplantation on postoperative delirium, cognitive impairment and prognosis in children with biliary atresia

Objective:To explore the changes of neuron-specific enolase (NSE) and S-100β protein (S-100β) during perioperative period in infants undergoing living liver transplantation and examine the effect of brain injury.Methods:From January 2015 to January 2016 in Department of Anesthesiology First Central Clinical College Tianjin Medical University, study group was composed of forty infants of congenital biliary atresia with an age range of (4-12) months, a body weight of (4-10) kg and American Society of Anesthesiologists (ASA) class Ⅲ/Ⅳ. Another 40 infants undergoing general surgery were selected as control group. In study group, blood samples were harvested from central vein pre-operation (T0), before skin incision (T1), 30 min after anhepatic phase (T2), 1 h of neohepatic phase (T3) and 24h after hepato-reperfusion (T4). In control group, blood samples were collected at pre-operation (T0) and 24 h post-operation (T4). Serum levels of S-100β, NSE, heart rate (HR), mean arterial blood pressure (MAP), central venous pressure (CVP) and bispectral index (BIS) were monitored at T1-4 and end of surgery. All children were assessed by Bayley Scale of Infant Development (BSID) at Day 1 pre-operation and 2/4 weeks post-operation for observing mental and motor development status. The results were described with mental development index (MDI) and psychomotor development index (PDI). Pediatric anesthesia emergence delirium (PAED) was employed for evaluating the severity of delirium during the recovery stage at 30 min and 2/4h post-extubation.Results:In study group, serum levels of S-100β and NSE changed significantly during non-hepatic and neohepatic reperfusion phases. After inferior vena cava occlusion, serum concentrations of S-100β and NSE spiked ( P<0.05) and gradually recovered during neohepatic reperfusion period ( P<0.05). No significant inter-group difference existed in serum S-100β or NSE at T4 ( P>0.05). In study group, as compared with Day 1 pre-operation, MDI/PDI decreased at Week 2 post-operation ( P<0.05) and increased from Month 1 post-operation ( P<0.05). Both MDI and PDI were lower than control group before and at Week 2 post-operation ( P<0.05). MDI/PDI of study group basically reached the preoperative level at Month 1 post-operation ( P<0.05). In control group, no significant difference existed in MDI/PDI at Day 1 pre-operation and Week 2/4 post-operation ( P>0.05). In study group, the delirium rate was up to 30% post-extubation and decreased at 2/4h post-extubation. In control group, the incidence of delirium was low at 30 min and 2/4h post-extubation ( P<0.05). Conclusions:Perioperative evaluations of serum levels of NSE and S-100β are significant for predicting the postoperative onsets of delirium and cognitive impairment in children with living donor liver transplantation.
10.Allocation and rebuilt of vessels of donor pancreas

Objective To explore the technique of allocation and rebuilt of vessels of donor pancreas during pancreas-kidney transplantation.Methods Multiple abdominal organs were successfully retrieved from 40 donors.Results Excision of donor abdominal organs was successful in all the 40 cases.The average warm ischemic time was no 3.2 min,ranging from 2 min to 5 min.No injury occurred in any of the donor organs or vessels.After transplantation,2 patients lost the transplant,one because of abdominal infection,and the other because of venous thrombosis.Conclusion A good trim or rebuilt of donor pancreas vessels is one of the key points for successful combined pancreas-kidney transplantation.
