BACKGROUND:Eucommia ulmoides has a certain osteogenic effect, which can promote the proliferation and differentiation of osteoblasts. However,it is unclear whether Eucommia ulmoides has effects on alveolar bone formation and Wnt/β-Catenin signaling pathway. OBJECTIVE:To investigate the mechanism by which Eucommia ulmoides promotes alveolar bone formation in ovariectomized rats based on the Wnt/β-Catenin signaling pathway. METHODS:Sixty female Sprague-Dawley rats were selected and randomly divided into five groups:blank control group,sham-operation group, model group,low-dose group Eucommia ulmoides group, and high-dose Eucommia ulmoides group, with twelve rats in each group.Osteoporosis animal models were constructed by bilateral oophorectomy in the model group and the low-dose and high-dose Eucommia ulmoides groups. The sham-operation group underwent the same method to remove adipose tissue of equal mass around the bilateral ovaries.Three months after surgery, the low-and high-dose Eucommia ulmoides groups were given 2.1 g/kg/d and 4.2 g/kg/d Eucommia ulmoides by gavage, respectively. The sham-operation group and model group were given the same amount of physiological saline by gavage. After 12 weeks of drug intervention, the changes in alveolar bone mass of rats in each group were observed through Micro-CT;hematoxylin-eosin staining was used to observe the pathological structural changes of alveolar bone in rats;enzyme linked immunosorbent assay was used to detect the expression levels of alkaline phosphatase and osteocalcin in the serum of rats;western blot was used to detect the expression levels of β-Catenin and Frizzled9 receptor proteins in the alveolar bone of rats;and real-time fluorescence quantitative PCR was used to detect the expression of osteocalcin,Runt-related transcription factor 2(Runx2),alkaline phosphatase,β-catenin,and frizzled9 mRNAs in alveolar bone tissues of rats. RESULTS AND CONCLUSION:Compared with the blank control group, bone volume fraction, trabecular number, trabecular thickness, and bone mineral density were reduced in the model group(P<0.05),and trabecular separation was elevated(P<0.05).Pathological observation showed that the arrangement of trabeculae was disordered and irregular, the trabeculae were thinned or broken, and the marrow cavity was enlarged in the model group, with a significant reduction in bone volume;the level of alkaline phosphatase in the serum was increased(P<0.05),and the level of osteocalcin was decreased(P<0.05);mRNA expression of alkaline phosphatase,osteocalcin,Runx2,β-catenin,and frizzled9 were decreased(P<0.05);protein expression of β-Catenin and Frizzled9 was decreased(P<0.05).Compared with the model group, the low-and high-dose Eucommia ulmoides groups showed an increase in bone volume fraction, trabecular number, trabecular thickness, and bone mineral density(P<0.05)and a decrease in trabecular separation(P<0.05).In the low-and high-dose Eucommia ulmoides groups, bone trabeculae were slightly aligned and thickened, with a significant increase in bone mass.Compared with the model group, the serum level of alkaline phosphatase was reduced(P<0.05)and the serum level of osteocalcin was elevated(P<0.05)in the low-and high-dose Eucommia ulmoides groups.Compared with the model group, the mRNA expression of alkaline phosphatase,osteocalcin,Runx2,β-catenin,and frizzled9 were increased in the low-and high-dose Eucommia ulmoides groups(P<0.05).Compared with the model group, the protein expression of Frizzled9 was increased in the low-dose Eucommia ulmoides group(P<0.05),while the protein expression of β-Catenin and Frizzled9 was increased in the high-dose Eucommia ulmoides group(P<0.05).Compared with the low-dose Eucommia ulmoides group, the high-dose Eucommia ulmoides group had a more significant improvement in the above indexes. To conclude,Eucommia ulmoides can effectively promote the alveolar bone formation, and its mechanism of action might be related to the activation of the Wnt/β-catenin signaling pathway.

Chinese Journal of Biologicals 2025;38(01):14-21
Objective To obtain monoclonal antibodies against the glycoprotein Gn of severe fever with thrombocytopenia syndrome virus(SFTSV) based on immune repertoire sequencing and identify the biological characteristics. Methods Using purified SFTSV-Gn as bait, virus-specific B cells were captured from B cell library of immunized female BALB/c mice. Based on 10 × immune repertoire sequencing and bioinformatics analysis, the antibody sequences with potential binding activity were selected and constructed into eukaryotic expression vector pcDNA3.4. The monoclonal antibodies were expressed and purified, and identified for the biological activity. Results Fourteen antibodies were selected from 3 544 captured B cells, and the functional verification showed that 12 antibodies could bind to SFTSV-Gn glycoprotein, among which C9 and 9/10 antibodies had higher binding ability. Western blot, indirect immunofluorescence assay(IFA) and flow cytometry proved that they specifically recognized SFTSV, and the effective concentration was as low as 2 μg/mL.Conclusion The monoclonal antibodies against SFTSV-Gn glycoprotein were successfully obtained, which lays a foundation for the development of rapid diagnostic reagents for SFTSV.

Gastrointestinal tumors (GTs), including colorectal cancer, gastric cancer, liver cancer, pancreatic cancer, and esophageal cancer, are increasing in incidence worldwide and have become one of the major diseases threatening human health. Tumor stem cells (TSCs), an undifferentiated subpopulation within tumor tissues, possess biological characteristics such as self-renewal, multidirectional differentiation, high tumorigenicity, and resistance to radiochemotherapy. They play an important role in the occurrence, progression, recurrence, and metastasis of GTs and have increasingly become a research hotspot in GT treatment. Although modern medicine has made remarkable progress, there remain many problems in therapeutic approaches targeting TSCs. In this context, traditional Chinese medicine (TCM), with its favorable safety profile and multi-target mechanisms, has shown potential advantages and value in regulating TSCs. It can reduce TSC drug resistance, enhance the sensitivity of tumor cells to chemotherapeutic agents, inhibit tumor growth and metastasis, and has shown unique advantages in improving the quality of life and prolonging the survival of GT patients. Studies have found that active components of Chinese medicine, such as terpenoids, polyphenols, flavonoids, glycosides, and quinones, and Chinese medicine compound formulas, including Zuojin pills, Sijunzi decoction, Biejiajian pills, and Xuanfu Daizhe decoction, can inhibit TSCs-related signaling pathways such as the Notch signaling pathway, the Wnt/β-catenin signaling pathway, the phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, and the Hippo signaling pathway. They also reduce the expression of TSC surface markers, including sex-determining region Y-box 2 (SOX2), sex-determining region Y-box 9 (SOX9), octamer-binding transcription factor 4 (OCT4), prominin-1 (CD133), cluster of differentiation 44 (CD44), cluster of differentiation 24 (CD24), and thyroid transmembrane protein 1 (CD90), thereby hindering TSC differentiation, accelerating their metabolic processes, improving the tumor microenvironment, and consequently inhibiting GT growth. This study collects and analyzes recent research on the regulation of TSCs by TCM in the treatment of GT, aiming to provide a new theoretical basis for tumor therapy with TCM, expand its application in the comprehensive treatment of GT, and offer new therapeutic ideas and methods for clinical practice.

Objective:To explore the factors influencing adherence to voice therapy among patients with voice disorders in China. Methods:Patients with voice disorders who visited the Voice Therapy Center at Sun Yat-sen Memorial Hospital, Sun Yat-sen University, from February to May 2022 were enrolled in the study. Adherence was assessed using the URICA-Voice scale, while influencing factors were assessed through the Voice Handicap Index(VHI) scale and a general information questionnaire. Correlation analysis was conducted using univariate and multivariate logistic regression analysis. Results:A total of 247 patients were included in the study, comprising 57 males(23.08%) and 190 females(76.92%). The results revealed that: ①Female patients demonstrated higher likelihood of being in the contemplation stage(OR=0.22) compared to males. ②Patients with a monthly family income per capita>6 000 yuan were more likely to be in the contemplation stage than those with<3 000 yuan with an OR = 13.94. ③High vocal-demand occupations increased contemplation stage probability(OR=7.70) compared to moderate-demand occupations. ④Residence within 30-minute commute predicted action/maintenance stages(OR=7.14) versus≥60-minute commute. ⑤Patients whose occupations had high voice demands were more likely to be in the action and maintenance stages than those with average voice demands, with an OR of 16.20. Conclusion:Gender, monthly family income per capita, occupational voice demands, and distance to the hospital significantly impact the URICA-Voice compliance stages of patients. Patients who are female, have higher family income, have occupations with high voice demands, and live closer to the hospital exhibit higher compliance with voice training.

The cardioprotective effects of histone deacetylase (HDAC) inhibitors (HDIs) are at odds with the deleterious effects of HDAC depletion. Here, we use HDAC3 as a prototype HDAC to address this contradiction. We show that adult-onset cardiac-specific depletion of HDAC3 in mice causes cardiac hypertrophy and contractile dysfunction on a high-fat diet (HFD), excluding developmental disruption as a major reason for the contradiction. Genetically abolishing HDAC3 enzymatic activity without affecting its protein level does not cause cardiac dysfunction on HFD. HDAC3 depletion causes robust downregulation of lipid oxidation/bioenergetic genes and upregulation of antioxidant/anti-apoptotic genes. In contrast, HDAC3 enzyme activity abolishment causes much milder changes in far fewer genes. The abnormal gene expression is cardiomyocyte-autonomous and can be rescued by an enzyme-dead HDAC3 mutant but not by an HDAC3 mutant (Δ33-70) that lacks interaction with the nuclear-envelope protein lamina-associated polypeptide 2β (LAP2β). Tethering LAP2β to the HDAC3 Δ33-70 mutant restored its ability to rescue gene expression. Finally, HDAC3 depletion, not loss of HDAC3 enzymatic activity, exacerbates cardiac contractile functions upon aortic constriction. These results suggest that the cardiac function of HDAC3 in adults is not attributable to its enzyme activity, which has implications for understanding the cardioprotective effects of HDIs.

Therapeutic cancer vaccines have experienced a resurgence over the past ten years. Cancer vaccines are typically designed to enhance specific stages of the cancer-immunity cycle, primarily by activating the immune system to promote tumor regression and overcome immune resistance. In this review, we summarize the significant recent advancements in cancer immunotherapy based on the cancer-immunity cycle, including the effector cell function, infiltration, initiation, and exhaustion. We summarize the identification of tumor antigens and their delivery through cancer vaccines. We discuss how specific stages of the cancer-immunity cycle have been leveraged to augment anti-tumor immune responses and improve vaccine efficacy. Additionally, the impact of aging and myelosuppression, two prevalent forms of immunological stress, on the effectiveness of therapeutic cancer vaccines is deliberated. Finally, we summarize the current status of various therapeutic cancer vaccines at different clinical trial phases.

The Journal of Clinical Anesthesiology 2024;40(7):757-761
Apnoeic oxygenation(AO)is the uptake of oxygen from the lungs by the patient's blood to maintain oxygenation in the absence of spontaneous breathing or mechanical positive pressure ventilation.During the administration of general anesthesia, the use of AO prolongs the patient's safe apnea time on top of preoxygenation before voluntary respiration ceases after induction and a safe airway is established, which may reduce the number of attempts at endotracheal intubation, and decrease the incidence of airway injury, hypoxemia, and hemodynamic decompensation.Over the past decade, more and more AO techniques have been applied in different clinical settings, such as perioperative airway management and"tube-free anesthe-sia".In order to deepen the understanding of AO and its applications, this article reviews the latest research progress on the mechanism, method and clinical application of AO for clinical applications provide refer-ence.

Chinese Journal of Preventive Medicine 2024;58(3):389-393
With global climate change, the frequency and intensity of extreme weather events have increased, and the possibility of multiple extreme weather events occurring simultaneously or continuously forming compound extreme events has also significantly increased, bringing catastrophic damage to the ecological environment, socio-economic and human health. This study reviewed the changing trends and health risks of consecutive compound heavy precipitation and heatwave extremes. There is currently no unified definition for these types of events, which could be divided into two subcategories: heavy precipitation-heatwave and heatwave-heavy precipitation. Their historical frequency and intensity have increased, and future estimates are also on the rise. The majority of current studies on single extreme events such as heatwaves or heavy precipitation revealed significant adverse health effects. However, few studies focused on the association between compound events of heavy precipitation and heatwave and human health risk. This paper further summarized the current shortcomings in this field and proposed suggestions for future research directions.
