Journal of International Oncology 2014;41(6):408-411
GRP78,as endoplasmic reticulum resident chaperone,is highly induced by a variety of tumor microenvironmental stresses, such as hypoxia and glucose deprivation.GRP78 is implicated in tumor cell proliferation, apoptosis, invasion, metastasis and angiogenesis.Recent evidence indicates that GRP78 levels is correlated with pathological grade, stage and prognosis for the majority of solid tumors. In addition,GRP78 plays an important role in drug tolerance and knockdown of GRP78 has been shown to sensitize malignant cells.GRP78 could not only be a good biomarker to predict cancer progression and chemo-responsiveness,but also an appealing target for the development of a more selective chemotherapy.

Objective To investigate the relationship between apoptosis of peripheral blood lymphocytes (PBLCs) and impaired immunity by assessing the apoptosis and expression of Fas and Bcl-2 in PBLCs of patients with secondary syphilis. Methods Flow cytometry (FCM) was used to detect PBLC apoptosis as well as Fas and Bcl-2 expression in 33 patients with secondary syphilis and 30 normal controls. Results The Fas expression on PBLCs and CD4+ cells was significantly higher in patients than that in controls (P 0.05). The apoptosis levels were significantly higher in PBLCs and CD4+ cells in patients than those in controls (P 0.05). In patients group, the apoptosis levels of both PBLCs and CD4+ cells had a positive correlation with the Fas expression (r = 0.68, P

Objective To explore the effect of Schisandrae Sphenantherae on proliferation and differentiation of rat osteoblast in vitro. Methods Using the method of serum pharmacology, osteoblast was isolated from calvaria of newborn SD rats by means of modified sequential collagenase digestion and incubated in RPMI 1640 medium. The cell morphology was observed under a phase contrast inverted microscope. MTT assay, ALP activity, mineral node count were detected to determine the status and activities of proliferation and differentiation. Results In 10%, 15% concentration groups after cultured 48 h and 5%, 10% after cultured 72 h, 75% ethanol extracts of Schisandrae Sphenantherae significantly stimulated the proliferation (P

Objective To isolate and purify polysaccharide from Angelica sinensis by ultrafiltration technology. Methods Extract of polysaccharide of Angelica sinensis was ultrafiltrated with different aperture membrane, the separation parameters on membrane separation efficiency of polysaccharide of Angelica sinensis were optimized by and orthogonal test. Results Polysaccharide of Angelica sinensis was separated with 200, 100, 50, 20 kD membrane. The contents of polysaccharide was 30.47% before ultrafiltration. The main component of Angelica sinensis polysaccharide was the fraction with 200 kD (65.42%). The optimum condition of ultrafiltration was obtained as follow:one times of extract volume, 35 ℃, 0.3 MPa. Conclusion The technology was simple and available, and the content of Angelica sinensis polysaccharide was higher.

Objective To assess the application of Xpert MTB/RIF assay in rapid diagnosis of tuberculous lymphadenitis. Methods Lymph node samples were collected from 1 02 clinically diagnosed patients with lymph node tuberculosis and 65 patients with other lymph node diseases from Zhejiang Provincial Hospital of Traditional Chinese and Western Medicine during January 201 4 and February 201 5. Xpert MTB/RIF,pathological examination and mycobacterium tuberculosis culture were conducted in all specimens of two groups.Taking clinical comprehensive diagnosis as the gold standard, the diagnostic sensitivity, specificity, positive predictive value, negative predictive value and diagnosis efficacy of three detection methods were assessed. The sensitivity of Xpert MTB/RIF in determining rifampicin resistance was analyzed using drug susceptibility testing as gold standard. Results The mean detection time of Xpert MTB/RIF was (2.2 ±0.2)h.Among 1 02 patients,Xpert MTB/RIF achieved higher sensitivity (96.1 %, 98 /1 02 ) than pathological examination (76.5%,78 /1 02 ) and mycobacterium tuberculosis culture (33.3%,34 /1 02)(χ2 =1 6.558 and 87.91 9,both P <0.01 ).Among 65 patients with other lymph node diseases, the specificity of all three detection methods was 1 00%.The receiver-operating-characteristic curve analysis demonstrated that diagnostic performance of Xpert MTB/RIF was better than that of other two methods. In 8 patients resistant to rifampicin confirmed by drug susceptibility testing,Xpert MTB/RIF detected 6 resistant-resistant patients. Conclusion Xpert MTB/RIF shows higher sensitivity and specificity in diagnosis of lymph node tuberculosis with the advantages of easy and rapid performance.

The rapid mutation and widely spread of duck hepatitis A virus (DHAV) lead to the vast economic loss of the duck industry. To prepare and evaluate bivalent inactivated vaccine laboratory products of DHAV, 6 strains were screened from 201 DHAV-1 strains and 38 DHAV-3 strains by using serotype epidemiological analysis in most of the duck factory. Vaccine candidate strains were selected by ELD50 and LD50 tests in the 6 strains. Continuously passaged, the 5th passaged duck embryos bodies grinding fluid was selected as vaccine virus seeds. The virus seeds were treated with formaldehyde and water in oil in water (W/O/W) emulsions, making into three batches of two bivalent inactivated vaccine laboratory products. The safety test, antibody neutralization test, challenged protection and cross immune protection experiment suggested that the vaccines possessed good safety, and neutralizing antibodies were detected at 7th day and the challenged protection rate reached 90% to 100% at the 14th and 21st day. Moreover, immune duration of ducklings lasted more than five weeks. However, cross-immunity protection experiments with DHAV-SH and DHAV-FS only had 20%-30%. The two bivalent inactivated vaccine laboratory products of duck viral hepatitis were effective and reliable, providing a new method as well as a new product for DHAV prevention and control.
