1.Acetylation regulatory mechanism of vascular aging

Protein acetylation extensively regulates fundamental biological process, such as cardiovascular homeostasis and tu-morigenesis. Mechanism of protein acetylation and its abnormal changes during diseases has become one of the basic questions in medical research. Vascular aging is inseparable from the occur-rence and development of senile hypertension and coronary ather-osclerosis. Endothelial cell senescence plays a key role during this process. This paper reviews the research progress in acetyla-tion regulation during vascular aging, and the prospects of the re-search in this field.
2.Research Advance of Health Risk Assessment on Methylmercury Exposure

Methylmercury is a potent toxicant, bioaccumulated and concentrated through the aquatic food chain, placing risks to human on the top of the chain. Based on the researches of adverse health effects exposed to methylmercury, several health risk assessment indexes (reference dose, provisional tolerable weekly intake, occupational mercury exposure assessment indexes ) and experiences on methylmercury exposure prevention (mainly on fish consumption) from the developed countries are presented in this paper.
3.Investigation on Cognition of the Clinical Pharmacists on the Clinical Pharmacy Practice and Their Profes-sion in the Medical and Health Institutions in Shanghai

OBJECTIVE:To investigate the cognition of clinical pharmacists on the clinical pharmacy practice and their profes-sion,and provide reference for further developing clinical pharmacy practice. METHODS:A questionnaire was designed,and ran-dom sample was conducted for the clinical pharmacists from 30 secondary and tertiary hospitals in Shanghai,and the investigation results were analyzed statistically. RESULTS:Totally 130 questionnaires were sent out,and 102 were effectively received with valid response rate of 78.46%. 94.11% of the investigated subjects were willing to be a clinical pharmacist,but 17.65% of them consid-ered that they were incompetent for their work;29.41% of them thought the current situation of clinical pharmacy practice was not ideal;all the respondents considered that the clinical pharmacist system should be implemented in medical and health institu-tions;the cognition and evaluation of investigated subjects in tertiary hospitals on clinical pharmacy practice and their profession was generally higher than those in secondary hospitals(P<0.05). CONCLUSIONS:The clinical pharmacists in medical and health institutions in Shanghai showed high cognition on their profession,and the clinical pharmacy practice has already made some achievements,but many work still need to be improved. Therefore,clinical pharmacists should continuously study their profession-al knowledge and improve their professional skills and quality;hospitals and universities should strengthen the subject construction and personnel training of clinical pharmacy and deeply carry out the clinical pharmacist system;health administration departments should strengthen the related laws and regulations of clinical pharmacy;colleges and universities should strengthen the discipline construction and personnel training.
4.Determination of Dioxin in Waste Printed Circuit Boards by Accelerated Solvent Extraction and Soxhelt Extraction

Waste printed circuit boards(W-PCBs) were multiple smashed and separated, then passed through a 60-mesh screen, treated with hydrochloric acid (2 mol/L), ultrapure water and dehydrated with acetone successively.The filter residue and filter paper were filled into the extraction pool, or inserted into Soxhlet Extraction tube parceled with new filter paper.After addition of 5 μL of internal standard substance, the filter residue above was respectively extracted by Soxhlet Extraction (SE) method or Accelerated Solvent Extraction (ASE) method, cleaned with multi-layer silica gel column and activated-charcoal column to obtained the dioxins samples.The samples were analyzed by isotope dilution high resolution gas chromatography-high resolution mass spectrometry (HRGC-HRMS).The effects of SE and ASE method and number of chlorines atoms on recovery of 15 kinds of 13C-2,3,7,8 PCDD/Fs were investigated, and the accuracy and precision of the two extraction methods were compared.The results show that, the recovery of 15 kinds of 13C-2,3,7,8 PCDD/Fs using ASE method is 54.3%-113.0%, and that of SE is 28.3%-77.7%, and the Toxic Equivalent Quangtity (TEQ) in W-PCBs is 0.075 ng/kg (TEQ) and 0.266 ng/kg (TEQ) using ASE and SE method respectively.Under the premise that accuracy and precision meet with the international standard, ASE is simple, rapid, solvent-free and accurate.

6.Assessment of microcirculation in a hind limb ischemia model

Aim To establish the standards for evaluation of microcirculation in a rat hind limb ischemia model.Methods Rats were subjected to unilateral femoral artery ligation.After 4 weeks,the regional blood flow in skeletal muscles was measured by fluorescent microsphere assay.Formation of collateral vessels was determined by microangiography.Capillary density was examined by immunostaining for CD34.Results In ischemic limbs,regional blood flow decreased significantly,some collateral vessels formed with typical "corkscrew" appearance,and histological analysis showed lower capillary density.Conclusion Quantitative assessment of microcirculation in a hind limb ischemia model is established.
7.Effect of hemorrhage on cell apoptosis in hippocampal CA1 region of rats under controlled hypotension

Objective To explore the effect of hemorrhage on cell apoptosis in the hippocampal CA1 region of rats under controlled hypotension (CH).Methods A total of 36 Sprague Dawley rats were randomly divided into 2 groups: Group C (with no CH) and Group H (with CH).According to different ratios of blood loss to total blood volume, Group C and Group H were redivided into 6 subgroups (6 in each group):C_1,H_1(10%);C_2,H_2(20%);C_3,H_3 (30%). Induced by so-dium nitroprusside and esmolol, the mean aterial pressure in Group H was decreased to 50~55 mmHg and kept for 10 minutes, and then blood loss was started, keeping the pump speed. Without CH, the same style of hemorrhage was performed in Group C. The aterial pressure was increased 60 minutes later after the hemorrhage.Expression of bcl-2 and caspase-3 protein was detected by immunohistochemical method, and apoptosis cells were detected by TUNEL staining. Results The average optical density of bcl-2 and caspase-3 was higher in Group H_3 than that in Group C_3(P<0.05). There were more apoptosis cells examined by TUNEL in Group H_3 than in Group C_3(P<0.05).Conclusion Thirty percent blood loss under controlled hypotension can induce cell apoptosis in hippocampal CA1 region in rats.
8.Effect of ulinastatin on postoperative immune function of effective and regulatory T-lymphocytes in peripheral blood in patients undergoing laparoscopic operation

Objective To investigate the effect of ulinastatin on the postoperative immune function of effective T-lymphocytes (Teff cells) and regulatory T-lymphocytes (Treg cells) in the peripheral blood in patients undergoing laparoscopic operation.Methods Forty ASA physical status Ⅰ or Ⅱ patients,aged 20-40 yr,weighing 45-65 kg,scheduled for elective laparoscopic operation for infertility,were equally and randomly divided into 2 groups using a random number table:control group (group C) and ulinastatin group (group U).At 15 min before induction of anesthesia,ulinastatin 200,000 U (in 100 ml of normal saline) was infused intravenously in group U,while the equal volume of normal saline was infused instead in group C.Anesthesia was induced with iv injection of fentanyl,propofol,and cisatracurium and maintained with iv infusion of propofol and sevoflurane.Before surgery (T0),at the end of surgery (T1) and on 1st and 3rd days after surgery (T2.3),venous blood samples were collected from the peripheral blood for determination of the expression of Teff cells (CD4 + CD25-) and Treg cells (CD4 + CD25+) and serum interferon-γ (IFN-γ) and transforming growth factor-β (TGF-β) concentrations (by ILISA).Teff/Treg and IFN-γ/TGF-β ratios were calculated.Results Compared with the baseline value at T0,the expression of Treg cells was significantly up-regulated,and Teff/Treg ratio was decreased at T2 and T3 in group C,and the expression of Teff cells was up-regulated at T3 (P < 0.05 or 0.01),while no significant difference was found in the expression of Treg cells and Teff/Treg ratio at T3 in group U (P > 0.05).Compared with group C,the expression of Teff cells was significantly up-regulated at T3,the expression of Treg cells was down-regulated and Teff/Treg ratio was increased at T2 and T3,the concentration of serum TGF-β was decreased and IFN-γ/TGF-β ratio was increased at T2 and T3 in group U (P < 0.05 or 0.01).No significant changes were found in the serum IFN-γ and TGF-β concentrations and IFN-γ/TGF-β ratio at each time point in the two groups (P > 0.05).Conclusion Ulinastatin can enhance the immune function of Teff cells in the peripheral blood and decrease the immune function of Treg cells,thus maintaining the balance of the postoperative cellular immune function in patients undergoing laparoscopic operation.
9.The differences among the pass rate of intensity modulated radiation therapy planning in different tumors

Objective To investigate the difference in the pass rate of intensity?modulated radiation therapy ( IMRT) planning in patients with different tumors and its value in determining pass rate thresholds. Methods A total of 35 verified IMRT plans for each of esophageal cancer, nasopharyngeal carcinoma, breast cancer, cervical cancer, and lung cancer were selected consecutively, and a one?way analysis of variance was used to investigate the difference in pass rate. A single pass rate threshold was used to test all IMRT plans, and the pass rate thresholds of IMRT plans for different tumors were calculated based on normal distribution law. Results There was a significant difference in the means between the 5 groups of data ( F=35. 83, P<0. 01) , and there was also a significant difference between any two groups ( P=0. 000) . There were statistically significant differences between nasopharyngeal carcinoma group and other four groups ( P=0. 000) . The difference was not only significant between the breast cancer group and the esophageal cancer group ( P=0. 001) , but also between the breast cancer group and the lung cancer group ( P=0. 033) . The calculated results of each threshold were 93. 37%, 89. 34%, 97. 68%, 95. 99%, and 95. 42%, respectively. Conclusions Different thresholds should be used for IMRT plans for different tumors, and the normal distribution law can be used to calculate the threshold.
10.Participation of Clinical Pharmacist in Hypoglycemic Program for One Case of Diabetes Allergic to Insulin Preparations

Objective:To analyze one case of diabetes allergic to insulin preparations and investigate the role of clinical pharmacist in the treatment. Methods:Different insulin preparations were tried out after the patient was allergic to one kind of insulin preparation. However, the allergic symptoms were not improved significantly. Clinical pharmacist recommended to use glargine combined with oral hypoglycemic drugs instead of insulin containing protamine for the patient. Results:The allergic symptoms of the patient were improved significantly after the drug adjustment in the hypoglycemic program by clinical pharmacist. The blood glucose was controlled and aller-gic symptoms didn't show again during the hospitalization. Conclusion:Clinical pharmacist can participate in the treatment of diabetes allergic to insulin preparations through identifying allergens accurately and improving individual hypoglycemic program using pharmaco-logical knowledge. It is important to improve the clinical diagnosis and treatment, and ensure and promote the safety of medical treat-ment and drug use.
