1.Molecular Switch of T Cell Factor-4 in Carcinogenesis of Colorectal Cancer

Objective To investigate the role of T cell factor-4 (TCF-4) in the carcinogenesis of colorectal cancer. Methods Relevant references about TCF-4 and the carcinogenesis of colorectal cancer, which were published recently domestic and abroad, were collected and reviewed. Results For TCF-4 gene, multiple isoforms are generated by way of alternative splicing, which encode different proteins. TCF-4 protein is sequence-specific DNA binding protein and is incapable of activating or repressing transcription independently, but it can interact with distinct partners to lead to different effects through multiple domains. Conclusion TCF-4 might be viewed as nuclear vehicles targeting other auxiliary proteins to a specific set of promoters and functions as molecular switch during the carcinogenesis of colorectal cancer.
2.Clinical analysis of infantile laryngopharyngeal reflux disease with cyanosis

Objective:To analyze the clinical characteristics of infantile laryngopharyngeal reflux diseases(LPRD) with cyanosis as the main manifestation, improve diagnosis of the disease and avoid missed diagnosis.Methods:Clinical data of children diagnosed with LPRD at Shenzhen Children′s Hospital from September 2015 to October 2018 were retrospectively analyzed.Results:Cyanosis was the predominant symptom in all 5 cases.The course ranged from 3 days to 4 months.Laryngomalacia was observed in 4 cases.Two patients were treated with esophageal atresia, 1 with esophageal stenosis and 1 with severe tracheomalacia.All 5 cases had vocal cord and arytenoid mucosa edema.The Ryan index was 65.92-1 444.5 in the upright position and 3.73-431.44 in the supine position.Three cases were treated with erythromycin and ceftriaxone according to the etiology.Two cases were treated with motilium at the same time, and 3 cases were with omeprazole oral.One case was recovered after 8 weeks and the Ryan index became normal.Symptoms were alleviated in 3 patients after 4 weeks, drugs were stopped by parents.One case still had cyanosis during medication, the symptoms were significantly relieved after tracheal stent implantation.Conclusion:LPRD is closely related to children′s respiratory and digestive system diseases.LPRD should be taken into account when children show unexplained clinical symptoms of respiratory and digestive system, and laryngoscopy and DX-pH detection should be performed as soon as possible to avoid missed diagnosis.
3.Observation of curative effect of point electric stimulation for urinary incontinence after postoperative cerebral hemorrhage

Objective To study the effect of point electric stimulation for urinary incontinence after postoperative cerebral hemorrhage.Methods Eighty patients with urinary incontinence after postoperative cerebral hemorrhage were randomly divided into treatment group and control group(n=40 case). Point electric stimulation was given to the treatment group, and Oxybutynin hydrochloride sustained release tablets was given orally to the control group. Index of urinary diary and bladder capacity in both groups before treatment and after 3 weeks of treatment were observed.Results After 3 weeks of treatment,the total effective rate(87.2%vs. 65.8%,χ2=4.916) in the treatment group were better than the control group (P<0.05). The times of voided (10.15 ± 2.21 times/24 hvs. 18.63 ± 3.42 times/24 h,t=12.956), the times of urinary incontinence (2.18 ± 1.04 times/24 h vs. 6.19 ± 2.16 times/24 h,t=10.421), the average voided volume (293.38 ± 16.32 mlvs. 180.82 ± 15.43 ml, t=31.083), the initial sensory capacity of bladder (276.73 ± 20.95 mlvs. 186.43 ± 21.14 ml,t=18.825), the maximum capacity of bladder (492.81 ± 23.72 mlvs. 293.81 ± 24.46 ml,t=36.244), the residual urine volume(13.25 ± 5.83 mlvs. 34.63 ± 6.25 ml,t=15.527) in the treatment group were significantly better than those in the control group (P<0.01).Conclusions The treatment of point electric stimulation can reduce the symptoms of urinary incontinence.

Objective To study the effects of Ginkgo biloba extract (GBE) on free radical metabolism of different fiber types of quadriceps in exercised rats and its mechanism. 6.Clinical Observation of Suspending-needle Therapy Combined with Modified Zhengan Xifeng Decoction for Treatment of Primary Facial Spasm

Objective To observe the therapeutic effect of suspending-needle therapy combined with modified Zhengan Xifeng Decoction for the treatment of primary facial spasm,and to explore an effective traditional Chinese medicine therapy for primary facial spasm.Methods A total of 65 qualified patients were randomly divided into trial group (33 cases) and control group(32 cases),which was treated with suspending-needle therapy combined with modified Zhengan Xifeng Decoction,Carbamazepine tablets respectively.Twenty-eight days constituted one course of treatment.After treatment for one course,therapeutic efficacy of the two groups was evaluated,and the degree of facial spasm was assessed.Results (1) After one course of treatment,the markedly-effective rate of the trial group was 84.8%,and that of the control group was 62.5%,the difference being significant(P < 0.05).(2) After treatment,the degree of facial spasm in the two groups was much improved as compared with that of the control group (P < 0.05 or P < 0.01),and the improvement in the trial group was superior to that in the control group (P < 0.01).Conclusion Suspending-needle therapy combined with modified Zhengan Xifeng Decoction has better effect on relieving facial spasm of the primary facial spasm patients than Carbamazepine tablets.
7.The radiological manifestation of hypophosphatasia

ObjectiveTo explore the radiological features of hypophosphatasia.MethodsFive cases of hypophosphatasia were definitely diagnosed,which included 3 males and 2 females aged from 5 months to 23 years.The laboratory assays were analyzed,radiological appearances of bone were determined and differential diagnoses were made.ResultsThe alkaline phosphatase in blood serum of five patients decreased,which were 8,20,13,21,and 18 U/L respectively.Phosphoethanolamine increased in blood serum of the five patients,which were 16.5,13.5,21.6,18.7,and 28.9 μmol/L respectively.Phosphoethanolamine also increased in urine,which were 2350,9120,3520,5280,and 1820 μmol/L respectively.Calcium in blood serum increased,which were 4.2,5.6,4.9,6.1,and 3.5 mmol/L respectively.X-ray images displayed that the density of bone decreased in 5 cases,the metaphyses exhibited widening and cupping in 4 cases,the provisional calcification zone of the metaphysis became thinning or disappearing in 4 cases,long bone bended in 4 cases and pathologically fractured in 1 case.Conclusion Hypophosphatasia can be indicated by its relatively special radiological appearance,and it can be diagnosed and differentiated from rickets,osteomalacia and osteogenesis imperfect by the laboratory examination combined with its clinical presentation.
8.Differentiation of rabbit bone marrow mesenchymal stem cells labeled by green fluorescent protein into osteoblasts and lipoblasts

BACKGROUND:Labeling was commonly used to further trace bone marrow mesenchymal stem cells (BMSCs).OBJECTIVE:To label rabbit BMSCs by green fluorescent protein,and then to observe the potency of multi-directional differentiation by osteoblast and lipoblast formation induction.DESIGN,TIME AND SETTING:Cell observational study was performed at the Laboratory of the Department of Orthopaedics,Tongji Hospital Affiliated to Tongji Medical College,Huazhong University of Science and Technology from December 2007 to October 2008.MATERIALS:Totally 6 healthy New Zealand rabbits aged 4 months were purchased from the Animal Experimental Center,Tongji Medical College,Huazhong University of Science and Technology.METHODS:BMSCs were isolated from the femurs of rabbits,purified and cultured in vitro by density gradient centrifugation and adherent culture.BMSCs at the third passage were obtained.Green fluorescent protein plasmid was transfected into BMSCs using Lipofectamine.The stable transfection cells were obtained using G418 selection after transfection.The stable transfected BMSCs were induced to osteoblasts and lipoblasts by osteogenic inductor and adipogenic inductor.MAIN OUTCOME MEASURES:The following parameters were measured:BMSC morphology,surface marker expression,green fluorescent protein labeling,the differentiation potential of BMSCs into osteoblasts and lipoblasts.RESULTS:The BMSCs were mostly fusiform in shape,to be similar to fibroblast cell after cultivation of primary and subculture.The flow cytometer analysis showed that the membrane mark CD44 was positive,CD34 was negative.Following G418 screening,BMSCs labeled by green fluorescent protein were apparent calcium mineralization after 21 days osteogenic induction.Alizarin red staining showed positive with red.Intra-cellular lipid droplet appeared after 3 days adipogenic induction.2 weeks later,oil red O staining showed a great quantity lipid deposition.CONCLUSION:Rabbit BMSCs labeled by green fluorescent protein using Upofectamine have the potency of multi-directional differentiation after induction.
9.Axial pressure capacity of injured vertebrae witch in dogs treated with injectable artificial bone composite following thoracolumbar fractures

BACKGROUND: Injectable artificial bone composite has been reported to consider as a moulding filler of vertebrae at the histology level; however, the therapeutic effect needs to be further studied at the level of axial pressure capacity. OBJECTIVE: To explore the axial pressure capacity of dogs with thoracolumbar fractures treated by injectable artificial bone composite. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at the Animal Laboratory of Shenzhen Longgang Central Hospital from September 2008 to January 2009. MATERIALS: Type I and type II vertebral pedicle screw systems were provided by Tianjin Zhengtian Medical Device Company Limited; injectable artificial bone composite, which was made of coral-hydroxylapatite compound, recombinant human bone morphogenetic protein-2 (Academy of Military Medical Sciences), and 2% chitosan solution (the Second Military Medical University of Chinese PLA and Shanghai Qisheng Biomaterials Institute), was manufactured according to the methods provided by Yin et al. METHODS: A total of 20 1-year-old healthy dogs were randomly divided into treatment and control groups with 10 dogs in each group. The model of those dogs with thoracolumbar fractures was made by imitating falling accidents; thereafter, the dogs in the treatment group were treated with vertebral pedicle screw system internal fixation and vertebroplasty by filling the injured vertebrae with injectable artificial bones. The control group was treated with vertebral pedicle screw system internal fixation alone. MAIN OUTCOME MEASURES: The maximum pressure intensity of injured vertebra, upper and lower vertebra at vertebral body center was detected using micro-computer pressure testing system after three months. RESULTS: The maximum pressure intensity at vertebral body center was not significant differences in the treatment group (100% cases) between injured vertebral body and its neighboring vertebral body, and axial loading was recovered. 10.Salvia miltiorrhiza monomer IH764-3 protects human brain microvascular endothelial cell from hypoxia/reoxygention injury through inhibiting leukocyte activity

Objective To investigate the effect of IH764-3 on the leukocyte-mediated hypoxia-reoxygention injury of human brain microvascular endothelial cell (HBMVEC). Methods MTT assay was used to detect the survival of HBMVEC; gelatin zymography was used to check the activity of MMPs. The level of reactive oxygen species (ROS) in leukocyte was determined via commercially available kit,and the indirect enzyme-linked immunosorbent assay (ELISA) was used to quantify the contents of TNF-?,IL-1?,IL-2 and INF-? in leukocyte culture medium.Results Survival of HBMVEC was impaired by hypoxia-reoxygenation,which was aggravated by supernatant of activated leukocytes but was attenuated by IH764-3. Leukocytes produced high level of MMP-9,ROS and cytokines (TNF-?,IL-1?,IL-2,IFN-?) after hypoxia-reoxygenation,the process was inhibited by IH 764-3. Furthermore,IH764-3 could effectively reverse hypoxia-reoxygenation injury of HBMVEC with supernatant of activated leukocytes. Conclusion IH764-3 can protect HBMVEC from leukocyte-involved hypoxia-reoxygenation injury by attenuating the activation of leukocytes and inhibiting the pathogenic effects of leukocytes products.
