1.Protein MG29 and skeletal muscle aging

OBJECTIVE: To review research advances of Mitsugumin-29 (MG29) protein and its relationship with skeletal muscle aging. DATA SOURCES: A computer-based online search of CNKI (http://www.chkd.cnki.net), Duxiu (http://edu.duxiu.com) and SSReader (http://edu.sslibrary.com) with key words of MG29 and skeletal muscle aging in Chinese; Elsevier SD (http:llwww.sciencedirect.com) and Springer Link (http:llspringer.metapress.comlhomelmain.mpx) with key words of MG29, aging skeletal muscle, sarcopenia. In addition, related articles were manually searched.DATA SELECTION: Inclusion criteria: location, structure and function of MG29; MG29 and skeletal muscle aging; MG29 and exercise; related articles aimed to the content with large impact factor. Exclusion criteria: repetitive studies or Meta analysis.MAIN OUTCOME MEASURES: MG29 has close relationship with skeletal muscle aging.RESULTS: In recent years, extensive researches have been conducted to discover the mechanisms underlying skeletal muscle aging by investigators home and abroad from histology, biochemistry and molecular biology. It is universally accepted that the changes in the cellular homeostatic capacity for Ca~(2+) is a core factor in regulating excitation contraction coupling, and it dynamic balance is the major cause of skeletal muscle aging. MG29, which is located in the membrane of transverse tubule, is a protein that is newly found to modulate Ca~(2+) signal resembling the synaptophysins. Studies have shown that the level of MG29 reduced 50% in old skeletal muscle. Similar phenotypes to aging skeletal muscles have been observed in adolescent muscle null for MG29 which is involved in muscle weakness, increased susceptibility to muscle fatigue, fragmented sarcoplasmic reticulum networks, swollen transverse tubule system, altered calcium sparks, segregated Ca~(2+) reserve and so on. These suggest that MG29 and aging skeletal muscle are closely related.CONCLUSION: It has been confirmed that MG29 is highly correlated with aging skeletal muscle. However, studies concerning MG29 improvement to delay skeletal muscle aging should be conducted subsequently.
2.Clinicopathological significance and expression of Numb and α-catenin in gastric carcinoma

Objective To detect the expressions of Numb and α-catenin in the gastric cancer and matched normal gastric mucosa,and to analyze their relationship with clinicopathological factors of gastric cancer and explore their roles in gastric carcinogenesis and progression.Methods Immunohistochemical method was used to detect the expressions of Numb and α-catenin in 109 cases of tumor specimens and 97 cases of matched normal gastric mucosa.The correlations between Numb,α-catenin and clinicopathological factors were analyzed.Results The positive rates of Numb and α-catenin in gastric cancer were significantly lower than those in normal gastric mucosa (60.6% ∶ 100.0%,x2 =48.361,P =0.000;76.1% ∶ 100.0%,x2 =26.480,P=0.000).The expression of Numb protein was correlated with Lauren type (x2 =17.018,P =0.000) and tubular adenocarcinoma differentiation (x2 =17.586,P =0.000).The expression of α-catenin protein was correlated with Borrmann type (x2 =6.700,P =0.035),Lauren type (x2 =11.098,P =0.001),tubular adenocarcinoma differentiation (x2 =8.203,P =0.017) and lymph node metastasis (x2 =6.402,P =0.011).The expressions of Numb and α-catenin were statistically correlated in 109 cases of gastric cancer (rk =0.184,P =0.028).Conclusion Compared with normal gastric mucosa,both Numb and α-catenin expressions are down-regulated and the two expressions are correlated in gastric cancer.Numb and α-catenin may be involved in the regulation of histological differentiation of gastric cancer by certain passages,Numb protein may be adjusted by α-catenin pathway which is involved in Wnt-β-catenin pathway,and thus plays an important role in promoting cell proliferation,invasion and metastasis in human gastric cancer.

The growing number of cognitive dysfunction patients is bringing heavy mental and financial burdens to the society and families.Voltage-gated potassium channels (Kv), which consist of delayed rectifier potassium channels and transient outward po -tassium channels , are involved in the incidence of cognitive dysfunction .This review summarized the role of Kv channels in cognitive dysfunction and their relationship with N-methyl-D-aspartic acid receptors ( NMDARs) that play an important role in the process of learning and memory .
4.Telemetric Localization of Detecting Capsule in Gastrointestinal Tract

Objective To develop a method for localizing a detecting capsule in the gastrointestinal tract.Methods A three-axis magnetic sensor was sealed in the detecting capsule.Three flat coils were fixed on patients' back.The coils were excited one by one to generate electromagnetic fields.The position and the orientation of the capsule could be determined by telemetering the signals from the magnetic sensor incorporated in the capsule.Results The telemetric localization system worked successfully.A localization model describing the relationship between the magnetic field strength and the position and orientation of the capsule,was correctly established basing on the principle of magnetic dipoles,and neural network algorithm was employed to solve a non-linear equation set.Conclusion The results showed that the localization method is feasible and the precision is higher than existing methods.The localization device can perform consecutive tracking and can be made into a portable type.After putting into practice,the device can also be used in food selection for astronauts by monitoring their gastrointestinal parameters in space.

ObjectiveTo analyze the autophagy of mouse melanoma B16 cells induced by TNF-ct.MethodsMouse melanoma B 16 cells treated with TNF-α were used in this study.The expression levels of LC3- Ⅱand Beclinl were measured by western blot and mRNA levels of autophagy related gene atg5,atg7 and atgl2 were measured by reverse transcription polymerase chain reaction (RT-PCR) after TNF-α treatment.ResultsThe viability of B16 cells were obviously inhibited after incubation with TNF-α.The expression levels of autophagy related protein LC3- Ⅱ and Beclinl were elevated in TNF-α treated B16 cells compared with in control B16 cells.The mRNA expression levels of autophagy related gene Atg5,atg7 and atg12 showed consistent up regulation in TNF-a treated B16 cells compared with in control B16 cells.ConclusionTNF-α can induce autophagic cell death in B16 cells.
7.Study on the characterization and isolation of collagen from different shark skins

Objective To investigate the methods of isolating collagen from different shark skins and to study its biochemical properties.Methods The collagen from the Blue shark scalp was extracted and purified by enzymolysis after treatment with diluted caustic alkali,and the collagen from the Spanish mackerel skins was extracted and purified by phosphate buffer method,respectively.Subsequently,the relative molecular mass,thermal denatured temperature and secondary structure of extracted collagen were determined.Results The extracted collagen with a yield of 2.9%~4.1% showed three clear bands by SDS-PAGE,and the relative molecular mass were 205,134 and 118KDa.The collagen from Blue shark and Spanish mackerel skins maximal thermal denatured temperature was 69,47℃,respectively.The secondary structures of their collagen were primary with ?-sheet and the random structure,without ?-alix.Conclusion The purified collagen was also extracted by the method of enzymolysis after treatment with diluted caustic alkali and the method of phosphate buffer.The relative molecular mass and the composition of Blue shark collagen and Spanish mackerel collagen were limited.However,the different extracted conditions could affect the specifically biochemical properties of thermal denatured temperature and secondary structure of collagen.
8.Proteomics analysis of Sut melanocytes in response to xCT deficiency

Objective To investigate the mechanism of Sut melanocytes growth inhibition in response to xCT -deficiency. Methods TTotal proteins were extracted from xCT-deficient Sut melanocytes and wild melanocytes, respectively, and were separated by 2-dimensional electrophoresis. Altered expression profile of proteins of Sut melanocytes was analyzed by PDQuest software and compared with that of wild melanocytes.Proteins with significant change were chosen to be identified by mass spectrometry and database query.Results Twenty proteins in Sut melanocytes altered significantly compared with wild melanocytes. Ten of the proteins were up-regulated, while the other tens were down-regulated. Four proteins from both up-regulated and down-regulated were identified respectively: up-regulated proteins were Tubulin alpha-1b, S100-A6,Nucleoside, S-formylglutathione, and down-regulated proteins were Calumenin,NDRG1 ,DPYSL2, 14kDa unknown protein. Conclusion The identification of the xCT-deficiency related proteins may provide supporting evidence for the mechanism research of Sut melanocytes' growth inhibition caused by xCT-deficiency.
9.Effect of Enhanced Extracorporeal Counterpulsation on Intraocular Pressure

Objective To observe the effect of enhanced extracorporeal counterpulsation (EECP) on intraocular pressure (IOP). Methods 25 patients were measured their IOP bilaterally with Schoitz tonometer before and after EECP. Results The IOP decreased in both left and right eyes after EECP (P<0.01). Conclusion EECP can reduce intraocular pressure.
10.Analysis and Management of Off-label Use of Tamoxifen for Ovulation Induction

OBJECTIVE:To analyze the rationality of tamoxifen for ovulation induction,and to provide reference for the stan-dardization of off-label use of tamoxifen. METHODS:Takingtamoxifen,ovulation inductionandpolycystic ovarian syn-dromeas keywords,relevant literatures about off-label use of tamoxifen for ovulation induction were analyzed and summarized by a search of CNKI,Wanfang database,PubMed database up to July 2015. RESULTS:58 literatures were searched,among which clinical trial or systematic review about the effect of tamoxifen on ovulation induction were selected. 11 valid literatures were col-lected to confirm clinical efficacy of tamoxifen for ovulation induction. Referring to Diagnosis and Treatment of Polycystic Ovary Syndrome:An Expert Consensus,pharmaceutical affairs council of our hospital had agreed to off-label use of tamoxifen for induc-tion ovulation,and formulated the standard of off-label use of tamoxifen. CONCLUSIONS:Standard management of off-label use of tamoxifen could be better protect the rights both of patient and doctor.
