1.Application of Peripheral Blood SEPT9 DNA Methylation Assay for Screening of Colorectal Cancer

Colorectal cancer is a malignancy with poor prognosis and high mortality and is caused by multiple factors. Colonoscopy,flexible sigmoidoscopy,guaiac-based fecal occult blood test,immunochemical fecal occult blood test,fecal DNA test,CT colonoscopy and serum carcinoembryonic antigen test are the methods frequently used for screening of colorectal cancer,but they all have certain limitations. Elevation of methylation of SEPT9 is associated with the pathogenesis of colorectal cancer,and detection of the level of methylation of SEPT9 in peripheral blood can be used for screening of colorectal cancer in susceptible population. This article reviewed the application of peripheral blood SEPT9 DNA methylation assay for screening of colorectal cancer.
2.The time interval of using electric toothbrush after the adhesion of two common luting cements

Objective:To explore the time interval of using electric toothbrush after the adhesion of zinc polycarboxylate cement (ZPC) and PanaviaTM F(PF) respectively.Methods:The facial surfaces of 60 premolars were exposed and embedded in self-curing resin with custom made cylinder-shaped molds.Then the facial surfaces were cut with a diamond saw and polished with waterproof polishing papers to create standardized dentin surfaces.30 co-cr ally test-pieces and 30 zirconia ceramic test-pieces (4.0 mm in diameter and 2.0 mm in height) were adhered on 30 dentin surfaces respectively with ZPC and PF.The samples of each kind were divided into 5 groups (n =6) and brushed with an electronic toothbrush for 30 seconds immedietly (group 1),12 h (group 2),24 h (group 3)and 48 h(group 4) after adhesion,respectively.The samples in group 5 without brush were the controls.Shear bond strength of the samples was measured and the fracture patterns were observed under steriomicroscope.Results:In the ZPC adhered samples the bond strength of group 1,2,3 was lower than that of group 4 and 5 (P < 0.05),group 4 vs group 5,P > 0.05.In the PF adhered simples the bond strenth of group 1 and 2 was lower than that of group 3,4 and 5 (P < 0.05),group 3 or 4 vs group 5,P > 0.05.The fracture patterns were mostly cement cohesive failure.Conclusion:The time intervals of using electric toothbrush after the adhesion of ZPC and PF are 48 h and 24 h respectively
3.Study of the efficacy of xuebijing injection applied after abdominal operation

Objective To investigate the effect of xuebijing injection applied after abdominal operation on the inflammatory reaction,infection rate,length of stay and hepatorenal function. Methods Sixty patients received abdominal operations were randomly divided into therapeutic group (30 cases) and control group (30 cases). The inflammatory reaction indexes (temperature,heart rate,leucocyte) and hepatorenal function indexes(aminopherase, total bilirubin, creatinine, blood urea nitrogen) were observed in both groups before operation and after operation 3,7 days respectively. The infection rate and length of stay were compared in two groups. Results Temperature and leucocyte post operative 3 days [temperature:(37.0 ± 0.2) ℃ vs. (37.9 ± 0.4) ℃ ;leucocyte: (8.8 ± 1.1 ) × 109/L vs. ( 10.3 ± 1.7) × 109/L, P< 0.05] and length of stay in therapeutic group was obviously better than that in control group[ (8.7 ± 1.9 ) d vs. ( 10.9±1.6) d, P < 0.05 ]. The hepatorenal function indexes and infection rate in both groups had no significant difference(P > 0.05). Conclusion Xuebijing injection could significantly palliate the inflammatory reaction after abdominal operation, shorten the length of stay and has no hepatorenal toxicity in short term.
4.Analyzing the risk factors of root caries by Logistic regression analysis

Objective To analyze the risk factors of root caries.Methods With randomly stratified cluster sampling method,we selected 600 elderly in Tangshan city and performed oral examination.Data was analyzed by Logistic regression analysis.Results Gingival recession,age,oral calculus index,false tooth restoration were the risk factors of root caries.The vertical brushing tooth method and using fluoride toothpaste were the protective factors.Conclusion Gingival recession,age,oral calculus index(CI-S),false tooth restoration were the major risk factors of root caries.
5.Evaluating the clinical effects of light cure Dyractflow in pit and fissure enamel-plasty sealant technique during 3 years

Objective To evaluate the clinical effect of light cure Dyractflow (LCDF) in pit and fissure enamel-plasty sealant technique (EST). Methods A total of 400 first permanent molars in 200 patients aged from 6 to 10 years were treated with LCDF in occlusal pit and fissure enamel-plasty sealant technique. The opposite permanent teeth treated with LCDF in occlusal pit and fissure conventional sealant technique (CST) were employed as the controls. The patients were followed up for 3 years. Results Follow-up for 3 years showed that there was no significant difference in preservation rate between the EST and the CST groups in the first year, but in 2-3 years, there was significant difference in preservation rate (P

Objective: To explore the therapeutic effect of dexamethasone on proliferative retinopathy in different stages and its interaction with the vascular endothelial growth factor(VEGF).Methods: Models of oxygen-induced retinopathy(OIR) were established.Twenty C57BL/6J mice pups(7 days after birth) were randomized into 4 groups: normal control,pure OIR,early-treatment and late-treatment.Dexamethasone was administered at the dose of 0.5 mg/kg?d-1by subcutaneous injection to the early-treatment group from postnatal day 7 and to the late-treatment group from day 12,both for 5 days.Common pathological sections were made from both eyeballs of the C57BL/6J mice and detected by immunohistochemistry to count the nuclei of proliferative retinal vessel cells and investigate the expression of VEGF in the retina.Results: The nuclei of proliferative retinal vessel cells and the expressions of VEGF significantly increased in the pure OIR group as compared with the normal control(P
7.Antibacterial constituents of Senecio cannabifolius(Ⅱ)

Objective To study the antibacterial constituents of Senecio cannabifolius.Methods The antibacterial effect of all the extracts was tested in vitro with Staphylococcus aureus,Bacillus subtilis(Gram-positive bacilli),and Escherichia coli(Gram-negative bacillus).Chemical constituents were isolated by chromatography.Physicochemical characters and spectroscopic analyses were employed for their structural identification.Results Five compounds were isolated and their structures were elucidated by chemical and spectral methods as follows: methyl-5-hydroxy-2-pyridinecarboxylate(E-13),6-hydroxy-7,7a-dihydro-2(6H)-benzofuranone(E-14),2-(1,4-dihydroxycyclohexanyl)-acetic acid(E-16),3-hydroxycyclohexanecarboxylic acid(E-17),and 4-hydroxy benzaldehyde(B-14).Conclusion All of them are isolated from the plants of Senecio L.for the first time.
8.Efficacy of laparoscopic or open surgery for gastrointestinal stromal tumors and effects of different risk level on prognosis

Objective To compare the efficacy of laparoscopic or open surgery for gastrointestinal stromal tumors (GISTs) and investigate effects of different risk level on prognosis.Methods The retrospective cohort study was adopted.The clinical data of 192 patients with GISTs who were admitted to Zhongshan Hospital of Fudan University from January 2008 to December 2013 were collected.Among the 192 patients,88 undergoing laparoscopic surgeries were allocated into the laparoscopic surgery group,104 patients undergoing open surgeries were allocated into the open surgery group.The following indicators were observed:(1) operative status:surgical procedure,operation time,volume of intraoperative blood loss.(2) Status of postoperative recovery:time of gastrointestinal function recovery,time of drainage tube removal,complications and duration of hospital stay.(3) Follow-up status.(4) Prognosis of patients in different risk level.The follow-up using outpatient examination and telephone interview was performed to assess patients' survival,tumor recurrence and metastasis until June 2015.Measurement data with normal distribution were presented as (x) ± s and comparison between groups was evaluated by the t test.Comparison of count data was analyzed by the chi-square test.The Kaplan-Meier method was used to draw survival curve and calculate the overall survival rate and relapse-free survival rate.Results (1) Operative status:of the 88 patients in the laparoscopic surgery group,1 underwent laparoscopic wedge gastrectomy + cholecystectomy + appendectomy,6 underwent laparoscopic wedge gastrectomy + cholecystectomy,14 underwent laparoscopic assisted partial gastrectomy,67 underwent laparoscopic wedge gastrectomy.Of the 104 patients in the open surgery group,1 underwent partial gastrectomy + splenectomy,2 underwent partial gastrectomy combined with distal pancreatectomy + splenectomy,2 underwent total gastrectomy,7 underwent distal subtotal gastrectomy,7 underwent wedge gastrectomy + partial or total adjacent organ resection,8 underwent proximal subtotal gastrectomy,8 underwent wedge gastrectomy + cholecystectomy,69 underwent wedge gastrectomy.The operation time and volume of intraoperative blood loss were (105 ± 33)minutes and (43 ± 16)mL in the laparoscopic surgery group,(121 ± 52)minutes and (199 ± 81) mL in the open group,respectively,with statistically significant differences between the 2 groups (t =-2.104,2.632,P < 0.05).(2) Status of postoperative recovery:the time of gastrointestinal function recovery,time of drainage tube removal and duration of hospital stay were (4.6 ± 1.8) days,(5.8 ± 2.2) days,(7.1 ± 2.9) days in the laparoscopic surgery group and (5.2 ± 1.6) days,(7.1 ± 2.8) days,(8.7 ± 4.3) days in the open surgery group,respectively,with statistically significant differences between the 2 groups (t =-2.783,-3.891,-3.078,P < 0.05).Wound infection,gastric emptying delay,anastomotic leakage,lung infection and bleeding were detected in 1,3,0,0,0 patients in the laparoscopic surgery group and in 0,2,2,2,1 patients in the open surgery group,respectively,with no statistically significant difference between the 2 groups (x2=0.421,P > 0.05).(3) Follow-up status:Of the 192 patients,149 received follow-ups.Of 88 patients in the laparoscopic surgery group,68 were followed up for an average time of 39 months.Of 104 patients in the open surgery group,81 were followed up for an average time of 51 months.During the follow-up,tumor recurrence rate in the laparoscopic surgery group and open surgery group was respectively 8.8% (6/68) and 21.0% (17/81),with no statistically significant difference between the 2 groups (x2=1.888,P >0.05).Postoperative 1-,3-,5 year survival rates were 98.5%,92.9%,87.4% and 91.7%,85.2%,76.9% in the laparoscopic surgery group and open surgery group,respectively,with no statistically significant difference between the 2 groups (x2 =1.967,P > 0.05).(4) Prognosis of patients in different risk level:of the 149 who received the follow-up,the tumor recurrence rate of patients in low,intermediate and high recurrence risk was 7.0% (5/71),13.6% (6/44) and 35.3% (12/34),respectively,with a statistically significant difference among the above indexes (x2 =14.637,P < 0.05),showing statistically significant differences between low risk and high risk patients and between intermediate risk and high risk patients (x2=13.263,6.279,P < 0.05),while no statistically significant difference between low risk and intermediate risk patients (x2 =0.894,P > 0.05).Five-year relapse-free survival rate of low,intermediate and high risk patients was 94.2%,80.0% and 61.8% respectively,with a statistically significant difference (x2=13.547,P < 0.05),showing statistically significant differences between low risk and high risk patients,intermediate risk and high risk patients (x2 =4.357,12.336,P < 0.05),while no statistically significant difference between low risk and intermediate risk patients (x2 =0.696,P > 0.05).Conclusions Compared to open resection,laparoscopic GISTs resection offers better short-term outcomes,however,the two surgical techniques offer equal long-term outcomes.Patients of high risk have poor prognosis.
9.Relationship between mutated k - ras and biological behavior of colorectal cancer

AIM: To investigate mutations of oncogene k-ras in colorectal cancer tissues and the relationship between mutations of k - ras and biological behavior of colorectal carcinoma. METHODS:The specimens of 123 patients with colorectal cancer were collected. Real - time fluorescence quantitative PCR were performed to detect k-ras mutations at codon 12 and codon 13 of exon 1, and the results were analyzed with the corresponding clinical pathological data. RESULTS: Among 123 colorectal cancer cases, point mutations were detected in 53 cases (40.8% ) , point mutations at codon 12 were found in 42 (34.1 % ) cases, and 11(8.9% ) cases at codon 13.No closely relationship between mutations of k-ras and tumor size, location, invasive depth and differentiation extent was observed. The rate of k-ras mutation in the cases with more invaded lymph nodes was higher than that in the cases without invaded lymph nodes ( P < 0.05 ) , and the rate of k-ras gene mutation in the cases with hepatic metastases was higher than that in no hepatic metastases (P <0.05). The rate of k - ras gene mutation was higher in TNM staging Ⅲ/Ⅳ than that in Ⅰ/Ⅱ( P < 0.05 ). CONCLUSION: Mutation of oncogene k-ras plays an important role in the carcinogenesis and development of colorectal cancer, and it is closely associated with invaded lymph notes and hepatic metastases, suggesting that mutation of k- ras indicates a poor prognosis.
10.Reinforcement of disinfection and management of collecting and sending carts in the supply room

Objective To control the incidence of infection during transportation process of supply room,reinforce the management of collecting and sending carts in supply room and ensure the effect of disinfection and safety of use.Methods Available chlorine of 500 mg/L was used to disinfect thein ner face of the clean carts,available chlorine of 1000 mg/L for the outer face,irradiation for 1 hour by ultraviolet lamp before use.The inner and outer faces of the dirty carts were disinfected by available chlorine of 1000 mg/L and irradiation for l hour by ultraviolet lamp.The wheels were flushed by clean water and then sterilized by available chlorine of 1000 mg/L"Clean"or"dirty"was indicated on the carts and they were strictly used and stored.The carts were covered with water-proof cloth,also indicated with"clean"or"dirty".Results The bacteriaJ colony was ≤5 cfu/m2 after disinfection on the clean carts and≤15 cfu/m2 on the dirty carts.Conclusions Disinfection by available chlorine and irradia- tion by ultraviolet lamp can reach the standard of sterile area,clean area and contamination area set by the Ministry of Health.This method is an effective measure in the control of transportation process of supply room with simple,feasible,economical and effective characteristics.
