1.Effect of hyperthermia on the biological functions of human lung cancer cell line H1299 and its molecular mechanism

Objective To explore the influences on biological function induced by hyperthermia in human lung cancer cell line H1299, and to investigate the possible molecular mechanism. Methods H1299 cells were cultured in vitro and divided into 2 groups. The cells in culture flasks of hyperthermia group were immersed into a water bath at 43 ℃for 1 h, and the cells of control group were cultured at 37 ℃. The cell growth was detected by CCK8 assay, and the cell cycle and apoptosis rates were detected by flow cytometry [propidium iodide (PI) staining and PI/Annexin V staining]. The effects of hyperthermia on migration and invasion abilities of H1299 cells were determined by Transwell migration and invasion assays, respectively. The expression of LLGL1 was measured by Western blot. Results The cell cycle had no significant difference between the two groups, but the apoptosis rate was significantly higher in hyperthermia group [(24.81 ±2.80) %] than that in control group [(11.73 ±1.55) %] (t= 7.709, P= 0.0021). The migrating cell number was decreased in hyperthermia group (25.67±4.81) than that in control group (85.00±10.31) (t=5.182, P=0.0066). The invasive cell number was also decreased in hyperthermia group (22.00±2.08) than that in control group (108.3.0±10.14) (t=8.342, P=0.0011). The expression level of LLGL1 protein in hyperthermia group was 4.2 times that in control group(t=3.028, P=0.0389). Conclusion Hyperthermia induces the cell apoptosis and inhibits migration and invasion abilities of H1299 cells, which maybe associate with increasing LLG1 expression.
2.Efficacy analysis of polysomnography and improvement after nocturnal non-invasive ventilation in treating children of spinal muscular atrophy with moderate-severer sleep disordered breathing

Objective To learn about polysomnography (PSG) of the patients with spinal muscular atrophy (SMA),and the improvement of the moderate-severe sleep disordered breathing(SDB) patients after non-invasive ventilation (NIV) treatment.Methods The PSG examination of 11 SMA patients in Children′s Hospital Affiliated to Capital Institute of Pediatrics was performed.The PSG results and the levels of breathing disorder of these patients were collected.These patients were divided into 2 groups as follows.The mild group:apnea hypopnea index(AHI) was between 5 to 10 times/h,or average pulse oxygen saturation(SpO2)>95%;the moderate-severe group:AHI>10 times/h,and average SpO2≤95%.The moderate-severe levels of the patients were treated with NIV.Then the improvement of PSG after the treatment was evaluated.Results Within the 2 cases of type 1 SMA,1 was moderate-severe,the other 1 was mild.There were 2 cases moderate-severer and 6 cases mild SDB cases of type 2 SMA.The only 1 case of type 3 was categorized as mild disorder.The average hypopnea index(6.3 times/h) was higher than apnea index(4.9 times/h).The indicators to distinguish the moderate-severe and mild groups were as follows:AHI,average SpO2,hypopnea index,and arousal index associated with respiratory case,and there were statistically significant differences(all P<0.05).Significant improvement in PSG of the 3 NIV-treated patients was observed and it remained in normal range.The following indicators also remarkably improved after NIV treatment were hypopnea index [(1.5±0.1) times/h vs.(11.5±4.6) times/h,t=3.741,P<0.05],AHI [(2.4±1.1) times/h vs.(17.4±8.1) times/h,t=3.196,P<0.05],and average SpO2 [(96.7±0.6)% vs.(94.3±1.2)%,t=-3.130,P<0.05].Conclusions There are different levels of SDB within SMA patients.The major characteristic of SMA patients is hypopnea.There are more moderate-severer SDB in type 1 than those in type 2.The average SpO2,arousal index associated with respiratory incidents,AHI,and hypopnea index can reflect the levels of SDB of SMA patients.Besides,the results indicate NIV treatment can significantly improve SDB of SMA patients.
3.Telemetric Localization of Detecting Capsule in Gastrointestinal Tract

Objective To develop a method for localizing a detecting capsule in the gastrointestinal tract.Methods A three-axis magnetic sensor was sealed in the detecting capsule.Three flat coils were fixed on patients' back.The coils were excited one by one to generate electromagnetic fields.The position and the orientation of the capsule could be determined by telemetering the signals from the magnetic sensor incorporated in the capsule.Results The telemetric localization system worked successfully.A localization model describing the relationship between the magnetic field strength and the position and orientation of the capsule,was correctly established basing on the principle of magnetic dipoles,and neural network algorithm was employed to solve a non-linear equation set.Conclusion The results showed that the localization method is feasible and the precision is higher than existing methods.The localization device can perform consecutive tracking and can be made into a portable type.After putting into practice,the device can also be used in food selection for astronauts by monitoring their gastrointestinal parameters in space.
4.Effects of targeted treatment of the carboplatin-Fe@C nanocage-loaded chitosan nanoparticles on rats with transplanted liver cancer

Objective To investigate the effects of targeted treatment of the carboplatin-Fe@C nanocage-loaded chitosan nanoparticles (C-Fe@CN-CN) combining external magnetic field on rats with transplanted liver cancer.Methods Twenty-four model rats with transplanted liver cancer were established and divided into four groups randomly (n =6).Abdominal exposure was carried out through a midline incision,and a cannula was inserted into the hepatic artery and fixed.Group A:saline water was injected as control,group B:saline water with 10 mg/kg free carboplatin was given,group C:saline water with C-Fe@CN-CN (equivalent dose of free carboplatin 10 mg/kg) was injected in absence of magnetic field,group D:saline water with C-Fe@CN-CN (equivalent dose of free carboplatin 10 mg/kg) was injected in presence of magnetic field for 30 min.All the animals were sacrificed and abdominal exposure was done again after 7 days.After tumors were reselcted,tumor weight and volume was measured,the inhibiting rate of tumor weight was calculated.Tumor and liver tissues were examined for histological changes.Results The growth of tumor was significantly inhibited after therapy with different forms of carboplatin.There was significant difference in the tumor weight of A,B,C,D groups [(0.85±0.12) g,(0.61±0.10) g,(0.48±0.09) g,(0.33±0.06) g,P < 0.05,respectively].The inhibiting rates of tumor weight of B,C,D groups were 28.9 %,43.4 %,61.7 % respectively.The inhibiting rate of D group was highest which was 1.1 times higher than that of B group.There was also significant difference in the tumor volume of A,B,C,D groups [(1.06±0.24) cm3,(0.72±0.10) cm3,(0.50±0.07) cm3,(0.28±0.05) cm3,P < 0.05,respectively].The tumor volume of group A was largest which was 2.8 times larger than that of group D.In group D,tumor tissues from six rats presented severe necrosis,and nanoparticles were concentrated in the necrotic tissue.In group C,five rats presented middle necrosis,one rats presented severe necrosis.There was no concentration of nanoparticles in the necrotic tissue.In group B,four rats presented middle necrosis,two rats presented mild necrosis.In group A,six rats presented mild necrosis.Conclusion C-Fe@CN-C can significantly increase the therapeutic effects of carboplatin by hepatic artery injection combining with an external magnetic field on the tumor.
5.Perceived organizational support's moderating effect on the relationship between personality and emotional exhaustion of Chinese armed police force

Objective To test the hypothesis that perceived organizational support(POS) plays a moderate role in the process of personality' s effect on emotional exhaustion.Methods Using POS,Revised NEO Personality Inventory and Emotional Exhaustion Inventory,376 special combat soldiers were tested from the Chinese Armed Police Force.Then SPSS 16.0 was used to conduct descriptive analysis,correlational analysis and hierarchical regression analysis.Results The interaction of neuroticism and POS as well as the interaction of conscientiousness and POS were significant or marginally significant,respectively.POS significantly moderated the relationship between neuroticism and emotional exhaustion(β=-0.19,t=-2.82,P<0.01)as well as the relationship between conscientiousness and emotional exhaustion(β=-0.12,t=-1.95,P=0.052).Conclusion Neuroticism and conscientiousness are more strongly related with emotional exhaustion among individuals with low POS than high POS.
6.CT Contrast Enhancement in Infants : Observation of the Drug Application and Experience of Nursing

Objective To discuss drug application and nursing experience during CT contrast enhancement in infants.Methods CT contrast enhancement with Ultravist,one of the non-ionic contrast media,was performed on 202 infants.Before examination,all the infants were calmed with Diazepam or Chloral Hydrate(10% in concentration).The doses of the drugs were determined by the weight of these infants.Results Non-ionic contrast media was not absolutely safe and a few infants had side effects.Only 7 infants had positive allergic tests.The results of CT contrast enhancement on 202 infants were satisfied.Conclusion During CT enhancement,appropriate use of contrast media and sedatives as well as proper nursing according to the infants’ age will get a better result.
7.Study on Shattering Extraction with Solvent for Jingzhi Guanxin Granules

OBJECTIVE: To optimize the extraction technology of Jingzhi guanxin granules.METHODS: The extraction technology of Jingzhi guanxin granules were optimized using shattering extraction with solvent,refluxing extraction and primary extraction with the content of tanshinone ⅡA,salvianolic acid B,paeoniflorin,ferulate and hydroxysafflor yellow A and extraction rate as index.RESULTS: The optimal extraction process was as follows: Jingzhi guanxin powder,6 fold 95% alcohol,extracting for 10 min in the first time,6 fold 60% alcohol,extracting for 10 min in the second,6 fold 20% alcohol,extracting for 10 min in the third.CONCLUSION: Using shattering extraction with solvent for Jingzhi guanxin granules,extraction rate of each constituent are higher than that of primary extraction.This extraction process is simple in operation and save time.

The enzyme activity of α-Acetolactate Decaroboxylases (ALDC)from different microbes was studied, the results demonstrated that it was quite different among them. There were diversities of their enzyme reaction velocities. It was clear that the enzyme activity was affected by the pH of the enzyme reaction system, for example, the optimum pH of ALDC from Lactococcus lactis was 6.6, while for Aerobacter Aerogenes it was 5.8. Addition leucine,valine and isoleucine into enzyme reaction system obviously affected the enzyme activity of ALDC from different microbes.
9.Selection of Ginkgo biloba cell line for suspension culture and studies on stability of ginkgolide B in subculture

Object To select the cell line of Ginkgo biloba L that may produce high yield of ginkgolide B (GB) in suspension culture and to study the stability of GB in subculture Methods Calculus was induced with stem, root and leaf of selected elite species and high yield suspension cell lines chosen by hypoxia stress Results Seven suspension cell lines with improved yield of GB were obtained Among which cell line MH 3 gave a 3 71 fold increase of cellular bioproduct with an increased content up to 302 ?g/g DW after culturing for 18 d, a leading record country wide In shaking bottle culture, it showed a consistent yield of GB with a content of 291 ?g/g DW in 6 successive subcultures, coefficient of variation=0 131 Conclusion The suspension cell line selected by hypoxia stress gave a higher yield and stability in successive transfer culture
10.Research Advance on Diketopiperazines Produced by Marine Microorganism

The basical structure of diketopiperazines is a cyclic dipeptide condensed by two amino acids. Because of the stable framework of the six-member ring structure, and having two hydrogen bond donor and two hydrogen bond receptor, DKPs have become important chemical pharmacophores, with strong biological activities and pharmacological activities in the drug. A series of cyclic compounds were found from marine organisms in recent years, research showed that their functions are not limited on anti-bacterial, cytotoxic activity, and so on, but also playing an important role in regulatory mechanism of quorum sensing as signal molecules, they have become research hot point in ecological chemistry. This paper reviewed the research progress of diketopiperazines found in the marine microbial metabolites, and the future study trends was discussed and outlooked.
