2.Comparative study of the influence of silicone oil and heavy silicone on ultrastructure of rabbits' retina

Objective To investigate the medium and long-term influence of silicon oil versus heavy silicone oil on rabbit retinas.Methods 28 health standard rabbits were randomly divided into A,B and C groups,with 12,12 and 4 rabbits respectively.All rabbits received routine vitrectomy and tamponade with silicone oil (group A),or heavy silicone oil (group B) or balanced salt solution (group C).After 4,8,12 and 24 weeks,the retinal b-wave amplitude was measured by ERG,posterior retinal thickness was measured by optical coherence tomography (OCT).Retinal ultrastructure and tissue morphology were observed by transmission electron microscopy and optical microscopy.Results Compare to group C,the b-wave amplitude decreased at 4 weeks after surgery,and decreased at 8 weeks after surgery for group B,and decreased at 8 weeks after surgery,and decreased at 24 weeks after surgery for group A.The decreases were greater in group B than group A at 8,12,24 weeks after surgery,the difference was statistically significant (P<0.05).The posterior retinal thickness of group A and B was thinner than group C at 24 weeks after surgery (P<0.05).The decreases were greater in group B than group A,the difference was statistically significant (P<0.05).Transmission electron microscopy and optical microscopy revealed severe pathological changes of retinal ultrastructure and morphology in group A and B rabbit eyes,at 12 weeks and 8 weeks after surgery respectively.The changes were more severe in group B and group A,including edema and necrosis in cone/rod cells,in disk membranes,mitochondria,cytoplasm,nucleus and other organelles.The morphological changes were also more severe in group B and group A,including degenerations of ganglion cell layer,inner nuclear layer changes.Those changes became more severe when the tamponade time extended.Conclusion The heavy silicone influence on visual function,ultrastructures,histomorphology of rabbit retinas is much worse than the silicon oil,and the effect is more significant with its time prolong.
3.Study the animal model of experimental myasthenia gravis with peptide 125-147 of the acetylcholine receptor ?-subunit

Objective Investigate animal model of experimental myasthenia gravis with peptide 125-147(T?125-147)of the acetylcholine receptor(AchR)?-subunit.Methods 12 Lewis rats of experiment group were injected in hypo with synthesized peptide T?125-147 of AchR of electric eel.The muscle fatigability,repetitive nerve stimulation(RNS)for decrement response and ELISA assay for serum AchRAb titers were observed and compared with control group.Results A week after the second inoculated,6 rats of experiment group appeared weakness.The RNS and serum AchRAb titers of 11 rats of experiment group were positive,none of the control group appeared weakness,all RNS were negative,serum AchRAb titers of 7 rats were negative and of 1 rat was probable positive.There were significant differences between experiment group and control group(all P

5.Protective effect of both human TH and GDNF cDNA-engineered SH-SY5Y cells on primary dopaminergic neurons

Objective To construct genetically engineered cells that secrete human TH and GDNF at the same time by stable co-transfection pcDNA3.0/hTH and pcDNA3.1/hGDNF in SH-SY5Y cells and study their effects on the gene therapy of Parkinson′s disease (PD). Methods pcDNA3.0/hTH and pcDNA3.1/hGDNF were constructed by inserting human TH and GDNF cDNA into pcDNA3.0 and pcDNA3.1, respectively. SH-SY5Y cells were transfected with pcDNA3.0/hTH and pcDNA3.1/hGDNF and the positive cell clones of human TH and GDNF cDNA engineered cells could be identified by RT-PCR. The engineered cells were co-cultured with primary dopaminergic neurons in mouse. The number and growth condition of primary dopaminergic neurons were examined by Immunocytochemistry. 7.Observation of Tanreqing Injections Combined with Azithromycin in the Treatment of Pediatric Bronchial Pneumonia

Objective: To investigate the effect of Tanreqing injections combined with azithromycin in the treatment of children bronchial pneumonia. Methods:Totally 56 cases of pediatric bronchial pneumonia were selected in our hospital from September 2012 to September 2013 and randomly divided into the observation group and the control group with 28 cases in each. The control group was with azithromycin treatment, and the observation group was with Tanreqing injections additionally. The effect and adverse reactions of the two groups were studied and compared. Results:In the observation group, the effective rate was 92. 9%, which was significantly higher than that in the control group(P<0. 05). The hospitalization time and the symptoms disappearance time in the observation group were significantly shorter than those in the control group (P<0. 05). The adverse reactions showed no significant difference be-tween the two groups. Conclusion:Tanreqing injections in the treatment of children bronchial pneumonia is rapid, effective and safe.
8.Clinicopathologic characters and lymphatic metastasis of cancers at the gastroesophageal junction

Objective To summarize the clinicopathologic characters and the route of lymphatic metastasis of cancers at the gastroesophageal junction. Methods Clinicopathologic data of 86 cancer patients treated from October 2000 to December 2004 were analyzed retrospectively. Results There were 66 males and 20 females, the mean age was 60 years. Most patients were of Bormann typeⅢadenocarcinoma. The incidence of high differentiated adenocarcinoma in TypeⅠcancer was higher than that in other two types (P = 0. 002, P = 0. 004) , while the incidence of poor differentiated carcinoma in typeⅢcancer was higher than other two types(P = 0. 005 ,P = 0. 015). 9.Changes in expression of spinal aquaporin-4 during remifentanil-induced hyperalgesia in a mouse model of incisional pain

Objective To evaluate the changes in the expression of spinal aquaporin-4 (AQP4) during remifentanil-induced hyperalgesia in a mouse model of incisional pain.Methods Seventy-two pathogen-free healthy adult male CD1 mice,weighing 25-30 g,were divided into 4 groups (n=18 each) using a random number table:control group (group C),incisional pain (group I),remifentanil group (group R) and remifentanil plus incisional pain group (group R+I).Normal saline was infused subcutaneously in group C.An incision was made in the left hind paw in group I.Remifentanil 80 μg/kg was subcutaneously infused for 30 min at a rate of 0.8 ml/h in group R.Remifentanil was infused subcutaneously before establishment of the model in group R+I.The thermal paw withdrawal latency (TWL) and mechanical paw withdrawal threshold (MWT) were measured at 1 day before establishment of the model (T0) and 6 h and 1,2 and 7 days after establishment of the model (T1-4).After measurement of the pain threshold at T3,12 animals were sacrificed randomly,and the lumbar segment of the spinal cord was removed for determination of the distribution and expression of AQP4 by Western blot.Results Compared with group C,the TWL was significantly shortened at T1-3,and the MWT was decreased at T2-4 in R and R + I groups,and the expression of AQP4 was significantly up-regulated at T3 in I,R and R+I groups (P<0.05).Compared with group I,the TWL was significantly shortened at T2,3,and the MWT was decreased at T2.4 in group R,and the TWL was significantly shortened at T1-3,the MWT was decreased at T2.4,and the expression of AQP4 was up-regulated at T3 in group R+I (P<0.05).Conclusion The mechanism by which remifentanil induces hyperalgesia is related to up-regulation of AQP4 expression in the spinal cord in a mouse model of incisional pain.
10.Effect of lysophosphatidylcholine on cholesterol efflux from macrophage foam cells

AIM: To explore the effect of lysophosphatidylcholine(LPC) on cholesterol efflux from macrophage foam cells, and to offer experimental evidence for research on prevention and treatment of atherosclerosis.METHODS: Peritoneal macrophages and LDL were seperated from mice and serum of healthy volunteers, respectively. The foam cells were derived from macrophages in the presence of Acylated LDL (AcLDL). Cholesterol efflux from cells and LDH activity were measured by enzymetic fluorometry and LDH kit, respectively.RESULTS: After incubated with LPC for 24 hours, cholesterol efflux from macrophage foam cells increased significantly compared to control, and cellular cholesterol was lower than that in control group. At the same time, medium LDH activity of LPC group was not increased obviously. CONCLUSION: Within the dosage of 10-80 ?mol/L, LPC can promote cholesterol efflux from macrophage foam cells in a dose-dependent manner, and this effect has nothing to do with cytotoxity of LPC.
