1.THE PROTECTIVE EFFECT OF BERBAMINE ON CEREBRAL INFARCTION MODEL IN RATS

The cerebral infarction model was made according to Chen et al To evaluate the severity of infarction the quantity of Evan's blue was measured. The cerebral infarction size was determined by TTC stain. The results showed that ligating the right common carotid artery and the right middle cerebal artery and clamping the left common carotid artery could induce cerebral cortex in farction. Berbamine could decreased the level of Evan's blue and reduce the infarction size compared with control group. It suggests that berbamine possesses protective effect on experimental induced cerebral infarction.
2.STUDY ON THE MECHANISM OF RELAXATION OF BERBAMINE ON THE ISOLATED RABBIT RENAL ARTERIES

The contraction of the isolated rabbit renal arteries induced by noradrenaline, high K+ & calcium could be relaxed by berbamine ( BA ). BA shifted dose-response curve of noradrenaline, high K+ snd calcium to the right, but the maximal response was reduced. The antagonistic parameter pD' 2 of berbamine on calcium contrac-tion was 4.76, which is similar to the verapamil (pD'2=5.68). Therefore berbamine can be considered as a non-competitive calcium antagonist.

Objective To summarize the hot spots and cutting-edges by exploring Multidisciplinary Team (MDT) research in cancer.Methods CiteSpace is used to analyze MDT in cancer visually.Results 2 160 related literatures,10 key literatures,15 main clusters and 29 burst terms are detected.Conclusion MDT research on cancer mainly focuses on operation process,implemention and evaluation of effects.Improving efficiency,cost-effectiveness analysis and randomized controlled trial is the cutting-edge topics.This provides a new perspective for China.
5.Efffect of LincRNA-p21 on Proliferation of Renal Cell Carcinoma

Objective To study the effect of lincRNA-p21 on the proliferation of renal cell carcinoma.Methods The expression levels of lincRNA-p21in renal cell carcinoma tissues and their corresponding para-cancerous tissues from 17 patients in department of nephrology,Hua Xi hospital,was detected by RT-PCR.In addition,the expression of lincRNA-p21 was analyzed in human clear cell RCC cell line 786-O and human embryonic kidney cell line HEK-293.The cell proliferation,apoptosis and the expression of Bax、Noxa、Puma that participated in p53 signaling pathway were detected after lincRNA-p21 was downregulated with siRNA.Results The expression levels of lincRNA-p21was higher in renal cell carcinoma tissues and 786-O cells than para-cancerous tissues and HEK-293 cells respectively(P < 0.05).The ability of proliferation was significantly increased after lincRNA-p21 siRNA treatment in 786-O cells,whereas the cell apoptosis was decreased (P < 0.05).The mRNA expressions of Bax,Noxa,Puma were also dramatically decreased after lincRNA-p21 was downregulated(P <0.05).Conclusion LincRNA-p21 inhibits the cell proliferation of 786-O cells through participation in p53 signal transduction pathways.These data suggested that lincRNA-p21 maybe could be used as molecular targets for human RCC treatment.
6.THE MECHANISM OF DILATING VASCULAR SMOOTH MUSCLE BY N-METHYLBERBAMINE

The study showed that NMBA could inhibit the rabbit thoracic aorta contractions induced by KC1, CaCl2 and NA, and the pD' 2 values were 3.44, 3.67 and 3.73 respectively. NMBA could also antagonize the extracellular Ca2 + contraction of NA with no effect on the intra-cellular Ca2+ contraction. Comparing with verapamil, the mechanism of NMBA action was inferred to blocking Ca2+ influx during excitation. However, further study will be needed.
7.EFFECTS OF BERBAMINE HCL ON ISOLATED WORKING HEART OF GUINEA PIG

The isolated working heart of guinea pig perfused with Tyrode's solution could work normally for at least 60 min. It was shown that berbamine ( BA ) could depress the function of isolated working heart of guinea pig in dose-dependent manner. BA 3 mol/L could decrease the left ventricular pressure, aortic pressure, -dP/dtmax, aortic blood flow and coronary blood flow, and increase left ventricular end-diastolic pressure. BA 100 mol/L could result in the ventricular asystole, however, no obvious influence the contraction of atrium.It was also demonstrated that BA could antagonise the arrhythmias induced by the adrenaline in isolated working heart of guinea Pig.
8.Chemical constituents from chinese marine sponge Cinachyrella australiensis

Objective: To investigate the secondary metabolites from Chinese marine Sponge Cinachyrella australiensis. Methods: Column chromatography techniques including HPLC were used for the separation and purification of the compounds, and extensive spectral analyses including various 2D NMR spectra were employed for structure elucidation. Results: Nineteen compounds were obtained ,including 2 methyoxy 6,12,15 trien 8 yne octadecanoic acid(1), 2 benzenedicarboxylic acid dibutyl ester(2), 1,2 benzenedicarboxylic acid bis(2 ethylhexyl)ester(3), ( ) (3S) 1,2,3,4 tetrahydro ? carboline 3 carboxylic acid(4), L Tryptophan (5), p hydroxylbenzaldehyde (6), p hydroxyl benzylethanol(7), p hydroxyl benzyl propanol(8), cholesta 4 en 3 ol(9), 2 methyl 6 amino 9 (2 deoxy ? D ribofuranosyl purine(10), 2' Deoxyadenosine (11), 6 amino 9 ? D ribofuranosyl 9H purine (12),uracil(13), thymine(14), thymidine(15), 1 (2 deoxy ? D Ribofuranosyl)uracil(16), 1 ethyl ? (2 deoxy) ? D ribofuranos(17),isolumichrome(18),and zarzissine(19).Conclusion:Compounds 1 and 18 are new natural products,and compounds 2 to 17 as well as 19 are isolated from this species for the first time.
9.Manifestation of dynamic contrast-enhanced CT of hepatic injury in rabbits

Objective To analyze the features of dynamic contrast-enhanced CT of blunt hepatic injury in rabbits. Methods The model of blunt hepatic injury was established in 40 New Zealand white rabbits with a steel ball falling down to the xiphoid process of the animals. Plain CT scan and dynamic contrast-enhanced CT scan (Hispeed spiral CT/2i, GE, America) of the liver were performed. Arterial,portal and balanced phases were respectively at 8-10 s, 35-40 s and 120-150 s after initiation of the contrast medium injection. The non-enhanced and enhanced images were compared in aspects of location and range of injury, tear of the liver capsule, active bleeding, involvement of the main hepatic veins and CT features of abdominal hemorrhage, which was further compared with the results of gross anatomy. Results The rate of plain CT scan was obviously lower than that of dynamic contrast-enhanced CT scan,which defined single tear in 13 patients, multiple lacerations in 18, liver subcapsular hematoma in seven,liver hematoma in nine, liver coated gap in 17, active bleeding in nine and main hepatic vein injury in five, with coincidence rates with the results of gross observation for 13/13,18/18,7/9,9/9,25/30,9/5and 5/4 respectively. According to Moore' s classification, CT/Laparotomy performed from grade Ⅰ to grade Ⅵ were 5/4 patients at grade Ⅰ , 15/13 at grade Ⅱ, 9/11 at grade, 5/6 at grade Ⅳ, 1/2 at grade Ⅴ, O at grade respectively. Conclusion Dynamic contrast-enhanced CT scan, especially at portal and balanced phases, is of great value for diagnoses of liver injuries and determination of injury severity.
10.Pathological validation of a novel experimental chronic compressive cervical myelopathy rat model

Objective To establish a novel chronic compressive cervical spinal cord injury rat model,to validate the chronic pathological characteristic.Methods Fifty-four SD rats were randomly divided into 3 groups,including control group(n =6),acute compressive group(4 h,24 h,respectively.n =6),and chronic compressive group(4 h,12 h,24 h,48 h,72 h and 1week,respectively.n =6).Two sizes of water-absorbing polyurethane polymer sheets were implanted into C5~6 epidural space on postero-lateral side in acute and chronic compressive group respectively,to induce a consistent compression in the cord after expanded.While a laminectomy on C5~6 was performed only in control group.The neurological integrity,MRI signal change in the cords,large motoneuron number in the ventral horn,and myelin staining intensity on posterior funiculus were studied.Results In the acute group at 4-24 h,the compression was confirmed significantly on T2WI image,as well as hypointense signal change intramedullary.These changes were consisted with intramedullary bleeding,tissue necrosis.Large motoneuron number(P < 0.05),rather than myelin staining intensity (P > 0.05),was significantly decreased compared with the control group and chronic compressive group.BBB score was 6.0 at 24 h.In the chronic group:cord distortion with progressive compression was observed on T2WI image,but without intramedullary bleeding signal change.At 4-12 h,intramedullary edema,central canal distortion were seen in the cords.Large motoneuron number and myelin staining intensity decrement were not significant(P >0.05).BBB score was 20.6.At 24-72 h,central canal enlargement,venous congestion,and edema were observed.Large motoneuron number was less than that in the control group.In the compressive epicenter,nerve fiber disorganization or rupture was observed.Myelin staining intensity decreased significantly after 48 h and 72 h compression (P <0.05).BBB score was 19.3.At lweek,vacuolation changes were noted and large motoneuron decreased (P < O.05),as well as myelin density and staining intensity (P < 0.05),suggesting nerve fiber demyelination.BBB score was 17.5.In the control group,there was no neurological deficit and pathological change in the cords.Conclusion The pathology and MRI characteristic consistent with chronic compressive injury change,which proved this method is able to induce a chronic course on the rat model,and established a reliable model foundation for cervical myelopathy.
