1.Relationship between fetal karyotype and age of pregnancy

Objective To evaluate the possibility of elevating the age line for pregnant women with advanced age to conducted amniocentesis .Methods A retrospective cohort study was performed .The information of 5 176 advanced age pregnant women in July 2003 to August 2015 from Ningbo Women and Children Hospital were collected and analyzed .Grouped the patients by age , using chi-square analysis , comparing the difference of abnormal rate of target chromosomes (13, 18, 21) and sex chromosomes (X and Y) between the adjacent two age groups .Results Total of 15 408 cases were screened.Among these, 262 cases were diagnosed with abnormal chromosome .The positive rate was 1.70%.Total of 5 176 advanced age pregnant women were included , 82 women were found with abnormal target chromosomes (13, 18, 21) and sex chromosome.The positive rate was 1.58%(82/5 176).For age groups, the positive rates were 0.95%(11/1 156), 0.94%(9/954), 0.87%(10/1 144), 2.16%(14/648), 2.16%(10/464), 2.51%(10/399) and 4.38%(18/411) respectively.Chi-square analysis results showed that the positive rate between group of 37 and 38 was significant difference (chi-square value of 5.181,P value of 0.023).There were no significant differences among all remained groups ( chi-square value: <0.001, 0.028, <0.001, 0.117, 2.129 respectively, P value: 0.985, 0.868, 0.995, 0.733, 0.145 respectively ) .Conclusion It is worthwhile to elevate the age of pregnant women up to 38 years old as the age line for the indications of amniotic fluid chromosome analysis .
2.Distribution and Drug Resistance of Clinically Common Non-fermentative Gram-negative Bacilli

OBJECTIVE To explore the characteristics of the clinical distribution and drug resistance of non-fermentative Gram-negative bacilli in nosocomial infection.METHODS The bacteria were identified by API 20NE.Antibiotic susceptibility test was performed by K-B disk diffusion method.RESULTS The rate of non-fermentatives in Gram-negative bacilli was 17.6%.The dominant strains were Pseudomonas aeruginosa(43.8%),Acinetobacter spp(39.9%),Stenotrophomonas maltophilia(6.7%),and Burkholderia cepacia(2.7%).The resistant rates of P.aeruginosa to cefoperaxone/sulbactam,amikacin,cefepime and meropenem were less than 20%.The resistant rates of Acinetobacter spp to imipenem,meropenem,cefoperaxone/sulbactam and minocycline were less than 20%.The resistant rates of S.maltophilia to minocycline,cefoperaxone/sulbactam,piperacillin/tazobactam and levofloxacin were less than 20%.The resistant rates of B.cepacia to minocycline,piperacillin/tazobactam and cefoperaxone/sulbactam were less than 20%.CONCLUSIONS The sensitivities of non-fermentative Gram-negative bacilli to cefoperaxone/sulbactam,piperacillin/tazobactam,minocycline,imipenem and meropenem were the highest.These antibiotics may be chosen first for the clinical treatment.
3.Study on the mitochondria Ca 2+, expression of Bcl-2 and Bax and germ cell apoptosis in testes of rats with varicocele

AIM: The cytoplasm Ca 2+, expression of Bcl-2 and Bax and germ cell apoptosis of testes in adult male rats with varicocele were observed to investigate the relationship between apoptosis and infertility. METHODS: Thirty-five healthy male Wistar rats were divided into two groups randomly: varicocele group (VG, n=20) and sham operation group (SOG, n=15). Bilateral testes were removed after ten weeks. One part of it was homogenized, the other part was performed for tissue sectioning. The atomic absorption method was used to detect the cytoplasm Ca 2+. The germ cell apoptosis was detected by the TdT-mediated dUTP-biotin nick end labeling (TUNEL) technique. The expression of Bcl-2/Bax was detected by immunohistochemical method. RESULTS: The cytoplasm Ca 2+ in testes of rats with varicocele was decreased obviously. The apoptosis indexes of germ cells and Bax expression in VG were significantly increased as compared with those in the SOG, while Bcl-2 expression was obviously decreased. CONCLUSION: The results showed that apoptosis was increased in testes of VG rats, suggesting that male infertility with varicocele may be caused by some apoptosis agents such as high temperature, testes toxin and oxygen free radical. The cytoplasm Ca 2+ and change of expression of Bcl-2/Bax may contribute to germ cell apoptosis and cause male infertility.
4.Target genes of the hsa-miR-106b-25 cluster:Prediction and bioinformatic analysis

Objective The hsa-miR-106b-25 gene cluster is involved in various biological processes of carcinoma .This study aims at a prediction and function analysis of the hsa-miR-106b-25 gene cluster, miR-106b, miR-93, and miR-25, so as to provide some evidence for further studies on the functions of the three miRNAs and the mechanisms of their interaction . Methods We obtained the sequences of miR-106b, miR-93, and miR-25 from the miRBase, predicted their target genes with TargetScan , PicTar, and miRanda, and used 3 or more experimentally verified target genes from the miRTarbase as the gene set for further bioinformatic analysis .We predic-ted the biological processes of the target genes by GeneOntology analysis and enriched KEGG ( Kyoto Encyclopedia of Genes and Genomes) by pathway analysis, produced protein-protein interaction ( PPI) networks with STRING . Results The target genes of the miR-106b-25 gene cluster were significantly enriched in such biological processes as the regulation of macromolecule metabolism , regulation of metabolic process , and cell cycle process , while the KEGG pathway mainly in glioma, melanoma, prostate cancer , and gallbladder carcino-ma.The proteins encoded by the targeted genes of showed complicated interactions , and those encoded by the KAT2B, PTEN, TP53, CDH1, MDM2, E2F1, RB1, and SMAD7 plaid a core role in the interac-tion network. Conc lusoi n MiRNAs of the miR-106b-25 gene cluster regulate the downstream target proteins involved in tumorigenesis by participating in the cell cycle and cancer signaling pathway .
5.Analysis on clinical distribution and antimicrobial resistance of common gram-positive cocci

Objective To investigate the clinical distribution and antimicrobial resistance of common gram-positive cocci in author's hospital.Methods Identification of these bacteria were done with API analysis system,disc diffusion tests were employed to study the antimicrobial resistance.Results A total of 25 052 clinical isolates were collected in 8 years,of gram-positive cocci accounted for 7907(31.9%).Staphylococcus aureus(3549 strains,44.9%),enterococcus(1760 strains,22.3%)and coagulase-negative staphylococcus(1558 strains,19.7%)were the most common isolates.The prevalence of MRSA increased from 59.6% in 2001 to 76.3% in 2008,and MRSCoN increased from 64.2% to 77.0%.The resistant rate of MRSA to gentamicin,clindamycin,erythromycin and levofloxacin were over 90%,to trimethoprim/sulfamethoxazole and chloramphenicol were less than 20%.The resistant rate of MSSA to gentamicin,levofloxacin,trimethoprim/sulfamethoxazole and chloramphenicol were low 20%,and to beta-lactamase antibacterial agents except penicillin were 0.The resistant rates of MRSCoN to all antimicrobial agents were lower than MRSA,but to trimethoprim/sulfamethoxazole(71.2%)was higher than MRSA(21.2%).No staphylococcus strains were resistant to vancomycin,teicoplanin and linezolid.2.1% enterococcus feacium and 4.4% other enterococcus were resistant to vancomycin.No strains of enterococcus were found resistant to teicoplanin and linezolid.Conclusion The resistant rate of gram-positive cocci were increasing obviously,the prevalence of MRS was high.Vancomycin,teicoplanin and linezolid were the most active agents against severe infection induced by multidrug-resistant gram-positive cocci.
6.Effects of growth-discordant twin pregnancies on thyroid function of neonates

Objective To investigate the effects of growth-discordant twin pregnancies on neonatal thyroid stimulating hormone (TSH) level and congenital hypothyroidism (CH).Methods A total of 3 444 live-birth twin neonates born between January 1,2012 and December 30,2014 in Ningbo City were enrolled.Blood samples via heel puncture were collected and tested.Incidence of CH in singleton and twin neonates was compared.Deviation of birth weight larger than 25% in twin neonates was set as the criteria for discordant growth.TSH and 17 α-hydroxylase levels in CH twins and normal twins,with or without discordant growth,were compared.Chi-square and non-parametric statistics were performed for data analysis.Results The incidence of CH in twin neonates was 0.56% (19/3 444),higher than that in singleton neonates [0.09% (203/225 712),x2=76.225,P<0.01].Among nineteen CH twins,CH occurred in both twins in eight cases (four twins) and in one of the twins in eleven cases.The gestational age at birth in the eight CH twins were less than 37 weeks,with four males and four females;five were low birth weight infants;one twin were dichorionic,and three twins were monochorionic.In the eleven cases of CH occurring in one of the twins,the gestational age was less than 37 weeks in nine cases,eight were low birth weight infants,six were male and five female;seven were monochorionic and four were dichoronic twins.Five cases of temporary hypothyroidism were all low birth weight infants among the growth-discordant twins.CH cases in growth-discordant group had lower birth weight than their normal twins [M(P25-P75),2 100 (1 800-2 600) vs 2 770 (2 530-2 960) g,Z=4.369],and a higher TSH level [15.4 (11.8-18.5) vs 6.4 (4.8-7.9) mU/L,Z=6.339] (both P<0.05).In normal twins with or without discordant growth,the neonates with a lower birth weight had a higher TSH level [3.6(2.5-4.7) vs 2.4(1.8-2.9) mU/L,Z=0.962] in weight consistent group,compared with 6.0(4.4-7.8) vs 3.4(1.9-4.1) mU/L in weight inconsistent group (Z=4.369),both P<0.05.Conclusions In the growth-discordant twins,neonates with a lower birth weight have a higher TSH level and a higher risk of temporary hypothyroidism.
7.Effects of Guiqi Polysaccharide on Telomerase Activity in Premature Senescence WI-38 Cells

Objective To investigate the effects of Guiqi polysaccharide (GQP) on the telomerase activity in premature senescence of WI-38 cells;To discuss its mechanism of action.Methods MTT assay was used to observe the effects of different concentrations of H2O2 on the proliferation of WI-38 cells and screened the best concentration of H2O2 to induce premature senescence cellular model in WI-38 cells. ELISA and chemical staining method were used to evaluate the protection of GQP in telomerase activity and senescence-associated β-galactosidase activity in premature cellular senescence.Results 200μmol/L H2O2 treatment could induce premature senescence in WI-38 cells. There were no significant differences in senescence-associated β-galactosidase and telomerase activity between GQP group and normal control group (P>0.05), but had significant difference with model group (P<0.01).Conclusion GQP can increase the telomerase activity and inhibit the senescence-associated β-galactosidase activity in premature senescence of WI-38 cells.
8.Radioimmunoimaging of 131I Labeled Human scFv Against Anaplastic Thyroid Carcinoma in Tumor Bearing Nude Mice

Purpose To study the radioactive purity and activity of 131I labeled human single chain variable fragments antibodies (scFv) against anaplastic thyroid carcinoma (ATC),and to explore its distribution and radioimmunoimaging characteristics in tumor bearing nude mice model in vivo so as to provide a new method for anaplastic thyroid carcinoma diagnosis and treatment.Materials and Methods The nude mice model bearing human anaplastic thyroid carcinoma was constructed.The chloramine T method was used to label scFv with 131I and the Sephadex G25M was used for purification of labeled scFv.Labeling rate was determined by trichloroacetic acid method;radiochemical purity,room temperature stability and serum stability were examined using paper chromatography.131I-scFv was injected via tail vein in mice,and the distribution of 131I-scFv in body tissues and organs was analyzed at 12,24,48,72 h after injection.Static SPECT imaging was performed at 12,24,48,72 h after injection to observe the intratumoral accumulation of radioactivity.The SPECT/CT image fusion was performed when the tumor tissues were clearly visible.Results 131I-scFv was purified,and the labeling rate was 91.64%;the radiochemical purity was (93.3 ±0.3)%.The radiochemical purity of 131l-scFv placed at room temperature and the serum for 1,6,12,24 h were all >90%.The radioactive distribution of 131I-scFv in tumor,liver,kidney,intestine and blood was high.SPECT imaging showed 131I-scFv was selectively concentrated in tumor tissue;the target/non-target ratio was the highest at 48 h,and the imaging was most satisfactory.Conclusion 131I-scFv can be successfully prepared.SPECT imaging of 131I-scFv in nude mice model is satisfactory,which lays the foundation for further research in ATC diagnosis and treatment.
9.Prognosis of metabolic volume parameters by 18 F-FDG PET/CT in stage Ⅱ-Ⅲ non-small cell lung cancer

Objective To evaluate the prognostic value of 18F-FDG PET/CT-based metabolic tumor volume (MTV) and total lesion glycolysis (TLG) in stage Ⅱ-Ⅲ non-small cell lung cancer (NSCLC).Methods All of 47 patients with pathological stage on Ⅱ or Ⅲ NSCLC who underwent PET/CT before treatment were retrospectively reviewed.Kaplan-Meier method,Log-rank test and COX proportional hazard model were used to analyze the relationship between metabolic parameters and overall survival (OS),progression free survival (PFS).MTV were measured using a cutoff of 40% maximum standard uptake value (SUVmax).Results The median OS and PFS was 28.93 and 17.37 months,and the 1-,2 and 3-year overall survival rate was 80.85% (38/47),59.57% (28/47),38.30% (18/47),respectively.Univariate analysis suggested that SUV MTV and TLG had significant differences on OS and PFS (all P<0.05).On multivariate analysis,TLG was a risk factor for PFS (HR=5.591,P<0.001),and MTV was a risk factor for OS (HR=4.369,P<0.001).MTV was correlated with TNM-staging (r=0.449,P =0.012).Conclusion In patients with stage Ⅱ or Ⅲ NSCLC,MTV is a independent significant prognostic factor for OS,and TLG is able to predict the progression.
10.The mediating effect of resilience between family support and benefit finding in diabetic patients

ObjectiveTo investigate the status of benefit finding in diabetic patients,and mediating effect of resilience between family support and benefit finding in diabetic patients.MethodsFive hundred and thirteen diabetic patients from a hospital of Harbin were recruited as investigative subjects from September 2015 to June 2016.The resilience scale,family adaptability,cohesion scale and benefit finding scale were used in the study.ResultsThe scores of benefit finding,resilience and family adaptability and cohesion were 55.67±12.87,70.65±10.39 and 104.25±15.38,separately.The total score of family adaptability and cohesion was positively related to the total score of benefit finding(r=0.389,P<0.05).The total score of resilience was positively related to the total score of benefit finding(r=0.155,P<0.05).The total score of family adaptability and cohesion was positively related to the total score of resilience(r=0.112,P<0.05).Benefit finding as the dependent variable,the results of hierarchical regression analysis indicated that family adaptability and cohesion (β=0.382,P<0.01),resilience(β=0.098,P<0.01)entered the regression equation.There was a partial mediating effect of resilience between family support and benefit finding in diabetic patients.The mediating effect was 0.011,accounted for 2.8% of the total effect.The resilience was the important protective factor for benefit finding.ConclusionFamily support has a direct influence on benefit finding and an indirect effect mediated by resilience on benefit finding.
