1.Incision concrescence of a nano-sized hydroxyapatite-zirconia bioceramical scalpel

BACKGROUND: Hydroxyapatite is a kind of ideal orthopedic material, but its Iow strength and brittleness need to improve. The research suggested that the improvement of the toughness and strength of bioceramics which composite hydroxyapatite (HAP) and zirconia (3% mol yttria-stabilized cubic zirconia) and fabricated ceramic scalpel possesses of the wound recover faster with smaller side. So it is significant that research on the incision concrescence of nano-sized hydroxyapatite-zirconia bioceramical scalpel.OBJECTIVE: To observe the effects of incision using nano-sized HAP-TZP bioceramics as scalpel.DESIGN: A single sample study.SETTING: Second Department of Orthopaedics, the Second Affiliated Hospital of Harbin Medical University and Harbin Institute of Technology.MATERIALS: The experiment was carried out in the Animal Laboratory of the Second Affiliated Hospital of Harbin Medical University (Provincial Laboratory) from March to May 2006. A total of 54 SD rats, of 4 months old, weighing 120-180 g, of both genders, were selected in this study. The experiment materials included nano-sized hydroxyapatite Quartz Clock Company), ammonia (Suihua Chemical Reagent Company), and anhydrous ethanol (Tianjin Chemical Reagent Company).METHODS: The nano-sized Hydroxyapatite/zirconia composite powder was synthesis by rubbing according to 40% hydroxyapatite + 60% zirconia powders (mass ratio) were mixed and milled. The knife-edge biomaterials, which fabricated by hot pressed sintering, was grinding and used in incision experiment after autoclaving. The back of the rats was shaved and removing a patch of skin under ether anesthesia made a length of 2 cm open excision wound and stitched up the tear by 1# string. The each 18 rats were sacrificed on 3, 7 and 14 days post-wounding and used for analyses respectively. A full-thickness specimen containing the wound was dissected out from each of the surgical sites. The hematoxylin-eosin (HE) staining was applied in the pathological observation.MAIN OUTCOME MEASURES: The tissue sections of back and the results of pathological observation. RESULTS: All 54 rats were involved in the results analysis without any loss. There was long gap with minimal cellular reaction in the scalpel wounds that has not healed on day 3. There was inflamed cells infiltration at the wounded margin edge. By week 1, the tiny detritus which some of them appear the repaired response of epidermis rend were still obviously found in the area of the scalpel wounds where the detritus was found to be significantly less, the wounds changed smooth and started to heal. The optical microscopy observed indicated that there was incomplete epithelialization and hyalinization of dermal collagen at the wounded area. There were obvious inflamed cells to infiltrate in the margin subcutaneous tissue of the wounds where the newborn loose collagenous fibers filled in scalpel wounds was the tissue repairing response phase. There was a great deal of the infiltrated white blood cells in the subcutaneous tissue. The tiny detritus of epidermis by the optical microscopy observed rend could not be found in the area of the scalpel wounds which were healed by week 2. The newborn capillary nets appeared in the newborn repaired collagen fibers tissue. The inflamed cells to be infiltrated were reduced. The compact and strong collagenous fibers filled in scalpel wounds were the normal wound repairing and tissue. The white blood cells infiltrated obvious reduction in the subcutaneous tissue. The normal blood cells in the newborn capillaries could be seen. CONCLUSION: The experiments prove that a HAP-ZTP bioceramic scalpel, which possesses excellent bioactive, is nontoxic, nonallergenic and noncarcinogenic for incisions.
2.Expression of Human T Cell Receptor ? Chain Gene in Insect Cell

Objective: To clone human T cell receptor (TCR) ? chain gene and express its encoding protein by baculoviral expression system in insect cells. Methods: TCR ? chain cDNA was cloned from normal human peripheral blood mononuclear cells (PBMC) by RT-PCR and inserted into baculoviral transfer vector. This vector was co-transfected with baculovirus into insect sf 9 cells. The recombinant protein expressed was identified by SDS-PAGE and flow cytometry with mouse anti-human ? chain monoclonal antibody. Results: Human TCR ? chain cDNA cloned and inserted into baculoviral vector specifically expressed protein in the insect sf 9 cells, accounting for about 11% of the total protein yield in the supernatant of cell lysate. The molecular weight of the recombinant protein determined by SDS-PAGE was identical to what we anticipated. The insect cells transfected with recombinant baculovirus were demonstrated by intracellular labeling flow cytometry to express ? chain protein. Conclusion: Human T cell receptor ? chain gene was successfully cloned from human PBMC, and its encoding protein was highly expressed in insect cells. The TCR ? chain protein obtained by bioengineering technique is useful for in depth biological function study.
3.Studies on counteractive effects of WUZI SIWU GUASHITANG against GTW-induced genital system toxicity in male rats and its mechanism

Object To study the effects of WUZI SIWU GUASHITANG (WT) against multi glycosides of Tripterygium wilfordii Hook. f. (GTW) induced genital system toxicity in male rats and its mechanism. Methods Male rats in treatment groups were treated respectively with a 80 days oral administration of large , middle and small doses of WT plus a fixed dose of GTW. The rats as control were treated with GTW alone or saline. Thereafter, the changes in testis organ index, the number of spermatozoa, the rate of movable spermatozoa and the structure of spermatogenic cell epithlium were observed and the serum testosterone level was determined by IRMA. Then the comparison was made between treatment and control groups as to above variables. Results As compared with GTW group, large or middle dose of WT significantly increased the weight of testis, the number of spermatozoa and the rate of movable spermatozoa. Large dose of WT could relieve the damage of GTW to the spermatogenic cell epithelium completely and keep the serum testosterone at normal level. Conclusion The combined use of GTW with WT is very useful in relief of GTW induced adverse reactions of genital system in male rats. The mechanism by which WT exerts the counteractive effects is related to protecting the spermatogenic cell epithelium from being damaged by GTW, and keeping the serum testosterone at normal level.
4.Progress of Personalized Interior Femoral Anatomical Plate with 3D Printing Technology

Currently,independent clinical medial femoral plate is more needed to apply in more complex C3 distal femoral fractures and knee valgus orthopedic surgery.The doctors always pre-bent the contralateral femoral lateral plate in order to achieve close to the natural shape of the bone.The cold deformation of the plate can change the metallographic structure and the biomechanics,and the position of the screw holes in the plate may also affect the healing of fractures.Compared with traditional manufacturing techniques,3D printing technology as a core technology in third industrial revolution,instead prior manufacture of molds,removing the large amount of material in the manufacturing process and the complex process of forging a final product,it is more suitable for difficult to machine materials,manufacture,inspection shape design,assembly,testing and fast reverse engineering.Therefore,a personalized interior distal femoral plate with 3D printing technology was designed,and the application of the plate,the current situation and prospects of 3D printing technology and other aspects were summarized in this article.
5.Botulinum Toxin-A Injection into Detrusor to Treat Neurogenic Detrusor Overactivity in Patients with Spinal Cord Injury

Objective To evaluate the effectiveness and safety of Botulinum toxin-A (BTX-A) injection into detrusor to treat neurogenic detrusor overactivity in patients with spinal cord injury (SCI).Methods A total of 78 patients with SCI were treated with transurethral injection of BTX-A (300 IU dissolved in 15 ml of saline) into 30 different points of detrusor with 15 ml in every patients. Urodynamic parameters and voiding diary were assessed at baseline and 3 weeks and 3 months after the injections. Adverse events were recorded after the injection if present.Results After the first injection, 78 patients showed that the mean frequencies of incontinence decreased from 13.5 to 2.7 times per day, the mean volume of intermittent catheterization (IC) increased from 131 ml to 389 ml per time, the mean volume of incontinence decreased from 1 690 ml to 281 ml per day, the mean getting effect time was 7.6 days. 10 patients received second injection at 8.9 months after first injection, the results showed that the mean frequencies of incontinence decreased from 9.7 to 3.7 times per day, the mean IC volume increased from 108 ml to 387 ml. 6 patients received third injection at 5.8 months after second injection, the results showed that the mean frequencies of incontinence decreased from 9.2 to 3.9 times per day, the mean IC volume increased from 116 ml to 364 ml. No side effects were observed during the follow-up.Conclusion BTX-A injection into detrusor to treat neurogenic detrusor overactivity in patients with SCI seems to be an effective, safe and miniinvasive solution.
6.Determination of Fumonisin B1 and Fumonisin B2 in Formula Feeds for Livestock and Poultry by High Performance Liquid Chromatography-Tandem Mass Spectrometry

A method was developed for the determination of fumonisin B1 ( FB1 ) and fumonisin B2 ( FB2 ) in livestock and poultry formula feeds by high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS. After extracted by acetonitrile-water (50: 50, V/V) and purified with MAX solid phase extraction column, the fumonisins were separated by Thermo C18(100 mm×2. 1 mm, 5 μm) column with 0. 1% formic acid in water and methanol as the mobile phase. Multiple reaction monitoring (MRM) was used to acquire mass spectrometric data under electrospray positive ionization mode ( ESI+) . The results showed that the linear correlation coefficients (R2) of fumonisin FB1 and FB2 were all greater than 0. 999 in the range of 1-500 μg/L. The limits of quantitation (LOQ) were 0. 098 and 0. 197 μg/L, and the limits of detection ( LOD) were 0. 328 and 0. 656 μg/L, respectively. At different spiked levels, the recoveries of FB1 and FB2 were ranged from 89. 7% to 95. 1%, and the relative standard deviation ( RSD) was ranged from 3. 2% to 8. 6%. Additionally, the detection rate reached 98. 11% screening through the established method in the 106 livestock and poultry formula feeds collected from markets. This result indicates that the method is suitable for accurate quantitative analysis of FB1 and FB2 in different complicated livestock and poultry formula feeds.
7.Effect of Holmium Laser Urethrotomy under Ureteroscopy on Urethral Stricture:30 Cases Report

Objective To investigate the effect of Holmium laser urethrotomy under ureteroscopy on urethral stricture.Methods 30 men with urethral stricture who accepted urethrotomy with Holmium laser under ureteroscopy were observed.Results Operation was performed successfully in 21 cases;7 cases underwent 2~3 endoscopic surgical treatments.Endoscopic surgical treatment failed in 2 cases,and open surgery were performed on them.21 cases were followed up for 3~37 months,and 8 of them need urethral dilatation termly.Conclusion Endoscopic surgery with ureteroscopy and Holmium laser may be effective on urethral stricture with slight trauma.
8.Augmentation enterocystoplasty on treatment of neurogenic bladder dysfunction : a single center outcomes and experience with 77 patients

Objective To assess clinical and urodynamic results of augmentation enterocystoplasty (AE) in patients with ncurogcnic hladdcr dysfunction (NBD).Methods Retrospectively reviewed our database between 2005-2011 to identify 77 patients who underwent AE ( sigmoid 74 patients,ileum 3 patients).Postoperativc complications,renal function,urodynamics parameters and quality of daily life were evaluated.Results The mean follow-up length was 24 months.Compared with pre-operative condition,the mean bladder capacity significantly increased from (160.6 ± 128.3) to (468.5 ± 60.6) ml (P <0.001 ) and the maximum detrusor pressure decreased from ( 31.1 ± 26.4) to ( 10.9 ± 4.5 ) cm H2O ( P =0.002 ).Serum creatinine level decreased from (270.3 ± 113.6 ) to ( 174.4 ± 81.3 ) μmol/(l) ( P =0.00 1 ).There were significant decrease on mean number of incontinence episodes and pads used per day ( P <0.01 ).Post-operative complications included metabolic acidosis in two patients (2.6%),adhesive intestinal obstruction in four patients (5.2%),deteriorating renal function in one patients ( 1.2% ) and recurrence of vesicoureteral reflux in three patients (3.9%).Conclusions The results suggest that AE is safe and effective in treating patients with NBD.Concomitant URI is considerèd on patients with long illness history,vesicoureteral reflux at low intravesical pressures and upper urinary tract dilation.Patients with moderate and severe upper urinary tract deteriorations benefit from this procedure and the benefit can maintain a long time.
9.Long-term follow-up of reconstruction for urinary continence function using implantation of artificial urinary sphincter

Objective To evaluate the long-term outcome of reconstruction for urinary conti-nence function by using implantation of artificial urinary sphincter (AUS). Methods From 2002 to 2005,15 incontinent patients (14 men and 1 woman) with age range of 19-75 years treated with im-plantation of AUS were followed up. In 15 eases, there were 5 cases with traumatic urinary inconti-nence, 1 with neurogenic stress incontinence, 1 with neurogenic urgent incontinence,6 with postpros-tatectomy incontinence and 2 with neurogenie voiding dysfunction. Three eases had taken urethro-stenotomy, 3 had sphineterotomy and 1 had enterocystoplasty and ureterovesieostomy before the im-plantation for AUS. The patients were followed up for 13-55 months with mean of 37 months. The information about the continence status and pads usage, also the complications were collected. Results After implantation, 13 cases(87%)used AUS device normally, 12(92%)beeame dry and 1 (8%) got social continence. Eleven cases (85%) got continence depending on the original implanted AUS and 15% did continence by reoperation. There were 4 cases(27%) with complications including erosion of skin and urethral in 1, graft rejection in 1, urethral atrophy in 1, and voiding dysfunction in 1. Durability of 11 cases with original implanted devices was 13-55 months with mean of 38 months.Conclusion The implantation of AUS is a long-term reliable method in reconstruction for lower uri-nary tract function.
10.Preliminary Observation on the Influence of Traditional Chinese Medicine Injection Adjuvant Chemoradio-therapy on the Quality of Life of Tumor Patients

Objective: To explore the influence of traditional Chinese medicine injection adjuvant chemoradiotherapy on the quality of life of cancer patients. Methods: Totally 76 cases of cancer patients were selection from PLA 307 hospital and randomly divided into the observation group and the control group with 38 ones in each group. The control group was given chemoradiation, and the observa-tion group was given Aidi injection or Shenqi Fuzheng injection intravenously additionally. The treatment course was 4 weeks. Before and after the treatment, cartesian score (KPS), simplified Chinese quality of life assessment (QLQ C30) were used to evaluate the quality of life. Results: After the treatment, the KPS was higher than that before the treatment, while QLQ C30 score decreased and was better than that in the control group (P<0. 05). Conclusion: The traditional Chinese medicine injection adjuvant chemoradio-therapy is more beneficial to the quality of life of cancer patients than chemoradiortherapy only.
