1.Relationship between Energy Storage and Clinical Practicability of Prosthetic Feet YANG Wen-bing

Objective To investigate the relationship between energy storage and clinical practicability of prosthetic feet.Methods Six different prosthetic feet were selected and tested the ratio between released energy and absorbed energy of these feet through a specially-designed equipment. Then, the differences on total cycles of heartbeat of the amputee were tested when he wore the six feet separately to walk and run.Results The total cycles of heartbeat of the amputee with feet of higher energy storage were apparently lower during running, but might be equal or even a little higher during walking.Conclusion The ability of energy-saving of a foot is more important than the ability of energy-storing.
2.Determination of Plasma Diclofenac Sodium by SPE-HPLC

OBJECTIVE: To establish an SPE(solid phase extraction)-HPLC method for the determination of diclofenac sodium in human plasma.METHODS: The separation of diclofenac sodium was performed on ODS-2 column with a detection wavelength of 280nm.The mobile phase was composed of phosphate buffer(pH6.5)-methanol(40∶ 60) at a flow rate of 1.0mL? min-1.RESULTS: The linear range of diclofenac sodium was 16.2~ 2 024.0ng? mL-1(r=0.998 9).The methodological recovery ranged between 90% to 110% and the extraction recovery was above 90%.Both intraday RSD and inter-day RSD were less than 5%.CONCLUSION: The method is simple,accurate and reliable,and suitable for the pharmacokinetic study of diclofenac sodium.
3.Preventive effects of moulded cervical orthosis on the flexion deformity in burn pateints

BACKGROUND: Cervical orthosis is one of the non-surgical therapeutic methods for the remedy of cervical deformities. The introduction of cervical orthosis into the clinical remedy of cervical deformities resulted by burn can maximally restore the cervical function of the patients.OBJECTIVE: To analyse the clinical applicative mode and the effects of moulded cervical orthosis (MCO) on cervical burn.DESIGN: A before-and-after verification study by employing pateitns as subjects.SETTING: Prosthetics and orthotic department in a provincial artificial limb center.PARTICIPANTS: Totally 19 cases including 13 males and 6 females after skin-transplantation due to serious burn in head, neck and chest admitted by the outpatient department of prosthetics center between January and September 2003.METHODS: Nineteen patients with cervical burn wore MCO. Return visits were made afterwards for the evaluation of the treatment effects of MCO.MAIN OUTCOME MEASURES: Resutls of therapeutic effectiveness evaluation.RESULTS: All nineteen patients with cervical deformities due to burn reached the effects of correcting cervical flexion and restoring cervical functions through wearing orthosis. The cervical functions restored entirely in seven cases, almost restored in eight cases and improved in four cases.CONCLUSION: MCO can be clinically used on cervical deformities resulted by burn and provide good effects on remedy and rehabilitation.
4.Magnetic Resonance Imaging in the Evaluation of Leptomeningeal Metastasis of Gliomas

Objective To evaluate the characteristics of leptomeningeal metastasis of gliomas on magnetic resonance(MR) imaging and to provide evidence for clinical early diagnosis and treatment.Methods Twenty-one cases of intracranial gliomas with leptomeningeal metastases were scanned with conventional spin echo(SE) and fast spin echo(TSE),8 cases with fluid attenuated inversion recovery(FLAIR),and all of cases were operated with enhancement.Results ⑴ With MR plain scan,18 cases revealed vary range of distortion and illegibility structure of cerebral cistern and sulcus,and 6 cases revealed the asymmetrical enhancement of the wall of ventricle.Eleven meningeal neoplasms nodes were found in subarachnoid space in 7 cases,and some of the appearances could be seen in several cases.Eight cases of them were added with FLAIR,and the neoplasms nodes were showed more clearly and higher signal than TSE-T2W imaging.⑵ With MR enhancement,all cases with encephalic gliomas showed obvious enhancement with vary features of the enhancement of leptomeningeal metastasis,including tail sign(5/21),line sign(10/21),strip sign(3/21),node sign(7/21) and a few of cases included two signs.Conclusion It was very important for clinician to improve the cognition of MR imaging characteristics of leptomeningeal metastases of glioma,and to choose the proper MR imaging scan technique to increase the detection rate of early meningeal diseases.
5.Clinical Study of Effect of Magnesium ion on Patients with Severe Craniocerebral Injury

Objective To investigate the curative effect of magnesium ion on patients with severe craniocerebral injury. Methods According to the standards,80 patients with severe craniocerebral injury were chosen and divided into control group and experimental group randomly. The control group received routine therapy and the experimental group received routine therapy plus magnesium sulfate. Serum endothelin(ET) and Glasgow coma score(GCS)of all the patients were measured at different time respectively,and Glasgow outcome scale(GOS)of all the patients were determined after 3 months,and then these data were analyzed statistically. 6.Diagnosis value of 128-slice spiral CT in patients with adult intussusception

Objective To investigate the diagnosis value of 128-slice spiral CT in patients with adult intussusception. Methods Direct features of 128-slice spiral CT of 63 adult intussusception patients confirmed by operation and pathology were analyzed retrospectively. Results In 63 cases of intussusceptions, 1 case had idiopathic intussusception, and the other 62 cases had secondary intussusception (1 case with multiple intussusceptions). Direct signs included:target signin 60 cases,double intestines signin 59 cases,blood vessel curling signin 56 cases, andcomet-tail signandkidney signin 51 cases. Lipoma was the most common benign lesions of intussusception, and colon cancer was the most common malignant lesions of intussusception. Conclusions The 128-slice spiral CT combined with multiple plane restructuring has the important value in diagnosing adult intussusception.
7.Study on stability of N-terminal site-specific PEGylated uncase

Purpose The research was conducted to evaluate the stability of N-terminal site-specific PEGylated uricase. Methods The enzyme activity is used as an index to evaluate the thermal stability (at 4-80 ℃) , the pH stability, the ability of avoiding the trypsin digestion and half-life in vivo of PEG-uricase, then it is compared with uricase. Results PEGylated uricase is thermally more stable than uricase between 4 ℃ and 60 ℃, but at 70 ℃, the enzyme activity of both PEG-uricase and uricase decreases sharply. In the test of pH stability, the curve of uricase shows an obvious decrease of enzyme activity at 5 .2-6.0 and 9.2-10.0. The behavior of PEG-uricase indicates that the destabilizing was prevented effectively by PEGylation. The test of anti-trypsin digestion suggests that PEG-uricase retains 70% of its enzyme activity,but uricase only 20% ,200 minutes after being reserved in trypsin solution. Stability in vivo indicates that the half-life of PEG-uricase is 1 530 min and uricase, only 45 min. Conclusion PEGylated uricase has improved thermal stability, the pH stability, ability of protecting from trypsin digestion and stability in vivo.

9.Secreted expression of recombinant human autoantigen Sm B' in methylotrophic yeast Pichia Pastoris

Objective To clone and express human autoantigen Sm B'in methylotrophie yeast Pichia Pagtoris.Methods The gene Sm B' was cloned bv PCR The PCR product wag inserted into the vector pPIC9k.The recombinant plasmid pPIC9k.Sm B' was transformed into yeast Sm D1168 by electroporation.The positive clones were screened in MD plates.The high copy number transformants were rapidly selected by using G418 and were induced by methan01.Supematants after induction were analyzed by SDS-PAGE and western blot.Sera collected from thirty patients with SLE.thirty patients with mixed connective tissue disease(MCTD)and thirty healthy volunteers were detected by immunodot and immunoblot.Results The PCR product wag about 700 bD in size which Wag in accordance with predicted 657 bp.The pPIC9k-Sm B'showed the same seqencing result with GenBank's report and restriction enzyme analysis confirmed our prediction.The pPIC9k-Sm B' positive clone produced a 32 000 protein which had natural immunogenicitv of human autoantigen Sm B'by SDS-PAGE and western blot.The positive rate of immunodot and IBT were 46.7%(42/90)and 51.1%(46/90),respectively.The agreement between immunodot and IBT was very close(Kappa value=0.911 2,P<0.01).Conclusion Successfully cloning and expression of human autoantigen Sm B' in methylotmphic yeast Pichia Pagtoris hid a foundation for further research work.
10.Three-dimensional kinematic analysis of the ability in the activities of daily living of hemiplegic patients and normal subjects

Objective To study the kinematic characteristics of normal subjects and hemiplegic patients in activities of daily living (ADL) by three-dimensional (3D) kinematic analysis. Methods A Vicon motion capture system was used to record 3D kinematic data on 15 normal subjects ( the healthy group) and 10 hemiparetic patients (the hemiplegic group) performing 5 ADL tasks: reaching up, combing, drinking, touching the opposite shoulder and touching the back pocket. The movement times and the 3D motion angles of the sternoclavicular joint,the acromioclavicular joint, the shoulder joint, the elbow joint and the wrist joint were recorded. Three-dimensional joint angles and movement times were compared between the normal group and the hemiplegic group. Results Compared with the normal group, the hemiplegic group had significantly more lateral rotation and abduction of the sternoclavicular joint, medial rotation and extension of the acromioclavicular joint and flexion of the elbow joint.The hemiplegics used significantly less medial rotation of the sternoclavicular joint and adduction/abduction, flexion/extension and lateraL/medial rotation of the shoulder joint. Additionally, the hemiplegic group showed significantly longer movement times. Conclusion Hemiplegics use different joint motor patterns from healthy subjects in completing ADL actions.
