1.Bcl-2 protein and acute renal ischemia-reperfusion injury

Ischemia-reperfusion after neonatal asphyxia is a key factor in renal injury,which often can lead to apoptosis of tubular epithelial cells.Apoptosis is an important form of injury for renal tubular epithelial cells after asphyxia.Large number of cancer genes involve in regulation of renal ischemia-reperfusion injury in the process of apoptosis.Bcl-2 protein which are expression products of Bcl-2 oncogene act on the mitochondrial pathway in apoptosis.Furthermore they can inhibit the caspase cascade of apoptosis via the cells "cross-talk",which contribute to attenuate renal ischemia-reperfusion injury and improve renal function.
2.Effect of interferon gamma on immune factor and CD69 , CD107 a in patients with delayed type hypersensitivity

Objective To investigate effect of interferon gemma on immune factor and CD69, CD107a in patients with delayed type hypersensitivity by interferon gamma.Methods 76 cases with delayed type hypersensitivity were selected and divided into 2 groups.38 cases in control group were treated conventional therapy, 38 cases in experiment group were treated by interferon gamma.Peripheral blood CD69, CD107a, immune factor, T cell subsets and the treatment efficiency were compared after the treatment.Results Compared with the control group, the serum CD69 and CD107a levels were lower in the experimental group (P<0.05), serum IgG and IgA levels were higher, the serum IgE level was lower (P<0.05), and the CD3 +,CD4 +,CD4 +/CD8 +level was higher, serum CD8 + was lower (P<0.05) .The effective rate of the treatment group was significantly higher than that of the control group (P<0.05) .Conclusion Interferon gamma has good clinical effect in the patients with delayed type hypersensitivity, and can effectively reduce the levels of CD69 and CD107a in the peripheral blood, and regulate the immune function of the body.
3.Relationship between lifestyle actual-ideal self-discrepancy and self-regulatory fatigue in undergraduate students

Objective To investigate the relationship between the lifestyle actual-ideal self-discrepancy and the self-regulatory fatigue.Methods A sample of 434 healthy undergraduate students were recruited from September to October 2015.They were inquired the demographic information questionnaire,the lifestyle actual-ideal self-discrepancy of Chinese Adolescent Lifestyles Scale(CALS) and the Self-Regulatory Fatigue Scale (SRF-S).Results The scores of lifestyle actual-ideal self-discrepancy in study habits ranked the highest(1.00(0.60,1.40)),followed by physical exercise(0.86(0.43,1.29)),sleep status(0.80(0.40,1.00)),diet (0.75 (0.50,1.25)),recreational activities (0.63 (0.00,1.00)),coping style (0.50 (0.25,0.75)),interpersonal relationship (0.46(0.17,0.67)) and health habits(0.25(0.00,0.50)).Furthermore,the scores of actual-ideal self-discrepancy in interpersonal relationship,study habits,sleep,physical exercise,coping style and diet were positively correlated with the scores of self-regulatory fatigue (r=0.22,0.19,0.19,0.16,0.13,0.12,P<0.05).There was no relationship between the scores of actual-ideal self-discrepancy and the scores of self-regulatory fatigue in health habits and recreational activities(r=0.31,0.04).Conclusion The actual-ideal self-discrepancy in interpersonal relationship,study habits,physical exercise,coping style,sleep and diet were positively correlated with the self-regulatory fatigue.The reduction in these lifestyle actual-ideal self-discrepancy may help college students ease their self-adjustment fatigue.
4.Association study on lifestyle and internet addiction in adolescents

Objective To investigate the lifestyles of internet addiction disorders and explore the relationship between lifestyles and internet addiction among Chinese adolescents.Methods In this cross-sectional stratified cluster sampling study,Demographic Information Questionnaire,Chinese Adolescent Lifestyles Scale (CALS)and Young's Internet Addiction Test were applied.Results (1) Adolescents with internet addiction were 275,accounting for 11.20%.Boys(15.45%) addicted the internet more than girls(7.48%).The prevalence rate of IAD was higher in students from urban(13.55%) than that in students from rural area(9.67%).The ratio of IAD was the highest in junior school students (14.01%),the second in university students (13.84%),the third in senior high school students(9.66%),and the lowest in primary school students(7.67%) (P<0.01).(2) The internet addicts scored significantly lower on sleep,study habits,physical exercise,interpersonal relationship and coping style,compared to the normal group(P<0.01).(3) Sleep status,study habits,physical exercise,interpersonal relationship and coping style were negatively correlated with internet addiction (r=-0.193,r=-0.319,r=-0.175,r=-0.225,r=-0.105,P<0.01).(4)Multiple logistic regression analysis indicated learning habits and interpersonal relationship had negative predictive effects on internet addiction; whereas,the coping style showed a weak positive prediction effect on Internet addiction.Conclusion Internet addict teenagers have poor lifestyles which should be suggested to start the good behavior habit and insist on a healthy lifestyle to prevent the internet addiction disorder.
5.Optimization of Extraction and Purification Technology of Total Flavonoids from Engelhardia roxburghi-ana and Content Determination of 3 Kinds of Effective Components

OBJECTIVE:To optimize the extraction and purification technology of total flavonoids from Engelhardia roxburghi-ana,and to establish the method for the content determination of 3 kinds of effective components. METHODS:Using the extrac-tion transfer rate of astilbin as index,single factor test was used to investigate extraction solvent,extraction method,volume frac-tion of percolation solvent ethanol,percolation material-liquid ration,soaking time before percolation and percolation rate of extrac-tion technology,and volume fraction of eluant ethanol in AB-8 resin purification technology. The contents of 3 effective compo-nents as astilbin,texifolin and engelitin in total flavonoids from E. roxburghiana were determined by HPLC. RESULTS:The opti-mal extraction technology was using 70% ethanol as extraction and percolation solvent,percolation extraction,soaking for 8 h be-fore percolation,percolation material-liquid ratio of 1∶16(g/ml),percolation rate of 30 ml/(min·kg). The purification technology was diluting the solution to 0.5 g (crude drug)/ml with water,ethyl acetate extraction,dissolved extract with 50% ethanol after evaporated to dryness,AB-8 resin for sampling,eluted with 50% ethanol,concentrating and drying. In verification test,extraction transfer rate of astilbin was more than 80%(RSD=0.42%,n=3). The contents of astilbin,taxifolin and engeletin in total flavo-noids from E. roxburghiana by purified were 57.94%,3.72% and 2.83%,respectively;the contents of 3 components accounted for 64.00% of total flavonoids. CONCLUSIONS:The extraction and purification technology is stable,rational and reliable;the content determination method of 3 effective components in total flavonoids of E. roxburghiana is accurate,simple and producible.
6.Mechanism of retinoic acid and mitogen-activated protein kinases regulating hyperoxia lung injury.

To investigate the protective effect of retinoic acid (RA) on hyperoxic lung injury and the role of RA as a modulator on mitogen-activated protein kinases (MAPKs), gastation 21 d Sprague-Dawley (SD) fetuses (term = 22 d) were delivered by hysterotomy. Within 12-24 h of birth, premature rat pups were randomly divided into 4 groups (n=12 each): air-exposed control group (group I); hyperoxia-exposed group (group II), air-exposed plus RA group (group III), hyperoxia-exposed plus RA group (group IV). Group I, III were kept in room air, and group II, IV were placed in 85 % oxygen. The pups in groups III and IV were intraperitoneally injected with RA (500 microg/kg every day). All lung tissues of premature rat pups were collected at the 4th day after birth. Terminal transferase d-UTP nick end labeling (TUNEL) staining was used for the detection of cell apoptosis. The expression of PCNA was immunohistochemically detected. Western blot analysis was employed for the determination of phosphorylated and total nonphosphorylated ERKs, JNKs or p38. Our results showed that lungs from the pups exposed to hyperoxia for 4 d exhibited TUNEL-positive nuclei increased markedly throughout the parenchyma (P<0.01), and decreased significantly after RA treatment (P<0.01). The index of PCNA-positive cells was significantly decreased (P<0.01), and was significantly increased by RA treatment (P<0.01). The air-space size was significantly enlarged, secondary crests were markedly decreased in hyperoxia-exposed animals. RA treatment improved lung air spaces and secondary crests in air-exposed pups, but had no effect on hyperoxia-exposure pups. Western blotting showed that the amounts of JNK, p38 and ERK proteins in hyperoxia-exposure or RA-treated lung tissues were same as those in untreated lung tissues (P>0.05), whereas activation of these MAPKs was markedly altered by hyperoxia and RA. After hyperoxia exposure, p-ERK1/2, p-JNK1/2 and p-p38 were dramatically increased (P<0.01), whereas p-JNK1/2 and p-p38 were markedly declined and p-ERK1/2 was further elevated by RA treatment (P<0.01). It is concluded that RA could decrease cell apoptosis and stimulate cell proliferation under hyperoxic condition. The protection of RA on hyperoxia-induced lung injury was related to the regulation of MAP kinase activation.
7.Effect of acute exposure to high altitude on the pharmacokinetics of propranolol

Objective:To study the pharmacokinetics of propranolol in Wistar rats after acute exposure to high altitude.
Methods:Fourteen male Wistar rats (200±20) g were selected. After administration of propranolol tablets (0.05 g/kg, i.g.), blood samples (3 mL) were collected at 0, 20, 40 min,1, 1.5, 2, 4, 6, 8, 12 and 24 h, respectively. The pharmacokinetic parameters were determined by LC-MS/MS and DAS 2.0 software.
Results:The main pharmacokinetic area under concentration-time curve (AUC), mean retention time (MRT), half-life (t1/2) and peak plasma concentration (Cmax) of propranolol were increased by 442.61%, 47.45%, 73.13%and 352.97%, respectively, whereas Tmax and clearance (CL) were decreased by 80.87%and 68.94%, respectively.
Conclusion:This study displays significant changes in the pharmacokinetics of propranolol under high altitude, which may provide evidence for clinical rational application of propranolol at high altitude.
8.Total hip arthroplasty with subtrochanteric femoral shortening osteotomy for Crowe Ⅳ developmental dysplasia of the hip

Objective To evaluate the clinical efficacy of total hip arthroplasty with subtrochanteric femoral shortening osteotomy for Crowe Ⅳ developmental dysplasia of the hip.Methods From September 2003 to September 2012,21 patients (24 hips) underwent total hip arthroplasty with subtrochanteric femoral shortening osteotomy for Crowe Ⅳ developmental dysplasia of the hip in our hospital.There were 3 males and 18 females,aged from 28 to 71 years (average,54±10 years).The ceramicpolyethylene articulation was used in 17 patients (20 hips),and metal-polyethylene articulation in 4 patients (4 hips).The osteotomy site was treated with autologous bone graft in 18 patients (21 hips) and allogeneic bone graft in 3 patients (3 hips).The Harris hip score was used to assess the clinical results.Results A total of 18 patients were followed up for 0.5 to 9 years (average,3.5 years).The Harris hip score was improved from preoperative 47.9±9.1 to 88.4±3.5 at 6 months postoperatively.For most patients,hip pain relieved significantly; range of motion of the hip was improved,and the gait returned to normal.Sciatic nerve palsy occurred in 1 patient.There was no wound infection.X-rays 6 months after operation showed that the position of prostheses was satisfactory,without loosening of prostheses and bone block resorption.Conclusion Total hip arthroplasty with subtrochanteric femoral shortening osteotomy can achieve good clinical effect in Crowe Ⅳ developmental dysplasia of the hip.Moreover,it can improve leg length discrepancy and decrease the risk of sciatic nerve injury.
9.Optimizing a method of establishing colon cancer liver metastasis models

BACKGROUND:The metastasis ways of colon cancer include blood metastasis, peritoneal metastasis and lymph node metastasis. Among them, the most common metastasis organ is the liver. It is necessary to establish a simple, suitable animal model to study the biological behavior of the colon cancer liver metastasis, the heterogeneity of cancer cells, and the mechanism of tumor metastasis. OBJECTIVE:To establish animal model of colon cancer liver metastasis with splenectomy injection method. METHODS:Human colon cancer celllines HT29 were cultured, and subcutaneously transplanted tumor was produced in BALB/c mice. Models of colon cancer liver metastases were established through spleen injection and excision of spleen method. The tumor growth and metastasis, the pathology of transplanted tumor, DNA content and cellcycle of the transplanted tumor were al determined. RESULTS AND CONCLUSION:In this study, we successful y established the model of colon cancer liver metastases through the spleen injection via the dorsal midline incision, the tumor formation rate reached 8/8 (100%). Observation of tissue slice showed that the transplanted tumor was adenocarcinoma I-II. Flow cytometry analysis results showed that DNA ploid was heteroploid DNA. This model provides a good technical platform for biology research for colon cancer metastasis.
10.Effects on the pharmacokinetics of furosemide after acute exposure to high altitude at 4010 meters in rats.

The paper is to report the pharmacokinetics of furosemide in rats living at plain area and high altitude. After intragastric administration of furosemide (2.87 mg x kg(-1)), serial blood samples (0.5 mL) were collected by retro-orbital puncture at 0, 20 min, 40 min, 1, 1.5, 2, 3, 4, 6, 8, 12 and 24 h, samples were determined by LC-MS/MS, and plasma concentration-time data were analyzed by DAS 2.0 software to get the related pharmacokinetic parameters. The main pharmacokinetic parameters: area under curve (AUC), mean residence time (MRT), the biological half-life (t1/2) and the peak concentration (C(max)) of furosemide, were significantly increased at high altitude, the time to reach peak concentration (t(max)) and clearance (CL) was significantly decreased. This study found significant changes on the pharmacokinetics of furosemide under the special environment of high altitude. This finding may provide some references for clinical rational application of furosemide at high altitude.
