1.Rapid Genomic DNA Fragments Extraction From Serum Denatured With High Temperature

Objective To develop an approach for a rapid extraction of genomic DNA fragments from serum.Methods Serum specimen from human was processed using high temperature denaturation and was centrifuged, then using various primer sets to amplify the DNA fragment extracted, so as to verify the efficiency of this DNA extraction method.Results The templates preparing from this DNA extraction method were able to guarantee the amplification of fragments shorter than 400 base pairs.Conclusion This method is simple, rapid and inexpensive.These genomic DNA fragment extracted can be utilized to amplify definite DNA template for polymerase chain reaction.
2.Application of photoacoustic imaging in the diagnosis and treatment of skin tumors

Photoacoustic imaging is a kind of non-invasive technology without ionizing radiation that can be used to image optical absorbers in tumor tissues according to the principle of "light in and sound out". It has the advantages of high contrast of optical imaging as well as high penetrability of acoustic imaging. However, current researches on photoacoustic imaging are still limited in the laboratory. This technology can be used for detection of cutaneous melanoma in situ and non-melanoma skin cancer, sentinel lymph node imaging, detection and eradication of circulating tumor cells by combining real-time laser ablation technique, and versatile imaging and treatment with the aid of exogenous imaging agents. This review summarizes preclinical experiments about photoacoustic imaging technology in skin tumors, with a view to promoting its clinical application at an early date.
3.Stability analysis of allelopathic effects of Panax notoginseng on main crops by AMMI model.

This paper is aimed to study the differences of allelopathic effects of Panax notoginseng under different allelopathic chemicals resources and selection of appropriate rotation crops. The additive main effects and multiplicative interaction ( AMMI) model had been used to evaluate the stability of allelopathic effects of P. notoginseng on the varieties of corn, wheat and rice properly. The model could use not only to evaluate the stability of non-regional trial data but also explore the interaction between the rotation crop genotypes and donor substances more efficiently. Meanwhile, correspondence analysis can be used in the AMMI to evaluate genotype stability and donor substances. Ejingza No. 1 (g6) had stronger allelopathic effects with high stability, but Yunrui No. 1 (g9) which was appropriate rotation crop genotype, had weaker allelopathic effects with high stability. These findings will aid in choosing appropriate rotation crops and establishing proper rotation system.

6.Design of pulsed magnetic fields stimulation instrument

In this paper,a pulsed magnetic fields stimulation instrument is designed and realized,which provides a pulsed magnetic field with the range of maximal intensity from 0.01~2T,frequency from 0.2~100Hz and time width of pulse from 0.01~1ms.The instrument,controlled by the hand,foot or itself,can display stimulus intensity and times and output trigger signals with different waveforms to make measuring devices operate synchronistically.

Objective According to Kaschin-Beck Disea se monitory standardization that had been adjusted by our country,we monitored the state of Kaschin-Beck Disease in Gansu province.Methods So as to understand change of illness,we took methods of epidemiological investigation,clinical examination and X-ray diagnosis.Results It is not detected in the clinical that patient suffered from more than I of KBD among 7~12 years old in Qingyang monitory netw ork.X-ray detectable rate is 3%,but 12 cases patients were showed in Zhangjiach uan.X-ray detectable rate is 22.22%.Conclusions Illness was showed steady state and was con trolled in Qingyang region,but illness recurred clearly in Zhangjiachuan region.
8.Effect of Ligusticum wallichii-containing serum on expressions of Toll-like receptor 4 and myeloid differentiation factor 88 in hepatic stellate cells.

To observe the effect of Ligusticum wallichii-containing serum on the expressions of Toll-like receptor 4 and myeloid differentiation factor 88 in hepatic stellate cells. Clean-grade SD rats were randomly divided into 5 groups and orally given L. wallichii decoction, colchicine and normal saline for 7 d to prepare L. wallichii-containing serums. Except for the blank group, all of the remaining groups were stimulated with LPS 1 mg x L(-1) for 24 h. After being intervened, the L. wallichii-containing serums were cultured in 5% CO2 incubator at 37 degrees C for 24 hours. The expression of TLR4 and MyD88 were detected by RT-PCR and Western blot. After HSC was stimulated with LPS, TLR4 and MyD88 mRNA and protein expressions were significantly higher than the blank control group (P < 0.01). After being intervened with L. wallichii-containing serum, TLR4 and MyD88 mRNA and protein expressions were notably lower than the model group (P < 0.05 or P < 0.01). In conclusion, L. wallichii-containing serum could regulate the TLR4 signaling pathway and show the anti-fibrosis effect by inhibiting the expression of TLR4 and MyD88 in LPS-induced HSCs.

9.Effects of standardized three-stage rehabilitation treatment on neurological deficit scores and ADL performance of ischemic stroke patients

Objective To observe the effects of standardized three stage rehabilitation treatment on the neu- rological deficit scores (NDS) and ADL performance of ischemic stroke patients.Methods A total of 164 ischemic stroke patients were recruited and randomly divided into a rehabilitation group and a control group.The neurological function and ADL performance of the patients were assessed by using NDS and Modified Barthcl Index (MBI) at the admission,at the end of 1st,3rd and 6th months post stroke.Results No significant differences were found be- tween the rehabilitative and the control groups with regard to NDS and MBI at admission.The NDS demonstrated a decreasing tendency,while the MBI score an increasing tendency in both groups.In the control group,significant difference of NDS was found between admission and the end of 1st month as well as between the end of the 1 st and the 3rd months.In rehabilitation group,significant difference was revealed between all the time points with regard to NDS and MBI scores.At the end of the 1st,3rd and 6th months,the MBI scores of the rehabilitation group were signifi- cantly higher than those of the control group,indicating that the ADL performance of those treated with standardized three-stage rehabilitation protocol was improved quicker than those without the protocol.Conclusion Standardized three-stage rehabilitation treatment could improve the neurological function and ADL performance of the ischemic stroke patients.
10.Effects of stromal cell-derived factor-1 on proliferation,migration,and odontoblastic differentiation of human dental pulp stem cells

Objective:To compare the effects of stromal cell-derived factor-1 (SDF-1 )and granulocyte colony-stimulating factor (G-CSF)on proliferation,migration,and odontoblastic differentiation of human dental pulp stem cell (DPSC)in vitro.Methods:DPSCs were cultured in vitro and treated with either 1 00 μg/L SDF-1 or 1 00 μg/L G-CSF.Cell counting kit-8 (CCK-8 )and colony-forming unit (CFU ) were used to detect the effect of SDF-1 and G-CSF on the proliferation ability of DPSC.Cell migration of DPSC was determined by wound healing assay and Transwell migration assay.The effects of SDF-1 and G-CSF on odontoblastic differentiation of DPSC were evaluated by alkaline phosphatase (ALP)staining, ALP activity and alizarin red S staining.The expression of odontoblastic-related genes such as dentin ma-trix protein 1 (DMP-1 )and dentin sialophosphoprotein (DSPP)were quantified by real-time RT-PCR. Results:SDF-1 and G-CSF promoted the proliferation of DPSC slightly,but the difference was not statis-tically significant.Wound healing assay showed that SDF-1 and G-CSF promoted cell migration of DPSC significantly (P<0.01 ),but there was no significant difference between the two factors.In Transwell migration assay,the number of migrated cells of the control group was 5 .0 ±1 .4 per sight,while the SDF-1 group was 24.3 ±6.8 per sight and the G-CSF group was 1 1 .8 ±3.3 per sight,suggesting that cell migration of DPSC was improved significantly after being treated with SDF-1 or G-CSF,and SDF-1 was more effective than G-CSF (P<0.05 ).Significantly greater odontoblastic differentiation potential was found in SDF-1 group and G-CSF group based on the ALP staining.Higher ALP activity,more mineralization nodule formation and higher expressions of DMP-1 and DSPP were also found after SDF-1 or G-CSF treatment.Conclusion:SDF-1 had no significant effect on the proliferation of DPSC,but could significantly promote cell migration and odontoblastic differentiation of DPSC.Its effect on DPSC was bet-ter than G-CSF.
