1.Experimental study of mustard seeds prevent colorectal tumor by antioxidation and immune deviation

Background and purpose:Recently, a large number of researches have shown that cruciferous plants have the chemopreventive effect on tumor. Mechanisms of antitumorigenesis were investigated on antioxidation, antimutation, immunity and inducing apoptosis, and so on. Mustard seeds (MS) are the seeds belong to the cruciferous plants. This study aimed to investigate antioxidation and immune deviation of MS on colorectal tumor in rats induced by 1, 2-dimethylhydrazine (DMH). Methods:A total of 48 male Wistar rats were randomly divided into four groups:DMH alone, DMH+5%MS, DMH+7.5%MS, and the untreated control group(Saline). Colorectal tumorigenesis was induced by intraperitoneal injecting 30 mg/kg DMH once a week for 20 weeks. At the end of 32 weeks, the rats were sacrificed, then colorectal tumor incidence was observed and histological type was determined by HE staining. A colorimetric assay was used to detect levels of the lipid peroxidation product malondialdehyde (MDA) and the activity of antioxidant enzymes in the serum of all rats. The levels of Th1 and Th2 cytokines were detected with Luminex200. Results: No tumorous lesion was found in the untreated control group. However, the total tumor incidence in DMH+5%MS group and DMH+7.5%MS group was signiifcantly decreased 33.3%and 58.3%respectively, compared with the DMH group’s (100%, P<0.05). As DMH induced colorectal tumorigenesis, MDA and Th2 cytokines in the serum were signiifcantly higher in the DMH group than those in the untreated control group (P<0.05), but the activities of antioxidant enzymes were signiifcantly lower (P<0.05). While the MS treatment, compared with the DMH group, signiifcantly suppressed the MDA level but enhanced the activities of antioxidant enzymes and levels of Th1 cytokines (P<0.05). Conclusion: MS significantly decrease prevalence rates of DMH-induced colorectal tumor in rats. The mechanism may be related with the antioxidation and immune balance deviation.
2.The effect of mustard seed on azoxymethane-induced colorectal tumor in mice

Background and purpose:Colorectal cancer is one of the most common cancers worldwide as well as a prevalent cause of morbidity and mortality in China.Mustard seed and its components have been reported to possess anticancer properties.The purpose of this study was to investigate the effect of a diet mixed with mustard seeds (MS)on the prevention of colorectal tumorigenesis in Kunming mice induced with azoxymethane.Methods:Sixweek-old female Kunming species mice of were randomly divided into 4 different groups:AOM alone,AOM+5%MS,AOM+10%MS,and the untreated control group.Colorectal tumorigenesis was subcutaneously induced by injecting 10 mg/kg of AOM once a week,for 3 weeks.Different doses of MS were administered in the diet during this study.Thirtytwo weeks after initiation,the mice were put down through cervical dislocation.The colon and recta of the mice were isolated and flushed with nonnal but ice-cold saline.The site,size,and number of colorectal tumors were recorded.The incidence of colorectal tumor in each group was determined.Results:No tumors were found in the control group.Compared to AOM-treated control group(86.7%),the occurrence of colorectal tumors was reduced to 60.0%when administered with a 5%diet of mustard seeds and to 41.7%(P<0.05)with a diet of 10%mustard seeds.Tumor burden (tumors/mouse)was 2.2±1.2 in the AOM-treated control group whereas it was reduced to 1.1±1.1(P<0.05)in the 5%diet of mustard seeds and 0.7±0.9(P<0.05)in the 10%diet of mustard seeds.Conelusion:The results indicate that a diet of mustard seeds is able to inhibit the development of colorectal cancer in mice induced by AOM,suggesting possible future application of MS as a chemopreventive agent for colorectal cancer.
3.Isolation and characterization of ?_2m~-/Thy-1~+ bone marrow-derived liver stem cells from cholestatic rats in vitro

Objective To explore the in vitro isolation of ?2m-/Thy-1+ bone marrow-derived liver stem cells(BDLSCs) which bear double features of stem and liver cells from bone marrow stem cells(BMSCs)as so to provide suitable donor cells for the treatment of liver diseases by cellular transplant. Methods ?2m-/Thy-1+ BDLSCs were isolated by magnetic bead cell sorting(MACS) method from cholestatic rats in vitro,and cell purity was detected using flow cytometry.Liver associated phenotype markers were characterized by RT-PCR and immunofluorescence staining. Results BDLSCs isolated by MACS were purified and viable,and possessed hepatocyte-like features at gene and protein levels. Conclusion ?2m-/Thy-1+ BDLSCs are special subsets of BMSCs which may have promising potentials in the stem cell-based treatment of liver diseases.
4.Empirical study of learning experience based on medical MOOC teaching design system

Objective:To explore the learning experience of the medical massive open online course (MOOC) teaching design standard system constructed by the research group in the early stage.Methods:In this study, the questionnaire was adapted from four dimensions: academic analysis, curriculum content design, teaching process design, and teaching evaluation design, including 519 students majoring in clinical medicine of a university who had studied MOOC cases like "Ultra-early Diagnosis and Treatment of Acute Cerebral Infarction" based on the system. SPSS 25.0 was used for statistical analysis.Results:The system had a high degree of recognition in all dimensions, with 64.5% of academic analysis, 57.6% of content design, 54.5% of teaching design process, and 59.3% of teaching evaluation design.Conclusion:The study has found that the medical MOOC teaching design system has good learning experience effect. According to the data feedback, the key teaching design points such as the core factors of learning experience analysis and the suitability of teaching content in the practical operation of teaching design has been explored, providing the practical basis and method reference for the design of medical MOOC teaching design.
5.Design, synthesis and pharmacological investigation of isoindoline derivatives as 5-HT/NE double reuptake inhibitors.

A series of isoindoline derivatives were designed, synthesized, and evaluated for their double inhibitory activities. All of them were new compounds, and their structures were confirmed by 1H NMR and HR-MS. Preliminary in vitro pharmacological tests showed that all compounds exhibited 5-HT or NE reuptake inhibition activity. Among the tested compounds, compound I-3 exhibited potent inhibitory activity against 5-HT and NE reuptake in vitro, and exhibited potent antidepressant activity in vivo. These compounds designed can be further optimized for finding more potent 5-HT/NE dual reuptake inhibitors and antidepressant candidates as well.

6.Establishment and Application of Electronic Data Capture System in Drug Clinical Trials of Our Hospital

OBJECTIVE:To ensure the stability of electronic data capture(EDC)system in drug clinical trials and to improve the quality of drug clinical trials. METHODS:The quality control system for EDC system was established and introduced from the formulation of quality control process,establishment of data standard,trial project management,daily management,trial project design,system operation,system function,etc. RESULTS & CONCLUSIONS:Data standard have been achieved through estab-lishing EDC quality control system by our hospital based on attributable,legible,contemporaneous,original and accurate principle. The management of trial project and daily management are conducted through data registration,staff training,the formulation of da-ta management plan,fault emergency treatment,database backup;multiple verification of support data,data lock and export,trial report autogeneration and other functions have been realized by formulating related standard operation instruction,program file,op-eration manual and quality record. Those aspects improve facticity,accuracy and integrality of data in clinical trials,and lay a foun-dation for further data mining.
7.Structure,Function and Operation Practice of Clinical Trial Management System in Our Hospital

OBJECTIVE:To promote the supervision and management of clinical trial by institution. METHODS:The structure and function of clinical trial management system(CTMS)developed by our hospital and other enterprise together were analyzed to evaluate the application and operation result of CTMS. RESULTS & CONCLUSIONS:CTMS of our hospital is made up of foun-dation,efficiency and strategy. It is equipped with role allocation,information exchange and report,information warning,drug tracking,clinical trial process control,quality control of electronic record,electronic signature and integration and connection with other system,etc. Relevant operation procedure is established to promote standardization and institutionalization of CTMS. Due to the application of CTMS,the cooperation among departments become smoother,and management level have been enhanced in dai-ly management,pharmacy management,subjects and document administration. It also simplifies the work of researcher and reduc-es the human error by the autogeneration of trial records and tables with the system. Consequently,the monitor coveraged through-out all the trial process.
9.Analysis of the occurrence of delayed traumatic intracerebral hematoma

Objective To discuss the pathogenesis,diagnosis and treatment of delayed traumatic intracerebral hematoma.Methods The clinic data of traumatic delayed intracranial hematoma patients in this hospital were retro- spectively analyzed.According to clinic observation and CT re-examination,47 cases were diagnosed as delayed trau- matic intracranial hematoma(45 cases by operative treatment,and the other 2 by conservative treatment).Results There were 21 cases of recovery,10 cases of slight disability,8 cases of severe disability,8 cases of death.The total mortality rate was 17 %.Conclusion Brain contusion,subarachnoid hemorrhage and skull base fracture were impor- tant factors of DTICH.Fine-observation and prompt CT re-examination offered excellent results for DTICH.
10.Studies on chemical constituents of Ranunculus ternatus.

OBJECTIVETo study the active constituents of Ranunculus ternatus.
METHODThe constituents were isolated with silica gel and Sephadex LH -20 gel column chromatography and purified by HPLC. Their structures were elucidated by spectroscopy.
RESULTSixteen compounds were obtained and identified as Stigmasta4, 6, 8 (14), 22-tetraen-3-one (1), 5-hydroxymethyl furaldehyde (2), gamma-keto-delta-valerolactone (3), pantolactone (4), 5-hydroxymethyl-dihydro-furan-2-one (5), methyl 5-hydroxy-4-oxopentanoate (6), methyl hydrogen succinate (7), succinic acid monoethyl ester (8), 3, 4-dihydroxybenzoic acid methyl ester (9), p-hydroxycinnamic acid (10), 4-oxo-pentanoic acid (11), succinic acid (12), nonanedioic acid (13), 4-hydroxybenzoic acid (14), 4-hydroxybenzaldehyde (15), stigmasterol-3-O-beta-D-glucopyranoside (16).
CONCLUSIONCompound 1, 4 -15 are obtained from this plant for the first time.
