1. The effect of lysophosphatidic acid on growth, invasion and metastasis of ovarian cancer cells
Tumor 2008;28(9):746-750
Objective: To investigate the effect of lysophosphatidic acid (LPA) on growth, invasion and metastasis of ovarian cancer cell lines SKOV3, 3AO and CAOV3. Methods: The ovarian cancer cell line 3AO, SKOV3 and CAOV3 were cultured and incubated with LPA at different concentrations for 2, 4, 6, 16 and 24 h. The control group was incubated with PBS instead of LPA. The effect of LPA on cell proliferation was detected by MTT assay. The invasion ability of ovarian cancer cells was evaluated by the Boyden chamber invasion test in vitro. The secretion of cytokines in the supernatant of cultured CAOV3 was measured by ELISA after being stimulated by LPA. Results: LPA interference greatly enhanced cell growth in a concentration-dependent manner. Boyden chamber invasion test showed that the invasion indices of 3AO and CAOV3 test groups were all higher than those of corresponding control groups, respectively. The difference was significant between different concentration groups. But the difference was not significant between the 3AO and CAOV3 cell lines. The expression of cytokines in supernatant fluid of CAOV3 cells had corresponding changes after LPA interference at different concentrations. Conclusion: LPA interference significantly enhances the growth of ovarian cancer cells in vitro. The invasion indices and the levels of migration-related cytokines have corresponding changes after LPA interference at different concentrations. It indicates that LPA might have the biologic significance in invasion and metastasis of epithelial ovarian cancer.
2. The effect of realgar on apoptosis of transplanted ovarian SKOV3 carcinoma cells in nude mice
Tumor 2007;27(10):787-790
Objective: To observe the effect of traditional Chinese medicine, realgar, on apoptosis and angiogenesis of ovarian carcinoma cells. Methods: The anti-tumor effects of realgar were observed in nude mice bearing ovarian transplanted carcinoma. Apoptosis and the expression of vascular endothelial growth factor were determined by flow cytometry, fluorescence microscopy, transmission electron microscopy, and immunohistochemical methods. Results: Realgar induced apoptosis and inhibited the growth of ovarian transplanted tumors in nude mice. Realgar also blocked the angiogenesis, inhibited the DNA synthesis, and prolonged the survival time of nude mice. Conclusion: Realgar exerted its anti-tumor effect through multiple pathways such as inducing apoptosis, blocking angiogenesis, inhibiting DNA synthesis and had a wide foreground for application.

Objective To assess the reliability and validity of the Tree-Drawing Test in medical college students. Methods The study randomly selected 312 aged 19 to 23-year-old medical students to take part in TreeDrawing Test.In addition,a total of 275 college students were selected to receive re-test,30 days late and Pearson correlation coefficient of two tests were calculated. The three raters were invited to assess 30 trees painting score, analyzing the Kendall coefficient of concordance between the scores to verify raters' reliability; parts of students also participated in the 16PF test,SAS,SDS test, analyzing the correlation coefficient between the various test results,in order to assess the effectiveness of the Tree-Drawing Test. Results The re-test reliability in different time was 0.570-0.733 and 0.341-0.713 (P<0.05),the raters' reliability was 0.491 ~ 0.626(P<0.05),there are some correlations between Tree-Drawing Test and 16PF,SAS,SDS.Conclusion The Tree-Drawing Test has good reliability and validity; it can be applied to the detection of college students' psychological assessment and psychological problems.

Objective To study the protective effect of total flavones of hawthorn leaf (TFHL) on renal ischemia/reperfusion (I/R) in rats. Methods A model of renal I/R was made by clamping double renal pedical for 60 min and reperfusion for 24 h.TFHL (30 or 60 mg/kg) was injected before clamping renal ischemia. The level of serum urea nitrogen(BUN), serum creatinine(Scr), tumor necrosis factor-?(TNF-?),interleukin-1(IL-1) in serum were detected after reperfusion for 1 and 24 h,respectively, and the level of malonaldehyde (MDA) and erythrocuprein (SOD) in renal tissues were measured after reperfusion for 24 h.Results TFHL significantly decreased the level of BUN and Scr, decreased the concentration of TNF-? and IL-1 in serum. Pretreatment with TFHL also enhanced the activity of SOD reduced the concentration of MDA in renal after I/R.Conclusions TFHL has significantly protective effect on renal I/R and markedly improve the renal function. The mec-hanisms are associated with decreasing the lipid peroxidation reaction and reducing the level of TNF-? and IL-1.

Isoliquiritigenin (ISL) is a flavonoid compound isolated from licorice. It possesses excellent antioxidant and anti-diabetic activities. This study aims to investigate the molecular mechanism underlying the alleviatory effect of ISL on energy metabolism imbalance caused by type 2 diabetes mellitus (T2DM). 8-week-old male C57BL/6J mice were used in

Isoliquiritigenin (ISL) is an active chalcone compound isolated from licorice. It possesses anti-inflammatory and anti-oxidative activities. In our previous study, we uncovered a great potential of ISL in treatment of type 2 diabetes mellitus (T2DM). Therefore, this study aims to reveal the mechanism underlying the alleviatory effects of ISL on T2DM-induced glycolipid metabolism disorder. High-fat-high-sugar diet (HFD) combined with intraperitoneal injection of streptozotocin (STZ) were used to establish T2DM mice model. All animal experiments were carried out with approval of the Committee of Ethics at Beijing University of Chinese Medicine. HepG2 cells were used in
