1.Relationships between serum levels of adiponectin and essential hypertension in the people with overweight and obesity

Objective To explore the relationships between serum levels of adiponectin and essential hypertension in the people with overweight and obesity.Methods The body mass index(BMI),waist circumference(WC),waist-hip ratio(WHR) and blood pressure(BP) were measured among 21578 people from 21 to 60 years old in Shenzhen.According to BMI,the people were divided into the normal-weight group(BMI≤24kg/m~2)(n=14817),the overweight group(2495 cm,respectively(all P0.05).The serum levels of adiponectin were negatively correlated with BMI,WC,WHR and systolic blood pressure in the hypertension obesity cases.Conclusion Adiponectin may be intimately related to obesity and essential hypertension.Keeping the BMI and WC at normal range is one of the effective ways to prevent the risk of essential hypertension.
2.The effects of brushite coating extract with strontium on the osteoblast-related factor expression of MC3T3-E1 cells

Objective:To study the effects of brushite coating with strontium on the osteoblast-related factor expression of MC3T3-E1 cells.Methods:Brushite coating on tinanium surface was prepared by electrochemical deposition and the extract of the coating with stronium at 0.1%,0.5% and 1% were prepared respectively.MC3T3-E1 cells were cultured with the extracts,cell prolifera-tion,ALP activity,bFGF and VEGF mRNA were examined by MTT assay,ALP staining and RT-PCR respectively.Results:Stronti-um containing extracts promoted the proliferation,increased the ALP activity and mRNA expression of bFGF and VEGF(P <0.05)of MC3T3 cells.Conclusion:Brushite extract containing strontium may promote osteogenic function of MC3T3-E1 cells.
3.Expression and clinical significance of CD+8 CD-28 and CD+4CDhigh25 CDlow127 regulatory T cells in peripheral blood of lung cancer patients

Objective To observe the changes of CD+8CD+28,CD+8CD-28 and CD+4CDhigh25CDlow127 regulate T (Treg)cellsin peripheral blood of lung cancer patients,and to analyze the correlation between CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells to reveal the role and clinical significance of them in lung cancer patients.Methods Flow cytometry was applied to evaluate the level of CD+8CD+28,CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells in peripheral blood of 60 untreated lung cancer patients and 60 healthy controls group.The association of each term with clinical features was analyzed.Results The percentage of CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells in lung cancer group[(58.430:15.749) %,(7.365±2.025) %]was significantly higher than those in healthy group [(41.057±15.436)%,(6.648±1.669)%,(t=6.102,P<0.05;t=2.115,P<0.05)],while the percentage of CD+8CD+28cells is lower(41.570±15.739)% than that in healthy group[(58.700±15.298)%,(t=-6.043,P<0.05)].No close associations were found between three index and gender,age,and biological characteristics.With the increase of TNM stage,The percentage of CD+4CDhigh25 CDlow127 Treg cells increased gradually,which was remarkably higher in patients rith stage Ⅳ than that with stage ⅢA(t=-3.898,P<0.05).The percentage of CD+4CDhigh25 CDlow127 Tregcells was uncorrelated with CD+8CD-28 cells(r=-0.169,P>0.05).Conclusion The higher percentage of CD+8CD-28 and CD+4CDhigh25 CDlow127 Treg cells,the lower percentage of CD+8CD+28 cells may be the important reasons of immune suppression in lung cancer patients.Though there is no correlation between CD+8CD-28 and CD+4CDhigh25 CDlow127 Treg cells,it is may be helpful to understand immunologic function and it may look for more specific therapy and provide a new reference in the prognosis of lung cancer.
4.Manganese-enhanced MRI for detection of metastatic potential in colorectal cancer

Objective To study manganese superoxide dismutase (MnSOD) expression in colorectal cancer (CRC) cells and observe its relation to metastatic potential. To investigate the diagnostic performance of manganese-enhanced magnetic resonance imaging (MEMRI) for detecting metastatic potential of CRC. Methods High and low metastatic potential CRC cell lines SW620, HCT116, LoVo and SW480, DLD-1, HCT15, Caco-2, as well as normal colon mucosal cell CCD841 CoN were cultured. MnSOD expression level in cells was detected by western blot and the measurement was repeated for three times. HCT15, DLD-1, LoVo and SW620 cells were selected to perform in vitro MEMRI and subcutaneous xenografts were developed for subsequent in vivo MEMRI. MnCl2·4H2O solution was utilized as the contrast agent and T1 shortening was calculated. The differences of MnSOD expression level in cells, average T1 value shortening of cells and xenografts were separately compared by one-way ANOVA.Results The difference of MnSOD expression level in CRC cells were significant (P<0.05). All CRC cells have shown higher level of MnSOD expression than normal cell, except for Caco-2. The highest expression were shown in low metastatic potential cells (HCT15 and DLD-1). Whereas relatively low MnSOD expression was found both in high and low metastatic potential cells (LoVo, SW620, HCT116 and SW480). In vitro MEMRI demonstrated that the average T1 value shortening was greatest in high metastatic potential cells SW620 and LoVo [(289.33±0.57) and (268.45 ± 6.87) ms)], was significantly lower in low metastatic potential cells DLD-1 and HCT15, was lowest in CCD841 CoN [(65.12 ± 0.12)ms] (P<0.05). Twenty nine xenografts were successfully developed, high metastatic potential SW620 and LoVo tumors of 5 and 10 mm group showed greater T1 shortening, low metastatic potential DLD-1 and HCT15 tumors showed significantly less T1 shortening (P<0.05). However no significant difference was found in similar metastatic potential tumors (P>0.05). Conclusions MEMRI has the potential to noninvasively distinguish different metastatic potential CRC by revealing greater T 1 value shortening in more aggressive one. However the MnSOD expression in CRC cells is not corresponding to malignant potential.
5.Factors affecting dyslipidemia among residents in Chengdu City

Objective:
To investigate the prevalence and influencing factors of dyslipidemia among residents in Chengdu City, so as to provide insights into improving the prevention and control of dyslipidemia.
Methods:
Based on the baseline survey of the Natural Population Cohort Study in Southwest China, residents aged 30 to 79 years was selected from 34 towns (communities) in 5 counties (districts) of Chengdu City using the multi-stage stratified cluster random sampling method in 2018. Demographic information and lifestyle behaviors were collected through questionnaires. Blood pressure, fasting blood glucose, serum uric acid, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were collected through physical examination and laboratory tests. A multivariable logistic regression model was used to identify the factors affecting dyslipidamia.
Results:
A total of 21 113 participants were surveyed, including 9 331 males (44.20%) and 11 782 females (55.80%), and had a mean age of (50.80±12.32) years. The prevalence rate of dyslipidemia was 35.64%, and the prevalence rates of high TG, low-HDL-C, high TC and high LDL-C were 17.25%, 11.88%, 10.11% and 7.35%, respectively. Multivariable logistic regression analysis identified gender (male, OR=1.584, 95%CI: 1.463-1.716), age (50 to 79 years old, OR:1.221-1.444, 95%CI: 1.079-1.632), residence (urban, OR=1.123, 95%CI: 1.052-1.198), marital status (not married, OR=1.246, 95%CI: 1.128-1.376), educational level (high school and above, OR=0.914, 95%CI: 0.849-0.983), current smoking (OR=1.220, 95%CI: 1.121-1.327), drinking (1 to 2 d/week, OR=1.525, 95%CI: 1.368-1.700; 3 to 5 d/week, OR=1.857, 95%CI: 1.575-2.191; almost every day, OR=1.512, 95%CI: 1.269-1.801), sedentary time in leisure time (>2 h/d, OR=1.123, 95%CI: 1.046-1.206), central obesity (OR=2.212, 95%CI: 1.986-2.265), hypertension (OR=1.489, 95%CI: 1.388-1.598), diabetes (OR=1.998, 95%CI: 1.833-2.157) and hyperuricemia (OR=2.012, 95%CI: 1.848-2.192) as factors affecting dyslipidemia.
Conclusion
The prevalence of dyslipidemia among residents in Chengdu City was mainly associated with smoking, drinking, sedentary time, central obesity, hypertension, diabetes and hyperuricemia.

7.Cluster analysis on hypocrellin A content in anamorphs of Shiraia bambusicola.

OBJECTIVEBy examining the colony growth rate, colony texture, substrate mycelium color and separation method of 40 anamorph strains of Shiraia bambusicola to investigate the relationship between these indexes and content of hypocrellin A.
METHODIn UV-spectrophotometry and microscopic observation were applied in the experiments. The results were analyzed by the clustering and variance analysis.
RESULTSPSS clustering analysis showed that the colony characters of S. bambusicola anamorph strains had significant differences with hypocrellin A content.
CONCLUSIONIn the tested conditions, substrate mycelium color of S. bambusicola anamophs has the most distinct correlation with the yield of hypocrellin A.

8.Isolation and determination of anamorph of Shiraia bambusicola.

OBJECTIVETo isolate and identify the anamorph of Shiraia bambusicola.
METHODFungus strains were isolated from mature ascospores and stroma. They were identified by means of morphological identification including colony and microscope characteristic, the molecular identification was done by 5.8S-ITS rDNA.
RESULTThe strains GZDXIFR-171 and GZDXIFR-181 were isolated and obtained with the separation of different methods, which had the same colony morphology. With the universal primers of the ITS1-5.8S rDNA-ITS2, the 5.8S-ITS rDNA sequence of GZDXIFR-171 and GZDXIFR-181 were obtained by the PCR amplification and sequencing. Compared with the published nucleotide sequence of 5.8S-ITS rDNA in NCBI (National Center for Biotechnology Information), GZDXIFR-171 and GZDXIFR-181 were highly identical with S. bambusicola.
CONCLUSIONThe isolated strains GZDXIFR-171 and GZDXIFR-181 were confirmed to be the true anamorph of S. bambusicola.

9.Comparison of head dual energy computed tomography angiography and neuro digital subtraction angiography.

OBJECTIVETo investigate the advantages and disadvantages of dual energy with dual-source computed tomography (CT) in cerebral angiography by comparing those of neuro-digital subtraction angiography (Neuro-DSA) .
METHODSTotally 300 patients with normal CT angiography (CTA) images underwent dual energy CT angiography (DE-CTA groups, n=200) and Neuro-DSA scan (Neuro-DSA groups, n=100) using dual source CT. The comparison of two scan approaches was based on image quality, radiation does, post-processing methods, and duration of scanning and subtraction.
RESULTSThe image quality of intracranial blood vessels in DE-CTA group showed no significant difference with that in Neuro-DSA group (P>0.05) , while the number of Grade I images was significantly higher than those of other three grades (P<0.01) . However, the effect of the skull base bone removal with Neuro-DSA is superior to DE-CTA (P<0.01) . The duration of scanning was significantly longer in DE-CTA group than in Neuro-DSA group (P<0.01) . However, the radiation dose, time of entire examination, and time of subtraction were significantly lower/shorter in DE-CTA group than in Neuro-DSA group (P<0.01) . The radiation dose of DE-CTA group is 26.3% lower than Neuro-DSA group.
CONCLUSIONSThe image quality of intracranial blood vessels with head DE-CTA is same as Neuro-DSA, while head DE-CTA has lower radiation doses and higher inspection efficiency than Neuro-DSA. However, the effect of the skull base bone removal needs to be improved.
