1.Anti-light aging effect of Polygala tenuifolia saponins on the skin

Objective To study the anti-aging effect of total saponins of Polygala tenuifolia (PTS). Methods The effects of total saponins of Polygala tenuifolia on oxygen free radicals were investigated by the methods of scavenging hydroxyl radical, superoxide radical and DPPH oxygen free radicals in vitro. The photoaging model of mice skin was established to observe the effect of total saponins of Polygala tenuifolia on mouse skin pathology. Results In the experiment of antioxidation in vitro, the total saponins of Polygala tenuiflora significantly scavenged hydroxyl free radicals (P<0.05, P<0.001) and superoxide radical (P<0.01) at 6.0 and 12.0mg/mL, DPPH free radicals (P<0.001). In the light aging experiment, the light aging model of mice skin was established successfully in group B (model + medium tea oil). Compared with group B, the density of the skin epidermis in D group (30 min before irradiation, 6mg/mL PTS) was thickened, the elastic fibers were arranged uniformly and tightly, the collagen fibers proliferated significantly and densely, and the inflammatory cells infiltration was not found. The treatment effect was most remarkable in group G (30min after irradiation, PTS 12mg/mL PTS), and the skin of the mice was thickened, the elastic fibers were evenly arranged, the collagen fibers had some hyperplasia, evenly distributed, and there was no infiltration of inflammatory cells, which had significant therapeutic effect. Conclusion The total saponins of Polygala tenuifoliahas significant antioxidant activity and anti - light aging effect.
2.Prophylactic effect of chimera on acute graft-versus-host disease after xenogeneic bone marrow transplantation

Objective To explore the effect of chimera on acute graft-versus-host disease (aGVHD) after xenogeneic bone marrow transplantation. Methods Chimera was produced by exposing rats to sublethal total body irradiation and mouse bone marrow transplantation plus intraperitoneal injection of CTX. Mice were divided into three groups. Mice in group A were transplanted with marrow cells of rats without chimera, mice in group B with marrow cells with chimera, and mice in group C with C57BL/6 of rat marrow cells with chimera. Prophylactic effect on the model of aGVHD in the mice induced after xenogeneic bone marrow transplantation was observed. Results All transplantated mice showed typical aGVHD signs. Animals in group A developed typical aGVHD, and death occurred in 10-12 days after transplantation. The mean survival time of group B were longer than that of group A and C, and the signs and histopathological changes of aGVHD in group B were less severe. Conclusion Xenogeneic chimera can prevent aGVHD, alleviate its symptoms and histopathological changes, and prolong mean survival time. The prophylactic effect is specific.
3.Nursing experience for patients of esophageal water-perfused high-resolution manometry

Objective To investigate the key points in the nursing work of esophageal waterperfused high-resolution manometry (WHRM).Methods Summarized the nursing experiences before,during and after WHRM of 153 patients.Results Only 5 cases could not finish the test due to failure of intubation.23 cases complained about nasopharyngeal discomfort but follow-up monitoring was unremarkable.Conclusion In order to ensure the completion of WHRM,nursing cooperation requires serious assessment and preparation before manometry,intimate observation of patients' condition during manometry,and keep the catheter disinfection after manometry.

5.Study on the relationship between the pathologic change of chronic atrophic gastritis and helicobacer pylori

Objective To investigate the correlation between helicobacter pylori (H.pylori) infection with gastric muco?sa pathologic changes in chronic gastritis. Methods A total of 250 patients with chronic gastritis who came to Tianjin Union Medicine Center from November 2011 to March 2013 were collected in this study. Electronic gastroscope examina?tions, pathology and Urea-14C breath tests were performed in patients. There were 153 cases with chronic atrophic gastritis (CAG group), and 97 cases without chronic atrophic gastritis (non-CAG group). The positive rate of H.pylori was compared between two groups. At the same time the positive rate of H.pylori was compared between different parts of CAG patients. The positive rates of H.pylori were compared between different pathologic features of chronic gastritis (active degree, the de?gree of inflammation, atrophy and intestinal classification). Results The positive rate of H.pylori was higher in CAG pa?tients than that of non-CAG patients (70.6%vs 35%,χ2=30.552). The positive rate of H.pylori was higher in antral and cor?pus atrophy of CAG group than that of antral atrophy (82.6%vs 65.4%,χ2=4.578). With the aggravating activity of gastritis, the inflammation, chronic gastritis, atrophy and intestinal classification, the positive rate of H.pylori was increased (χ2=200.643, 206.715, 73.286, 218.432). Conclusion H.pylori infection is related with chronic gastritis, chronic atrophic gas?tritis. And antral and corpus atrophy CAG is closely related with H.pylori infection.
6.Effects of dexamethasone sodium phosphate on the proliferation of in vitro cultured rabbit articular chondrocytes

BACKGROUND: Studies have demonstrated that dexamethasone can reduce type Ⅱ collagen and increase type I collagen in articular cartilage surface matrix, but the action mechanism of glucocorticoid to articular chondrocyte proliferation remains unclear. OBJECTIVE: To investigate the effects of dexamethasone sodium phosphate on rabbit articular chondrocytes cultured in vitro. DESIGN: Comparative observation.MATERIALS: New Zealand rabbits, 1 month old, were used for chondrocyte isolation and culture.METHODS: Rabbit articular chondrocytes cultured in vitro were randomly divided into control and experimental groups. Cells of the control group were cultured in DMEM media without dexamethasone sodium phosphate. Cells of experimental groups were cultured in DMEM media with different concentrations of dexamethasone sodium phosphate (0.02, 0.1, 0.5 g/L), respectively. MAIN OUTCOME MEASURES: Primary and passage chondrocytes were observed. Flow cytometry and immunocytochemistry were used to observe the effect of dexamethasone sodium phosphate on cell proliferation and type Ⅱ collagen synthesis. The ultrastructural changes of cultured chondrocytes were observed by transmission electron microscopy. RESULTS: The attachment and proliferation of experimental group chondrocytes was slower than control group. There average gray scale values of the experimental groups were significantly greater than the control group (P < 0.05). The cellular proportions of S phase and G_2+M phase of the experimental groups decreased but the cellular proportions of G0/G1 period increased. Under transmission electron microscope the amount of rough endoplasmic reticulum and mitochondria of the experimental groups decreased. CONCLUSION: Dexamethasone sodium phosphate inhibited articular chondrocyte proliferation, possibly due to the decrease of type Ⅱcollagen and protein synthesis ability of chondrocyte.

8.Efficacy observation of sequential plasma perfusion of low dose plasma exchange in patients with acute on chronic liver failure

Objective To investigate the clinical efficacy of sequential plasma perfusion of low dose plasma exchange in the pa-tients with acute on chronic liver failure (ACLF).Methods The patients with HBV related ACLF in the infection department of the Affiliated Hospital of Luzhou Medical College were divided into the plasma exchange therapy group (group A,n=114)and the low dose plasma exchange sequential plasma perfusion therapy group (group B,n=144).The group A was treated only by plasma exchange of fresh frozen plasma 2 400 mL;while the group B adopted the sequential plasma perfusion of plasma exchange,with fresh frozen plasma dosage of 1 400 mL.The changes of the liver and renal function,coagulation function and electrolyte indicators were observed in the two groups.The differences in the clinical effects and adverse reactions were compared between the two groups.Results The serum levels of ALT,TBIL and INR after treatment in the two groups were significant decreased than before treatment(P<0.05);ALB and PTA were significantly improved compared with before treatment(P<0.05);there were no signif-icant differences in serum levels of CRE,K+ and Na+ between before and after treatment.The changes of various indexes before and after treatment had no statistical differences between the two groups.The total effective rate and the mortality at 24 weeks in the group B was 66.66% and 47.22% respectively,which had no statistical differences compared with the group A.The total oc-currence rate of adverse reactions in the group B was 23.75%,which was significant lower than 35.26% in the group A with statis-tical difference (P=0.011).Conclusion Sequential plasma perfusion of plasma exchange has better clinical effect and few adverse reactions for treating the patients with ACLF,which can reduce the plasma dosage significantly.
9.Expression of hepatitis C virus H strain envelope glycoprotein in insect cells

Aim To express hepatitis C virus glycoprotein E (gE) deleting carboxy-terminal 31 amino acids, and detect anti-E antibody in HCV patients using expressed gE. Methods E gene derived from HCV H strain was inserted into baculovirus transfer vector containing a polyhedrin promotor. The recombinant plasmid was cotransfected into insect cell sf9 with a viral expression vector. The expression of gE was analyzed with Western blot, and the cells were used for dectecting antibodies against E1 and E2 in 35 hepatitis C patients by indirect immunofluorescence. Results A series of proteins with different relative molecular masses(Mr) could be detected by Western blot. Results from indirect immunofluorescence staining showed and only 4 patients were anti-E antibody positive gE was located in cytoplasm. Conclusion HCV gE is expressed successfully in insect cells, the study lay the foundation for further developing HCV vaccine.
