3.Influence of TAMs on monocyte chemoattractant protein-1 and macrophage inflammatory protein-1 in NSCLC
Chinese Journal of Primary Medicine and Pharmacy 2008;15(5):796-798
Objective To observe cocultured TAMs cell U937 and NSCLC cell A549 effecting on MCP-1, MIP-1 expression under different conditions in vitro. Methods The cell UM937 of different concentrations (0,1,2,4,8×105/ml) was respectively selected to coculture with the cell A549 0f certain concentration (1.0×105/ml) for 24 hours, in situ hybridization was carried out to detect MCP-1, MIP-1mRNA positive expressions in cell A549 of every concentration group. Under the condition that both concentrations of cell A549 and cell U937 are l.0×105/ml, the same method was used to measure MCP-1, MIP-1mRNA positive expressions in cell A549 in different periods (4h, 8h, 16h, 32h, 48h). Results The positive expression ratios of MCP-1, MIP-1mRNA in cell A549 significantly increased along with addition of the cell U937 concentration and delay of the cocultural time, apparently higher than that of primary cocultured group that the cell U937 was 0×105/ml and that of contrast group at every period (all P<0.05). Conclusion The mutual interaction between TAMs and NSCLC cells has closely positive correlation with MCP-1, MIP-1mRNA positive expressions in cell A549, and depends on cocultural concentration and time.

Chinese Journal of Blood Transfusion 1988;0(03):-
Objective Study on fetomaternal immuno state and RHD type of a pregnant woman of weak D phenotype.MethodsThrough polymerase chain reaction (PCR)、direct genomic DNA sequencing and flow cytometry.ResultsIn both sequence specific promer (SSP) PCR and the sequencing PCR tests, the sample was detected negative in exons 3 6 of the RHD gene, whereas all other exons (exons 1 2,7 10) were tested positive. And the sequence of detected exons (exons 1 2,7 10) are the same with normal RHD in GenBank (accession no. AJ299020 1 and AJ299026 9). Serologically and genetically, the sample can be designated as D category VI type Ⅲ. Through a duce tube PCR method, the RhD zygosity of this individual was typed CD VI e/cde。In flow cytometry, a few fetal erythrocytes were detected in peripheral blood of the mother. However there were no anti D detected in sera and hemolytic disease of the newborn(HDN) observed at all.ConclusionSevere cases of HDN have occurred in D positive babies born to partial D mother with anti D, although HDN don't take place in this case. We may still consider D VI phenotype individuals as D positive donors and D negative receiptions in our transfusion practice and in clinical anti D allo immune prophylaxis and monitoring.
