Chinese Journal of Integrated Traditional and Western Medicine 2016;36(5):602-607
OBJECTIVETo observe the effect of Wenyang Huoxue Lishui Recipe (WHLR) containing serum on the expression of cathepsin L (CatL) in puromycin aminonucleoside-induced injury of mouse glomerular podocytes.
METHODSMouse podocyte cells (MPCs) in vitro cultured were divided into the normal control group, the model group, the dexamethasone (DEX) group, 10% WHLR containing serum group, 20% WHLR containing serum group, the vehicle serum control group. MPCs in the normal control group were cultured at 37 degrees C culture solution for 24 h. 45 mg/L puromycin was acted on MPCs in the model group for 24 h. On the basis of puromycin intervention, 1 limol/L DEX was co-incubated in MPCs of the DEX group for 24 h; 10% or 20% WHLR containing serum was co-incubated in MPCs of the 10% WHLR containing serum group and 20% WHLR containing serum group for 24 h. The vehicle serum control group was also set up by incubating with WHLR containing serum alone for 24 h. The expression of CatL and its substrate Synaptopodin in podocytes were detected by cell immunofluorescence staining. FITC-conjugated phalloidin was used to stain F-actin. A cortical F-actin score index (CFS index) was designed to quantify the degree of cytoskeletal reorganization in cultured podocytes.
RESULTSCompared with the normal control group, the expression of synaptopodin significantly decreased and the expression of CatL significantly-increased in the model group. F-actin arranged in disorder, gradually forming pericellular F-actin ring. CFS index was obviously elevated (P < 0.01). Compared with the model group, the epression of synaptopodin increased, the expression of CatL decreased, and CFS index also decreased in the DEX group, 10% WHLR containing serum group, and 20% WHLR containing serum group (P < 0.05, P < 0.01). Compared with the DEX group, the expression of synaptopodin decreased in 10% WHLR containing serum group, CFS index also decreased in 20% WHLR containing serum group (P < 0.05).
CONCLUSIONSWHLR could up-regulate the expression of synaptopodin, down-regulate the expression of CatL, and alleviate cytoskeletal reorganization of F-actin. It was helpful to stabilize the cytoskeleton of F-actin and improve the merging of podocytes.

5. Effect of bromocriptine on the expression of Pit-1 in prolactinoma rats
Chinese Pharmaceutical Journal 2013;48(7):527-530
OBJECTIVE: To investigate the effect of bromocriptine on the expression of Pit-1 in prolactinoma rats. METHODS: Firstly, to prepare prolactinoma model in rats. Adult Wistar rats were divided into two groups at random. The rats in control group were subscutaneously implanted with a blank implant. Rats in 17β-estradiol group were implanted with 17β-estradiol-containing implants. Secondly, rates in 17β-estradiol group were divided into two groups at random: model group and bromocriptine group. Water was administrated to rats in model group. Bromocriptine (0.225 mg · kg-1 · d-1) were orally administrated to rats in bromocriptine group, the rats in control group were orally administrated with water. After four weeks of treatment, all the animals were executed. Each pituitary gland was weighed. Serum prolactin (PRL) levels were measured by RIA method. Pit-1 mRNA levels in pituitaiy tissue were measured by RT-PCR method. RESULTS: The weights of pituitary gland and PRL levels in 17β-estradiol group were individually higher than those in control group (P<0.001). The expression levels of Pit-1 mRNA in bromocriptine group was obviously lower than that in model group (P≤0.001). CONCLUSION: Bromocriptine has potential preventive effect to estrogen-induced rat prolactinoma. The decrease of Pit-1 mRNA level may be involved in the mechanism of anti-prolactinoma effect of bromocriptine.

Objective To investigate the etiology and prognostic factors of acute kidney injury (AKI) in pediatric patients . Methods The clinical data of children patients with AKI in this hospital ,including the clinical features ,causes and outcomes ,were retrospectively analyzed .The relationship between the risk factors and prognosis was analyzed .Results Infants were dominated by the prerenal factors and the other age groups were dominated by the renal factors .The univariate analysis revealed that the AKI stage ,etiology ,mechanical ventilation ,sepsis/septic shock ,MODS ,acidosis ,creatinine initial value ,creatinine peak value and serum potassium were the factors affecting prognosis .The Logistic regression analysis showed that mechanical ventilation ,MODS and sep-sis/septic shock were the independent risk factors affecting prognosis .Conclusion The etiology of AKI in children is diverse and its distribution has the age characteristics .Mechanical ventilation ,MODS and sepsis/septic shock are the independent risk factors af-fecting prognosis .The early diagnosis and the active treatment conduces to improve prognosis .

Chinese Pharmacological Bulletin 1987;0(03):-
Aim To evaluate retrospectively the effect of genetic polymorphisms in CYP3A4 and CYP3A5 on cyclosporine pharmacokinetics during the early period in renal transplant patients. Methods Polymerase chain reaction-restriction fragment length polymorphism was performed to genotype for CYP3A4*18B and CYP3A5*3.Cyclosporine whole blood levels were measured by fluorescence polarization immunoassay. Results The frequencies of CYP3A4*18B and CYP3A5*3 variant alleles were 0.235(95 CI:0.235~ 0.241)and 0.770(95 CI:0.767~0.773),respectively in 63 of renal transplantation patients, and these alleles exhibited incomplete linkage disequilibrium. The median (range) value of cyclosporine dose adjusted blood trough concentrations (C0/D) and C0 in CYP3A4*1/*1 genotype patients was higher than that in CYP3A4*1/*18B and CYP3A*18B/*18B patients, the same result was found in CYP3A5 expression (CYP3A5*3/*1 and CYP3A5*1/*3) (P

Objective To evaluate the efficacy and safety of the InVance bulbourethral sling for stress urinary incontinence in male patients. Methods Five male patients with incontinence after prostatectomy were treated with the perineal sling(InVance). The patients aged from 62 to 76 years(mean,67 years),and the history of the incontinence ranged from 2 to 6 years(average 4 years).Four of the cases were due to TURP,and one radical prostatectomy. All the cases had been failed to respond to conservative therapies. Before the procedures, urodynamic examination showed a mean abdominal leak point pressures(ALPP)at 25.5 cm H2O(20-32.5 cm H2O).Under spinal anesthesia,a 3-to 5-cm vertical incision was made between the scrotum and the anus with the patients at the lithotomy position to expose the right and left ischiopubic branches. We put the three screws with its sutures on each bone and fixed the sling.A cough test was performed systematically and ALPP was set over 60 cm H2O.Results The operation time was 50-85 min(mean,60 min)and intraoperative blood loss was 20-50 ml(average 30 ml).The catheter was removed at day 5 and none of the patients had urination difficulty. The cases were followed up for 6 to 24 months(mean,12.6 months),during which urodynamic examination showed an ALPP at 65 cm H2O(55-70 cm H2O).Among the patients, the 4 who had received TURP were cured, and the other one undergone radical prostatectomy was improved. Conclusions InVance bulbourethral sling is suitable for mild-to-middle urinary incontinence after prostatectomy.
