1.Preliminary study on morphology of proximal femoral medullary cavity and design of artificial furcal interior locking joint

The irregular anatomic morphology of proximal femoral medullary cavity does not match the current regular prosthetic stem,which is a primary cause of prosthesis loosening post-surgery. The newly designed artificial furcal interior locking joint features irregular prosthetic stem, which adapts to the irregular medullary cavity in the proximal femur. The artificial furcal interior locking joint is anchored in a multi-plane and multi-point manner. Together it can prevent prothetic early peristalsis and late loosening in the medullary cavity post-surgery. Therefore, artificial furcal interior locking joint is promising for prosthetic replacement.
2.Efficacy Observation of Jiangu Tongxiao Pills in the Treatment of Osteoarthritis

OBJECTIVE:To observe therapeutic efficacy of Jiangu tongxiao pills in the treatment of osteoarthritis. METH-ODS:72 patients with osteoarthritis were randomly divided into treatment group and control group with 36 cases in each group. Treatment group was given Jiangu tongxiao pill orally,5 g/time,3 times/d;control group was treated with Glucosamine sulfate capsules orally. 0.5 g/time,3 times/d. A treatment course lasted for 7 days,and both group received 4 treatment courses. Clinical ef-ficacy,WOMAC score and VAS score were observed and compared between 2 groups. RESULTS:The total effective rate of treat-ment group(94.44%)was obviously superior to that(83.33%)of control group,with statistical significance(P<0.05). WOMAC score and VAS score of treatment group were lower than those of control group,with statistical significance(P<0.05). CONCLU-SIONS:Jiangu tongxiao pills has good therapeutic efficacy for osteoarthritis.
3.Laparoscopic splenectomy: a single-center clinical study on 55 patients

Objective To study the safety and efficacy of laparoscopic splenectomy for splenic diseases.Method We retrospectively studied the outcomes of 55 patients who underwent laparoscopic splenectomy from May 2007 to December 2009.Splenic diseases included idiopathic thrombocytopenia purpura (n=11),autoimmune hemolytic anemia (n=6),hereditary spherocytosis (n=1),splenic lymphoma (n =1),splenic cyst (n=10),splenic angioma (n=5),vascular tumor of spleen (n=2),cirrhosis,portal hypertension and hypersplenism (n=9),cirrhosis and hyperplenism (n=9),and idiopathic splenomegaly (n=1).Results All patients underwent laparoscopic splenectomy,and there was no conversion to open surgery.The operation time (mean±S.D.) was (119.7±33.0) min.The intraoperative blood loss (mean± S.D.) was (83.8± 65.2) ml,and the postoperative hospital stay (mean±S.D.) was (5.7±1.1) days.One patient developed postoperative ascites,and 7 patients had drain fluid rich in amylase.There was no perioperatively death.Conclusion Laparoscopic splenectomy was safe and efficacious for splenic diseases.
4.Protective Effects of Epidermal Growth Factor on Pancreas of Rats with Acute Pancreatitis

Objective To investigate the protective effects of epidermal growth factor (EGF) on pancreas of rats with acute pancreatitis(AP). Methods Seventy-two male Sprague-Dawley rats were randomly divided into 3 groups: Control group, AP group and AP-EGF group. Subcutaneously injection of EGF (0.1 ?g/g) were given to animals in the AP-EGF group after the establishment of the model of AP. The other two groups of animals received the same volume of saline. At 6 h, 12 h and 24 h after induction of AP, 8 animals in each group were sacrificed respectively, 4 ml of blood sample was withdrawn from heart,2 ml for the analysis of amylase activity and 2 ml for MDA content in serum. Ascites was sucked with dry gauzes and was weighed thereafter. Changes of pancreas morphology were evaluated at every time point. The same part of pancreas was removed for measurement of MDA content, apoptotic index (AI) and histologic changes. Results Histologic injury of the animals in the AP-EGF group was milder than that in the AP group. 5.Protective mechanism of ischemic preconditioning on ischemic/reperfusion injury after pancreas transplantation in rats

Objective To investigate the protective mechanism of ischemic preconditioning on ischemic/reperfusion injury after pancreas transplantation in rats.Methods The model of diabetic SD rat was established. Twenty-four diabetic SD rats were randomly assigned to ischemic/reperfusion group (I/R group, n=6) and ischemic preconditioning group (IPC group, n=18). The rats in group IPC were averagely assigned to 3 sub-groups: group IPC_ 1 (5 min ischemic and 5 min reperfusion), IPC_ 2 (5 min ischemic and 5 min reperfusion twice) and IPC_ 3 (5 min ischemic and 5 min reperfusion thrice). Six normal SD rats whose abdomen was opened only served as control group, and they did not receive pancreas transplantation. I/R group and IPC group received pancreas transplantation. The superoxide dismutase (SOD) and myeloperoxidase (MPO) activity of grafts were monitored 2 h after reperfusion, the apoptotic cells in grafts were observed by TUNEL method, and the expression of Bcl-2 and Bax gene of the grafts was detected by Western blot.Results As compared with I/R group, the SIOD activity and the expression of Bcl-2 gene of grafts were significantly increased, while MPO activity, apoptotic index and the expression of the Bax gene in the grafts were markedly reduced in IPC group (P
6.Influence of Heat Shock Treatment upon the Expression of Membrane HSP70 and Its mRNA of H22 Cell

To identify the best inducing condition, we studied the expression of membrane protein HSP70 and mRNA of H22 cell at various temperature. Using MTT,RT-PCR, immunofluorescence and FCM techniques, we observed H22 cell survival rate, the expression of HSP70 mRNA and membrane HSP70. No effects of H22 cell survival rate under 42 ~ 43℃ was observed, but cell survival rate declined with increasing stress time at 44 ~ 45 ℃; the level of HSP70 mRNA decreased initially (0.5~4.0) hours but gradually resumed and increased from 8 to 12 hours at 42℃. Objective To assess the long-term efficacy and safety of mid-dosage (13~15 mg?kg-1?g-1) ursodeoxycholic acid (UDCA) treatment for primary biliary cirrhosis (PBC). Methods A systematic review of all randomized controlled trials (RCT) comparing UDCA with placebo was performed. Results Seven trials including 1038 patients were assessed. UDCA could significantly improve liver biochemistry, but had no effect on pruritus and fatigue. In the patients with initial stage Ⅰ-Ⅱ, there was a significant decrease in the histologic progression after treatment with UDCA for 2 years compared with the placcebo group(P=0.03), but there was no significant difference between the two groups when considering overall patients with initial stage Ⅰ-Ⅳ (P=0.08). Meta-analysis showed no significant difference in the incidence of death (odds ratio 0.99, 95% confidence interval 0.62-1.58), liver related death (1.05, 0.53-2.05), liver trans-plantation (0.87, 0.53-1.41), death and/or liver transplantation (0.92, 0.64-1.31) and liver decompensation (0.94, 0.60-1.49) between the UDCA and placebo groups. Conclusions The analysis of RCTs of UDCA versus placebo shows improvement of liver biochemistry, but not improvement of clinical symptoms and survival. UDCA may slow down liver histologic progression in the early-stage patients with PBC.
8.Effect of lymphatic reconstruction on pharmacokinetics of FK506 after small bowel transplantation in rats

Objective To investigate the effect of immediately lymphatic reconstruction (LR) after heterotopic small bowel transplantation on pharmacokinetics (absorption and transportation) of FK506. Methods Male Brown-Norway (BN) and Lewis (LEW) rats were divided to two groups. Group A: BN→LEW+LR+FK506 ( n =12); Group B: BN→LEW+FK506 ( n =12). FK506 at a dose of 5?mg/kg per day was taken through stoma. At 1, 4, 7 and 14 postoperative day (POD), venous blood 0.5?ml was obtained on the 1.2 , 2 and 3?h after administration of FK506. Whole blood concentration of FK506 was analyzed by microparticle enzyme immunoassay and a kit of monoclone antibody. 9.Determination of Aripiprazole in Human Plasma by HPLC

OBJECTIVE: To establish a HPLC method for the determination of aripiprazole in human plasma by HPLC. METHODS: The plasma sample was determined by HPLC after subjected to liquid- liquid extraction .The separation was carried out on C18 column with clolumn temperature at 40℃. The mobile phase consisted of 0.03mol?L-1 ammonium acetate-acetonitrile(34:66) with a flow rate of 0.8mL?min-1 and detective wavelength of 257nm.The sensitivity was 0.01 AUFS. RESULTS:The linear range of aripiprazole was 5.0~600.0ng?mL-1(r=0.999 5) .The recovery rate was above 90% .CONCLUSION:The method is sensitive,accurate and rapid,and suitable for the determination of aripiprazole in human plasma.
10.Impact of nicotine on rat fracture healing callus content and process maturity

Objective To explore the effect of nicotine on rats callus content and maturity in the process of fracture healing.Methods Sixty male Sprague-Dawley rats were randomly divided into model group,mild nicotine group and severe nicotine group (n =20/each group).The 3-mm bone defects fracture models were made in the junction of the lower 1/3 of the rat left radial.Five rats of each group were sacrificed randomly in the 3,7,14,21 days after surgery,respectively.The left radial were collected as the observed object.The callus thickness and maturity of the specimens were detected by HE staining.Results At the 3rd days after modeling,the difference in specimens callus thickness between each treatment group and the model group was not statistically significant (P > 0.05),no difference in the maturity of the callus under the microscope; callus thickness in mild and severe nicotine groups and model group was (1.59 ± 0.09) mm,(1.43 ± 0.12) mm,(1.39 ± 0.09) mm at the 7th day after modeling,(1.98 ± 0.12) mm,(1.78 ± 0.08)mm and (1.68 ± 0.09) mm at the 14th day after modeling,and (2.39 ± 0.09) mm,(1.93 ± 0.11) mm,(1.89 ± 0.09) mm at the 21 st day after modeling; The difference of callus thickness in specimens between each treatment group and the model group had statistical significance (P < 0.05,P <0.01),callus thickness and maturity of the treatment group were lower than that in the model group.Conclusions Nicotine affects the proliferation and differentiation of callus,reduces callus formation,inhibits maturity transformation of bone,and delays the healing process of fracture.
