2.Ultrasound-guided percutaneous drainage for pancreatic pseudocyst

Objective To investigate the therapeutic value of ultrasound-guided percutaneous drainage in the treatment of pancreatic pseudocyst. Methods Twelve patients with pancreatic pseudocyst underwent ultrasound-guided percutaneous drainage from December 2000 to October 2003 in this hospital, including 1 case of simple puncture aspiration and 11 cases of drainage placement. Results A conversion to open cyst-jejunum Roux-en-Y anastomosis was required in 1 case because the cyst and the main pancreatic duct opened into each other. Pseudocysts disappeared in the remaining 11 cases. The drainage time was 7~90 days, with a mean of 28 days. No complications occurred. Follow-up in the 12 cases for 6~34 months (mean, 18 months) found 1 case of recurrence of cyst which decreased remarkably in size. Conclusions Ultrasound-guided percutaneous puncture aspiration and drainage for pancreatic pseudocyst is simple and feasible, offering advantages of minimal invasion and fewer complications. The procedure may be applied repeatedly at early stage to multiple sites.
3.Ultrasound-guided percutaneous transhepatic gallbladder or bile duct drainage in hepatobiliary diseases

Objective To investigate the practical value of ultrasound-guided percutaneous transhepatic gallbladder drainage (PTGD) and percutaneous transhepatic bile duct drainage (PTBD)in the treatment of hepatobiliary diseases. Methods A total of 68 times of ultrasound-guided PTGD/PTBD were performed in 60 patients from December 2000 to December 2003, including 15 cases of acute pyogenic cholecystitis, 8 cases of acute severe cholangitis and 37 cases of malignant obstructive jaundice. Results Out of the 15 cases of acute pyogenic cholecystitis, PTGD was completed successfully in 14 cases and failed in 1 case which then required a conversion to open operation. The 37 cases of malignant obstructive jaundice had undergone 45 times of PTBD, which consisted of 42 times of successful drainage tube placement and 3 times of failure (2 cases of conversions to open operation and 1 case of quitting treatment). Out of the 37 cases, resection of bile duct carcinoma or palliative cholangiojejunostomy was conducted in 14 cases, stents were inserted into the bile ducts under radioscopy in 8 cases, and the drainage tubes were left in place permanently in 15 cases (the maximum survival time was over 2 years and the mean survival time was 9 months). Of the 8 cases of acute severe cholangitis, successful drainage was achieved in 6 cases, a conversion to open surgery was required in 1 case because of blocked drainage, and 1 patient died of the multiple organ failure (MOF). Complications included 2 cases of bile leakage, 3 cases of hemorrhage, 1 case of portal vein-bile duct fistula, and 7 cases of drainage tube dislocation or blockage. Conclusions Ultrasound-guided PTGD/PTBD is an effective alternative for bile duct drainage, with advantages of minimal invasion, simplicity and accurateness of performance, safety and fewer complications. It presents much clinical value for hepatobiliary diseases, especially acute cholecystitis.
4.Study the differentiation of human bone marrow mesenchymal stem cells into dopaminergic neuron-like cells in vitro

Objective To explore the differentiation of human bone marrow measenchymal stem cells (hMSCs) into neuron-like and dopaminergic neuron-like cells in vitro.Methods The hMSCs were isolated from adult human bone marrow and expanded on the flask undifferentiated state for 2 passages. After pretreatment with WHI-P131 for 48 h, the hMSCs were cultured at the medium containing 10 ng/ml basic fibroblast growth factor for 24 h, then incubated with all-trans-retinoic acid and glial-derived neurotrophic factor in serum-free media for 5 h. The surface markers of differentiated neuron were detected by immunocytochemical method and the transdifferentiation process was observed under light microscope.Results Under induction conditions, hMSCs progressively resumed typical neuronal morphological characteristics. After hMSCs were incubated in induction medium for 5 h, the percentage of NSE, nestin, GFAP, TH and DAT positive cells were (77.0?5.7)%, (54.2?3.7)%,(8.8?2.4)%, (36.5?15.8)% and (26.0?14.2)%, respectively. There were no positive expressions in the control group.Conclusion The hMSCs are able to differentiate into neuron-like and dopaminergic neuron-like cells in vitro.
5.Angiogenesis and mosaic vessel in colorectal carcinoma.

Objective To investigate angiogenesis and mosaic vessel in colorectal carcinoma.Methods Forty-nine patients suffering from colorectal carcinoma were studied.Endothelial cell was labelled with monoclonal anti-human CD_~31 antibody,tumor cell was labelled with monoclonal anti-human CEA antibody.Endothelial cell and tumor cell were labelled by double-stained immunohistochemistry.The quantity of microvessel was estimated by counting microvessel density (MVD) at hot spots,and the quantity of mosaic vessels (MV) and double-stained cells were estimated at the same visual field.The relationship between mosaic vessels,microvessel density and Dukes staging were analyzed.Results The MVD in colorectal carcinoma "hot spots" was counted.The MVD of Dukes staging B was 43.98?21.46,staging C was 59.54?26.95,and staging D was 70.80?19.04.MVD increased gradually accompanied by clinical staging.There was statistical difference between staging B and D (P

Objective To investigate the effects of Huoxue yiqi runchang decoction in the treatment of chronic intractable constipation in colonic dynamics.Methods 68 cases patients of chronic obstinate functional constipation from October 2013 to June 2016 in linhai traditional chinese medicine hospital were selected and randomly divided into two groups,34 cases in each group,two groups received reasonable diet and lifestyle guidance and general treatment, and the control group received oral mosapride,and the experiment group received more with Huoxue yiqi runchang decoction.Levels of serum MTL and SP,the score of line Wexner constipation and GIQLI,colon motility examination,and clinical efficacy were compared.Results Compared with before treatment,levels of serum MTL and SP in two groups were increased (P<0.05),score of Wexner constipation decreased (P<0.05),and score of GIQLI increased (P<0.05),and transmission time of RCTT,LCTT and RSTT decreased (P<0.05).Compared with the control group,levels of serum MTL and SP in the experiment group were higher,score of Wexner constipation were lower (P<0.05),score of GIQLI were higher(P<0.05), and transmission time of RCTT,LCTT and RSTT was lower ( P <0.05 ) , and the clinical efficacy was higher ( P <0.05 ) .Conclusion Huoxue yiqi runchang decoction can improve clinical symptoms in patients with chronic constipation, and enhance the gastrointestinal motility,and improve the clinical efficacy.
7.Drug-resistance and Disinfectant-resistant Genes of Chlorhexidine-sulfadiazine-resistant Escherichia coli Isolates

OBJECTIVE To investigate the resistance and the disinfectant-resistant genes of chlorhexidine-sulfadiazine-resistant Escherichia coli isolates from hospitalized patients in the Lishui People's Hospital.METHODS Microdilute tests were performed to detect susceptibility to 19 kinds of antimicrobial agents in 60 E.coli strains,and the qacE△1 gene was detected by PCR.RESULTS The resistant rates to other antimicrobial agents arranged in 5.0-90.0%.The prevalence of ESBLs in isolates was 45.0%.qacE△1 Gene was 56.7%.CONCLUSIONS The 60 strains are multiple-drug-resistant.There are very high positive percentages of qacE△1 gene in E.coli isolates from the hospital.
8.Detection of five common cell culture contaminant Molllcutes using rolling circle amplification

Objective To develop a novel padlock probe and rolling circle amplification(RCA) to detect 5 common cell culture contaminant Mollicute( Mycoplasma arginini, Mycoplasma fermentans, Mycoplasma hyorhinis, Mycomplasma orale, and Acholeplasma laidlawii ). Methods "Universal" primers ( SPS1, SPA2 and SPS2, SPA1) were used to amplify the Mollicute 16S-23S rRNA intergenic spacer region.Amplicon was ligate Mollicute specific padlock probe. Probe amplified and monitored using a Corbett RotorGeneTM 6000 machine. Results Five reference Mollicute strains were correctly detected by RCA method.There was no cross-reaction. RCA method can sensitive detect 10 copies templates and show strong positive signal. Sixty-two cell culture specimens were detected. Thirty-seven samples were contained single specie Mollicute and 14 samples contained two Mollicutes. Eleven samples did not contained Mollicute. RCA detection results were concordant with previously species-specific PCR. Conclusion RCA can rapid, sensitive and specific detect contamination Mollicute.

Object To optimize the preparation procedure for WEI'AN GRANULES Methods The optimum conditions of WEI'AN GRANULES were selected with the active components: magnolol, polysacchride of Rhizoma Atractylodis Macrocephalae and berberine hydrochloride as mark by orthogonal test Results The optimum preparative procedure was as follow: Cortex Magnoliae officinalis and Fructus Aurantii Immaturus were extracted with water and precipitated with alcohol (adding 20 fold water as much as medicine, extracting three times, 0 5 h per time and precipitated with 95% alcohol to remove impurities); Rhizoma Coptidis was extracted with 95% alcohol by refluxing; Rhizoma Atracylodis Macrocephalae and other herbs were extracted with water (adding 24 fold water as much as medicine, extracting twice, 2 h per time, its granule size of Rhizorna Afracylodis Macrocephalae was 2 mm to 4 mm) Conclusion The optimum preparative procedure is reliable and the extracting ratio of active component is high
