2.Exploration on new assessment methods for cell biology among postgraduates

This paper summarized the assessment model reform practiced in the postgraduate cell biology course for several years and successfully obtain a new testing method based on research papers re-porting and questioning. Before the assessment, each student searched and selected one required research paper to read and make a PowerPoint reporting document. During assessment , some students were ex-tracted to speak and comprehensive score, which was regarded as student's final examination results was made by teaching group. The rest of the students listened to speeches , questioned and filled the questions on the record sheet.Teaching group reported a score , which was regarded as part of student's final exami-nation result based on questioning record sheets and printed PowerPoint documents.This method can well evaluate students' scientific thinking and strengthen the training of scientific thinking of postgraduates.
3.Study on the Preparation and in Vitro Dissolubility of Ofloxacin Hollow Suppository

OBJECTIVE:To prepare ofloxacin hollow suppository(OHS) and to observe its in vitro dissolubility.METHODS:OHS was prepared with PEG 6000,PEG 400 and Carbopol-940 as wase material and wax as retardant.The content of ofloxacin in OHS was determined with the first order derivative UV-spectrophotometry.RESULTS:The wax,as a retardant,could retard the release of drug.The in vitro dissolution of OHS revealed the first order dissolution pattern:K0~1=27.81/h,indicating a speedy effect,K2~6=5.94/h,indicating sustained-release effect.CONCLUSION:This preparation is feasible in technology and controllabe in quality.The preparation of OHS extends the kinds of dose-form of ofloxacin
4.Comparison of the Dissolubility of Chlorphenamine Maleate Tablets Produced by 4 Different Factories

OBJECTIVE:To compare the in vitro dissolubility of four kinds of commercial chlorphenamine maleate tablets. METHODS: To determine the dissolubility of four kinds of domestic commercial chlorphenamine meleate tablets by paddle method and to analyse the dissolution parameters, T50, Td, m, by variance analysis method. RESULTS: The dissolution parame- ters of the different tablets were T50(30. 3 197, 17. 3 695, 20. 1038, l4. 3 651), Td(34. 6 088, 26. 7 162, 28. 0 514, 22. 1 593)and m (2. 7 676, 0. 8 505, 1. 0 992, 0. 8 448). 5.Preparation and Quality Control of Compound Chlorphenamine Gel

OBJECTIVE:To prepare a compound gel composed of chlorphenamine(CH) and ephedrine hydrochloride(EP) METHODS:The gel was prepared by taking carbomer 940 as emulgator,trolamine as pH adjuster and propylene glycol as preservative The contents of CH and EP in gel were determined with the first order derivative spectroanalysis and dual-wave_length spectrophotometry RESULTS:The prepared gel exhibited fine consistency and moderate viscosity The content of CH was 101 0% with an average recovery of 100 4%,RSD 0 99% and the content of EP was 100 4% with an average recovery of 101 3%,RSD 1 7% CONCLUSION:This preparation is stable and non-irritative It is a newly developed preparation for tre_ating allergic rhinitis
6.The effect of helicobacter pylori eradication by doxycycline,levofloxacin,bismuth potassium citrate combined ;with omeprazole

Objective To observe the effect and safety of helicobacter pylori eradication by doxycycline, levofloxacin,bismuth potassium citrate combined with omeprazole.Methods 240 patients who infected helicobacter pylori were selected,and were randomly divided into four groups:treatment group(doxycycline,levofloxacin,bismuth potassium citrate,omeprazole),control group A(doxycycline,levofloxacin,omeprazole),control group B(amoxicillin, clarithromycin,bismuth potassium citrate,omeprazole),control group C (amoxicillin,clarithromycin,omeprazole), 60 patients in each group.Helicobacter pylori eradication treatment for 7 days,patients with gastritis stop after the course of the treatment,peptic ulcer patients continue taking omeprazole 5 weeks after 7 days treatment.All patients stopped medicine 2 weeks accepted 14 carbon breath test.And adverse events were investigated.Results In the treatment group and the control group A,B,C,the helicobacter pylori eradication rates were 93.3%,80.0%,91.7%, 75.0%.Compared with group A,the treatment group helicobacter pylori eradication rate increased significantly(χ2 =4.61,P <0.05 ).Compared with group C,the treatment group helicobacter pylori eradication rate increased significantly(χ2 =7.57,P <0.05).But compared with group B,the treatment group helicobacter pylori eradication rate had no significant difference(χ2 =0.12,P >0.05).Compared with the treatment group,group B had high inci-dence of oral odor(χ2 =6.56,P <0.05).Compared with the treatment group,group C had high incidence of oral odor (χ2 =5.46,P <0.05).Conclusion Doxycycline,levofloxacin,bismuth potassium citrate combined with omeprazole can improve the helicobacter pylori eradication rate significantly,and with low price,has little side effect,no need of penicillin skin test,can used for the patients poverty or allergic to penicillin.
7.Clinical study of stereotactic gamma-ray body radiation therapy for patients with stage Ⅰ / Ⅱ nonsmall cell lung cancer

ObjectiveTo further evaluate the value of the stereotactic gamma-ray body radiation therapy ( γ-SBRT ) for patients with stage Ⅰ / Ⅱ non-small cell lung cancer ( NSCLC ) basing on the preliminary studies.MethodsTwenty-nine eligible patients with stage Ⅰ / Ⅱ NSCLC who is unable or unwilling to receive surgery underwent treatment prospectively with γ-SBRT (OUR-QGD).Patients were fixed by vacuum bag.Each patient underwent slow CT simulation at 5 s/slide with thickness of 5 mm and interval of 5 mm to take into consideration tumor motion.A total dose of 50 Gy was delivered at 5 Gy/fraction to 50% isodose line covering 100% of the PTV,with 60 Gy at 6 Gy/fraction to 60% isodose line covering 90% of the CTV,70 Gy at 7 Gy/fraction to 70% isodose line covering 80% of the GTV,and 10 times finished in 2 weeks,5 fractions per week.ResultsThe follow-up rate was 97%.The 6 month local tumor response rate was 93%,with CR 86%,and PR 7%.The 1-,2-year local control rates were both 93%.The 1-,2-year overall survival rates for the whole group,stage Ⅰ and stage Ⅱ were 97% and 89%,100% and 92%,67% and 67%,respectively.The 1-,2-year progression-free survival were 90% and 86%,respectively.34% of the patients had acute radiation toxicities ( grade 1,2 and 3 in 6,2 and 2patients,respectively),and 38% late radiation toxicities ( grade 1 and 2 in 10 and 1,patients).Conclusions γ-SBRT is a safe and effective treatment regimen,resulted in promising local control and survival with minor toxicity.
8.Clinical outcome of whole body γ-knife for limited pancreatic carcinoma

Objective To evaluate the efficacy and side effects of whole body γ/-knife in patients with limited pancreatic carcinoma. Methods 111 patients with limited pancreatic carcinoma treated with the Stereotactie Gamma Ray Whole-Body Therapeutic System (Whole Body T-knife) were retrospectively an-alyzed. Patients were supine, fixed with a stereotactic body frame and vacuum bag, and then simulated by low-speed computed tomography. GTV, CTV and PTV were defined on the contrast-enhanced CT scans. It was required that 50% isedose line covered 100% of PTV and 70% isedose line covered more than 80% of GTV. The prescription dose was defined as 50% isodose. All patients were treated 5 fractions per week. The fractionated dose was 3-4 Gy for pancreatic head carcinoma, and 4-5 Gy for pancreatic body/tail carcino-ma. Irradiation of 40-51 Gy and 60-70 Gy were delivered to PTV and GTV margins, respectively. Re-suits The complete response rate, partial response rate and overall response rate of the primary tumors were 29.7%, 42.3% and 72.1%, respectively. The follow-up rate was 95.5%. The number of patients fol-lowed-up at 1-,2- and 3-year was 105,89 and 60. The 1-, 2- and 3-year overall survival rates were 49.3%,24.5% and 18.1%. For patients with stage Ⅰ/Ⅱ disease,the number of patients followed-up at 1-,2-,3-,4-and 5-year was 55,44,29,16 and 11 ;The 1-, 2-, 3-,4- and 5-year overall survival rates were 68%,34%, 30%, 21% and 17%, respectively. For patients with stage Ⅲ disease,the number of patients fol-lowed-up at 1-,2- and 3-year was 50,45 and 31 ;The 1-, 2- and 3-year overall survival rates were 28%,14% and 4%, respectively (χ~2=16.67, P=0.000). The acute side effects including nausea, vomiting and diarrhea were 71.2% of RTOG grade 1 -2 and 3.6% of RTOG grade 3. No treatment delay occurred.Conclusions With fractionated dose of 3-5 Gy,5 fractions per week and 40-51 Gy as total dose to PTV,whole body γ-knife is safe and effective to treat limited pancreatic carcinoma. The local control and overall survival could be improved.
9.Changes in ischemic myocardium following marrow mesenchymal stem cells implantation by echocardiography in rabbits

BACKGROUND: Recent studies indicate that bone marrow messenchymal stem cells (MSCs) can repairinjured myocardium and improve cardiac functions. However, there are still no effective non-invasive methods to detect ventricular wall motion, survival status of transplanted cells, and microcirculation improvement after MSCsimplantation. OBJECTIVE: To detect the effects of MSCs implantation on the cardiac function of ischemic myoeardium and investigate changes of tissue acoustics characteristics by using integrated backscatter (IBS) and Doppler tissue imaging (DT1). DESIGN, TIME AND SETTING: Completely randomized grouping design, and randomized controlled study, which was carried out in the Department of Cardiology, Renmin Hospital of Wuhan University from December 2004 to June 2005 (important laboratory of 211 government). MATERIALS: Forty-eight male rabbits with irrespective of gender were used for study. Anti-troponin T (TnT) antibody was provided by Maxim Company, USA; anti-bromodeoxyuridine (BrdU) antibody by Sigma Company, USA; Sonos 5500 echocardiography by HP Company, USA.METHODS: Forty-eight rabbits were randomly divided into model group and implantation group. Model of myocardium infarction was created in rabbits by left anterior descending coronary artery (LAD) ligation. Three days after modeling, autologous bone marrow MSCs were injected into crossed myocardium between infarction and non-infarction regions based on 5 injected points.MAIN OUTCOME MEASURES: Indicators including ventricular size, ventricular wall thickness, and ejection fraction (EF) in the left ventricle (LV) were measured with routine echocardiography one day before myocardial infarction, two weeks after myocardial infarction, and four weeks after implantation. IBS and DTI might detect the anterior backscatter parameters (AII, or IB) and ventricular wall motion parameters (PPI, or CVIB) to calculate the average backscatter scores and backscatter cycle changes. DTI was used to measure peak systolic flow velocity (VS), peak early diastolic flow velocity (VE), and peak late diastolic flow velocity (VA) of the middle and basal segments of the anterior wall of left ventricle. Finally, animals were sacrificed by anesthesia, and myocardial infarction region was stained with 5-Brdu immunohistochemical staining, and anti-TnT staining. RESULTS: Twelve rabbits were excluded because of death after operation.① Echocardiography: Imaging of 36 rabbits were clear in 1 day before myocardial infarction, 2 weeks after myocardial infarction, and 4 weeks after implantation. The sizes and systolic function of LV in 36 rabbits were normal before operation. Two weeks after operation, the sizes of LV became larger, the anterior wall of LV became thinner, and systolic range became weaker even disappeared. EF, CVIB, Vs, and Ve were lower (P < 0.05) and All was higher in both groups than pre-operation (P < 0.05). Four weeks after implantation, compared with control group, left ventricular diastolic diameter (LVDd) was smaller and systolic range became larger, All descended. EF, Vs, Ve and CVIB increased (P < 0.05).② Myocardial immunohistochemical detection: Numerous of Brdu-labeled positive cells were scattered, and anti-TnT was positive, which clearly showed horizontal stria of myocardium. However, normal myocardium was negative. CONCLUSION: Bone marrow MSCs after implantation can survive in ischemic myocardium and improve cardiac function. IBS and DTI can real-timely and exactly detect functional changes of infarction myocardium after bone marrow MSCs implantation.
10.Target in target radiotherapy for 90 patients with spinal metastases

Objective To analyze the result of dose-escalated radiotherapy vs conventional radiotherapy for spinal metastases. Methods Ninety patients (118 lesions) underwent radiotherapy..There were 52 patients (63 lesions) in target in target radiotherapy (TIT) receiving PTV 30~40 Gy, GTV 50~60 Gy/15~25 f. 38 patients (55 lesions) received PTV 30~40 Gy/10~20 f in the conventional radiotherapy. Results The overall responses were 90.4% and 81.6%,respectively.but no significant difference was found. There were 31 and 25 improved lesions, 28 and 20 steady lesions, 4 and 10 worsening lesions,in two goups, respectively. 1-year local control rates and survival were 84.7%vs 68.6%and 49.4%vs 32.2%, respectively. The significant differences were found in 1-year local control rates and survival. The main acute adverse reactions were mainly hematologic toxicities , no significant difference was found. Conclusion TIT for spinal metastases is a more safe and effective treatment, with better efficacy and fewer side-reactions.
