1.Culture and identification of microvascular endothelial cells from human endometriosis

Objective To establish the methods of isolating and culturing human ovarian endometriosis-derived microvascular endothelial cells (OEMEC).Methods The tissues of human endometriotic cyst of ovary were finely minced with scissors,then digested by collagenase Ⅰ,Ⅱ and trypsinethylene diamine tetraacetic acid (EDTA).The cells were purified by using centrifugation of 2000 r/min speed.OEMEC were identified by light microscope and transmission electron microscope observing CD34,FⅧ-Rag and Weibel-Palade in microvascular endothelial cells.Results The OEMEC grew as confluent monolayer like cobblestones under light microscope.CD34 and FⅧ-Rag were expressed strongly,and the percentages of CD34 and FⅧ-Rag positive cells were 91.4% and 92.5%.Weibel-Palade bodies could be observed under transmission electron microscope.The time of cell doubling proliferation was 4.5 days.Conclusion The established system of isolating OEMEC would provide lab base for studying the mechanisms of angiogenesis in endometriosis lesions.
2.Estimation of patient doses in paediatric cardiovascular interventional radiology under specific exposure conditions

Objective:To estimate the organ doses and effective doses to different-age children during cardiovascular interventional radiological procedures under some specific exposure conditions, and explore the main influencing factors on the doses.Methods:Based on the paediatric reference computational phantoms recommended in the ICRP Publication 143, several specific exposure models of cardiovascular intervention were built, and the Monte Carlocook MCNPX 2.7.0, was used to calculate the organ doses and effective doses for 1-, 5-, 10- and 15-year-old children. To validate the simulation result , an experiment was implemented by putting the thermoluminescent dosimeters in a 5-y old phantom (ATOM 705-D) manufactured by the CIRS Inc. in the USA.Results:Both the height and weight of the reference children for 1-, 5- and 10-year-old provided for by Chinese national standards are nearly in consistency with those recommended by ICRP, and even for the 15-year-old, the maximum relative deviations of the height and weight are only -1.9% and -5.7%, respectively. Under the exposure condition where the focal spot to image receptor distance (SID) was 90 cm, the length of square field of view (FOV) was 30 cm with a dose area product (DAP) of 45 Gy·cm 2, the relative deviations between simulated and measured doses to main organs/tissues within the irradiation filed were within ±6.7%. Under the same exposure conditions, the younger the children, the larger the organ doses and effective doses, and the effective doses could vary by a factor of about 5 among the 4 age groups. The conversion coefficient between the organ dose and the value of DAP was not only closely related to the age of children, but also affected by the FOV. Conclusions:In combination with the paediatric reference computational phantoms and the exposure models of cardiovascular intervention, the Monte Carlo method can be used to calculate the doses to children undergoing cardiovascular interventional radiological procedures. The information on the values of DAP and FOV as well as the directions of projection are needed for more accurate estimation of the exposure doses.
3.Respiratory pathogen distribution characteristics in children with acute respiratory disease

Objective To investigate the respiratory pathogen distribution characteristics in children with acute respiratory disease(ARD).Methods Distribution of respiratory pathogen in 28 600 children with ARD,treated from January 2011 to December 2014 in this hospital,were analyzed.Results Among the 28 600 children,12 162 cases were pathogen positive,including 7 704 cases(63.34%) with single pathogen infection and 4 458 cases(36.66%) with more than two kinds of pathogens infection.Time,season,sex and age distribution of pathogen infection were with statistical difference(P<0.05).There was significant difference in infection rate of pathogens between different time(P<0.05).Infection rate of Mycoplasmal pneumonia(MP) was the highest,and the infection rate of MP and influenza B virus(IFB) increased year by year.Except Legionella pneumophila(LP),the season distribution of infection rate of MP,Coxiella burneti(COX),Chlamydia pneumoniae(CP),adenovirus(ADV),respiratory syncytial virus(RSV),influenza A virus(IFA),IFB and parainfluenza virus(PIVs) were with statistical difference(P<0.05).The infection rate of MP and IFB were higher in the whole year.Conclusion Distribution of respiratory pathogen in children with ARD might be related with the changes of gender,age and season.Detection of respiratory infection pathogen could be with guiding value for clinical diagnosis and drug selection.
4.A preliminary study on the system construction for evaluating the professional capability of provincial radiological health institutions

Objective:To construct a system for evaluating the professional capability of provincial radiological health institutions.Methods:Based on the Donabedian model and the main professional responsibilities of provincial radiological health institutions, the logical framework and indicator database of the capability evaluation system were initially constructed, the Delphi expert consultation method and analytic hierarchy process were further used to determine each indicator and its weight. The self-assessment test was carried out throughout the provincial radiological health institutions by using the system established in this study.Results:The evaluation system included 3 primary-class indicators, 11 second-class indicators, 30 third-class indicators and 76 fourth-class indicators. Taking 100 points as the full score, the self-assessment scores of the 29 provincial institutions ranged from 28.7 to 97.7 with an average of 78.7, and the scores conform to the normal distribution.Conclusions:The system established in this study are scientific, comprehensive and operable, which can be used as an effective tool to evaluate the professional capability of provincial radiological health institutions.
5.Effects of grid methods on precision and efficiency of specific absorbed fraction calculation in Monte Carlo simulation

Objective To compare the precision and efficiency of computing the specific absorbed fraction (SAF) of a reference human with two grid methods in MCNP6.0. Methods Based on the adult female reference voxel phantom provided by the International Commission on Radiological Protection, assuming the liver as the source organ emitting single-energy photons (0.5 MeV), the SAF of each target organ/tissue was calculated by using the mesh method and repeated structure lattice method with the F4, F6, and *F8 tally cards in MCNP6.0. We compared the methods by assessing the relative deviation of SAF and computing time for 27 organs/tissues. Results Compared with reported data, the absolute values of relative deviations of SAF values for all the organs/tissues were less than 5%, except for the eye lens and skin. By using the repeated structure lattice-based *F8 tally, the relative deviations of SAF values of the organs/tissues were all smallest, but with the longest computing time. The computing time of the mesh-based F4 tally was slightly longer than that of the repeated structure lattice-based F6 tally, which was shortest. Conclusion The *F8 tally simultaneously simulating primary and secondary particle transport showed the highest precision. The mesh tally requireda longer computing time than the lattice tally when using the same tally card.
6.Performance of modified Hodge test on the detection of carbapenemase among Enterobacteriaceae

Objective To evaluate the performance of modified Hodge test on the detection of carbapenemases among Enterobacteriaceae with decreased susceptibility to carbapenems. Methods Fortynine Enterobacteriaceae isolates with decreased susceptibility to carbapenems ( MIC of imipenem, meropenem or ertapenem was ≥ 2 μg/ml ) were collected from 16 teaching hospitals from 2004 to 2008. MICs of imipenem, meropenem and etapenem were determined by agar dilution method. Carbapenemases were detected by modified Hodge test. Carbepenemase-causing positive results and AmpCs-causing positive results were differentiated by phenyl boronic acid and oxacillin. Beta-lactamases encoding genes including blaNDM-1were detected by PCR and sequencing. Results Thirty-six of 49 isolates were non-susceptible to imipenem (MIC >4 μg/ml), 31 were non-susceptible to meropenem (MIC > 4 μg/ml) and 47 were non-susceptible to ertapenem (MIC > 2 μg/ml). Twenty-three isolates showed positive modified Hodge test result, including 9 weak-positive results and 14 strong-positive results. Through PCR detection and sequencing, 2 out of 9 isolates showing weak-positive results carried blaKPC-2 and other 7 did not carry any carbapenemase genes but AmpCs/ESBLs genes. Among the 14 isolates showing strong-positive results, 4 carried blaKPC-2, 8 carried blaIMP-4 and 2 caried blaIMP-8. All 26 isolates with negative modified Hodge test result didn't carry any carbapenemase genes. No isolate carried blaNDM-1. Carbapenemases genes PCR detection was regarded as a gold standard, and the sensitivity, specificity, positive predictive value and negative predictive value of modified hodge test was 100%, 79%, 70% and 100% on the detection of carbapenemases among Enterobacteriaceae with decreased susceptibility to carbapenems. Conclusions Modified Hodge test revealed great sensitivity but showed a few false positive results. True and false positive results can be effectively differentiated by phynel boronic acid and oxacillin.
