1.Effects of Shenqifuzheng injection on T-cell subsets and oxidative stress in patients with diabetes mellitus

Objective To investigate the effects of Shenqifuzheng injection on T-cell subsets and oxidative stress in patients with diabetes mellitus . Methods 90 cases of 2 diabetes patients from June 2015 to June 2016 in our hospital were selected and randomly divided into control group and observation group,45 cases in each group,the control group treated with conventional treatment,the observation group was given Shenqifuzheng injection based on the control group.The levels of peripheral blood T cell subsets,serum oxidative stress metabolites glutathione peroxidase,superoxide dismutase and catalase,the control of blood glucose were compared between the two groups.Results After treatment,the levels of Th17,Treg and Th17/Treg in control group compared with before treatment had no significant difference, the levels of Th17 and Th17/Treg in observation group were obviously decreased and lower than in control group (P<0.05);the content of Treg in observation group was increased significantly and higher than the control group (P<0.05).After treatment,the levels of CAT in observation group was lower than control group,GSH-PX and SOD were higher than control group,the difference was statistically significant (P<0.05).After treatment,the GLU,2h PG,HOMA-IR and HbA1C (%) in observation group were significantly decreased than before treatment and lower that in control group, the difference was statistically significant ( P<0.05 ). Conclusion Shenqifuzheng injection can effectively improve the T cell subsets and oxidative stress levels in diabetic patients , help to control blood sugar.
2.Fluid-based thin-layer method for screening of squamous intraepithelial lesions in pregnant women

Objective To investigate the clinical significance and safety of liquid-based cytology (thinprep cytologic test, TCT ) in pregnant women and cytological characteristics of the smears during pregnancy. Methods TCT were performed in 1951 pregnant women (group A), 3752 gynecological out-patients(group B)and 5069 routine screening women (group C ). The age of the women was ranged from 20 to 39 years. The cytological preparations were analyzed using TCT method following the Bathesda system (TBS) standard. Results (1) The incidence of abnormal epithelial patterns in groups A, B and C was 23.58%(460/1951), 19.22%(721/3752) and 13.40%(679/5069) respectively (P0.05). Conclusions TCT smear screening is necessary, safe and acceptable for routine prenatal tests. There is no difference in the cervical squamous intraepithelial lesions between pregnant and non-pregnant women.
3.The effects of rosmartnic acid on the urine toxins,urinary protein ,serum MDA and SOD in the rats with the 5/6 nephrectomy

Objective To investigate the effects of rosmarinic acid on the kidney function,urinary protein,serum MDA and SOD in rats with the 5/6 nephrectomy.Methods We established the rat model with chronic renal dysfunction and randomized them into normal control group,model control group and rosmarinic acid treatment group.There were 6 rats in each group.The blood creatinine level was recorded 2 weeks after operation.We detected the effects of rosmarinic acid on urinary protein,urine creatinine,blood creatinine,serum MDA and SOD in rats with the 5/6 nephretomy which was established by Ponseau S method.The treatment group received rosmarinic acid as an intervention.Results ( 1 ) Compared with control group,the creatinine and The creatinine clearance rate ( Ccr ) of rosmarinic acid treatment group increased ( F =38.912,P < 0.01 ;F =19.968,P < 0.05 ) after treatment,; (2) Rosmarinic acid improved the 24 h urine protein in rats with renal dysfunction significantly(F =16.288 P <0.01); (3) Rosmarinic acid had no effect on serum MDA and SOD in rats.Conclusion Rosmarinic acid can decrease urine protein and blood creatinine,increase the Ccr and alleviate kidney failure,but has no effects on MDA and SOD in rat model.
4.Respiratory pathogen distribution characteristics in children with acute respiratory disease

Objective To investigate the respiratory pathogen distribution characteristics in children with acute respiratory disease(ARD).Methods Distribution of respiratory pathogen in 28 600 children with ARD,treated from January 2011 to December 2014 in this hospital,were analyzed.Results Among the 28 600 children,12 162 cases were pathogen positive,including 7 704 cases(63.34%) with single pathogen infection and 4 458 cases(36.66%) with more than two kinds of pathogens infection.Time,season,sex and age distribution of pathogen infection were with statistical difference(P<0.05).There was significant difference in infection rate of pathogens between different time(P<0.05).Infection rate of Mycoplasmal pneumonia(MP) was the highest,and the infection rate of MP and influenza B virus(IFB) increased year by year.Except Legionella pneumophila(LP),the season distribution of infection rate of MP,Coxiella burneti(COX),Chlamydia pneumoniae(CP),adenovirus(ADV),respiratory syncytial virus(RSV),influenza A virus(IFA),IFB and parainfluenza virus(PIVs) were with statistical difference(P<0.05).The infection rate of MP and IFB were higher in the whole year.Conclusion Distribution of respiratory pathogen in children with ARD might be related with the changes of gender,age and season.Detection of respiratory infection pathogen could be with guiding value for clinical diagnosis and drug selection.
5.Diagnosis and Differential Diagnosis of MRI in Periampullary Carcinoma

Objective To explore the value of magnetic resonance cholangiopancreatography (MRCP) and MRI in the diagnosis and differential diagnosis of periampullary carcinoma. Methods Plain and dynamic multi-phase enhanced MRI data of 54 patients with periampullary carcinoma proved by pathology were retrospectively analysed. χ~2 test and two independent samples t test were used to examine the relative results. The results of MRI were compared with that of pathology. Results Of 32 cases with carcinoma of head of pancreas, 7 cases (21.9%) exhibited "four-duct sign". 16 cases were carcinomas of the lower part of common bile ducts, 9 cases (56.3%) showed "three-duct sign". The difference between carcinomas of head of pancreas and carcinomas of the lower part of common bile ducts was significant (P<0.05). Additionally, the ratio of the largest area of masses and the diameter of com-mon bile duct in pancreatic cancer tumors was larger than that in common bile duct cancer (P<0.05) for whether the intraoperative or MRI measurement. Conclusion MRI and MRCP are very helpful in diagnosis of periampullary carcinoma.
6.Dynamic contrast-enhanced MRI in the evaluation of blood-brain barrier permeability following acute cerebral ischemia in rats

Objective To investigate the dynamic changes of blood-brain barrier(BBB) permeability after acute cerebral ischemia in rats with middle cerebral artery occlusion (MCAO) by dynamic contrast-enhanced(DCE)-MRI. Methods Sixty MCAO rat models were established by suture-occlusion method. All rats were divided randomly into twelve groups with different ischemia duration (3 hours, 6 hours, permanent) and reperfusion times (2, 6, 12 and 24 hours after reperfusion). Each group was examined by MRI at the time points. The BBB permeability parameters(Ktrans, Ve, Kep, rKtrans, rVe, rKep) were calculated by Siemens workstation and compared with Evans blue(EB) extravasation results. Multivariate analysis of variance (M-ANOVA), one-way analysis of variance (one-way ANOVA), Pearson analysis were respectively used to verify the influences of ischemia duration and reperfusion time on BBB permeability parameters, EB extravasation and relationships between parameters. Results In 3 hours and 6 hours ischemia duration groups, change of BBB permeability after reperfusion appeared biphasic. At 2 hours and 6 hours after reperfusion, BBB permeability increased, while rKtrans values and rVe values rose and rKep values dropped. BBB permeability decreased at 12 hours and increased again at 24 hours after reperfusion. The highest BBB permeability was observed at 6 hours after reperfusion. However, BBB permeability in permanent ischemia groups had uniphasic change, as its increase was rather mild as ischemia time went on. rKtrans values(1.99± 0.79)were positively correlated with rVe values(2.88 ± 1.78) (r=0.93, P<0.01) and negatively correlated with rKep values(0.66 ± 0.21) (r=-0.84, P<0.01). The negative correlation between rVe values and rKep valueswas also significant(r=- 0.80, P<0.01). EB extravasation results were consistent with MRI findings. Conclusions BBB permeability change was biphasic in reperfusion groups, while it was uniphasic in permanent ischemia groups. DCE-MRI may accurately reflect the changes of BBB permeability after acute cerebral ischemia. Both ischemic duration and reperfusion time had influences on BBB permeability. With prolongation of ischemic time, the duration of BBB permeability increase became shorter, BBB damage appeared earlier, with increased degree of ischemic damage.
7.Evaluation on the performance of MicroScan WalkAway in detecting carbapenem-resistant Enterobacteriaceae

Objective To investigate the performance of MicroScan WalkAway 96 Plus (MSW) system in detection of carbapenem-resistant Enterobacteriaceae (CRE).Methods A total of 81 stock CRE strains were used in this study. Bacterial identification and antimicrobial susceptibility test were performed by MSW system. Beta-lactamases genes blaKPC,blaIMP,blaVIM, blaOXA-48 and blaNDM were amplified by PCR and subjected to sequencing analysis. Disk diffusion method and PCR were used as gold standard to evaluate the performance and reliability of MSW system in identifying carbapenem-resistant and carbapenemase-producing Enterobacteriaceae.Results Overall, 69.1 % (56/81) of the Enterobacteriaceae strains were identified as CRE by the MSW system. The results of PCR showed that 48 strains were carbapenemase-producing Enterobacteriaceae. When carbapenemase-producing Enterobacteriaceae strains were identified by the instrument using an advanced expert system, the sensitivity was 93.8 % and specificity was 42.4 %. The positive predictive value was 70.3 %, the negative predictive value was 82.4 % and the predictive accuracy value was 72.8 %.Conclusions The MicroScan WalkAway 96 Plus system has shown good performance in detection of CRE.
8.Analysis of composition differences of intestinal microbiota in patients with colon cancer and rectal cancer

Objective:To analyze the composition differences of intestinal microbiota in patients with colon cancer and rectal cancer.Methods:The fecal samples of 72 patients in the Second Affiliated Hospital of Harbin Medical University from July 2018 to January 2019 were collected, and they were divided into colon cancer group and rectal cancer group, 36 cases in each group. DNA from fecal samples was extracted, and then high-throughput sequencing was performed on DNA. Bioinformatics was used to analyze the diversity and composition differences of intestinal microbiota between the two groups, and the potential cancer-promoting mechanisms of the differential flora were also discussed.Results:From high-throughput sequencing, 2 356 560 original sequences and 32 730 high-quality sequences were obtained from 72 samples. The average length of the sample sequence was mainly in the interval of 401-460 bp. And 1 409 operational taxonomic units (OTU) were acquired after OTU species taxonomy annotation of all the sequences. Alpha diversity analysis showed that Shannon index of the rectal cancer group and the colon cancer group was 2.61±0.56 and 2.43±0.67, respectively, and the difference was statistically significant ( t = 1.229, P = 0.223); Simpson index of the rectal cancer group and the colon cancer group was 0.17±0.09 and 0.21±0.16, respectively, and the difference was statistically significant ( t = 1.449, P = 0.151). Differences analysis of both groups and linear discriminant analysis (LDA) showed at the phylum level, Firmicutes were more abundant in the intestine of patients with rectal cancer (LDA = 4.67, P = 0.014), while Proteobacteria were more abundant in the gut of colon cancer patients (LDA = 4.49, P = 0.042). From the perspective of class level, the abundance of Gammaproteobacteria was higher in the intestine of patients with colon cancer (LDA = 4.50, P = 0.033), while the abundance of Erysipelotrichia was higher in the intestine of patients with rectal cancer (LDA = 3.50, P = 0.035). At the order level, the abundance of Erysipelotrichales was higher in the intestine of patients with rectal cancer (LDA = 3.50, P = 0.035); at the family level, the abundance of Porphyromonadaceae was higher in the intestine of patients with rectal cancer (LDA = 3.97, P = 0.033). Conclusion:The compositions of intestinal microbiota in patients with colon cancer and rectal cancer are significantly different, indicating that the different floras may contribute to the progression of colon cancer and rectal cancer.
9.Comparison of high-resolution diffusion-weighted imaging and diffusion kurtosis imaging for prediction of radiotherapy response in patients with nasopharyngeal carcinoma

Objective In this prospective study,the performance between high-resolution diffusion-weighted imaging ( DWI) and diffusion kurtosis imaging ( DKI) for prediction of radiotherapy response in patients with nasopharyngeal carcinoma was compared. Methods Forty-one patients pathologically diagnosed with NPC received IMRT. All patients underwent conventional MRI,high-resolution DWI and DKI before and after radiotherapy (1-2 d after the plan dose was administered).All patients received conventional MRI during follow-up at 3,6,9 and 12 months after radiotherapy. According to the RECIST 1. 1( response evaluation criteria in solid tumors),all patients were divided into the response group (RG;n=36) and non-response group (NRG;n=5). The mean kurtosis coefficient (Kmean) and the mean diffusion coefficient (Dmean) of DKI and apparent diffusion coefficient ( ADC) of DWI were analyzed before and after radiotherapy. Results Among 41 patients,36 cases were assigned into the RG group and 5 in the NRG group. Before and after radiotherapy, all parameters significantly differed between two groups ( P=0. 000-0. 013) except for the Dmeanand ADC prior to radiotherapy. At the end of radiotherapy,the sensitivity of Kmeanwas calculated as 87. 5% and the specificity was 91. 3% for predicting local control (optimal threshold=0. 30, AUC: 0. 924; 95%CI: 0. 83-1. 00 ). Conclusion Kmeanvalue after radiotherapy is a potential biomarker for the early evaluation of clinical efficacy of radiotherapy in NPC patients.
10.Influence of Japanese enciphalitis virus capsid protein on the self-replicate ability of JEV replicon vectors.

To optimize a self-replicate Japanese enciphalitis virus (JEV) replicon, and to make it as an efficient vector to express the heterologous protein, we constructed three JEV replicons by PCR-based shortening the length of capsid genes. The vectors remained full or part of C gene, based on the JEV replicon pCTCJEV. Lac Z was selected as the reporter gene to verify the self-replicate ability of these DNA-based replicons. While transfected into the cell lines CME-4, which continuously expressing the JEV structure proteins C-prM-E, the JEV replicons pCMW-2M-1LACZ, pCMW-2M-3LACZ, which remained the first 23aa and 68aa of C protein, can express the reporter protein as the same level as pCMW-2M-LACZ with the full-length C protein. These results illustrated that the JEV replicon vector with 69-nt of the C gene can retain the self-replicate ability, and provide valuable tools to construct a possible vector for a long-lasting JEV RNA virus expression system.
