1.Effect of Compound Fuling Gancao Decoction on Transforming Growth Factor ?_1 in Pulmonary Artery of Rats with Pulmonary Hypertension

Objective To observe the effect of Compound Fuling Gancao Decoction(CFGD) on transforming growth factor ?1(TGF-?1) in pulmonary artery of rats with pulmonary hypertension(PH),and to explore its therapeutic mechanism for PH and pulmonary vascular remodeling.Methods Thirty SD male rats were randomized into the normal control group,model group,and CFGD(5.46g?kg-1?d-1) group.PH rats models were induced by intraperitoneal injection of monocrotaline(50mg/kg).The mean pulmonary artery pressure(MPAP) in rats was detected with the method of right cardiac catheterization,right ventricular hypertrophy index was calculated,histological changes of lung small artery were analyzed with pathological image analysis,and TGF-?1 expression in pulmonary artery was detected by immunohistochemical method.Results MPAP and right ventricular hypertrophy index were enhanced obviously,the number of muscular arteries and partially-muscular arteries was increased,ratio of media area and luminal thickness in the muscular arteries became bigger,and TGF-?1 expression in pulmonary artery of the model group was increased.The above indexes were improved in CFGD group.Conclusion CFGD is effective on decreasing pulmonary pressure and improving pulmonary vascular remodeling,and its mechanism is probably related with the inhibition of TGF-?1 expression in pulmonary artery.
2.Simultaneous Determination of Hesperidin and Phillyrin in Siji Ganmao Capsules by HPLC

Objective:To establish an HPLC method for the simultaneous determination of hesperidin and phillyrin in Siji Gamao capsules. Methods:HPLC was applied with the chromatographic conditions as follows:the chromatographic column was Agilent-XDB C18 (250 mm × 4. 6 mm,5μm) at 30℃, the mobile phase was methanol-0. 1% phosphoric acid solution with gradient elution, the flow rate was 1. 0 ml·min-1 , the detection wavelength was 277nm and the injection volume was 5μl. Results:The linear range of hesperi-din and phillyrin was 12.000-60.000 μg·ml-1(r=0.999 6) and 18.000-90.000 μg·ml-1(r=0.999 3),respectively; the average recovery was 96.61%(RSD=1.40%) and 97.66%(RSD=1.27%), respectively. Conclusion: The method is simple, accurate and repeatable, which can be used in the quality control of Siji Ganmao capsules.
3.Regulation of osteopontin expression by its antisense RNA in renal epithelial cells

AIM: To investigate the effect of antisense RNA on osteopontin (OPN) expression in renal tubular epithelial cells. METHODS: Cell clone expressing stably OPN antisense RNA was formed by transfering retroviral vector expressing OPN antisense RNA into renal tubular epithelial cells, NRK52E cells, using liposome, with cell clones transfected by empty vector and vector expressing OPN sense RNA as controls. Ribonuclease protection assay(RPA), Western Blot, ELISA and assay of OPN activity were performed to detect expression of OPN mRNA and protein in above clones cultured with or without epidermal growth factor(EGF). RESULTS: The antisense RNA was only expressed by antisense clone. Antisense clone, sense clone and empty clone all expressed OPN mRNA. EGF enhanced expression of OPN mRNA, but not OPN antisense RNA or OPN sense RNA in above clones. OPN protein was not expressed in antisense clone cultured with or without EGF and empty clone cultured without EGF, but was expressed in sense clone cultured with or without EGF and empty clone cultured with EGF. CONCLUSION: Antisense RNA can inhibit OPN protein expression by means of preventing OPN mRNA translation, but not inhibit OPN mRNA transcription in renal tubular epithelial cells.
4.Detection of Virulence Factors of Vibrio parahaemolyticus by Real-Time PCR

OBJECTIVE To develop a real-time fluorescence PCR assay to detect the genes encoding thermolabile(hemolysin)(TLH),thermostable direct hemolysin(TDH) and TDH-related hemolysin(TRH) of Vibrio(parahaemolyticus).METHODS The genes of TDH and TRH were selected as target ones of thermostable direct and TDH-related hemolysin,and TLH gene as a specific genomic marker for V.parahaemolyticus.Designed and synthesized the primers and Taqman probes,we investigated 487 stool samples of doubt foodborne illness patients by real-time fluorescence PCR.RESULTS The sensitivity of the assay for TLH and TDH was 1.0?10~2copies,but the sensitivity of TRH was 1.0?10~3copies. Among 487 samples,112 V.parahaemolyticus strains were found;101 samples of these strains showed the production of TDH;none of them was positive for TRH.CONCLUSIONS The Taqman PCR is a rapid and sensitive method to detect the TLH,TDH and TRH of V.parahaemolyticus,it is well suited for screening large numbers of samples at the same time;and TDH is one of the primary virulence factors in clinical isolated V. parahaemolyticus.
5.Construction and application of a management scheme for nursing assistants in unaccompanied hospitals

Objective To construct a management scheme of nursing assistants in unaccompanied hospitals and explore its application effect.Methods The management team of nursing assistants in unaccompanied hospitals was set up.On the basis of investigating the current situation of nursing assistants'team management,the management scheme of nursing assistants was constructed by ORTCC management model for references,including 5 parts:determining management objectives,perfecting rules system,building hierarchical training system,checking and assessing,and shaping cultural system.Convenience sampling method was used to select 92 nursing assistants in an unaccompanied hospital in Xiamen,Fujian Province from June 2021 to June 2022 as application subjects.The post competence of nursing assistants and the satisfaction of patients and nurses to nursing assistants were compared before the application of the management scheme(June-December 2021)and after the application(January 2022-June 2022).Results After the application of ORTCC management model,the total score of post competency of medical nursing assistants increased from[65(64,67)]to[91(90,92)];the excellent rate of post competency level increased from 1.09%to 67.39%;the satisfaction of patients with nursing assistants increased from 61.99%to 88.62%;the scores of nurses'satisfaction with nursing assistants in all dimensions were significantly higher than before(all P<0.05),and the overall evaluation was improved from 92.73%to 99.24%.Conclusion The use of the ORTCC management model for nursing assistants can effectively improve their post competence,improve the satisfaction of patients and nurses,and help to further stabilize and develop the medical nursing assistants staff.
6.Autoantibody screening for the diagnosis of autoimmune cerebellitis

Objective To establish a test of autoantibody-panel for the diagnosis of autoimmune cerebellitis (AC) and determine the prevalence of AC in patients with cerebellar ataxia of unknown etiology.Methods Autoantibody screening tests with indirect immunofluorescence were performed in serum and cerebrospinal fluid (CSF) samples of 400 previously'idiopathic'Chinese patients with cerebral ataxia (inpatients and outpatients in Peking Union Medical College Hospital or referred from hospitals of Beijing Encephalitis Group from 2016 to 2018).Immunotherapy was given to autoantibody positive patients and the effectiveness of immunotherapy was assessed.Detailed AC autoantibodies panel included anti-glutamate decarboxylase 65 (GAD65) antibody,anti-Tr (delta notch-like epidermal growth factor-related receptor (DNER)) antibody,anti-zinc finger protein 4 (ZIC4) antibody,anti-inositol 1,4,5-trisphosphate receptor 1 (ITPR1) antibody,anti-homer protein homolog 3 (Homer 3) antibody,anti-neurochondrin (NCDN) antibody,anti-carbonic anhydrase-related protein (CARP) antibody and anti-Purkinje cell antibody 2 (PCA2) antibody.Results Eight out of 400 (2%) ataxia patients were positive for this AC panel tests,of whom two were positive for anti-GAD65 antibody,two for anti-Tr antibody,one for anti-PCA2 antibody,one for anti-Homer 3 antibody and two were positive for serum anti-NCDN antibody.Autoantibodies against ZIC4,ITPR1 and CARP were not detected in this cohort.Two of the eight ataxia patients also presented with limbic encephalitis,and only one anti-GAD antibody patient was screened with underlying small cell lung carcinoma (SCLC).All the eight patients received immunotherapy and four experienced partial response.Conclusions Autoimmune cerebellitis is the cause of acquired cerebellar ataxia.Tests of autoantibodies associated with AC have diagnostic value for paraneoplastic and non-paraneoplastic cerebellar ataxia.Immunotherapy may yield partial response in patients with AC.
7.Research progress in calcium phosphate-based biomacromolecules antigen delivery system

Nanometer materials have attracted much attention in the nanomedical field due to their unique physicochemical properties. Calcium phosphate nanoparticles have many advantages, such as non-toxicity, unique biomolecular protection, good biodegradability, and good biocompatibility. It has been widely studied as an ideal delivery system for biomacromolecules. In this review paper, the synthesis methods of calcium phosphate nanoparticles were summarized, including the conventional hydrothermal/solvothermal method and the aqueous wet chemical precipitation method. Moreover, the ways of binding calcium phosphate nanoparticles to biomacromolecule antigens were discussed from the perspectives of covalent and non-covalent binding, and their application in biomacromolecule delivery systems was described. The above review comments have important reference value for understanding the potential and application prospects of calcium phosphate nanoparticles in nanomedicine.
