1.Expression and significance of P-JNK and mutlidrug resistance protein in gastric cancer

Objective To investigate the clinicopathogenical significance of phosphorylation type of c-jun N-terminal kinase(p-JNK)and muhidmg resistance protein P-glycopretein(P-gP),multidrug resistance proteinl(MRP1)and lung resistance protein(LRP)in gastric cancer. Methods The expression of p-JNK,P-gP,MRP1 and LRP was detected in a tissue microarray containing 168 spots of gastric cancer tissue and 27 spots of normal gastric tissue by immunohistochemistry.Results The positive expression rate of p-JNK,P-gp,MRP1 and LRP in gastric cancer wag 45.8%,51.8%,45.8%and 55.4%respectively,which Was significantly higher than that in normal gastric tissue(P<0.05).The p-JNK expression correlated with depth of invasion(P<0.01),histology grade(P<0.05),vessel invasion (P<0.01),lymph node metastasis and distant metastasis(P<0.01).The expression of p-JNK,P-gp and MRP1 in a positive relationship(P<0.05).Kaplan-Meier analysis revealed a significant impact on survival by p-JNK,P-gp and MRPl(P<0.05). Conclusion The p-JNK expression in gastric cancer is correlated with malignant biological behavior and may be involved in the chemotherapeutic resistance by upregulating the expression of P-g[ and MRP1.
2.Research of functional segments of chicken B-FA molecule binding with Ii chain

Objective: To research the functional segments of B-FA molecule binding invariant chain and their characters. Methods:The DNA segments (α1α2, sα1α2 and α3TC ) of B-FA genes were respectively cloned and inserted into prokaryotic or eukaryotic expression plasmids,then they were singly or co-transfected with Ii gene into the engineering bacteria E. coli (BL-21)or 293T cells. After induction of expression,affinity chromatography and SDS-PAGE identification,the binding between B-FA segments and Ii molecule and co-localization in cells were observed with Pull-down and Western blot. Results:First three recombinant prokaryotic expression plasmids and four recombinant eukaryotic expression plasmids were constructed. The single molecules expressed by B-FA segments were observed after an affinity chromatography. Secondly the complexes of Ii/B-FA-α1α2 and Ii/B-FA-sα1α2 were detected by a Pull-down from the co-transfected corresponding prokaryotic expression plasmids,but no complex of Ii andα3TC,also in the western blot it was detected that B-FA-α1α2 or B-FA-sα1α2 as functional segment could bind Ii to form complex. Finally in eukaryotic expression 293T cells B-FA-sα1α2 kept localization, the same as B-FA. Conclusion: Chicken B-FA-α1α2 is function segment to bind with Ii molecule and keeps the location characters same as B-FA. The results of this research first time provide experimental evidence about B-FA functional region binding segment to Ii molecule.
3.Development of an assessment tool for nursing professional values

Objective The purpose of the study was to develop an instrument to measure nursing professional values held by Chinese nurses.Methods 381 nurses were enrolled for the survey on their professional values by the draft of nursing professional values assessment tool(NPVAT).With the item analysis,such as dispersion degree,correlation efficient,discriminability,alpha coefficients index and exploratory factor analysis,the items were selected.Results As a result of the item analysis,the final version of NPVAT was composed of 20 items selected from a total of 32 items.Exploratory factor analysis resulted in a four-factor solution explaining 51.99% of the common variance.Conculsions The NPVAT developed in this research can be used for measuring the nursing professional values among nurses.
4.Synthesis and antitumor activity of ent-kaurene diterpenoids

Aim: To synthesize the ent-kaurene diterpenoid and its derivatives from natural available stevioside.Methods: The crucial pharmacophore of exo-methylene ketone was designed and the carboxyl group at C-4 was modified.The cytotoxic inhibition activities in vitro of target compounds were evaluated against human cancer cells BEL-7402,HO-8910,MCF-7 and HL-60 by a MTT method.Results and Conclusion: The struc-tures of new target compounds were identified by ~1H NMR,FT-IR and EI-MS.The preliminary experimental re-sults showed that some derivatives of ent-kaurene diterpenoid possessed fair inhibitory activity against tumor cell lines compared with that of their parent steviol.
5.EFFECTS OF SOYBEAN PHOSPHOLIPIDS ON LEARNING AND MEMORY ABILITIES AND FATTY ACIDS CONTENTS IN HIPPOCAMPUS OF MICE

Objective:To study the effects of soybean phospholipids on learning and memory abilities and contents of fatty acids in hippocampus of mice and its mechanism. Method:Mice were randomly divided into four groups:low dose(2.5g/kg) ,middle dose(5.0g/kg) and high dose(10.0g/kg) groups of soybean phospholipids and control group. The treatment groups were fed with soybean phospholipids every day while the control group with distilled water for 30 d.After learning and memory abilities were measured,the mice were killed and contents of fatty acids in hippocampus of mice were determined by high performance liquid chromatography(HPLC) . Results:(1) The learning and memory abilities of mice in middle and high dose groups were significantly improved,but only memory abilities in low dose groups were stronger than control group.(2) The contents of protein in hippocampus in middle and high dose groups were increased extremely.(3) The contents of oleic acid(C18:1) and docosahexaenoic acid(DHA,C22:6 ) were obviously enhanced in hippocampus of high dose group;but decreased in eicosapentaenoic acid(EPA,C20:5) and linoleic acid(C18:2) . The content of DHA was increased and EPA decreased significantly in middle dose group.The increase of arachidonic acid(C20:4) was not significant.Conclusion:Soybean phospholipids may improve learning and memory abilities,which might contribute to change of fatty acids contents in hippocampus of mice.
6.Clinical application of transcatheter closure of secundum atrial septum defect with Amplatzer occluder in patients over 50 years of age

Objective To elucidate the efficacy and safety of transcatheter closure of secondary atrial septal defect (ASD) with Amplatzer septal occluder (ASO) in people over 50 years of age. Methods Retrospective comparison of 41 patients (14 males, 27 females) with ASD over 50 years of age ranging from 50 to 75(mean age 55.5?7.6, elder group) was conducted with 132 patients younger than 50 years(control group). After diagnosis of ASDs and evaluation of pulmonary artery systolic pressure and right atrial and ventricular dimensions by transthoracic echocardiography (TTE), all patients underwent transesophageal echocardiography (TEE) for complete assessment of ASD size, margins and anatomic relationship of the defect before closure of ASD. Each case was treated with ASO through the percutaneous transcatheter procedure under fluoroscopy and TTE or TEE. Early follow up(3 months) by echo was taken after the intervention. Results There was no difference of mean defect diameter measured by TEE and the balloon-stretched defect diameter of the ASDs between 2 groups. Objective To study the effect of health education on compliance of patients undergoing anticoagulant therapy after cardiac valve replacement? Methods One hundred and six patients undergoing anticoagulant therapy after cardiac valve replacement received enhanced health education after cardiac valve replacement? A self-designed questionnaire was used to investigate their treatment compliance and mastery of related knowledge pre-and postoperatively? Result The mastery of the knowledge related to anticoagulant therapy and the compliance were significantly improved compared to pre-health education(bothP < 0?05)? Conclusion The health education may improve the recognition of anti-coagulant therapy as well as compliance of patients?
8.CT and MRI findings and classification study of brain schistosomiasis granuloma

Objective To study CT and MRI features and classification of brain schistosomiasis granuloma. Methods CT and MRI data of 30 cases of brain schistosomiasis granuloma were reviewed.All cases were proved by the surgery or pathological examination and clinical laboratory test.There were 20 males and 10 females, and their age ranged from 5 to 58 years, mean 29.2 years.Plain and enhanced CT were performed in all patients with GE MAX 640 scanner.Ten patients were examined by plain and enhanced MRI with GE Signa profile 0.2 Tesla open scanner.Results The lesions located in supratentorial region in 27 cases and in infratentorial region in 3cases.The nodules were isodense or slight hyperdense on CT plain scan, iso or hypointense on T_1WI, hyperintense on T_2WI, Slight hyperintense on FLAIR.After the contrast material was injected intravenously, CT and MRI findings were multiple or single enhanced nodules at the cortical or subcortical area.There were four types of imaging features: (1) multiple small nodules in 5 cases (presenting as bright stars in the dark sky); (2) single large nodule in 8 cases; (3) mixed nodules in 14 cases; (4) circle-enhanced nodules in 3 cases.Conclusion The brain schistosomiasis granuloma has typical CT and MRI findings.CT and MRI classification is not only helpful to its diagnosis and differential diagnosis, but also might be useful for the choice of clinical treatment.
9.The effect of iterative times on the segment weight optimization in IMRT plan of postoperative cervical cancer

Objective To explore the use of Swedish medical company CMS 4.6.4 radiotherapy XIO system for the design process of cervical cancer postoperative IMRT plans in different iterative times of the same case,and to observe the Segment Weight Optimization(SWO)effect generated after the plan results.Methods Ten cases of cervical cancer patients were chosen ,the use of XIO 4.6.4 CMS treatment system was performed for the development of the intensity modulated radiation therapy .In the SWO process , iterative times were used to generate different plans ,without affecting the clinical dose target area requirements , and compared the organs at risk( OAR) by determining whether there were differences between dose and the number of segments ,monitor units (MU).Results For the same patients,different iterative times within a certain range of SWO IMRT plan were selected to compare the different iterative times plan ,femoral head,rectum and the bladder dose did not change the basic plan.The total number of segments and MU did not change significantly (P>0.05).Conclusion IM-RT plans in the use of CMS XIO 4.6.4 design of cervical cancer after operation in the process of using the itera-tive times below 100 times for SWO is the most suitable method for ensuring the organ dosimetry in radiotherapy . The total number of segments and the MU reduces the radiotherapy plan time so as to improve work efficiency .
10.Effects of Huang-Lian-Jie-Du-Decotion containing serum on expressions of autophagy related gene in macrophages

Objective:To investigate the effects of Huang-Lian-Jie-Du-Decotion containing serum for the autophagy of RAM264.7 macrophages ,and to explore its possible anti-atherosclerosis mechanism .Methods:The SD rats were intervened with large , middle and small dose Huang-Lian-Jie-Du-Decotion to prepare containing drug serum .Cell proliferation was tested by MTT assay .The gene and protein expressions of Beclin 1, mTOR was assessed by fluorescent quantitative PCR and Western blot method .Results:Compared with the normal serum group ,Huang-Lian-Jie-Du-Decotion containing serum enhanced expression of the mRNA and protein level of Beclin1,and decreased the expression of mTOR and p-mTOR(P<0.05 or P<0.01).Conclusion:Huang-Lian-Jie-Du-Decotion containing serum could regulate the expressions of Beclin 1 and mTOR.These might be one of the mechanisms for preventing atheroscle-rosis.
