1.Aplastic anemia and clonal evolution:germline and somatic genetics

Clonal progression to myelodysplastic syndromes (MDS) or acute myeloid leukemia (AML) remains a dreaded complication for a subset of patients with bone marrow failure (BMF). Recognizing risk factors for the development of MDS or AML would inform individualized treatment decisions and identify patients who may benefit from early or upfront hematopoietic stem cell transplantation. Now that next-generation DNA sequencing is available in the clinical laboratory, research has focused on the implications of germline and somatic mutations for diagnosing and monitoring patients with BMF. Most germline genetic BMF disorders are characterized by a high propensity to develop MDS or AML. Recent studies of somatic variants in marrow failure revealed a high frequency of clonal hematopoiesis with the acquisition of mutations in genes associated with MDS or AML. The evaluation and implications of germline and somatic mutations for the development of clonal disorders in patients with BMF and challenges of clinical genetic testing will be summarized in this paper based on the reports from the 58th American Society of Hematology (ASH) Annual Meeting.
2.Grape seed procyanidin extract arrests cell cycle of human bladder cancer cell line BIU87

Objective To study the effect of grape seed procyanidin extract(GSPE) on the cell cycle arrest in human bladder cancer BIU87 cells and investigate the underlying molecular mechanism.Methods BIU87 cells were treated with different concentrations of GSPE(50,100 and 200 ?g/ml) and cultured for 24 h.The untreated group served as control.Flow cytometry was used to evaluate the cell cycle,and RT-PCR and Western blotting were used to detect the mRNA and protein expressions of CyclinD1 and cyclin-dependent kinase 4(CDK4).Results GSPE inhibited the cell growth through cell cycle arrest at G1 phrase in a dose-dependent manner.Semi-quantitated RT-PCR and Western blot analyses indicated that GSPE decreased CyclinD1 and CDK4 expressions in a dose-dependent fashion(P
3.THE DYNAMIC CHANGE IN ICE mRNA EXPRESSION AND ITS EFFECT ON IFN-? IN THERMAL INJURED RATS

To investigate the dynamic changes in ICE mRNA expression and its relationship with IL 18 and IFN ? mRNA expression in thermal injured rats.The animals were subjected to a 30% total body surface area full thickness thermal injury,and fluid resuscitation was performed at 6 hours post burn. 54 male Wistar rats were randomly divided into three groups: normal controls,30% Ⅲ degree burn group,and selective decontamination of the digestive tract (SDD) treatment group. P1asma endotoxin was measured by limulus amebocyte lysate test,and tissue ICE,IL 18 and IFN ? mRNA expression was determined by reverse transcription po1ymerase chain reaction (RT PCR).The results showed that ICE, IL 18, and IFN ? mRNA expression significantly increased in the intestine,1ung,liver,and kidney at 2 hours,peaking at 8 hours,and a high level was maintained till 24 hours. However, treatment with SDD could lower plasma endotoxin levels,and markedly inhibit ICE, IL 18, and IFN ? mRNA expression in various organs.This study suggests that major burns can lead to a marked elevation of tissue ICE mRNA expression in vital organs,which might be associated with IL 18 and IFN ?mRNA expression induced by gut derived endotoxemia.

5.The CorreIation of Serum BiIirubin and Hearing Loss of Neonate with Hypercholesterolemia

Objective To study the relationship between hearing loss and serum bilirubin of neonates with hypercholesterolemia .Methods The serum bilirubin level and auditory brainstem response thresholds of 70 hyper_cholesterolemia neonates of full term pregnancy and normal weight were tested ,and the correlations of serum biliru_bin ,and occurrence time of jaundice with hearing threshold were analyzed .ResuIts Among the 70 neonates ,there were 17 cases with normal ABR thresholds and 53 with abnormal ABR thresholds ,including 27 with middle and se_rious hearing loss .In the cases of serum bilirubin less than 342μmol/L ,the percentage of cases with middle and se_rious hearing loss and with occurrence time of jaundice less than 48 hours was higher than theses cases with occur_rence time of jaundice more than 48 hours (P<0 .05) .In the cases of serum bilirubin higher than 342μmol/L ,there was no obvious difference between the percentage of cases of middle and serious hearing loss with occurrence time of jaundice less than 48 hours and those with occurrence time of jaundice more than 48 hours .Among the 53 cases with abnormal hearing thresholds ,positive correlation was observed between serum bilirubin and ABR threshold (r=0 .041) ,whereas negative correlation was observed between occurrence time of jaundice and ABR threshold (r=-0 .291) .ConcIusion The higher probability of hearing loss for hypercholesterolemia neonates with earlier occur_rence of jaundice ,the more serious hearing loss with higher serum bilirubin .
6.Generation and application of three-dimensional cell traction force

BACKGROUND: Traction force generated by adherent cells exerts on the extracellular matrix or cell substrate through the interaction between actin and myosin, which involves a variety of physiological and pathological processes.OBJECTIVE: To review the fundamental principles and application of several traction force techniques.METHODS: The first author retrieved the databases of CNKI, WanFang, PubMed and SpringerLink for the research addressing the measurements of traction force using the keywords of cell traction force, biomechanics in Chinese and English, respectively. Repetitive articles were excluded, and 30 literatures were enrolled for analysis, including 1 Chinese and 29 English articles.RESULTS AND CONCLUSION: There are three kinds of measurements of traction force, such as silica gel membrane, traction force microscopy, and micromachined levers. The precise measurement of traction force contributes to further understand the cell-cell and cell-microenvironment mechanical interactions.
7.Determination of berberine hydrochloride in Fuyanping capsules by HPLC

Objective:To establish the quality standard of berberine hydrochloride in Fuyanping capsules by HPLC. Methods:HPLC with Phenomenex C18(250?4.6mm,5?m)column was used.Acetonitrile-0.033mol/l potassium dihydrogen phosphate(30:70)was used as a mobile phase,flow rate was 1.0ml/min and detection wavelength was set at 346nm.Results: Berberine hydrochloride showed a good linear relationship at a range of 0.16-0.83?g,r=0.9999.The average recovery was 99.09%,and RSD was 0.20%.Conclusion:This method is simple,fast and accurate for the quality control of Fuyanping capsules.
8.Pulsed radiofrequency and radiofrequency thermocoagulation in the treatment of idiopathic infraorbital nenralgia

Objective To study the effects of pulsed radiofrequency(PRF)and radiofrequency thermocoagulation(RFTC)in the treatment of idiopathic infraorbital neuralgia.Methods 16 patients suffering idiopathic infraorbital neuralgia were randomly divided into PRF(n=8)and RFTC(n=8).Numeral rating scale(NRS),pain relief(PR)and effective rate of treatment was performed before operation and at 1 hour、1 st day、7th day、15th day、1 st month、3rd month and 6th month after operation.Results The preoperative and postoperative impedance of thefirst,second and third operation in RFTC group were(490±86)Ω and(424±82)Ω,(492±90)Ω and(432±84)Ω,(481±98)Ω and(409±85)Ω respectively and the postoperative impedance was significantly decreased(P<0.05).The preoperative NRS in group PRF and RFTC were 9(7~10)and 8.5(7~10)respectively and the postoperative NRS at all points in two groups were significantly decreased as compared with the preoperative NRS(P<0.05).The PR at 7 and 15d in group PRF were 3(0~4)and that in group RFFC were 4(3~4)(P<0.05).There were no significant differences in effective rate of treatment at each time between two groups(P>0.05).Numbness appeared after operation in group PRF and disappeared in a week,however,that was persistent and all sense disappeared in group RFTC.Conclusion Curative effect of PRF for idiopathic infraorbital neuralgia is as well as RFTC and it is a kind of safe and effective technology that causes less side effects.
9.Analysis of Irrational Prescriptions and Pharmacists’Intervention in the Pharmacy Intravenous Admixture Service of Our Hospital

OBJECTIVE:To provide some references for rational application of intravenous infusion drugs to ensure the safety of drug use. METHODS:Statistical analysis was made to irrational prescriptions intercepted by the pharmacy intravenous admixture service of our hospital from 2009 to 2013. RESULTS:There were 5,802 groups of irrational prescriptions from 2009 to 2013,accounting for 0.12% of total prescriptions. For the irrationality,the top three reasons were improper solvent dosage, drug overdose and improper solvent selection in order,and the involved types of drugs were mainly antimicrobial agents,anti-neoplastic drugs,common drugs and parenteral nutrition drugs (arranged from more to less). Under the pharmacist’s interven-tion,the percentage of irrational prescriptions of intravenous drugs reduced from 0.169 4% in 2009 to 0.082 8% in 2013. The decrease in the irrational prescriptions involving with improper administration was the most significant,followed by those related to drug overdose,frequency,prescribing errors errors improper solvent dosage,incompatibility and improper solvent. CONCLU-SIONS:Pharmacists’intervention can effectively reduce the irrational prescriptions and significantly promote rational use of in-travenous drugs.
10.Significance of S-100 and CD83+DC Expression in Laryngeal Mucosa Lesions

Objective To investigate the infiltration of dendritic cell (DC) in Vocal cord polyp, laryngeal leukopla-kia and glottic squamous cell carcinoma,and to observe laryngeal mucosal lesions of the state of the immune microenviron-ment, and to research significance of DC on the development of glottic squamous cell carcinoma. Methods The infiltration of S-100+and CD83+DC in 20 cases of Vocal cord polyp, 47 cases of laryngeal leukoplakia, 45 cases of glottic squamous cell carcinoma tumor and 20 cases of laryngeal normal mucosa were examined using immunohistochemistry. Results The num-ber of S-100+and CD83+DC were significantly higher in glottic squamous cell carcinoma, laryngeal leukoplakia and vocal cord polyp than that in laryngeal normal mucosa (P<0.05). The number of S-100+and CD83+DC were higher in laryngeal leukoplakia than that in glottic squamous cell carcinoma (P<0.05). The number of S-100+and CD83+DC in mild dysplasia and moderate dysplasia were less than that in severe atypical hyperplasia (P<0.01). In glottic squamous cell carcinoma, S-100+ and CD83+DC with poor-differentiated was significantly less than that with well-differentiated (P < 0.01). Conclu-sion Changes in the number of dendritic cell was found in vocal cord polyp, laryngeal leukoplakia and glottic squamous cell carcinoma, which indicated that there were an abnormal immune status. Changing of dendritic cell in laryngeal mucosa plays an important role in laryngeal cancer development.
