Objective To explore the impact of four different anesthetic drugs commonly used in animal experi-ments on cardiovascular system in rats.Methods Electrocardiogram ( ECG) and blood pressure were dynamically recor-ded by a BioPac MP150 system after anesthesia.In addition, the blood glucose at different time points and hepatic func-tion, kidney function, cardiac enzymes and electrolytes at the end of the test were collected.Rusults Chloral hydrate caused severe ventricular arrhythmia.Isoflurane had inhibitory effect on the heart rate.Pentobarbital sodium induced a in-crease of ECG P wave.Urethane caused J point elevation of ECG.Blood pressure in the urethane-and pentobarbital sodi-um-treated groups were increased.Chloral hydrate caused CK to be raised, while isoflurane showed the opposite effect on CK and CKMB.Alanine aminotransferase and aspartate aminotransferase in the pentobarbitol sodium and isoflurane groups were decreased.Creatinine in the chloral hydrate, pentobarbital sodium and isoflurane groups were lower, and the serum sodium and potassium were decreased in the four groups.Conclusions Chloral hydrate has obvious effect on the cardio-vascular system, and is not suitable for animal studies on cardiovascular diseases.Pentobarbital sodium, urethane, isoflu-rane can be chosen for animal studies on cardiovascular diseases.
3.Effects of different serum microenvironments on culture of rat bone marrow mesenchymal stem cells in vitro

BACKGROUND:During culture of bone marrow mesenchymal stem cells(BMSCs),a certain serum is commonly added in the basic medium,such as calf serum and fetal bovine serum,but there are potential biological safety risks.OBJECTIVE:To study the effects of different serum microenvironments on in vitro culture of rat BMSCs.METHODS:BMSCs were harvested from adult rat bone marrow,and cultured in vitro by whole bone marrow adherence method.The cells were cultured under the following serum microenvironment.The primary cells of autoserum group were cultured with autoserum,changing the medium with fetal bovine serum after passage.The primary calls of homogeneity foreign serum group were cultured with homogeneity foreign serum,changing the medium with fetal bovine serum after passage.The primary cells of fetal bovine serum group were cultured with fetal bovine serum,and cultured with fetal bovine serum after passage.The primary cells of Dulbecco's modified Eagle's medium(DMEM)group were cultured with serum-free DMEM,changing the medium with fetal bovine serum after passage.The morphologic changes in BMSCs were detected under an inverted phase contrast microscope.Attachment rate and growth curve were measured.Surface marker CD11b,CD45 and CD90 expression was analyzed by flow cytometry.RESULTS AND CONCLUSION:In autoserum and homogeneity foreign serum groups,the homogenicity and degrees of fusion of call morphology were improved in comparison with other two groups and the day of first passage was less than other groups.The attachment rate was greater in the autoserum,homogeneity foreign serum and fetal bovine serum groups than the DMEM group at 24,48,72 hours(P<0.01).Doubling rate was fastest in the growth curve of autoserum group,followed by homogeneity foreign serum group and fetal bovine serum group.However,no doubling phenomenon was found in the DMEM group.Flow cytometry results demonstrated that the rates of CD11b-positive and CD45-positive cells at passage 3 were above 98% under medium containing serum,and CD90-positive rate was less than 2%.We could obtain BMSCs of higher purity.However,CD11b-positive rate was 95.83%,CD90-positive rate was 2.07%,but CD45 positive rate was only 64.79% under serum-free microenvironment.BMSC purity was significantly lower under serum-free microenvironment than under serum microenvironment.Results indicated that the microenvironment of rat autoserum can improve the attachment rate,growth and purification of BMSCs.
4.Effect of the Recipe of Effective Components from Yangxin Tongmai Formula on the Angiogenesis of Chick Chorioallantoic Membrane

Objective To investigate the therapeutic mechanism of the recipe of effective components from Yangxin Tongmai Formula (YTF) for myocardial ischemia by observing its effect on angiogenesis. Methods The breeding egges were divided into 6 groups: YT Ⅱ group treated with serum containing the recipe of effective components from YTF, YT Ⅰ group treated with serum containing YTF, SBW group treated with serum containing Shexiang Baoxin Pill, bFGF group treated with serum containing recombinant bovine basic fibroblast growth factor, KX group treated with blank control serum, and blank control group, 25 eggs in each group. The survival rate of chicken embryo, growth shape and vessel count of CAM model in each group were observed after 0, 24, 48, 72 and 96 hours. Results After treatment for 96 hours, the survival rate in b-FGF group and YT II group was higher than that in the other groups. The vasculature growth was rapidly in the medication group after culturing for 72 hours, and more apparent after 96 hours, especially in the b-FGF group and YT-Ⅱ group. The vessel count in b-FGF group, YT-Ⅱ group, YT-Ⅰ group and SBW group was in-creased as compared with that in KX group and blank control group (P < 0. 05), and ranged in the order as follows: b-FGF group > YT-Ⅱ group > YT-Ⅰ group > SBW group > KX group > blank control group. Conclusion YT-Ⅱ has the sim-ilar effect on improving angiogenesis as YT-Ⅰ, SBW and b-FGF, and its effect is better than that of YT-Ⅰ and SBW.
5.Analysis on Plasma Metabolicomics Pathway in Rats with Heart Blood Stasis Syndrome

This study was aimed to analyze the plasma metabolicomics pathway in rats with heart blood stasis syn-drome. KEGG database was used in the signal pathway analysis. HMDB was used in the analysis of molecular metabolite annotation, enzyme or transporter associated and its related properties. The metPA network software was used in the visualization of metabolite path. The results showed that 9 metabolites involved in 15 metabolic pathways. Among them, the P-value of metabolic pathway of pantothenate and CoA biosynthesis, propanoate metabolism, biosynthesis of unsaturated fatty acids was less than 0.05. It was concluded that the metabolic pathways of pan-tothenate and CoA biosynthesis, propanoate metabolism, biosynthesis of unsaturated fatty acids were involved with the pathological process of rats with heart blood stasis syndrome.
6.Bioinformatics Analysis on Proteomics of Rat Bone Marrow Mesenchymal Stem Cells Intervened by Active Principle Region of Yang-Xin Tong-Mai Formula

This study was aimed to analyze the bioinformatics of proteomics of rat bone marrow mesenchymal stem cells (MSCs) intervened by active principle region of Yang-Xin Tong-Mai Formula (apr-YTF). The latest versions of bioinformatics tools including DAVID (http://david.abcc.ncifcrf.gov/) and GO (http://www.geneontology.org/) were combined to assign a precise function to rat bone marrow MSCs intervened by apr-YTF. KEGG and VISANT were assigned with a precise function to rat bone marrow MSCs intervened by apr-YTF. The results showed that a total of 102 biological processes were mainly involved, with 35 cellular components and 6 molecular functions. These proteins interacted in 3 signal transduction pathways. It was concluded that the following proteins and signal transduction pathways played an important role in the process of apr-YTF inducing BMSCs differentiation into cardiomyocytes. Presenilin-1 and Presenilin-2 were in the Notch signaling pathway. And syntaxin-4 protein was in soluble N-ethylmaleimide sensitive fusion protein attachment protein (SNARE). The apr-YTF played a role on MSCs from multiple sites, with multiple links through different biological processes. The bioinformatics of proteomics can predict action mechanism of traditional Chinese medicine (TCM) from the holism concept. The validation in combination with molecularbiology was a good way for TCM modernization.
7.Functional MRI analysis of deception among people with antisocial personality disorders.

OBJECTIVE:
To investigate the functional magnetic resonance imaging (fMRI) data of deception in antisocial personality disorders (ASPD).
METHODS:
A total of 32 criminals meeting the criteria for ASPD underwent fMRI at 1.5T while responding truthfully questions or lying. We compared the brain activities between truth-telling and lie-telling, and then computed the correlation coefficient between the contrast brain activities and the inclination to deception.
RESULTS:
The left anterior cingulate gyrus, the bilateral dorsolateral prefrontal cortex, and left inferior parietal lobule were associated with the executive aspects of deception among people with ASPD. But with the greater inclination to deception, the blood oxygen level dependent (BOLD) activities in those regions decreased.
CONCLUSION
Evaluations of truthful and untruthful communications pertaining to ASPD subjects may be differentiated in terms of brain BOLD activities, though those activities may decrease in habitual liars, which remains a challenge to the diagnostic accuracy in lie detection.

8.Platelet Metabolomics Analysis in Rats of Coronary Heart Disease with Blood Stasis Syndrome by Overexpression of Fibrinogen

ObjectiveTo analyze the metabolomic characteristics of platelets in fibrinogen(FIB) overexpression rats of coronary heart disease with blood stasis syndrome(CHD-BSS), explore potential biomarkers, and investigate the mechanism of FIB overexpression on CHD-BSS. MethodsSD rats were randomly divided into BSS group and BSS+FIB overexpression group(BSS+FIB group), with 10 rats in each group. Both the BSS+FIB group and the BSS group were fed a high-fat diet combined with oral administration of vitamin D3 and subcutaneous injection of isoproterenol, but rats in the BSS+FIB group were overexpressed with FIB during the initial modeling stage by transfection with adeno-associated virus(AAV). The overexpression level of FIB in rat liver and plasma samples was detected by enzyme-linked immunosorbent assay(ELISA) and real-time fluorescence quantitative polymerase chain reaction(Real time PCR), as well as the expression level of liver FIB A(FGA) mRNA. The characteristics of metabolites in rat platelet samples were analyzed by ultra-high performance liquid chromatography-quadrupole-electrostatic field orbital trap high-resolution mass spectrometry(UPLC-Q-Exactive Orbitrap-MS), and the differential metabolites between groups were screened by principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA), and the enriched pathways were analyzed. The accuracy of potential biomarkers in the diagnosis of CHD-BSS was evaluated by receiver operating characteristic(ROC) curve. The expression of autophagy related proteins phosphorylated adenosine monophosphate(AMP) activated protein kinase(p-AMPK)/AMPK, phosphorylated mammalian target of rapamycin(p-mTOR)/mTOR, microtubule-associated protein 1 light chain 3(LC3) Ⅱ/Ⅰ and p62 in platelets were detected by Western blot. ResultsCompared with the BSS group, the expression levels of FIB in liver and plasma samples of the BSS+FIB group were significantly increased(P<0.05, P<0.01), and the expression level of FIB mRNA in the liver was remarkably increased(P<0.01), indicating successful overexpression of FIB. Platelet metabolomics results showed significant differences in metabolic profiles between the BSS+FIB group and the BSS group, and a total of 25 significantly enriched metabolic pathways and 8 metabolites involved in these metabolic pathways, among which uric acid, guanosine and ribose 1-phosphate levels were up-regulated, while adenosine diphosphate(ADP), AMP, guanosine diphosphate(GDP), adenylosuccinate and norepinephrine levels were down-regulated. The diagnostic ability analysis of differential metabolites showed that all 8 differential metabolites had good diagnostic ability, with an area under the curve(AUC)>0.85. Western blot results showed that compared with the BSS group, the expression levels of p-mTOR/mTOR and p62 proteins in platelets of the BSS+FIB group was significantly reduced(P<0.01), while the expression levels of p-AMPK/AMPK and LC3Ⅱ/Ⅰ proteins were increased, but the difference was not statistically significant. ConclusionOverexpression of FIB can change the metabolic characteristics of CHD-BSS rat model, involving multiple aspects such as vascular endothelial injury, platelet activation and myocardial function damage. Among them, overexpression of FIB may enhance the occurrence of platelet autophagy, thereby inducing platelet activation and promoting thrombus formation.
9.Brain structure analysis for patients with antisocial personality disorder by MRI.

OBJECTIVE:
To investigate the structural abnormalities of brain in patients with antisocial personality disorder (ASPD) but without alcoholism and drug abuse.
METHODS:
Volunteers from Hunan Reformatory (n=36) and the matched healthy subjects (n=26) were examined by high-spatial resolution magnetic resonance imaging (MRI) and diffusion tensor imaging (DTI). Voxel-based morphometry and fractional anisotropy (FA) maps were generated for each subject to reveal structural abnormalities in patients with ASPD.
RESULTS:
Compared with the healthy controls, ASPD patients showed significantly higher gray matter volumes in the inferior parietal lobule (P≤0.001, uncorrected), white matter volumes in the precuneus (P≤0.001, uncorrected), FA in the left lingual gyrus, bilateral precuneus, right superior frontal gyrus and right middle temporal gyrus (P≤0.01, uncorrected).
CONCLUSION
Our results revealed the abnormal neuroanatomical features in ASPD patients, which might be related to the external behavioral traits in ASPD patients.

10.Strategies of Traditional Chinese Medicine Standardized Treatment Based on Syndrome Differentiation of Bipolar Disorder

Bipolar disorder (BD) is a serious chronic emotional disorder with a high suicide rate and a common psychiatric disease. Traditional Chinese medicine (TCM) treatment based on syndrome differentiation of BD has unique advantages and good safety, which is expected to become a breakthrough in the treatment. Based on Expert Consensus on TCM Syndrome Differentiation Criteria for Bipolar Disorder by Professor Yin Dongqing and Professor Jia Hongxiao, this study collated the treatment protocols of BD with various syndrome types according to Meta-analysis of the existing literature in the database and evaluated the evidence level according to the evidence evaluation standard issued by the US Agency for Healthcare Research and Quality (AHRQ). (1) Depression attack. ① Liver depression and spleen deficiency syndrome: Xiaoyaosan pills or Shugan Jieyu capsules, ② Phlegm dampness and spleen stagnation syndrome: Wendantang modified with Tianwang Buxindan, ③ Heart and spleen deficiency syndrome: Jiuwei Zhenxin Granules or DANG's Ganmai Dazhaotang, ④ Fire heat and internal depression syndrome: Danzhi Xiaoyaosan Granules or Chaihu Longgu Mulitang, ⑤ Liver and kidney deficiency syndrome: JIANG's Buganshen Decoction. (2) Mania episode. ① Heart and liver fire hyperactivity syndrome: Zhengan Ningshen Formula, ② Phlegm heat harassing spirit syndrome: Huatan Xiehuo Dingshen decoction, Lianzhi Tongqiao Anshen decoction, Qingshen Dingkuang decoction or Qingshen Xingnao decoction, ③ Liver and gallbladder dampness-heat syndrome: Longdan Xiegantang.
