1.Lung cancer and chronic obstructive pulmonary diseases
Journal of International Oncology 2014;41(6):439-443
Lung cancer and chronic obstructive pulmonary disease (COPD) are two high-mortality diseases in the world.Numerous epidemiological studies have demonstrated that presence of COPD increases the risk of lung cancer.Habitual cigarette smoking frequently develops lung cancer as well as COPD,However the links between the two diseases should be more than smoking alone.The underlying mechanisms may include genetic predisposition,inflammation and cell injury,oxidative and noxious stress,extracellular matrix and proteinases,some of which might represent the targets for chemoprevention or chemotherapy.

Thymic carcinoid is an uncommon component of Multiple Endocrine Neoplasia Type 1 (MEN1).The low penetrance and the high mortality made TH-NET one of the most fatal tumor components of MEN1.Here we are discussing the pathology and pathogenesis, as well as epidemiology, screening strategy, diagnosis, therapies, and analysis of prognostic factors of thymic carcinoid associated to MEN1.

Objective To investigate the effects of direct current electric fields on the orientation responses of vascular endothelial cells and the influence of the cytoskeleton filament actin on the orientation responses of cells in direct current electric fields. Methods Cultured vascular endothelial cells, with or without treatment with the actin inhibitors cytochalasin B or Y27632, were exposed to a direct current electric field of 200 mV/mm, and cell images were taken at 0, 4 and 8 hours during the exposure. Cells not exposed to the electric field were used as a control.Cell orientation was quantified using an image analyzer. Immunofluorescence staining of the cells for F-actin was observed through confocal microscopy. Results Cells in the control cultures oriented randomly with no predominant polarity. Cells exposed to the direct current electric field showed significant re-orientation to align their long axes perpondicular to the field vector. Neither cytochalasin B nor Y27632 reduced the re-orientation induced by the field, and earlier orientation response was observed in Y27632-treated cells. F-actin staining showed that the orientation of F-actin stress fibres was at random in control cells and perpendicular to the field vector in the field-exposed cells without any drug treatment. Although the formation of stress fibres was inhibited in the cytochalasin B-or Y27632-treated cells, the cells in the direct current electric fields kept their re-orientation responses, similar to the cells without any drug treatment. Conclusions A direct current electric field can induce vascular endothelial cells to re-orient, but the re-orientation response is independent of actin polymerization and actin stress fiber formation.

Objective To observe the expression of heparan sulfate proteoglycan(HSPG)-Agrin in rat cortex of postnatal time in order to understand the relation between the development of cortex and agrin. Methods We performed immunohistochemical analysis of agrin expression in the postnatal rat cortex with agrin-specific polyclonal antibody. Results Agrin expression was shown to have significant differences in the different periods of postnatal time.It increased from postnatal 2 day(p2) to postnatal day 6(p6),then declined steadily after p6 and reached adult levels by 4w.In this periold of time,agrin has different expression peak following to the cortex layer development.Conclusion:Agrin is related to the development of cortex and may play an important role in the developing of central nervous system.;

Objective:To investigate Th17,Treg and related factors before and after myocardial infarction ,and explore the cor-relations among them.Methods:Extracted mononuclear cell in peripheral blood ,and used fluorescence-activated cell sorting analysis to detect the contect of Th17 and Treg.Enzyme-linked immunosorbent assay was used to detect cytokine production of IL-17 and TGF-β;real-time PCR was used to detect the contect of RORγT and Foxp3.Results: Compared with the control group , experimental group (both before and after thrombolysis) had increased expressions of Th17,IL-17 and RORγT(P<0.05);the expressions of Treg,TGF-βand Foxp3 had decreased(P<0.05).Compared with the group of before thrombolysis ,the group of after thrombolysis 7 d had decreased expressions of Th17,IL-17 and RORγT(P<0.05);increased expressions of Treg,TGF-βand Foxp3(P<0.05).Conclusion:Compared with the control group,patients with myocardial infarction have the high levels of Th 17 and the related factors,the decreased levels of Treg and the related factors.Thrombolytic therapy can decrease the levels of Th 17 and the related factors,and increase levels of Treg and the related factors.

Objective To clarify the mechanisms of total saponins of panax ginseng (TSPG) in induction of apoptosis of K562 cells and to provide the theoretical basis and the experimental evidence for its clinical application. Methods The effects of TSPG on apoptosis of K562 cells were studied by morphometry, flow cytometry, morphological observation, and immunocytochemistry. Results The results indicated that TSPG could markedly inhibit the proliferation of K562 cells and induce the apoptosis of K562 cells. Our experiment also showed that after induction by TSPG, the ratio of positive K562 cells with Fas expression increased. Conclusion The mechanisms of TSPG in the induction of K562 cell apoptosis may be related to the increased expression of Fas in K562 cells.

AIM: To detect dystrophin expression in skeletal muscles of mdx mice after bone marrow transplantation (BMT), and to evaluate the effect of BMT on Duchenne muscular dystrophy (DMD). METHODS: Bone marrow cells were cultured for three days, and then transplanted into mdx mice irradiated lethally through tail veins. After 4 and 6 months, dystrophin expression on myocytes membranes in mdx mice was detected by fluorescent immunohistochemical staining. The centrally nucleated fibers (CNF) were calculated by HE staining, and the physiologic parameters measured and the motor function detected by traction test, rotating rods test and rotating wheels test were also observed. RESULTS: Until 4 and 6 months after BMT, dystrophin was expressed partly on myocytes membranes in mdx mice, and the ratio of CNF decreased, physiologic functions improved, the motor ability reinforced in treated group. CONCLUSION: After BMT, marrow stem cells settled in injured skeletal muscles and bone marrow, then differentiated into myocytes with dystrophin expression and caused the improvement of pathology, physiology and motor function in treated group finally. These results give a powerful proof for the treatment of DMD with BMT.

Objective To study the clinical efficacy of free anterolateral femoral skin flap on the repair of skin and subcutaneous soft tissue defects caused by excision of oversized malignant tumour in the skin. Methods A retrospective review was performed of free anterolateral femoral skin flap reconstructions for oversized malignant tumor cut on body surface since April 2007 to November 2010. There were 6 patients with head and limb squamous cell carcinoma because of bum scar and 3 patients with recurrence of dermatofibrosarcoma protuberans in wall of belly. The area of soft tissue defects ranged from 19 cm × 15 cm to 24 cm × 21 cm, and skin flaps was 20 cm × 16 cm to 25 cm × 22 cm in size. Three cases received radio therapy after operation. Results Nine flaps survived perfectly, one flap survived with partial necrosis and healed after changing dressings. No complications were observed in the donor site, including wound dehiscence, hernia and weakness. Follow-up survey of 12-24 months after the operation showed that the appearance and function in the repaired sites were normal, and norecurrence of the tumors. Conclusion Free transplantation of anterolateral femoral skin flap is relatively an ideal operative type for the repair of soft tissue defects caused by excision of oversized malignant tumour in the skin.
