1.The expression of stem cell pdx-1 in different purity pancreatic islets before and after transdifferentiated culture

Objective:To obtain the stem cells in different pancreatic islets purity utilizing free-serum culture medium and investigate the PDX-1 mRNA expression distinction before and after transdifferentiation culture.Methods:Pancreatic islets of male SD rats(islet donors)were isolated by injection of collagenase into pancreatic ducts,and maintained in serum Ham's F-10 for 36h.Then the cells were kept in free-serum Ham's F-10 for 14 days.The last stage was to shift culture media from free-serum Ham's F-10 to free-serum DMEM/F12 that contained KGF,BSA,glucose and ITS.On the 28d,Dithizon was added into the culture flask for observation of 3D cell.The PDX-1 positive cells was investigated by immunocytochemistry.Results:After transdifferentiation culture,some 3D cells were red.The pdx-1 was remarkable in cell nucleus.Conclusion:After transdifferentiation culture,some pancreatic stem cell display the character of islet cell.The daughter cells of pancreatic stem cells have character of stem cells after transdifferentiation.KGF may have effect on promoting the expression of PDX-1.
2.Investigation on candida infections in pregnant women and drug resistance analysis

Objective To investigate the vulvaginal candidiasis(VVC)incidence in pregnant women,identify the candida species and analyze the drug resistance of isolated strains.Methods Candida isolation,sample cultivation,species identification and drug susceptibility test were performed for VVC patients.Seven hundred and forty-five samples were collected.Results The incidence of VVC was 10.20%,and the recurrence of VVC was 18.5%.The proportion of C.albicans,C.glabrata,C.krusei,C.tropicalis and other candida were 80.26%,8.95%,3.95%,1.32% and 6.58% respectively.C.albicans was susceptible to amphotericin and 5-flucytosine,the susceptibility for fluconzazole,itroconazole and voriconazole were 7.89%,6.58% and 9.21% respectively.C.gla-brata,C.krusei,C.tropicalis,and other candida were susceptible to those antifungal drugs.Conclusion VVC was an usual infection among pregnant women,C.albicans was the major species,followed by C.glabrata.The resistance of amphotericin and 5-flucytosine were lower,the Candida were resistant to fluconzazole,itroconazole and voriconazole in certain extent.

6.Dose-effect of HIFU in tumor therapy

8.Pseudotemporal lobe epilepsy

Pseudotemporal lobe epilepsy refers to an electroencephalogram (EEG) ictal pattern that is localized to the temporal region and the clinical ictal symptoms like the temporal seizure, especially mesial temporal seizure. But the epileptogenic zone is on the extratemporal regions. It is not easy to diagnose pseudotemporal lobe epilepsy. There are difficulties to detect by scalp EEG, and stereoelectroencephalography is usually required for epileptogenetic zone localization. Pseudotemporal lobe epilepsy almost is refractory for antiepileptic drugs. But the situation is illustrated by the failure of temporal lobe surgery resection alone. Therefore, the good result is often obtained after an epileptogenic zone and symptomatic zone resection.
9.Effects of Compatibility of Astragalus and Arctium on Glycolipin Metabolism and Renal Lesion in Diabetic Rats

Objective To investigate the effects of combination of aniso-dosage of Astragalus and Arctium on the glycolipin metabolism and renal lesion in diabetic rats.Method Streptozotocin (STZ)-induced diabetic rats were established as experimental model.The diabetic rats were randomized into model group and mixture-treated group.Astragalus and Arctium were divided into three dosages (low,moderate,high) respectively and combined according to orthogonal experimental design and were administrated to the mixture-treated group.At the 4th and 8th week,parameters of blood,urine and renal tissue were examined.The blood sugar,cholesterol,triglycerol,HbA1c were detected by analysis meter of biochemistry,urine microalbumin by radioimmunoassay (RIA) and 24 h urine protein by colorimetry.The HE and PAS staining were used to observe the change of glomerular ECM.Results At the 4th weekend,the combination of Arctium high-dosage had better effects on decreasing blood sugar and cholesterol.While at the 8th weekend,the effects of mixture were gradually augment with the Astragalus dosage increasing on decreasing triglycerol,24 h urine protein and ECM.Conclusion The compatibility of aniso-dosage of Astragalus and Arctium could rectify the metabolic disturbance of glycometabolism and lipid metabolism,and lessen the renal lesions.The compatibility had different effects on various experiment phases.
10.Application of three-dimensional cell culture model in cancer research

Compared to the conventional two-dimensional (2D) culture,in vitro three-dimensional cell culture (3DCC) can mimic the microenvironment more exactly.Compared to in vivo animal models,3DCC is less time-consuming and can be real-time recorded.Thus,3DCC fills the gap between 2D culture and animal models,which is used for oncologic research quickly.In 3DCC,the characteristics of cancer cells,such as morphology,the capability of invasion and migration and the resistance to chemo-radiotherapy,are quite different from those in 2D culture.The development of 3DCC technology provides a platform for oncology,organization engineering and gene or drug screening.
