ObjectiveObserving the distribution and the change of the quantity of SP,VIP in the colon tissue of 'cathartic colon'rats.MethodsWe divided the 32 wistar rats weighted (200 ±10)g into 2 groops randomly:the blank group( n =8) and model group( n =8).we induced the 'cathartic colon'rats with the suspension of dahuang.The blank group were given the distilled water for contrast.After one week during the time Dahuang was stopped.We killed them,measured the carbon powder propulsion rates in the intestine and then took Specimen after the 8 rats were fasted 24 hours as well as the blank group,but water were given.We compared the variation of SP,VIP distribution between two groups.ResultsThe carbon powder propulsion rates in the model group were slower than that in the saline control group.The distribution of SP,VIP are all more than that in the blank group.ConclusionThe distribution of SP,VIP are all more than that in the blank group.

3.Hero642 nickel-titanium rotary instrument for single-visit root canal treatment

BACKGROUND:Several studies have shown that Hero642 is a rotary nickel-titanium file with high safe performance, and its ability of root canal shaping is good.
OBJECTIVE:To observe the clinical effect of single-visit root canal treatment using Hero642 nickel-titanium rotary instrument on teeth with chronic pulpitis and apical periodontitis.
METHODS:Total y 400 teeth with chronic pulpitis and apical periodontitis were operated by Hero642 nickel-titanium rotary instrument and nickel-titanium hand file, respectively. The efficiency of root canal preparation was taken. Pain reactions were recorded 1 week after operation and curative effects were evaluated 2 years after root canal treatment. Rates of pain and success were calculated at 1 week and 2 years after treatment, respectively.
RESULTS AND CONCLUSION:The average time for root canal preparation was (3.57±0.76) minutes in the Hero642 group and (7.62±0.25) minutes in the hand file group, and there was a significant difference between the two groups (P<0.01). After 1 week of treatment, the pain rates were 3.5%and 22.8%in the two groups, respectively, showing a significant difference (P<0.01). Two-year fol ow-up was performed, and ratios of success were 94.4%in the Hero642 group and 92.4%in the hand file group, but there was no difference between the two groups. Hero642 nickel-titanium rotary instrument for single-visit root canal treatment showed excellent efficiency on clinical treatment, reduced treatment period and had less postoperative reactions.
4.Research on Fingerprint of Radix Dipsaci by HPLC

Objective To establish chromatographic fingerprint of water and ethanol soluble component of Radix Dipsaci by using HPLC for identifying and evaluating its quality. Methods HPLC method was set up with Agilent HC-C18 column. The mobile phase was comprised of acetonitrile(A) and 0.05mol/L KH2PO4-H3PO4 buffer solution pH=3.0(B) in gradient elution mode. UV detection wavelength was at 220 nm,with a flow rate of 1.0 mL/min. Results Ten batches of Radix Dipsaci samples from different habitats and harvested in different times were analyzed. HPLC fingerprint of medicinal material of Radix Dipsaci was established preliminarily. Seventeen characteristic peaks were confirmed and the fingerprints of different samples were compared by similarity evaluation software. Conclusion This method shows high precision and good repeatability,so it can be used to assess the quality of Radix Dipsaci.
5.Chemical constituents of Fructus Aurantii and Fructus Aurantii Immaturus by HPLC-ESI-MS

Objective To analyze the chemical components of Fructus Aurantii (FA) and Fructus Aurantii Immaturus (FAI).Methods HPLC-ESI-MS with Surveg mass spectrometer was used in the study.Chromatographic column: Symmetry Shield TM RP_ 18 (150 mm?3.9 mm, 5 ?m) (Waters, Milford, MA, USA); mobile phase: (A) water (0.6% HAc, pH=2.5), (B) methanol. Gradient elutions: 20%- 40% B (0-48 min); 40% B (48-54 min); 40%-55% B (54-60 min); 55%-95% B (60-75 min); 95% B (75-85 min); 95%-20% B (85-90 min).Flow rate and wavelength were 0.7 mL/min and 283 nm at room temperature, respectively.Results Four kinds of flavonoids were identified as naringin, neohesperidin, naringenin, and hesperidin, synephrine was also identified in FA and FAI. Furthermore, the contents of them were determined individually.The results showed that the chemical constituents in FA and FAI were the same but the contents were different.Conclusion HPLC-ESI-MS method can be efficiently used to study FA and FAI.
6.Expression of lipocalin-type prostaglandin D synthase in glioma

Objective To investigate the expression level of lipocalin-type prostaglandin D synthase (L-PGDS) mRNA and its protein in glioma.Methods The L-PGDS mRNA was determined by real-time quantitative RT-PCR in 23 glioma and 5 healthy brain specimens. L-PGDS protein was detected by Western blot in cerebrospinal fluid (CSF) and brain tissue specimens.Results In 23 glioma specimens L-PGDS mRNA expression ranges from 1.5?10 3~7.0?10 4 copy/?g RNA, mean value is 1.6?10 4 copy/?g RNA. In 5 normal brain specimens, the mean value is 8.1?10 5 copy/?g RNA, statistic analysis indicates a distinct discrepancy ( P

Objective To compare the effect of silica-extraction method and Silico membrane based method in DNA purification from bones and teeth.Methods DNA samples were purified respectively with the silica-extraction method and MinElute PCR Purification kit from 6 bones and 8 teeth,then tested STR types by GlobalFiler? kits. And evaluated the two methods with the success rate and the peak height. Results Both of the two purification methods can successfully obtain the STR markers of the 14 samples. And there was no statistical difference between the two methods in the average peak height from bones and teeth. Conclusion The Silico membrane based method which have more advantages in operation is an efficient method to purify DNA from bones and teeth, and there is no significant difference compared with the silica-extraction method. But the cost is higher. It can be selectively used in forensic practice.
8.Nano-hydroxyapatite/polyamide 66 as a direct pulp capping agent has effects on histological changes of odontoblasts and microvessels

BACKGROUND:Previous studies have demonstrated that nano-hydroxyapatite/polyamide 66 (nHA/PA66) composites as a root canal filing material possess the basic physical and chemical properties, have antibacterial properties against the infection of medulary cavity, and have no cytotoxicity. However, it is rarely reported as a direct pulp capping agent.
OBJECTIVE: To observe the histological reaction of nHA/PA66 composites as the pulp capping agent.
METHODS:The first and second molars from 12 Wistar rats were selected to prepare class I cavities by exposing the dental pulp. Then, these rats were randomized into three groups and subjected to dental pulp capping using nHA/PA66 composite, nHA and calcium hydroxide, respectively, folowed by filing with glass ionomer cement. At 7 and 30 days after filing, histological changes of odontoblasts and microvessels in the dental pulp were observed using ink perfusion method.
RESULTS AND CONCLUSION: At 7 days after filing, significant changes in dental pulp fibroblast proliferation, predentin thickening, dilation of blood vessels were observed in the nHA/PA66 group; only dental pulp fibroblast proliferation with no predentin thickening and dilation of blood vessels was found in the nHA group; in the calcium hydroxide group, the pulp tissues in the pulp chamber were mostly damaged, and obvious hyperplasia in dental pulp fibroblasts and certain inflammatory cel infiltration with vascular necrosis were observed. At 30 days after filing, predentin thickness was increased further, dental pulp fibroblasts proliferated, the number of odontoblasts increased, and dilation of blood vessels was observed in the nHA/PA66 group; predentin thickening and dilation of blood vessels were obviously visible in the nHA group; pulp necrosis was found on the pulp surface below the pitting pore, odontoblasts grew actively at the pulp junction, with no obvious thickening of the predentin, and there were dense vessels below the necrotic area. These findings indicate that the nHA/PA66 composite can induce the regeneration of dental pulp cels.
9.Effect of SonicFill ultrasonic resin on rat odontoblasts and dental pulp vessels

BACKGROUND:The mechanical properties of SonicFil ultrasonic resin are better than those of the traditional light-cured composite resin, but its effect on odontoblasts and dental pulp vessels is unclear.
OBJECTIVE:To study the effect of SonicFil ultrasonic resin on odontoblasts and dental pulp vessels of rats using histological methods.
METHODS:Twenty-five Wistar rats are selected for experiment. Ninety-six teeth from the first and second molars were selected and randomly divided into SonicFil ultrasound resin group and Filtek Z350 nano resin group with 48 teeth in each group. Experimental teeth in each group were equaly subdivided into 7 days group and 30 days group. Besides, randomly selected five teeth with no intervention served as normal control group. After anesthesia and disinfection, class I cavities on the occlusal surface of the maxilary first and second molars were prepared and filed with SonicFil ultrasound resin and Filtek Z350 nano resin group, respectively. The rats were sacrificed by ink perfusion method, and tooth specimens were taken to observe the changes of odontoblasts and pulp microvessels.
RESULTS AND CONCLUSION: At 7 days after filing, two groups had similar changes in the pulp presenting with mild inflammation. Besides, there were scattered inflammatory cels distributing in the cave pulp tissue, and in most of the specimens, odontoblasts arranged disorderly with little vacuoles and microvascular dilatation in the pulp. At 30 days after filing, in the SonicFil ultrasonic resin group, odontoblasts arranged orderly, inflammatory reaction and microvascular dilatation disappeared basicaly. In contrast, in the Filtek Z350 nano resin group, disordered odontoblasts decreased, inflammatory reaction weakened, as wel as microvascular dilatation became less. These results show that under the experimental condition, both of the SonicFil ultrasonic resin and Filtek SonicFil Z350 nano resin play certain roles in the formation of odontoblasts and dental pulp vessels, but over time al of these effects can automaticaly disappear.
10.Aspirin resistance and vascular events in secondary prevention of cerebral infarction

Objective To investigate the incidence of the aspirin resistance in secondary prevention of cerebral infarction, and the relationship between the aspirin resistance and the cerebral infarction recurrence or other vascular events during the follow-up periods.Methods Aspirin were taken from the first day of admission in 600 patients with cerebral infarction.The platelet aggregation rate was measured after 7-10 days to screen the patients with aspirin resistance or aspirin sensitivity.All patients were followed up for 6 to 24 months and the cerebral infarction recurrence and other vascular events were recorded.Logistic regression model was used to estimate the risk factors of aspirin resistance, vascular events and prognosis.Results Of 600 patients, 150 (25.0% ) patients were resistant to aspirin and 450 (75.0% ) patients were sensitive to aspirin.The proportion of female and diabetes patients, and the level of low density lipoproteins (LDL) in the aspirin resistance group were higher than those in the aspirin sensitivity group.Diabetes (OR = 2.58, 95% CI 1.37-4.85, P=0.003) and high LDL level (OR = 1.89, 95% CI 1.21-2.93, P = 0.005 ) were independent risk factors of aspirin resistance.The incidence of cerebral infarction recurrence and myocardial infarction and all-cause mortality in the aspirin resistance group were all higher than those in the aspirin sensitivity group.Diabetes ( OR = 2.47, 95% CI 1.36-4.65, P = 0.003 ) , atherothrombosis cerebral infarction (OR = 2.13, 95% CI 1.24-3.95, P = 0.023) and aspirin resistance (OR = 3.86,95% CI 1.79-5.87, P = 0.002) were independent risk factors of vascular events during the following-up period.In the patients with aspirin resistance, the risk of the recurrence of vascular events increased 3.86 times.Conclusions The incidence of aspirin resistance is high in secondary prevention of cerebral infarction.Aspirin resistance is closely correlated with cerebral infarction recurrence and other vascular events.
