1.Protective effects of curcumin on myocardial ischemia reperfusion injury in rats

Aim To study the protective effects of curcumin on myocardial reperfusion injury in rats.Methods Myocardial ischemia reperfusion injury model was established through occluding the left anterior descending branch of coronary artery for 60 min and removing the ligation later to reperfuse for 30 min in vivo.Different doses of curcumin were administered through intravenous injection 5 min before the onset of ischemia.The changes of hemodynamics were recorded and myocardial infarct sizes were measured.Results Curcumin(20,40 mg?kg~(-1))improved the hemodynamics during myocardial ischemia reperfusion in a dose-dependent manner,as shown by the significant increase in LVSP and?dp/dt_(max) and the decrease in LVEP.Moreover,curcumin reduced the myocardial infarct sizes.Conclusion Curcumin exerts protective effects on myocardial ischemia reperfusion injury.
2.Experimental studies on blood-activating and stasis-eliminating actions of Danshensu Capsules

Objective: To investigate the effect of Danshensu Capsules on platelet aggregation, experimental thrombosis and hemorrheology in rat. Methods : The effects of Danshensu Capsules were observed on platelet aggregation induced by ADP. Occlusive thrombotic time was determined by electrostimulating; “blood stasis” model was prepared by ice water stimulation and the main hemorheological indexes were detected. Results : Danshensu Capsules could greatly inhibit the platelet aggregation in rat induced by ADP and delay occlusive thrombosis time after electrostimulating rat common carotid artery as well as decrease whole blood viscosity, plasma viscosity, red-cell specific volume, red-cell electrophoresis time, yield stress and red-cell aggregation indexes in experimental rat with the blood stasis syndrone. The rat's hemorheological properties were improved. Conclusion : Danshensu Capsules could decrease obviously the blood state with “dense、viscosity、aggregation、coagulation” and have obvious actions of blood-activating and stasis-eliminating.
3.Effect of total flavones isolated from Metasequoia glyptostroboides on platelet aggregation and hemorrheological properties in rats and research of its mechanism

Object To observe the effect of total flavones isolated from Metasequoia glyptostroboides Hu et Cheng (FMG) on platelet aggregation, 5 HT of release, content of NO in plasma and hemorrheological parameters in rats. Methods Platelet aggregation was quantified by turbidimetry, release of 5 HT was assayed by fluorescence technique, and content of NO in plasma was determined by spectrophotometry. Results Inhibition of platelet aggregation and release of 5 HT, increase of NO content in plasma, as well as decrease of whole blood viscosity, plasma viscosity, red cell electrophoresis time, red cell specific volume, red cell count and K value in ESR equation were observed in the acute blood stasis rat, thereby the rat's hemorrheological properties were improved. Conclusion FMG is able to inhibit the platelet aggregation and improve the hemorrheological parameters in acute blood stasis rat. The inhibition of release of 5 HT, increase of NO content in plasma and antagonistic action of Ca 2+ may be the mechanism of its actions.
4.Effects of total flavones metasequosia on ischemia reperfusion injury in guinea pig working heart

AIM To investigate the effects of t ot al flavones metasequosia (TFM) on ischemia reperfusion injury in guinea pig work ing heart. METHODS Ischemia reperfusion injury model was used to evaluate the effects of TFM. Myocardial func tion, the content of CK、LDH and the activity of MDA were measured. RESU LT TFM improved the cardiac function, decreased the release of LDH and CK and inhibited the increase of MDA content. CONCLUSION TFM had a protective effect on ischemia reperfusion injury of myocardium which could be re lated to its inhibitory effect on lipid peroxidation.
5.The protective effects of total flavones metasequosia on experimental myocardial infarction in anaesthetized dogs

AIM To study the protective effects of total flavones metasequosia(TFM) on experimental acute myocardial infarction (AMI) in anaesthetized dogs. METHODS The parameters of coronary circulation and myocardial oxygen consumption, the degree and range of myocardial ischemia, myocardial infarct size, the activity of serum CK,LDH and the content of FFA were determined by using the AMI model of ligature of LAD in the anaesthetized open-chest dogs. RESULTS In dogs treated with TFM(in a dosage of 25, 100 mg?kg -1 , id), the myocardial oxygen consumption, degree and range of myocardial ischemia, myocardial infarct size, the activity of serum CK,LDH and the content of FFA were decreased. At the same time, myocardial blood flow was increased and coronary vascular resistance decreased significantly. CONCLUSION TFM possesses a protective effect on myocardial ischemia by modifying dysfunction of FFA, decreasing myocardial oxygen consumption and increasing myocardial blood supply.-
