1.Expression of embryonic stem cells key transcription factor Nanog in bladder cancer and its relationship with cancer stem cells

Objective:To study the expression of nanog in human bladder cancer tissue,and demonstrate its significance in bladder cancer formation and relationship with bladder cancer stem cells.Methods:The expression of nanog was examined in the pathological specimens from 32 cases of bladder cancers,15 apparently nontumor tissues taken from the margin of tumors and 10 normal urothelial tissues by using semiquantitative RT-PCR and immunohistochemistry respectively.Results:The expression rate of nanog in bladder cancers was 84.38%(27/32)but at much lower level(P0.05). Conclusion:The expression of nanog showed there might exist bladder cancer stern cells.This has significant aplications to uncover the existence of CSC and to isolate and identificate bladder cancer stem cells.The rate and level of nanog expression in bladder cancer showed there might exist significant correlation between embryonic gene and occurrence and development of tumor.
2.Studies on "floating sugar" mechanism in root of Achyranthes bidentata

Object To study the "floating sugar" mechanism in the root of Achyranthes bidentata Bl. Methods The important influent factors were analyzed by orthogonal test. Results The primary and secondary orders of influent factors were surrounding temperature, relative humidity, raw drug moisture. The best preservative condition: surrounding temperature is 25 ℃, relative humidity 60% and drugs moisture 11%. Conclusion Raw drugs can be stored safely when surrounding temperature is below 35 ℃, relative humidity below 70%, raw drug moisture between 9% and 13%.
3.Identification of Fat-soluble Constituents of Cornus officinalis and Its Preparation by LC-MS

OBJECTIVE:To establish a method for identification of Cornus officinalis and its Preparations. METHODS: LC-MS method was adopted. The separation was performed on Hypersil C18 column (150 mm?2.1 mm,5 ?m) with a mobile phase of methanol-acetic acid(84 ∶ 16) at a flow rate of 0.2 mL?min-1. The column temperature was set at 30℃. Mass spectra were equipped with an electrospray ionization (ESI) source with negative ion detection model as ion scanning. The conditions of determination were sheath gas flow rate of 1.5 L?min-1,sweep gas flow rate of 0.45 L?min-1,spraying voltage of 4.5 kV, high temperature capillary temperature of 350℃ and scan range of 110~600 amu. RESULTS: The three-level mass spectrometry of the ursolic acid and oleanolic acid was well separated. The results were stable. Three-level mass spectrometry of ursolic acid and oleanolic acid were established and identification of C. officinalis preparations was carried out.CONCLUSION:Three-level fragment peaks of fat-soluble constituents of C. officinalis can be used for indentification of C. officinalis and its preparations.
4.Radiosensitization of nitroindazole derivatives on HeLa cells

Objective To investigate the cytotoxicity and radiosensitization of 5-nitroindazole-3-formyliminodiacetic acid on HeLa cells.Methods HeLa cells in exponential growth phase were incubated in culture media with different doses and the survival rate was determined by MTT assay.The survival rate of cells receiving radiation combined with different doses of medicine was compared with that of the control.Results The cytotoxicity of S-nitroindazole-3-formyliminodiacetic acid on HeLa cells was very low.The drug had hypoxia radiosensitizing effect on HeLa cells.At doses of 0,6,12,24,48 and 96 μg/ml under hypoxia ,the survival rate were 0.91 ,0.87,0.84,0.81 ,0.76 and 0.60,respectively.At the dosage of 48 and 96 μg/ml,the survival rate were 0.85 and 0.73 under oxygenous).Conclusions 5-Nitroindazole-3-formyliminodiacetic acid has low cytotoxicity and rediosensitizing effect on HeLa cells.
5.MRI characteristics of solid papillary carcinomas in situ of breast

Objective To evaluate MRI characteristics of solid papillary carcinomas (SPCs) in situ of the breast.Methods A retrospective study included 5 patients with pathologically confirmed SPC in situ was performed.MRI data before operation including conventional MRI,dynamic contrast enhanced MRI (DCE-MRI) and DWI were analyzed.Results All the lesions showed iso/hypointensity on T1 FSPGR sequence,iso/hyperintensity on FSE T2WI sequence and STIR sequence.Mass enhancements were observed for all lesions with oval or irregular shapes on DCE-MRI.The margin of lesions were circumscribed,and internal enhancements were homogeneous or heterogeneous.Time intensity curve appeared a rapid increase in initial contrast phases and platform or outflow types in delayed phases.All the lesions on DWI showed slightly hyperintensity with the ADC value range from 1.34 × 10-3 mm2/s to 1.96)× 10-3 mm2/s.Conclusion MRI manifestations of SPC are characteristics,which may provide valuable information to distinguish SPC in situ from other invasive breast carcinomas.

7.Optimization of extracting technology for flavonoids in Selaginella tamariscina with orthogonal design

Objective To optimize the extraction technology for total flavonoids in Selaginella tamariscina.Methods To determine the content of total flavonoids by UV and the content of index constituent of amentoflavone by HPLC.The optimum extraction condition was investigated by orthogonal design and the extraction quantity was regarded as the investigated index.Results The optimum extracting condition was A1B2C2D2 with the extraction quantity of total flavonoids as the investigated index.The optimum extracting condition was A1~3B1~3C2D2 with the extraction quantity of amentoflavone as the investigated index.The optimum extracting condition was A1B2C2D2.That is adding ten times amount of 95% alcohol and refluxing twice,2 h once.Conclusion The optimum technology is stable and feasible for the extraction of S.tamariscina.
8.Effect of Total Flavonoids from Litchi chinensis Sonn on Expression of TGF-β1 Receptor and Collagen in Rats with Liver Fibrosis

Objective To observe the prevention and treatment of the total flavonoids from Litchi chinensis Sonn( TFL) on hepatic fibrosis induced by dimethylnitrosamine(DMN)in rats, and to explore its mechanism. Methods Ninety SD rats were randomly divided into six groups, normal control group, model control group, colchicine group, high-, medium- and low-dose TFL group(n=15).Expect for normal control group, the other groups were given intraperitoneal injection of 2 mL.kg-1 of 5% dimethylnitrosamine for 4 weeks as the model group. The rats in the normal control group and model control group were given 5 mL.kg-1of 0.9% sodium chloride solution, colchicine group was treated with 0.1 mg.kg-1 colchicine.High-, medium-and low-dose TFL groups were given 200, 100 and 50 mg.kg-1 of TFL.The rats were sacrificed and the livers were harvested and stained with HE and Masson staining to observe pathological changes and liver fibrosis in the same part 6 weeks after all the medicine was given to the rats each day. Immunohistochemistry and Western blotting were used to detect the expression of the transforming growth factor β-Ⅰ/type Ⅱ receptor ( TβRⅠ/Ⅱ) , collagen Ⅰ( Col Ⅰ) and Ⅲ collagen ( Col Ⅲ) . Results Compared with the normal control group, the semiquantitative score of liver fiber and the protein expression of TβRⅠ, TβRⅡ, ColⅠ and Col Ⅲ in the model control group were significantly increased(P<0.01).Compared with the model control group, the protein expression levels of TβR, TβRⅡ, ColⅠand ColⅢwere significantly decreased( P<0.01) in the high-,medium-and low-dose TFL group.The semiquantitative score of liver fiber was significantly decreased( P<0.01) with a dose-effect relationship. Conclusion TFL can inhibit formation of DMN-induced liver fibrosis in rats, which may be related with reduction of expression of TβRⅠ/Ⅱ of hepatic fibrosis promoting factor TGF-β1 , inhibition of the activation and increase of hepatic stellate cells, reduction of the collagen content.
9.Impact of hyperglycemia on hypersensitive C-reaction protein, B-type natriuretic peptide and ventricular remodeling of elder patients with acute myocardial infarction

Objective To explore the impact of hyperglycemia on Hypersensitive C-reaction protein (hsCRP),B-type natriuretic peptide(BNP) and ventricular remodeling in elder patients with acute myocardial infarction(AMI).Methods One hundred and twenty elder patients with AMI are divided into hyperglycemia group(blood sugar in admission > 7.8 mmol/L) and non-glycemia group(blood sugar in admission ≤7.8 mmol/L).The levels of blood sugar in admission,hsCRP,BNP were detected.LVEF,LVEDD,LVESD,WMS were observed by cardiac echo in admission and two weeks after therapy.Results The blood sugar in admission,hsCRP and BNP of the patients in hyperglycemia group were (9.6 ± 0.6) mmol/L,(1 750.6 ±677.1) ng/L,(56.1 ±38.6) ng/L,higher than those in non-glycemia group (t =38.679 and P =0.000; t =11.941 and P =0.000; t =3.288 and P <0.01,respectively).LVEF,LVEDD,LVESD and WMS of patients in hyperglycemia group didn't show statistical significance before and after therapy.However,there were significant in hyperglycemia group (t =2.049 and P =0.043,t =2.836 and P =0.005,t =3.814 and P =0.000,t =2.086 and P =0.039,respectively).Conclusion Hyperglycemia can reduce the increase of hsCRP and BNP,which has an impact on hsCRP,BNP and ventricular remodeling in elder patients with acute myocardial infarction,but the mechanism need to further research.
10.Early Hematoma Growth in Patients with Intracerebral Hemorrhage

Objective:To explore the incidenee of early hematoma growth in patients with intracerebral hemorrhage,and the correlation between early hematoma growth and early neurological deterioration.Methods:The evaluation of nervous system and CT scan in patients with intracerebral hemorrhage within 3 hours after the omet of symptoms were performed respectively at baseline,and 1,2,3,and 20 hours after the baseline.Results:In 106 patients,CT scan shelwed that the volume of intraparenehymal hemorrhage inereased significantly in 26% of patients at baseline and within 1 hour.CT scan showed that the bleeding volume increased in 12% of patients at 1,2,3,and 20 hours.By measuring baseline,1-hour Glasgow Coma Scale (GCS)score,and the changes of the National Institutes of Health Stroke Scale(NIHSS),it had found that the baseline status and the increased bleeding volume within 1 hour vdgre closely correlated with the clinical neurological deterioration.Conclusions:The early bleeding volume often inereases significantly in patiems with intracerebral hemorrhage,which is complicated with neurological deterioration.In order to determine whether these therapeutic measures improve the natural course of this early hematoma growth and decrease its occurrence,it is necessary to make a randomized clinical trial.
