1.Establishment of Two Cell Lines with Double Markers and Comparison between Their Abilities for Detecting Replication-Competent Retro viruses

In this study, we use retroviral vectors, LXSN (containing internal promoter) and GCEN(containing internal ribosomal entry site,IRES), which cany ?galactosidare gene (LacZ) and neomycin phosphotransferase gene (Neo_R) as double markers, to establish two cell lines, LG-3T3 and GG-3T3. The sensitivities of the two cell lines for detecting replication-competent retroviruses(RCR) in marker rescue test were compared. Our results indicated that both cell lines can be used in marker rescue test, the sentivity of GG-3T3 was 100 times higher than that of LG-3T3.
2.Effects of external substance P on scalding wound healing and neovascularization in diabetic rats

Objective To evaluate the effects of external substance P (SP) on scalding wound healing and neovascularization.Methods Eighty-four Wistar rats were induced into diabetic models by intraperitoneal injection of streptozotocin (STZ), and deep partial thickness scalding wound on the back with diameter of 2 cm was prepared. Rats were randomly divided into experiment group (n=42, local injection of SP) and control group (n=42, local injection of PBS). The process of wound healing was observed, and the percentages of wound closure were calculated on D0, D1, D3, D7, D10, D14, D21 post scald. The expression of CD31, surface marker of neovascular endothelial cells, was detected within the wound sites by immunohistochemical staining, and the microvessel density was calculated. Results The percentage of wound closure in experiment group was significantly higher than that in control group on D7 post scald [(42.69±3.26) % vs (30.24±1.17)%, P<0.01]. Immunohistochemical detection revealed that the expression of CD31 and the microvessel density in experiment group were significantly higher than those in control group from D7 post scald (P<0.01). ConclusionExternal SP may promote scalding wound healing in diabetic rats, the mechanism of which may be associated with upregulation of expression of CD31 and acceleration of neovascularization.

Purpose:To study the analgesic role of Shikangning(SKN) injection (containing scorpion venom protein) on cancer pain. Methods:In the clinical trial, controls were used(41 cases,who were divided into two groups by the use of randomized double blind methed: SKN group 21 cases and placebo group 20 cases) and the open test (total of 88 cases, 67cases were given drug for 7 days, and 21 cases, for 14 days). 4.Immunologic Classification of Acute Lymphocytic Leukemia: A Report of 19 Cases

Immunologic classification of acute lymphocytic leukemia (ALL) was carried out by using 11 - 13 kinds of monoclonal antibodies against human leucocyte differentiation antigens in 19 cases. The results showed that of these 19 cases, there were T-ALL in 4, B-ALL in 4, C-ALL in 7, AUL in 2 and hybrid type of ALL (H-ALL) in 2. The 5 types mentioned above could also be classified, according to the immunologic criteria described by Foon in 1986, into 3 major groups: T-ALL, non-T-ALL and H-ALL. The Foon's classification method is useful for judgement of malignant cell source and differentiated stage, and for further understanding the nature of ALL cells.

6.Effect of granulocyte/macrophage colony-stimulating factor on expression of vascular endotllelial growth factor in human dermal fibroblasts

Objective To study the effects of granulocyte-macrophage colony stimulating factor (GM-CSF) on the expression of vascular endotllelial growth factor (VEGF) in human dermal fibroblast. Methods In vitro human dermal fibroblasts in good status were incubated with GM-CSF (GM-CSF group) or non-GM-CSF (control group) culture medium for different periods of time. The mRNA, protein expression of VEGF in derma fibroblast were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting, respectively, and the secretion of VEGF in supernatant was measured by enzyme linked immunosorbant assay (ELISA). Results The expression of VEGF mRNA from dermal fibroblasts was increased significantly after l or more hours of incubation with GM-CSF comparing with the control (P<0.05). 6 hours of stimulation by GM-CSF caused maximal expression of VEGF mRNA. The expression of VEGF protein in dermal fibroblasts was increased from 12 hours and was peaked at 24 hour after stimulation by GM-CSF. VEGF protein from the supernatant of the dermal fibroblasts was also raised persistently from 12 hour after stimulation by GM-CSF and was improved remarkably compared with the control. Conclusions GM-CSF can up-regulate directly the expression of VEGF in human derma fibroblast, which may be one of the mechanisms that GM-CSF accelerates neovascularization in wound healing.
7.Production of VEGF induced by GMCSF via ERK-NF-KB singling 'pathway in human fibroblasts during wound healing

ObjectiveTo observe production of vascular endothelial growth factor (VEGF) induced by granulocyte/macrophage colony-stimulating factor (GMCSF)via ERK nerve growth factor (NF)-κB singling pathway in human fibroblasts during wound healing and explore relating mechanism.MethodsHuman fibroblasts from the injured skin were used for this study and treated with GMCSF.RT-PCR was used for analyzing the protein and mRNA levels of VEGF and Western blotting was employed to determine the phosphorylation of ERK. The fibroblasts were pre-treated with ERK specific inhibitor PD98059 and further treated with GMCSF, then the fibroblasts and the supernatant were collected for detection of protein level of VEGF by means of Western blot. ERK signal pathway was inhibited to detect the activation of NF-κB by means of immunofluorescence staining. Furthermore, the nuclear and cytoplasmic extraction kit was used to separate the cytoplasm and nucleus and Western blot employed for observation of the NF-κB activation. ResultsThe mRNA level and protein level of VEGF were increased significantly with treatment with higher concentration of GMCSF in a dose-dependent manner. VEGF mRNA level was increased two hours after administration with GMCSF and reached peak at 4-6 hours. GMCSF could remarkably activate the ERK phosphorylation. Compared with GMCSF, the ERK specific inhibitor PD98059inhibited significantly the effect of GMCSF in inducing VEGF expression (P < 0.05). Western blot and immunofluorescence staining analyses showed that the activation of NF-ΚB was inhibited with reduced production of VEGF after GMCSF treatment.Conclusion GMCSF up-regulates production of VEGF through activating NF-κB via ERK signal pathway in the human fibroblasts.
8.Investigation and Analysis of the Family Influence to the Undergraduate Students' Psychology

Objective:To master the information of the communication between undergraduate students and parents,so as to provide basis for strengthening university student's psychology work.Methods:We used the investigation questionnaire designed by ourselves,investigated 841 undergraduate students in ShanXi Medical University and established SPSS 11.5 database carrying on statistical analysis.Results:62.2% students ring up their parents once a week,5% students call once a day,22.7% students call once a month,and 10.1% students wait for their parents to call them.8.4% students think that they have generation gap of communication with their parents,10.3% students fully obey their parents,81.3% students use parental words to encourage themselves.Conclusion:Family is an important factor for students' psychological development.
9.Clinical Observations of Treating 40 Cases of Bronchial Asthma with Chuankezhi Injection

Objective: To prove the clinical therapeutic effect of acute and chronic bronchial asthma with Chuankezhi(CKZ) Injection. Methods: 40 cases of bronchial asthma, including inpatients and outpatients were selected, divided into the attack period group (20) and the relief period group (20). The former was intramuscularly injected CKZ injection 4mL, twice a day, 14 days being a course of treatment. The latter was intramuscularly injected CKZ injection 4mL, twice a day for 15~30 days and 2mL, every other day for 31~90 days; 90 days being a course of treatment. The symptoms and signs, such as asthma, cough, expectroation, pulmonary wheeze were observed. and pulmonary function, absolute eosinophilic granulocyte count and immunogobulin E were measured before and after treatment. 10.Preparation of nasal thermosensible gels of Chinese medicine Xingbi and release behavior in vitro.

OBJECTIVETo prepare the nasal thermosensible gels of Chinese medicine Xingbi and study the release mechanism.
METHODThe gels were prepared by using P407 as the gel matrix and P188 and PEG 6000 were used to adjust the gelatination temperature. The formulations were screened by the method of orthogonal test. Mathematic models were used to imitate the drug release.
RESULT20% poloxmar 407, 2% poloxmar 188 and 2% PEG 6000 were suitable to Chinese medicine Xingbi thermosensitive gel in situ. Nasal thermosensible gels of Chinese medicine Xingbi was gelated at the temperature between 32-34 degrees C and the dissolution curves in vitro showed that the drug release could be best described by the Higuchi equation.
CONCLUSIONThe formulation of the nasal thermosensible gels of Chinese medicine Xingbi is reasonable and it is worth doing further research.
