Life system and medicine are both complex systems. Recently, traditional biologists have acquired tremendous data from high-speed development of all kinds of species group. However, how to use these data to interpret the mechanisms of diseases and health has become the research target of current investigators. The booms of system biology provide new thought and research methods to tackle this problem. This paper emphasizes the relationship between system biology and traditional Chinese constitutional medicine.

Objective To investigate the role of human leukocyte antigen-G ( HLA-G ) on the invasion and the molecular mechanism involved in this cellular progress in HTR-8/SVneo cell line. Methods There were three groups: groups of transfection, negative control and blank control, which corresponding to treatment by HLA-G specific siRNA, negative siRNA and only lipofectamine 2000 using lipofection technology in HTR-8/SVneo cell line. The efficiency of down-regulated of HLA-G was detected by reverse transcription-polymerase chain reaction and western blot analysis in mRNA and protein level,respectively. Changes of p38 mitogen-activated protein kinases (p-p38MAPK)/p38MAPK protein levels and the cell invasion were respectively detected by western blot analysis and transwell test. Results ( 1 ) The mRNA levels of HLA-G transfection group, negative control group and blank control group were 0. 26 ±0. 08, 0. 71 ±0. 11, 0. 79 ±0. 07, respectively. There was significant difference between transfection group and negative control group ( P ＜ 0. 01 ), while there was no significant difference between negative control group and blank control group ( P ＞ 0. 05 ). The efficiencies of down-regulated of HLA-G were ( 69. 8 ±6. 3)%, ( 14. 9 ± 2. 2 )%, 0 in transfection group, negative control group and blank control group respectively in mRNA level. (2)In protein levels, HLA-G were 0. 20 ±0. 15, 0. 75 ±0. 12, 0. 76 ±0. 21 in transfection group, negative control group and blank control group, respectively. There was significant difference between transfection group and negative control group ( P ＜ 0. 01 ), whereas there was no significant difference between negative control group and blank control group ( P ＞ 0. 05 ). The efficiencies of down-regulated of HLA-G were (81. 1 ± 14.4)%, ( 18.0 ± 7.7)%, 0 in transfection group, negative control group and blank control group respectively. ( 3 ) The invasive number of transfection group, negative control group and blank control group were 57 ± 38,364 ± 79 and 260 ± 84, respectively, with a significant difference between transfection group and negative control group (P ＜ 0. 01 ). There was no significant difference between negative control group and blank control group ( P ＞ 0. 05 ). ( 4 ) The p-p38MAPK/p38MAPK values of the HLA-G transfection group, negative control group and blank control group were 0. 74 ±0.04, 0. 47 ± 0. 09 and 0. 36 ± 0. 21, respectively. HLA-G transfection group was significantly different compared with the other two groups( P ＜0. 01 ). (5)Without or with SB203580, the p-p38MAPK/ p38MAPK values of the HLA-G transfection group were 0. 89 ± 0. 09 and 0. 16 ± 0. 04, the values of negative control group and blank control group were 0.76 ±0.08, 0. 14 ±0.03 and 0.51 ±0.05, 0.03 ±0.01, respectively. There was significant difference between without SB203580 and with SB203580 ( P ＜ 0. 01 ). (6)Without or with SB203580, the invasive number of transfection group were 51 ± 13 and 90 ± 21 ,respectively,which was significantly different ( P ＜ 0. 01 ). The invasive number of negative control group and blank control group were 290 ± 52, 298 ± 33 and 290 ± 73, 264 ± 64, respeczively, which was no significant difference between without SB203580 and with SB203580 (P ＞ 0. 05 ). Conclusions HLA-G gene may regulate invasion of trophoblast-derived cell line HTR-8/SVneo via p38MAPK signaling pathway. The lower expression of HLA-G in trophoblast cells may lead to the occurrence of pathologic pregnancy.

AIM: To study the effect of Lomerizine on the activity of P-glycoprotein (P-gp) in the bloodbrain barrier(BBB) and search for novel effective P-gp inhibiting agent against multidrug resistance. METH-ODS: Rhodamine123 (Rh123) was used to examine the activity of P-gp and RT-PCR to study the mdr mRNA expression in cultured rat brain microvessel endothelial cells (RBMECs). RESULTS: Lomerizine could increase the cellular Rh123 in RBMECs in a concentration-dependent manner. RT-PCR indicated that lomerizine could not down-regulate the expression of mdr mRNA. Objective To investigate the effect of human leukocyte antigen-G(HLA-c)on the growth and invasion of JEG-3 cell line and the role of HLA-G in the onset and development of pre-eclampsia.Methods The experiment was composed of three groups:groups of transfection,negative control and blank control.which corresponded to groups of HLA.G siRNA transfection,negative siRNA transfection and no transfection HLA-G overexpressed choriocarcinoma cell line JEG-3 was used.The role of HLA-G in JEG-3cell monolayer was examined by RNA interference technology using HLA-G specific small interfering RNA (siRNA).Expression of HLA.G was detected by reverse transeriptase-polymerase chain reaction and western blot analysis.Changes of cell cycle,apoptosis,proliferation and invasion were respectively detected by methvl thiazolyl tetrazolium(Ma r).flow cytometry assay and transwell test.Results (1)The mRNA and protein levels of HLA.G control group and blank control group were 0.0013±0.0014.0.0163 ±0.0007 and 0.1923 ±0.0384.0.2184 ±0.0153,respectively,which were both significantly different(P<0.05);the number of negative transfcction group was 0.1606±0.0133 and 0.2020±0.0132.which had no significant difference compared with blank control group(P>0.05).(2)The integral absorbance(IA)valUCB of the HLA-G transfecfion group and blank control group were 0.44±0.04 and 0.75±0.13 respectively.which was significantly different(P<0.01);the/A value of negative control group was 0.69±0.10.which was not significantly different compared with blank group(P>0.05).(3)The ratios of G2/M and S phase cells in transfection group were(10.9±2.2)%and(58.6±0.8)%respectively,significantly different compared with the blank control group[(15.4±1.9)%and(52.9±2.3)%respectively;P<0.01].(4)The ratio of early apoptosis cells in transfection group[(14.5±2.7)%]Was significantly increased compared with neg~ive[(5.3 ±1.1)%]and blank control group[(4.7±0.6)%;P<0.01].(5)The invasion number of transfecfion group and blank control group were 121±12 and 452±17 respectively.with a significant eclampsia by regulating proliferation and invasion of trophoblast.

AIM: To observe protective effects of sanguis draxonis flavones on myocardial ischemia in rats and dogs. METHODS: Acute myocardial ischemia in rats was produced by iv pituitrin and ECG indexes were observed. Myocardial ischemia was induced by ligating coronary artery in anaesthetized dogs, and then EEC, CK,LDH, and LD in serum were determined respectively. RESULTS: J point and T wave in rats changed evidently after iv pituitrin, which was reversed by sanguis draxonis flavones (360, 180 mg?kg~ -1). In coronary artery ligation model, infraction range, △N-ST, △?-ST and some serum indexes (such as CK, LDH and LD) was decreased after ig sanguis draxonis flavones (120, 60, 30 mg?kg~ -1). CONCLUSION: Acute myocardial ischemia is protected effectively by sanguis draxonis flavones.

The diagnosis related groups( DRGs) method is applied in hospital management at both hospital and clinical department level. This technology enabled us to evaluate hospital medical services in general, objectively evaluate discipline development, and formulate individualized discipline guidelines, supporting scientific distribution of hospital resources. At the department level, the technology supported clinical departments in their discipline construction and quality management. The application of DRGs can improve fine management of hospitals.

This study was designed to investigate inhibitory effects and possible mechanisms of snake venom tripeptide (pENW) on platelet adhesion in order to promote the development of a novel anti-platelet therapy. To study the inhibitory effects of pENW on platelet adhesion, washed platelets pre-incubated with pENW (116.5-466.2 μmol x L(-1)) were used to test the ability of platelet adhesion to fibrinogen. Effect of pENW on fibrin clot retraction was also tested. Effect of pENW on platelets viability was tested by MTT assay. Effect of pENW on reactive-oxygen species (ROS) levels of platelet was studied by flow cytometry assay. Calcium mobilization in Fura-2/AM-loaded platelets was monitored with a spectrofluorimeter. Cyclic guanosine monophosphate (cGMP) and cyclic adenosine monophosphate (cAMP), thromboxane A2 (determined as its metabolite thromboxane B2) were measured using enzyme immunoassay kits. Akt, ERK and p38 phosphorylation were tested by Western blot. The results showed that pENW inhibited platelet adhesion and fibrin clot retraction in a concentration-dependent manner without cytotoxicity. Intracellular cGMP and cAMP in both resting and thrombin-activated platelets were increased by pENW. In addition, pENW attenuated intracellular Ca2+ mobilization and TXA2 production in platelets stimulated by thrombin. As shown by Western blot assay, Akt, ERK and p38 phosphorylation in thrombin-induced platelet were attenuated by pENW. However, inhibitory effects of pENW had nothing to do with ROS. Thus, pENW exhibited a significant inhibition on platelet adhesion to fibrinogen, which means pENW could block the first step of thrombosis as while as retard the more stable clot formation. The mechanisms of pENW on inhibition platelet adhesion might be related to instant regulations, such as protein kinases.

Eighty-three patients with transient ischemic attack (TIA) were randomly divided into study group (n =42) and control group (n =41) from July,2011 to Mar,2015.Patients in both groups were given aspirin 100 mg/d ; in addition,patients in study group received intravenous infusion of argatroban 10 mg,q12h,for 7 days and those in control group received subcutaneous injection of low molecular weight heparin (LMWH) 2 050 AXa IU,b.i.d for 7 days.The cases with cure,significantly effectiveness and effectiveness in study group and control group were 10,14,16 and 6,12 16,respectively.Compared with the control group,argatroban shows superiority in treatment of TIA with higher safety and lower side effect.

Objective To observe the therapeutic effect of beehive extract for the treatment of allergic rhinitis. Methods Sixty qualified subjects were evenly randomized into large dose group and small dose group, 30 in each group. The large dose group was given oral use of beehive extract 15 g per time, and the small dose group was given 5 g per time, three times per day. One week constituted a treatment period, and the treatment lasted 4 courses. After treatment, the therapeutic effect was evaluated in both groups. Follow-up was carried out one month after suspension of medicine. Results ( 1) The total effective rate was 90.00% in the large dose group, and was 93.00% in the small dose group, and the rank sum test results showed the difference was insignificant between the two groups (P>0.05) . (2) After treatment and during follow-up, the scores of general symptom Visual Analog Scale ( Uni-VAS) and the scores of Rhinoconjunctivitis Quality of Life Questionnaire ( RQLQ) were decreased in both groups ( P<0.01) , and the scores of symptoms in the small dose group were increased during the follow-up ( P<0.05) . The results of intergroup comparison showed that the differences of the decrease of Uni-VAS scores, symptom scores, non-nose/eye symptom scores, practical issue scores, nasal symptom scores, eye symptom scores and emotional reaction scores were insignificant between the two groups after treatment and during the follow-up ( P>0.05) . Conclusion Oral use of large or small dose of beehive extract shows certain therapeutic effect for allergic rhinitis by obviously relieving the symptoms of patients. The effect of large dose is similar to small dose, but the long-term effect of large dose is better.

BACKGROUND: Borneol can open blood-brain barrier (BBB) but the mechanisms are not very clear. Histamine and 5-hydroxtryptamine can take part in regulation of permeability of BBB. There is not report on the rela tion between the effect of opening BBB of borneol and the regulation of permeability of BBB of histamine (HA)/5-hydroxytryptamine (5-HT). OBJECTIVE: To study the relation between the effect of opening BBB of bornool and the regulation of permeability of BBB of histamine (HA)/5-hydroxytryptamine (5-HT).DESIGN: A completely randomized and controlled study.SETTING: Clinical Pharmacological Institute of University of Traditional Chinese Medicine of Guangzhou.MATERIALS: The experiment was carried out at the Clinical Pharmacological Institute of University of Traditional Chinese Medicine of Guangzhou from September 2003 to February 2004. Totally 104 healthy male SD rats, weighting 230-27.0 g, supplied by Guangdong Medical Experimental Animal Center, were selected.METHODS: The rats were randomly divided into 13 groups: pre-medicine group and post-medicine group (high, moderate and low dosage group and 5, 20, 45 and 60 minutes group in each dosage group) with 8 in each group. Borneol was mixed as 10% millet oil suspension. Rats were fasted before experiment for whole night, and medicine was perfused on the next morning with the high, moderate and low dosage of 0.15, 0.12 and 0.09 g/kg respectively. Hypothalami of rats was selected at various time points to make biological samples. Contents of HA and 5-HT were assayed with HPLC system electrochemical detector.MAIN OUTCOME MEASURES: Changes of level of histamin (HA) and 5-HT in hypothalamus of rats after administration of borneol.RESULTS: Data of totally 104 rats was entered the final analysis. ① Contents of HA and 5-HT in hypothalami were 2.07±0.54 μg/g and 1.45±0.14 μg/g respectively. ② The level of HA in hypothalamus of rats after different doses of borneol were higher than that of before administration. Comparing the level of HA in 20 minutes after moderate dose with before administration, the level of HA in 20 minutes was increased significantly [(3.36±0.21) μg/g, P < 0.01], the others of moderate dose, the 45 minutes after high dose, the 20 and 45 minutes after low dose were also increased significantly than before administration (P < 0.05). ③ After administration of different doses of borneol, the level of 5-HT after high dose were higher than that of before administration [5, 20, 45 and 60 minutes after medicine: (1.90±0.32), (3.28 ±0.25), (2.66±0.46), (2.80±0.34) μg/g, respectively; (P < 0.05 or P < 0.01)]; the level of 5-HT after 5, 20 and 45 minutes of moderate and low dose were increased significantly too (P < 0.05 or P < 0.01).CONCLUSION: Borneol could open BBB by increasing levels of HA and 5-HT in hypothalamus of rats.Borneol mediates opening of BBB by increasing levels of HA and5-HT in rats.
