1.MEASUREMENT OF HUMAN GROWTH HORMONE: DEVELOPMENT AND VALIDATION OF A SENSITIVE AND SPECIFIC ASSAY

The determination of human growth hormor.e (hGH) in urine by means of LAB-ELISA technique was described. The major factors affecting the Sensitivity of LAB-ELISA were studied systematically. The optimized conditions have been established. The sensitivity of LAB-ELISA is at the level of pg/ml. There were little cross binding with human prolactin, thyroid-stimulating hormone, follicular-stimulating hormone, and luteinizing hormone. Human growth hormone levels in overnight urine in 50 normal children and 11 cases of GH-deficiency (GHD) were measurejl by the LAB-ELISA technique. Results of recovery test (recovery rate 100 ?1.4%) and precision test (intra-assay CV 4.72-12.00%, inter-assay CV 4.89-11.17%, and bias 0.20-12.00%)indicate that the method is highly specific,precise and stable.
2.Tolerance of patients with chronic liver diseases undergoing gynecologic laparoscopic operations

Objective To investigate the liver function of patients with chronic liver diseases undergoing gynecologic laparoscopic operations.Methods Twenty-six asymptomatic HBV carriers,28 mild chronic hepatitis,21 compensatory cirrhosis were assigned to undergo gynecologic laparoscopic operations.Liver function were tested and compared in these three groups before operation and on the 1st、3rd、7th day after operation.Results 1st day after operation,ALT and AST in asymptomatic carriers and mild chronic hepatitis groups were significantly higher than preoperation ( asymptomatic carriers:ALT P < 0.05,AST P < 0.05 ; mild chronic hepatitis:ALT P < 0.05,AST P < 0.05 ).Then they recovered on the 3rd day after operation ( asymptomatic carriers:ALT P > 0.05,AST P > 0.05 ; mild chronic hepatitis:ALT P > 0.05,AST P > 0.05 ).ALB,total bilirubin (Tbil) after operation were not significantly different from those before operation on every tested day in these two groups ( asymptomatic carriers:ALB P > 0.05,Tbil P > 0.05 ; mild chronic hepatitis:ALB P > 0.05,Tbil P > 0.05 ).In compensatory cirrhosis group,ALT,AST,Tbil were significantly higher on the 1st day,3rd day after operation than preoperation ( 1st day:ALT P <0.01,AST P <0.01,Tbil P <0.01 ;3rd day:ALT P < 0.05,AST P < 0.05,Tbil P < 0.05 ).ALB levels were significantly lower on the 1st day,3rd day after operation than preoperation ( 1st day:P <0.01 ;3rd day:P <0.05 ).All the indexes recovered to the levels of preoperation on the 7th day after operation( ALT P > 0.05,AST P > 0.05,Tbil P > 0.05,ALB P >0.05).Conclusions To some extent,gynecologic laparoscopic operations have some adverse effects on short-term liver function after operation in patients with chronic liver diseases.Compensatory cirrhosis patients present delayed recovery compared with asymptomatic carriers and mild chronic hepatitis patients.
3.THE PROTECTIVE EFFECT OF TGPs ON ACUTE LIVER INJURY INDUCED BY CARBON TETRACHLORIDE IN MICE

The protective effect of TGPs on acute liver damage induced by carbon tetrachloride ( CCl4 ) was investigated in mice . TGPs ( 40 or 80mg/kg?d ? 3d, ip ) reduced the elevation of plasma or serume gl- utamic pyruvic transaminase ( GPT ) and plasma lactate dehydroge-nase ( LDH ) in CCl4-intoxicated mice, but if had no significant influence on the elevation of plasma glutamic oxalacetic transaminase ( GOT).TGPs (40mg/kg?d ? 3d, ip ) showed a protective effect on the histopathologic changes of liver injury induced by CCl4. The results suggest that TGPs may a protect against hepatic damage.
4.Subchronic Toxic Effects of Exposure in Combination to Dibutyl Phthalate and Benzo[a]pyrene on Spermatogenesis in Male Rats

Objective To explore the combined effects of subchronic exposure of dibutyl phthalate (DBP) and benzo [a] pyrene (B[a]P) on spermatogenesis in male rats. Methods A total of 32 male Sprague-Dawley rats aged 4-5 weeks were randomly divided into 4 groups and treated respectively as follows, corn oil, 1 mg/kg of B[a]P, 50 mg/kg of DBP, 1 mg/kg of B[a]P+50 mg/kg of DBP , the gavage was used every other day, for 90 days. The rats were sacrificed and the testes, epididymis, livers and other visceral organs were collected, and the organ coefficients were calculated. Flow cytometry (FCM) was used to detect changes of cell cycle. The testes were prepared for histological examination, and cauda epididymides were isolated for the determination of progressive motility and density of stored spermatozoa. The blood samples were collected to assess the effect of B[a]P and DBP on plasma testosterone and luteinizing hormone (LH) concentrations with chemiluminescence immumo-assay. Results The percentage of progressively motile stored spermatozoa and stored sperm density were not changed when exposure to B[a]P and DBP. Objective To study the mechanism of killing microfilariae in host by detecting the change of amino acids in Brugia malayi microfilariae. This is the first report of the sensitizing effect of cation surfactant on the enhanced resonance light scattering (RLS) resulted from the interaction of organic dyes with Deoxyribonucleic acid (DNA). On the conditions of pH 10.4 ~11.8 and ionic strength being less than 0.050, the interaction of brilliant green (BG) with DNA results in enhanced RLS signals at 398.0 and 467.2 nm overloaded some shoulder peaks in the enhanced region. It was found that the interaction was sensitized by the presence of cetyltrimethylammonium bromide (CTMAB). If 2.0~3.2×10-5 mol/L CMAB was used in the interacting medium, DNA in the range of 0~1.2 (mg/L could be detected with the limit of detection being less than 5 μg/L. Objective To construct the pcDNA3. 1-Brugia malayi (Bm) glyceraldehyde-3-phosphate dehydrogenase (GAPDH) eukaryotic recombinant plasmid and to study its effect on mouse cellular immunity response. Methods Total RNA was prepared from periodic Bm. The target gene fragments were amplified by reverse transcription-polymerase chain reaction (RT-PCR) technique and then were inserted into the cloning vector. pGEM-T Easy, and sub-cloned into pcDNA3. 1. Purified pcDNA 3. 1-BmGAPDH recombinant plasmid and CpG were injected into the anterior tibial muscle of BALB/c mice in order to induce host immunity response. Mice injected with PBS and mice injected with blank plasmid were prepared as controls. The mouse models were immunized for 3 times with an interval of 2 weeks. RT-PCR was utilized to detect target gene expression in the muscle tissue. MTT method was used to measure the immunized mice T lymphocytes stimulation index, while enzyme-linked immunoassay (ELISA) was used to determine the serum interleukin (IL)-4 and interferon (IFN)-γ level. Means were compared using t test with SPSS software. Results The recombinant plasmid pcDNA3. 1-BmGAPDH was constructed sucessfully. The target gene was 1020 bp long and its homology with known gene sequence in database was 99%. BmGAPDH gene in the injected muscle of the immunized mice was detected by PCR. The proliferation of spleen T lymphocytes was higher in pcDNA3-BmGAPDH group than in the 2 control groups which were 1. 398, 1. 006 and 1. 017,respectively (P< 0. 05). The levels of IFN-γ and IL-4 in serums from the immunized mice were significantly higher than those of the PBS control group and blank plasmid control group which were 163.905, 58.589, 51. 317 and 107. 906, 27.111, 34.627, respectively (P<0. 05). Immune adjuvant CpG could accelerate and boost antigen-specific immune responses induced by vaccine, which presented as significant increase of IFN-γ and lymphocyte proliferation at 4 weeks after immunization.Conclusion The recombinant eukaryotic plasmid pcDNA3. 1-BmGAPDH is constructed and could elicit cellular immune responses in immunized mice.
9.Relationship between microvessel density and recurrence of hepatocellular carcinoma after hepatic resection in the elderly

Objective To study the effect of age on the recurrence-free survival rate after hepatic resection for hepatocellular carcinoma(HCC)and the relationship between microvessel density (MVD)and recurrence of HCC in the elderly. Methods Severty one cases of elderly patients with HCC were analyzed retrospectively with 352 cases of non-elderly HCC patients as control,and the effect of age on the recurrence-free survival rate was studied.The expressions of CD34 and endocan in HCC tissues were detected by immunohistochemistry in 30 elderly and 30 non-elderly patients.Results The 1-,3- and 5-year recurrence free survival rates were 75.7%,43.0% and 43.0% in the elderly group respectively,which were higher than those in the non-elderly group(53.6%,38.5% and 33.4%,respectively,Log Rank value=10.25,P<0.05).The positive rate of alpha fetoprotein (AFP)in the elderly group was 47.9%,which was lower than that in the non-elderly group(62.2%)(X2=23.68,P<0.05).The median survival times in the high CD34-MVD group and high endocan MVD group were shorter than those in the low CD34-MVD group and low endocan-MVD group(260 d vs.850 d,360 d vs.800 d,Log Rank value was 22.18 and 20.56 respectively,both P<0.05).Conclusions The long-term prognosis of hepatic resection for HCC is better in elderly patients than in non-elderly patients.The recurrence of HCC in the elderly is closely related with angiogenesis.

The dynamic variation of folate content in the whole blood from ?-irra-diated rats have been observed. The level of folate, which did not alter significantly 24 hours after whole-body exposure of 800 rads, decreased dramatically to about 50 percent at the 5th day and to about 67 percent at the 10th day. After irradiation the haemoglobin concentration in rats also decreased, but there was no coincidence between the dynamic variation of folate level and haemoglobin concentration. It is probable that ionizing radiation induces the folic acid malnutrition of rats.
