1.THE EFFECT OF VITAMIN B_(12) AND FOLIC ACID ON RADIATION DAMAGE 3. NITROGEN METABOLISM

Sixty male rats divided equally into three groups were fed with synthetic diet. In every group each rat was subcutaneously given with vitamin B12 and folic acid, but the administration dose for different group was different so that the first group showed nutritional insufficiency, the second almost normal and the third good nutritional condition.Having fed to four weeks, the nitrogen intake for the first group (0.36 ?0.01 g/day) was significantly lower than that for the second (0.45?0.02 g/day) or for the third (0.44 ?0.012 g/day). The same were true for urinary nitrogen and total nitrogen.Under histidine load of 50 mg per rat, the urinary nitrogen excretion in every group was increased, but the increased quantity was much more than the nitrogen content in histidine used for loading test. At the same while the urinary amino nitrogen and ammonia nitrogen for the first group were increased, but not for the second or for the third before and after the loading test.Twelve rats selected from every group were equally distributed into two subgroups, one being irradiated with 60Co ray 700 rad and other not irradiated taking as control. The loading tests of histidine were undertaken at 1st, 7th, 14th, 21th and 28th day. The results showed that the radiation damage, the decrease of food intake and histidine loading test might exaggerate the disturbance of nitrogen metabolism in rats suffered from nutritional insufficiency of vitamin B12 and folic acid, as shown in urinary amino nitrogen and ammonia nitrogen excretion, but higher administration of vitamin B12 and folic acid could eliminate the exaggeration and improve the disturbance of nitrogen metabolism as well.

3.STUDIES ON THE EFFECT OF VITAMIN B12 AND FOLIC ACID ON RADIATION DAMAGE 1. URINARY EXCRETION OF FORMIMINOGLUTAMIC ACID

Weanlimg male rats fed on synthetic diet containing 1% succinylsulfa-thiazole and devoid of vitamin B12 and folic acid for 3-5 weeks, developed syndromes of the vitamins deficiency, i.e. poor food consumption, low growth rate, decrease in white cell count and marked increase in formimi-noglutamic acid excretion.Nevertheless, the others had an intake of vitamin B12 (0.02 ?g/day/-rat) and folic acid (0.5?g/day/rat) for a 36-day period, also showed signs of vitamins deficiency. But when given a daily dose of 0.1 ?g of vitamin B12 and 2.5 ug of folic acid, the rats had a good appetite and grew well, and excreted more formiminoglutamic acid than normal rats, when they were given a loading dose (0.05 g/rat) of histidine. Under the similar feed- ing condition, rat ingesting a daily dose of 0.3 ug of vitamin B12 and 7.5ug of folic acid, showed no obvious syndomes of the vitamins deficiency.After the rats were fed on the same diet and given with vitamin B12 (0.02 ug/day/rat) and folic acid (0.5ug/day/rat) for a 36-day period, the animals developed increased radiosensitivity. After total-body irradiation (700 rad), the food consumption, growth rate and survival rate in this group were lower than those in other groups ingesting individually 0.1 ug of vitamin B12 and 2.5 ug of folic acid or 0.3ug of vitamin B12 and 7.5 ug of folic acid daily.After irradiation, in the group of 0.02 ug of vitamin B12 and 0.5 ug of folic acid or 0.1 ug of vitamin B12 and 2.5 ug of folic acid in daily in-gestion there was a considerable increase in formiminoglutamic acid with a loading test of histidine(0.05 g/rat) than before. The results showed that the radiation intensified vitamins deficiency in rats.
4.THE EFFECTS OF VITAMIN B_(12) AND FOLIC ACID ON RADIATION DAMAGEⅡ,THE LEVEL OF FOLATE IN RAT BLOOD

The dynamic variation of folate content in the whole blood from ?-irra-diated rats have been observed. The level of folate, which did not alter significantly 24 hours after whole-body exposure of 800 rads, decreased dramatically to about 50 percent at the 5th day and to about 67 percent at the 10th day. After irradiation the haemoglobin concentration in rats also decreased, but there was no coincidence between the dynamic variation of folate level and haemoglobin concentration. It is probable that ionizing radiation induces the folic acid malnutrition of rats.
5.THE NUTRITIONAL EFFECT OF GELATIN ON ACUTE RADIATION DAMAGE

Sixty male rats divided into the control and the gelatin group were fed with 20% protein synthetic diet. The diet for the former group contained 24% casein, whereas for the latter 11% gelatin and 12% casein. , Having been fed for some days, all rats were irradiated with 875 rad (LD85-90). The main results were summarized as follows;1. The gelatin group had higher survival rate than the control group. 2. The body weight of gelatin group was less declined than that of control group and then much restored than the latter. 3. The survival time of dying rats within 14 days following the exposure to 875 rad in gelatin group was more longer than that in control group. 4. The gelatin group had less biological lesions than the control group.It was suggested that the gelatin diet had distinctly prophylactic-therapeutic action on acute radiation damage and its mechanism was probably related to the prevention from disturbance of collagen metabolism.
6.compliance and unhealthy lifestyle of patients with diabetes mellitus

Objective To observe and study the influence degree of strengthen training for family members for the treatment compliance and unhealthy lifestyle of patients with diabetes mellitus. Methods A total of 64 patients with diabetes mellitus from January 2015 to March 2016 were selected and divided into two groups by the method of random number table,32 patients in control group were taken care with routine nursing of diabetes mellitus,32 patients in observation group were taken care with strengthen training for family members,then the treatment compliance,related knowledge cognition of unhealthy lifestyle and unhealthy lifestyle situation of two groups before the intervention and at 4th, 8th and 12th week after the intervention were compared. Results The treatment compliance, related knowledge cognition of unhealthy lifestyle and unhealthy lifestyle situation of two groups before the intervention all had no obvious differences (all P>0.05). The treatment compliance at 4th, 8th and 12th week after intervention were (6.26 ± 0.32), (6.95 ± 0.36), (7.28 ± 0.40) scores in observation group, and (5.23 ± 0.29), (6.01 ± 0.30), (6.30 ± 0.33) scores in control group, and the differences were significant (t=8.253, 9.201, 10.257, all P<0.05). The related knowledge cognition of unhealthy lifestyle at 4th, 8th and 12th week after intervention were (80.78±5.90), (89.44±6.02), (93.43±6.20) scores in observation group, and (73.38± 5.73), (77.93±5.84), (82.20±6.01) scores in control group, and the differences were significant (t=11.258,12.563, 13.324, all P<0.05 ). The unhealthy lifestyle situation at 4th, 8th and 12th week after intervention were 75.00%(24/32), 90.63%(29/32), 96.88%(31/32) in observation group, and 59.38%(19/32), 68.75%(22/32),78.13%(25/32) in control group, and the differences were significant (χ2=7.251, 7.468, 9.169, all P<0.05). Conclusions The influence of strengthen training for family members for the treatment compliance and unhealthy lifestyle of patients with diabetes mellitus are more active, so the value of strengthen training for family members of patients with diabetes mellitus is higher.
7.NUTRITION AND ANTIBIOTICS TREATMENT FOR ACUTE RADIATION DAMAGE

Eight adult dogs distributed equally into control and therapy groups were fed on stock diet for 20 days and then irradiated whole-bodily with 60Co 4.65 Gy. After irradiation the control group was not given any therapeutic measure but fed on the diet as usual, while the therapy group was given with a larger amount of vitamins and high-caloric and high-protein diet, and sometimes force feeding was taken if the latter showed sign of anorexia. Whether higher values of nonprotein nitrogen in serum of two dogs in restored period and diarrhea happened in the therapy group were attribute to The treatments such as high-protein diet and force feeding were still unknown, the curing effects of nutrition and antibiotics were distinct as shown in that the body weight was easily maintained, and that the anemia and leucopenia wereless severe and recovered early. Moreover, within 30 days both the ther apy andcontrol groups one dog died respectively but survival time of the former was longer than the latter.
8.Preliminary application of hand robot assisted rehabilitation of upper-limb motor function in patients with stroke and hemiplegia

Objective Toinvestigatetheeffectofrehabilitationrobothandontheupperlimbmotor functioninpatientswithstrokeandhemiplegia.Methods Theeffectsoftraditionalrehabilitation therapy or robot rehabilitation treatment of 15 stroke patients with hemiplegia were analyzed retrospectively. There were 4 females and 11 males. According to the group order,they were divided into either a robot rehabilitation group (n=8 )or a general rehabilitation group (n =7 ). The patients of the general rehabilitation group received general rehabilitation training at the Department of Rehabilitation Medicine for 4 weeks,3 times a week,and 30 min for each time. At the same time,they also conducted family rehabilitation training,5 times a week,once for 1 h;the patients of the robot rehabilitation group received the general rehabilitation training for the same length of time as the general rehabilitation group,and they also performed robot rehabilitation training,5 times a week,once for 1 h. The patients of both groups were evaluated with the modified Ashworth scale (MAS )scores,Fugl-Meyer motor assessment of the upper extremity(FMA-UE),andWolfmotorfunctiontest(WMFT)beforeandaftertreatment.Results Compared with before training,the MAS score of the robot rehabilitation group was decreased significantly after four weeks of training. The time in WMFT was shortened significantly and the function score was improved obviously. The scores of upper extremity,hand score,and total FMA score in FMA-UE were improved significantly,and there were significant differences (all P <0. 05 ),while there were no significant differences in various indicators in the general rehabilitation group (all P>0. 05). Comparing the effects after four weeks of rehabilitation in both groups,the robot rehabilitation group was superior to the general rehabilitationgroupforthehandscoreinFMA-UE(allP<0.05).Conclusion Therehabilitation robot hand effectively lowered the upper limb spasticity. It has significant effect for recovery motor function of hand;however,it did not have a significant effect for motor rehabilitation on wrist.
9.Possible potential impact of Three-gorges Project on population health in Kaixian County of Chongqing

BACKGROUND: In the past, the incidence rates of infectious diseases,natural epidemic diseases and endemic diseases in three gorges areas were always high. Because the flooded areas of Kaixian was enormous, the population of Kaixian was intensive density, the pollution may harm to the population health after the dam has been established.OBJECTIVE: To predict the influence of the Three-gorges dam on population health in Kaixian County of Chongqing and put forward to scientific evidence of preventive measures through analyzing the main health indicators and influence factors in Kaixian.DESIGN: Cross-sectional study.SETTING: Public Health College of Chongqing Medical University and Disease Controlled Center in Kaixian County of Chongqing.PARTICIPANTS: The survey was conducted in Kai County of Chongqing,from February to May 2004. All disease surveillance data of people in this county was investigated and other people lived in three-gorges area or around area were selected as the controls.METHODS: Collect and sort relevant indicators of social development,population health and public health system of Kaixian and related area to go deep into the representative townships to do some filed investigation,collect water samples of local drainage areas to carry out laboratory quantitative analysis; carry out comparative analysis based on the relevant materials; carry out comprehensive predicting and evaluation to the potential impact on population health of the three gorges engineering by combining the quantitative and qualitative analysis methods.general people in Kai county, Chongqing city and all over the nations durKai county, Chongqing city and all over the nations during 1999 and 2003;ty, Chongqing city and all over the nations during 1999 and 2001: Mortality rates of infants and under-five children were also higher than that average levels of whole country and Chongqing respectively; Multi-indicators showed that the general population health status of Kaixian was great bad,which stood a distinct gap among average levels of the whole country and Chongqing maintained about 250/100 000 or so, which was higher than the national level (190/10 000), however, the incidence rate of infectious diseases of Kaixian was rising year after year, even that of the recent two years has been reached up to more than 1.5 times higher than that of the whole of the water-based infectious diseases, especially the virus hepatitis, was severe. Another potential impact on population health of three-gorge dam is the emergence of natural epidemic diseases and insect-based infectious diseases. Malignant tumor such as hepatocarcinoma.CONCLUSION: We should strengthen current public health system, and cognition to the possible threat on disease epidemic and public health.With the construction of three-gorge dam, it is necessary to speed up establishment of effective measures to reduce predictable influence on population health.
10.The expression and its clinical significance of microRNA-301 in pancreatic carcinoma

Objective To study the expression of microRNA-301 in pancreatic carcinoma andvalidate the significance of miR-301 in invasion and metastasis of pancreatic carcinoma.Methods miR-301 expression were detected by FQ-PCR in 5 pancreatic cancer eell lines(PANC-1,PaCa-2,AsPC-1,Hs766T.BxPC-3).Further immunohistochemistry in pancreatic cancer tissue microarrays was detected miR-301 expression,which contained 60 pancreatic cancer specimens along with 10 normal adjacent tissues and 10 normal pancreas tissues.After high expression of miR-301 in pancreatic carcinoma being confirmed.the clinical significance of high expression of miR-301 in invasion and metastasis of pancreatic carcinoma were studed.Pancreatic cancer cell lines(PANC-1.PaCa-2)were transfected by 100 nmoml/L miR-301 inhibitor(anti-miR-301)or negative eontrol(Anti-miR~(TM) Negative Control#1).COX-2 and MMP-2 protein expression in pancreatic cancer cell lines were detected by WB.and cell migration assays were performed using transwell technology.Results FQ-PCR resuhs indicated that miR-301 expression was higher in pancreatic cancer cell lines than normal pancreatic cells.The relative level of miR-301 in 5 pancreatic cancer cell lines(PANC-1,PaCa-2,AsPC-1,Hs-766T,BxPC-3)and normal pancreatic cell were 33.09± 4.21,30.76±3.18,47.57±3.56,20.20 ±1.21,76.75±13.51 and 1.00±0.08 respectively.The miR-301 level in all 5 pancreatic cancer cells were significantly higher than those of normal pancreatic cell(t=8.86,9.53,6.39,6.77,11.18,P<0.01).Immunohistochemistry results also showed miR-301 expression was higher in pancreatic carcinoma tissues than those in the cancer adjacent tissues and normal pancreatic tissues.The relative levels of miR-301 in pancreatic carcinoma tissues.normal adjacent tissues and normal pancreas tissues were 0.88±0.09,0.22±0.04 and 0.14±0.05 respectively.The miR-301 levels in pancreatic carcinoma tissues were significantly higher than those of normal adjacent tissues and normal pancreatic tissues(t=15.1,10.6,P<0.01).There was no significant difference between normal adjacent tissues and normal pancreas tissues(t=1.32,P=0.22).After miR-301 inhibitor was introduced into pancreatic cancer cells PANC-1 and PaCa-2.miR-301 levels were reduced while the protein levels of COX-2 and MMP-2.which were invasion and metastasis related factors,were down-regulated.The cell migration assay indicated the numbers of PANC-1 and PaCa-2 cells,which migrated to lower chamber.were 587±27 and 363±13 respectively after miR-301 inhibitor was applied.The numbers of migrated cells were 1091 4-15.737±44 when the netative control was applied.The cell invasion ability was decreased significantly in the inhibitor group compared with the negative group(t=7.89,7.56,P<0.01).Conclusions miR-301 is highly expressed in pancreatic cancer cell lines and pancreatic cancer tissues.Inhibition of miR-301 expression can effectively supress the invasion of pancreatic cancer cells.miR-301 may serve as a new biomarker for early detection of pancreatic cancer and molecular target for early treatment of pancreatic cancer.
