1.Correlation of left ventricular geometry with left atrial structure and function in essential hypertension

Objective To investigate the effects of the left ventricular geometry on left atrial structure and function in essential hypertension (EH).Methods One hundred and thirty-eight patients with EH and 86 normal controls were studied with echocardiography.According to left ventricular mass index (LVMI) and relative wall thickness (RWT),a total of 138 patients with EH were divided into normal geometry group (58 cases),concentric remodeling group (22 cases),concentric hypertrophy group (21 cases) and eccentric hypertrophy group (37 cases).Correlation between left atrial diameter index (LADI),left atrial ejection force (LAEF) and all parameters was respectively obtained using linear regression analysis,and the stepwise regression analysis was used to assess the independent effect of each parameter.Results Compared with control group,LADI and LAEF were higher ( P
2.Quantitative Evaluation of the Left Ventricle Diastolic Function in Chronic Mitral Regurgitation by Pulsed Wave Doppler Echocardiography

Pulsed wave Doppler echocardiograpdy was performed in eighteen cases with chronic mitral regurgitation(CMR)and 21 normal individuals td detect the quantitative changes of the mitral diastolic flow spectrum.The results showed that the pseudonormalization of mitral flow spectrum in CMR was actually caused by impaired left ventricle(LV)active relaxation and decreased LV compliance:It thus masked the LV diastolic dysfunction in CMR.E-wave pressure gradiant(E-PG),A-wave pressure gradiant(A-PG),isovolumic relaxation time(IVRT),E deceleration time(E-DT)and LV filling time(LVFT)in CMR group were significantly different in from those in normal group(P<0.01).Therefore it is essential to integrate these parameters as the indexes to evaluate the LV diastolic dysfunction in CMR.
3.Peritumoral Diffusion Tensor Imaging of High Grade Astrocytoma and Metastasis

Objective To study the value and clinical significance of diffusion tensor imaging(DTI)in differentiating high-grade astrocytomas and solitary metastatic tumors of brain.Methods 16 patients with intracranial solitary metastasis and 25 patients with high-grade astrocytoma confirmed pathologically were examined with MR DTI.Mean diffusivity(MD)and fractional anisotropy(FA)were measured in the regions of interest(ROI)of peritumor and cerebral parenchyma of the normal side on DTL Fiber tractography was reconstructed and the encountered patterns of tumor-related alteration to cerebral white matter were observed.Results The peritumor were all depicted as hyperintense or isointense signals on DTI.The FA values were 0.227±0.05 and 0.169±0.07 in the peritumor of high-grade astrocytoma and metastasis respectively,which was of significant difference(P<0.05).Furthermore,fiber tractography could display the course of white matter tracts in three-dimensional space.Conclusion DTI may be help to differential diagnosis of solitary metastasis and high-grade astrocytoma in clinical practice.
4.Expression of NF-κB and dendritic cells in renal tissue of lupus nephritis

Objective To explore expression of nuclear factor kappa B (NF-κB) and change of peripheral blood dendritic cells(DCs) surface. Methods The expression of NF-κB and DCs in kidney of 40 LN cases were measured,by ALP-anti-ALP mice immune complex law(APAAP) and density gradient separation centrifugal meth-ods. Results kidney tissue of LN patients, NF-κB in the glomeruli and tubules showed a high level of expression ; CD80+ ,CD86+, HLA-DR+ expression decreasedtacrolimus group than the control group Division before and after the shape of the DCs had no effect on tacrolimus after the DCs could inhibit the proliferation of T cells and T cells to Th2 cell transformation. Conclusinn In vitro Tacrolimus inhibits the DCs mature in LN patients, NF-κB in LN kidney LN local expression and the occurrence and development of related.
5.The Efficacy Analysis of U100plus Laser in Treatment of Incarcerated Gallbladder Stones

Objective To investigate the application of U100plus laser in treating incarcerated in gallbladder stones. Methods The clinical data of 52 patients accepted U100plus laser lithotripsy of gallbladder neck incarcerated were retrospective analyzed. Results All 52 patients were successful treated by surgery. Twelve patients with gallbladder wall edema, drainage tubes were used to prevent bile leakage after gallbladder surgery. After 2~4 d, the ultrasound examination showed that no abnormal drainage, and tube was removed. The mean operative time was 40 min. The mean intraoperative bleeding was 2 mL. Patients were able to get out of bed 1 day after surgery. The average length of hospital stay was 4 d. Pa-tients were followed up for 6 to 30 months, and no special discomfort, The ultrasonography showed no stone recurrence. Conclusion U100plus laser lithotripsy is a safe and effective method for treating incarcerated gallbladder stones.
6.Recombinant human granulocyte-macrophage colony stimulating factor in the treatment of drug induced skin ulcer

BACKGROUND: Recombinant human granulocyte-macrophage colony stimulating factor (rhGM-CSF) could stimulate the proliferation of fibroblasts, keratinocyte and skin mucosae cells to different degrees.OBJECTIVE: To observe the effect of rhGM-CSF on the healing of drug exosmose induced skin ulcers.DESIGN: A randomized and controlled animal experiment.SETTING: Department of Respiratory Medicine, Xijing Hospital, Fourth Military Medical University of Chinese PLA.MATERIALS: This experiment was carried out at the Department of Respiratory Medicine, Xijing Hospital, Fourth Military Medical University of Chinese PLA from June to November 2004. Totally 20 male Kunming white mice, with body mass of 18 to 24 g, were chosen.METHODS: Prepared skin ulcers animal models were randomly divided into control group and treated group with 10 white mice in each group.Mice in the control group were given 1mg phentolamine ,20 mg lidocaine and 1mg dexamethasone diluted by normal saline to 0.5 mL ,then sealed up , once a day for 7 days; 25 μg rhGM-CSF was diluted by normal saline to 0.5 mL , then the solution was injected into the periphery of ulcers of mice in treated group , once every other day, for 7 days. Healing time and histological change of skin tissue at ulcer were observed.MAIN OUTCOME MEASURES: To observe the effect of rhGM-CSF on the healing time of drug exosmose induced skin ulcer and anabrosis and histological changeRESULTS: Totally 20 mice entered the stage of result analysis. ①Healing time: the healing time of ulcer and erosion was significantly longer in control group than in treated group [(20-24,8-12)d,t=2.264,P=0.01];②Histological observation: hyperplasia of granulation tissue was not obviously on 7 days after treatment in control group; Hyperplasia of granulation tissue appeared and the newly born blood vessel was abundant on 7 days after treatment in the treated group.CONCLUSION: rhGM-CSF can promote the wound healing of drug induced anabrosis and ulcer.
7.Cloning,expression and characterization of sLAG-3-Ig

AIM: To obtain a vaccine with sLAG-3 as immunoadjuvant and investigate its biologic activity in order to establish the safe and effective way for asthma as one of the specific immunotherapy.METHODS: The coding sequence of LAG-3 was amplified by polymerase chain reaction,the expression vector pcDNA-sLAG-3-Ig was constructed by inserting the PCR products of sLAG-3 and Fc sequence of IgG.With electroporation transfection,pcDNA-sLAG-3-Ig was transfected into COS-7 cells and its biologic activity was investigated by Western blotting analysis.RESULTS: By temperature induction,the LAG-3-Ig was highly expressed in E.coli DH5?.LAG-3-Ig fusion protein was observed by SDS-PAGE and Western blotting,the results showed that the LAG-3-Ig protein was an antagonist of the IL-4-induced synthesis of IgE in B cells.CONCLUSION: A new vaccine with sLAG-3 as immunoadjuvant was obtained.It could inhibit synthesis of IgE in B cells.Thus,LAG-3-Ig would be hopeful to establish the safe and effective way for asthma as one of the specific immunotherapy.
8.The effect of instrumented respiratory training on the respiration of patients with stable chronic obstructive pulmonary disease

Objective To explore the effectiveness of a respiratory function training instrument with stable chronic obstructive pulmonary disease (COPD) patients.Methods Sixty-seven COPD patients in the stable period were randomly divided into a treatment group of 36 and a control group of 31 using a random number table.Both groups were given conventional pulmonary rehabilitation,including half-closed lip respiration,abdominal respiration and upper limb training.The treatment group was additionally provided with 30 minutes of respiratory training using a respiration function training instrument 5 times per week for 6 months.Both groups were assessed for their mobility,life quality and pulmonary function using the 6-minute walk test (6 MWT),a COPD assessment test (CAT),the BODE index,forced vital capacity (FVC),forced expiratory volume in one second (FEV1) and surface electromyography (SEMG) of the respiratory muscles before and after the 6-month intervention.Results Before the treatment there were no significant differences between the two groups in terms of any of the measurements.After the treatment,significant improvement was observed in the average 6 MWT,CAT,BODE index and SEMG results in both groups,but with significantly greater improvement in the treatment group.The average FVC and FEV1 results did not improve significantly,so after the intervention there was still no significant difference between the groups.Conclusions Respiratory training using the pulmonary function training instrument can improve the mobility,life quality and the functioning of the respiratory muscles of COPD patients in the stable period.
9.Expression of miR-590-3p in pancreatic cancer stem cells

Objectives To isolate cancer stem cells (CSCs) in pancreatic cancer cell lines PANC1 and ASPC-1 with serum-free medium( SFM ), and to detect the expression of miR-590-3p in CSCs. Methods PANC1 and ASPC-1 cells was cultured in serum-free medium. The monoclonal formation, differentiation and cell cycle, half inhibitory concentration ( IC50 ), and the expression of the surface markers CD24 + , CD44 + were detected. qRT-PCR was used to detect the expression of miR-590-3p. Results After SFM culture, (0.94 ±0.53 ) % of ASPC-1 and (0.57 + 0. 12 ) % PANC1 survived, and they formed spheres, and could continuously passage in vitro. Cell spheres differentiation recurred when serum was supplemented in SFM. The G0/G1 stage proportion, CD24+ , CD44 + , CD24+ CD44+ cells proportion, IC50 in ASPC-1 cell were (75.3 ± 5.4)%,0.96% ~ 2.01%, 27.52% ~ 34.47%, 0.35% ~ 0.44% and (224.37 ± 5.71 ) μg/ml, which were significantly higher than that those in parent cell [ (43.7 ± 3.8 ) %, 0. 38% ~ 0.42%, 17.65% ~ 18.25%,0.05% ~0.08%, (11.43 ±2.10)μg/ml, P<0.05]. The G0/G1 stage proportion, CD24+ ,CD44+ ,CD24 +CD44 + cells proportion, IC50 in PANC 1 cell were ( 80. 1 ± 4.7) %, 5.31% ~ 9.84%, 72.05% ~ 93.06%,4.91% ~5.21%, (296.58±4.27) μg/ml, which were significantly higher than that those in parent cell [ (46.1 ±5.3)%, 4.09% ~4.97%, 47.71% ~55.66%, 1.48% ~2.63%, (26.17 ±3.81)μg/ml, P<0.05]. The expression of miR-590-3p in ASPC-1, PANC1 spheres was 4.67 and 4.52 times higher than the expression in parent cell lines. Conclusions Pancreatic cancer cell spheres can be isolated from ASPC-1, PANC1 by culture with SFM. miR-590-3p is up-regulated and may play an important role in regulating biological characteristics of pancreatic cancer stem cells.
10.Advances in two-photon imaging technology.

As a new kind of advanced nonlinear imaging approach, two-photon fluorescence microscopy technology is wildly used in the field of live cell and tissue imaging, especially focusing on long-term dynamic three-dimensional cell imaging. This paper firstly presents the principle and characteristic of two-photon fluorescence microscopy. Then, the paper focuses on the three key aspects of the viability of the specimen, sensitivity of detection, as well as the speed of acquisition. In the end, the future prospect of development and application of two-phonon imaging technology are predicted.
