1.Study on aminoglycoside resistance and drug resistance gene of ESBLs-producing Escherichia coil

Objective To investigate the characteristics of aminoglycoside resistance of extend-ed-spectrum β-lactamases(ESBLs)-producing Escherichia coli(E, cold and expression of aminoglyco-side-modifying enzyme genes. Methods The minimal inhibitory concentrations(MICs) of gentamicin,amikacin, kanamycin, tobramycin, netilmicin and neomycin for 37 strains of ESBLs-producing E. Coli were detected by agar dilution. In addition, six aminoglycoside-modifying enzyme genes were amplified by polymersae chain reaction(PCR) and verified by DNA sequencing. Results MICand MIC90 of gentamicin, amikacin, kanamycin, tobramycin and netilmicin for 37 strains of ESBLs-producing E. Co-Il all excelled 256 μg/mL, the resistance rates of the above antibiotics were 78.4%, 45.9%, 72.9%,83.8%and 64.90%, respectively. However, neomycin still had powerful antibacterial activity. In ad-dition, five modifying enzyme genes, including aac(3)-Ⅱ , aac(6′)-Ⅰ b, aac(6′)-Ⅱ , ant(2″)-Ⅰ and ant(3″)- Ⅰ genes, were found in 37 isoaltes except aac(3)- Ⅰ , and their positive rates were 56.8%,27.0 %, 2.7 %, 5.4 % and 13. 5 %, respectively. Conclusion The aminoglycoside resistance of ES-BLs-producing E. Coil may be associated with the expression of aminoglycoside-modifying enzyme genes.
2.Effect of astragalus polysaccharides and soy isoflavones on glucose metabolism in diabetic rats

Objective To explore the effect of astragalus polysaccharides (APS) and soy isoflavones (SI) on glucose metabolism in diabetic rats. Methods Healthy 60 rats were equally and randomly divided into 5 groups, normal forage group (NF), high blood sugar model group(HBS), APS high dose group intervention group (APS-H), APS low dose group (APS-L), and SI intervention group (SI). Diabetic model was inflicted by intraperitoneal injection of streptozotocin (STZ). Ordinary feed and APS high dose,low dose APS and SI interfere were respectively given for 8 weeks. Blood glucose,glycated hemoglobin,glucose and lipid levels were determined in every group. Results After APS high dose,low dose and SI were given to feed diabetic rats for 8 weeks, their blood glucose, glycated hemoglobin, glucose, TC, TG, LDL, VLDL and high blood glucose was significantly lower than the HBS group (P
3.Analysis of the Antibiotic Resistance Rate of Helicobacter Pylori

Objective To investigate the antibiotic resistance rate of Helicobacter pylori,and to provide reliable evidence of choosing optimal drugs for clinical therapy.Methods Overall 3304 patients were enrolled from February 2006 to March 2007 who accepted gastroscopy followed by Helicobacter pylori culture of the gastric mucosa from biopsy.The antimicrobial susceptibilities of Helicobacter pylori strains for six antibiotics including metronidazole,amoxicilin,gentamycin,levofloxacin,furazolidone,clarithromycin were observed by Kirby-Bauer disk diffusion test.Results 1171(35.4%)biopsy clinical specimens were proved to be Helicobacter pylori positive when cultured in vitro and identified by conventional assays.The metronidazole resistance rate(99.32%)of Helicobacter pylori was significantly higher than that of any of the other 5 antibiotics,and the levofloxacin resistance rate(0.51%)was the lowest.Furthermore there were no significantly change in the tendency of resistance rate for the two above antibiotics from February 2006 to March 2007.However,Helicobacter pylori was resistant to amoxicilin,gentamycin,furazolidone,clarithromycin,and the resistance rate was 61.06%、4.78%、21.35% and 39.45% respectively.Meanwhile,the resistance rates corresponding to four antibiotics were different obviously.Conclusions With the application of antibiotics for Helicobacter pylori eradication,the antibiotic resistance rate increased perceptibly.The rate of resistance to metronidazole appeared to be the highest among six antibiotics applied to clinical treatment.Drug susceptibility testing was valuable to eradicate Helicobacter pylori in clinical therapy.

The incidence of thyroid cancer in South Korea increased 15 times over the past twenty years, that made South Korea became a country of the highest reported incidence in thyroid cancer all over the world. But the mortality of thyroid cancer was stable and unchanged. The incidence of thyroid cancer in our country was growing at an annual percentage change of 20. 1%. It is a generally accepted that the sharply increased incidence is attributed tooverdiagnosis. Here we introduced the definition of overdiagnosis of cancer and the current understanding in tumor field. The corresponding strategy in our country was proposed.

Objectives To study the effect of highway transportation on the quality of red blood cells. Methods Highway transportation environment was simulated by random vibration test. Red blood cell suspensions and whole blood were vibrated for 120 minutes based upon GB/T 4857.23-2003 with the RMSVA of 0.38 and 0.52grms, as well as GJB 2711-1996 with the RMSVA of 1.04grms and weight loading of 60kg. To study the lipoidal constituents of Setaria italica Beauv..Methods The constituents were isolated by chromatography and structurally elucidated on the basis of spectral analysis.Results The three glycerides were isolated as dilinolein (Ⅰ), monolinolenin (Ⅱ),α, β-digalactosyl-α′-linolenic-glyceride (Ⅲ).Conclusion These compounds were obtained from the plant for the first time. Compound Ⅲ is a new glycolipin.
10.IN VITRO ANTI-TUMOR EFFECTS OF fas GENE ON OESOPHAGEAL CANCER CELLS

To evaluate the inhibitory effects of fas gene transduction on oesophageal cancer cells in vitro, the eukaryotic expression vector fas pBK was constructed and transfected into oesophageal carcinoma cells EC109. Western blot results showed high expression of Fas protein in gene transfected cells Fas EC109. Cell growth curve and plating efficiency test revealed that Fas EC109 had longer population doubling time and lower plating efficiency compared with control cells. Results of MTT assay showed increased sensitivity of Fas EC109 cells to CDDP, VCR and 5 FU. All these data suggested that transgenic expression of fas gene could effectively inhibit the proliferation and increase the drug sensitivity of oesophageal carcinoma cells in vitro.
