1.Enhancing Effects of Intraperitoneal Injection with IL-2 Recombinant Adenovirus and IL-3 Recombinant Adenovirus on the Anti-Leukemia Activity of Chemotherapy

The Ad-IL-2 and/or Ad-IL-3 were i. p. injected into experimental leukemic mice after high dose chemotherapy. When Ad - LacZ was i. p. injected into leukemic mice, there were leukemic cells in bone marrow, vessal, liver and spleen. After i. p. injection of Ad-IL-2 and/or Ad-IL-3, the leukemia - bearing mice showed significant inhibition of tumor growth. As i.p. injection of Ad-IL-2, the splenic NK, CTL activity increased; as i. p. injection of Ad - IL - 3, the number and cytotoxicity of peritoneal macrophages were significantly higher than that of the control groups. After i. p. injection of Ad - IL - 2 and Ad - IL - 3 at the same time, the anti - leukemia activity was the best. Though there were less leukemic cells in the bone marrow, there were no leukemic cells in the liver and spleen. Our results suggested that i. p. injection of Ad - IL - 2 and Ad - IL - 3 could significantly enhance the anti - leukemia activity of chemotherapy.
2.Isolation and change of drug resistance of mucoid Pseudomonas aeruginosa in sputum from 2013 to 2015 in a hospital

Objective To understand the isolation and change of drug resistance of mucoid Pseudomonas aeruginosa in sputum from 2013 to 2015 so as to provide a scientific basis for reasonable clinical use of antibacterial drugs.Methods The isolation,culture and identification were carried out according to the fourth edition of National Clinical Laboratory Operation Rules.The drug susceptibility testing was performed with the use of Kindy-Bauer.The drug susceptibility testing results were judged according to the standards of CLSI 2014.The clinical data were analyzed by software WHONET 5.6.Results Among 1 653 strains of Pseudomonas aeruginosa isolated in the clinical sputum specimens from 2013 to 2015,including 255(15.4%)strains of mucoid Pseudomonas aeruginosa;during the three years,the drugs of highest drug resistance rate was piperacillin,which was 29.0%;the drug resistance rate less than 10.0% had amikacin and piperacillin/tazobactam;the drugs with larger increase range of resistance rate included imipenem,levofloxacin,ciprofloxacin and ciprofloxacin,their resistance rates were increased from 12.5%,9.7%,8.3% and 9.7% in 2013 to 20.5%,17.0%,19.3% and 15.9% in 2015.Conclusion The isolating rate of mucoid Pseudomonas aeruginosa in sputm is gradually increased year by year from 2013 to 2015.At the same time,the drug resistance rates show an increasing trend in recent three years.It is necessary to strengthen the surveillance of drug resistance of mucoid Pseudomonas aeruginosa.
3.Therapeutic efficacy in patients with infectious atypical pneumonia:comparison of different protocols

Objective To observe the therapeutic efficacy of different schemas in partients with contagious atypical pneumonia outbreaking in GuangZhou area.Methods Patient who were hospitalized in our ward from Feb,2003 to May,2003 were allocated to different groups according to the therapentic schema.To compare and analyse the efficacy.Results resnlts indicate that the effects of antivirus drugs of all groups have no difference.The use of glucocorticoid decreased the opportunity of mechanical ventilation and intubation.Conclusion Patients who have severe condition or present with adult respiratory dyspnea syndrome will relieve in pulmonary inflammatory bleeding at some degree if they get proper glucocorticoid and positive pressure nonivasive ventilation,the possibility of intubation and fatality will decrease
4.Detection and drug susceptibility of Ureaplasma urealyticum and Mycoplasma hominis in 8 546 cases of genitourinary tract specimen

Objective To investigate the mycoplasma separation rate in the patient with genitourinary tract inflammation in our hospital and the in vitro sensitivity to 12 kinds of antibacterial drugs for guiding clinical rational drug use.Methods 8 546 cases of genitourinary tract specimen were collected from our hospital STD clinical department and performed the culture and the in vitro drug susceptibility test by adopting the mycoplasma culture,identification and drug susceptibility integration reagent kit.Results Among 8 546 cases of patient specimen,3237 cases were mycoplasma positive,in which 2 897 cases (89.5%)were simple Ureaplas-ma urealyticum(Uu)positive,39 cases (1.2%)were simple Mycoplasma hominis (Mh)positive and 301 cases (9.3%)were Uu and Mh positive,there was statistically significant difference between any two groups (P <0.01 );the drug susceptibility test showed that the top 3 of sensitivity rates to antibacterial drugs in 2 897 cases of Uu infection from high to low were josamycin (98.4%),doxycline (97.9%)and minomycin (97.6%),the top 3 of drug resistance rates from high to low were ciprofloxacin (71.8%),spiramycin (46.8%)and ofloxacin (35.6%).The top 3 of sensitivity rates to antibacterial drugs in 39 cases of Mh infec-tion from high to low were josamycin (96.7%),doxycline (91.2%)and minomycin(90.8%),and the top 3 of drug resistance rates were ofloxacin (68.9%),ciprofloxacin (67.8%)and roxithromycin (54.2%).The top 3 of sensitivity rates to antibacterial drugs in 301cases of Uu and Mh infection from high to low were doxycline (95.2%),minomycin (94.8%)and josamycin (92.1%),and the top 3 of drug resistance rates from high to low were ciprofloxacin (83.3%),ofloxacin (80.1%)and levofloxacin (76.3%).Conclu-sion Uu and Mh are the common pathogens of genitourinary tract infection.Mycoplasma isolated from genitourinary tract speci-mens in our hopsital is dominated by Uu,so the drugs with higher sensitivity such as josamycin,doxycycline and minocycline should be used as the first choice in the treatment of mycoplasma infection.

6.The effect of mannose-6-phosphate on adipocyte differentiation and its underlying molecular mechanism

Objective Cathepsin K (CTSK) played an important role in adipocyte differentiation.The activation of CTSK needs to convey by mannose-6-phosphate receptors (M6PR) in osteoclasts. The aim of the present study was to identify the effects of mannose-6-phosphate (M6P) in adipocyte differentiation and its underlying molecular mechanism. Methods Oil red O staining, accumulation of cytoplasmic triglycerides and glycerine release were used to assess its effects on adipocyte differentiation in the 3T3-L1cell line. The enzyme activity of CTSK was observed by laser confocal microscopy. The proliferation of 3T3-L1 preadipocytes was detected by MTT methods. mRNA expression of M6PR was determined by RTPCR. Results M6P could prevent adipocyte differentiation in a dose-dependent manner as evidenced by absence of triglyceride accumulation and glycerol content. Statistical significance was showed when the concentrations of M6P were 5.0 mmol/L and 8. 0 mmol/L respectively(P <0. 05). The mRNA expression of M6PR was detected during the whole process of adipocyte differentiation. With the increase of M6Pconcentration, enzyme activity of CTSK was inhibited in a concentration-dependent manner. MTT method showed that the absorbance at 570 nm of 3T3-L1 preadipocytes was 0. 057 ±0. 091, increased about 62. 9%at 10. 0 mmoL/L compared with the control group (P < 0. 05 ). Conclusion M6P inhibits the terminal differentiation of adipocyte, which may be associated with its effect of blocking CTSK activity by competitive binding with M6PR.
7.Prognostic value of heart rate turbulence in patients with chronic heart failure

Objective To investigate the prognostic value of heart rate turbulence (HRT) in patients with chronic heart failure (CHF). Methods From October 2006 to May 2009, a total of 96 elderly CHF inpatients were selected, and the clinic data were recorded. Based on the echocardiogram and dynamic electrocardiogram, the differences of sinus HRT index of patients with different cardiac function classification were analyzed. During 9-28 months medical follow-up, the treatment endpoint was death from heart disease. Based on Logisitc regression, the prognostic values of HRT, age, hypertension, diabetes, myocardial infarction (MI), left ventricular ejection fraction, ACEI and β- adrenergic blocker for death of CHF patients were analyzed.Results There was no significant difference in HRT between grade Ⅱ and grade Ⅲ cardiac function (χ~2 = 1.60, P>0. 05), and between grade Ⅲ and grade Ⅳ cardiac function (χ~2 = 1.43, P>0. 05). But there was significant difference in HRT between grade Ⅱ and grade Ⅳ cardiac function patients χ~2 =9.84, P<0. 05), and HRT was weaken in grade Ⅳ group. The average follow-up time was (18. 0±9.6) months. Of all 96 patients, there were 34 dead of heart disease. There were correlations of death of CHF with HRT, low LVEF (≤ 45%), age (≥65 years), diabetes, MI and classification of heart function. Conclusions The sinus HRT in CHF patients has a favorable prognostic value.
8.Resistant Genes and Cluster Analysis in Multidrug-resistant Pseudomonas aeruginosa

OBJECTIVE To investigate resistant genes encoding ?-lactamases and aminoglycoside modifying enzymes in Pseudomonas aeruginosa isolated from clinical specimens,and phylogenetic analysis was performed.METHODS Antimicrobial susceptibility test was performed by PhoenixTM-100 system.Resistant genes encoding ?-lactamases,aminoglycoside modifying enzymes and antiseptic resistance were detected by PCR amplification and verified by DNA sequencer.RESULTS The resistant rates of ?-lactams including ampicillin/sulbactam,piperacillin,piperacillin/tazobactam,cefotaxime,ceftazidime,cefepime,imipenem and meropenem in 190 strains of P.aeruginosa were 98.9%,59.5%,45.8%,77.4%,34.2%,38.4%,15.3% and 6.8%,respectively.Ciprofloxacin and levofloxacin still showed powerful activities with resistance being 15.3% and 21.0%.The positive rates of blaVEB, blaGES and blaCARB genes were 9.5%,9.5% and 57.1% in 21 isolates.Twenty strains lost oprD2 genes.However,the ?-lactamase genes of TEM,SHV,OXA,PER,IMP,VIM,SPM,GIM and DHA were not found.Three resistant genes encoding aminoglycoside-modifying enzymes were found in 21 isolates,such as aac(6′)-Ⅰ,aac(6′)-Ⅱ and ant(2″)-Ⅰ,and they accounted for 9.5%,61.9% and 66.7%,respectively.The positive rate of qacE△1-sul1 genes was 66.7% in 21 isolates.CONCLUSIONS P.aeruginosa isolated in clinic has carried many resistant genes.The loss of oprD2 gene may be the important cause of P.aeruginosa resistant to imipenem.Cluster analysis indicates that the spread of clones occurred in our hospital.
9.Corticosteroids and hyaluronic acid effects on four growth factors in tendon tissues of rat rotator cuff injury models

BACKGROUND: Tissue engineering plays an important role in treating rotator cuff injury. Promoting the healing of injured rotator cuff and preventing adhesion by regulating growth factor expression are main functions. OBJECTIVE:To compare the effects of corticosteroids and hyaluronic acid on the expressions of epidermal growth factor (EGF),platelet-derived growth factor (PDGF),transforming growth factor ? (TGF-?) and basic fibroblast growth factor (bFGF) in injured rotator cuff. DESIGN,TIME AND SETTING: The randomized controlled animal experiment was performed at the Animal Experimental Center,School of Medicine,Shanghai Jiao Tong University from July to September 2007. MATERIALS: Totally thirty-six Sprague-Dawley (SD) rats were randomly assigned into four groups: normal group (n=6),rotator cuff injury group (n=6),hyaluronic acid group (n=12) and corticosteroids group (n=12). METHODS: Full-thickness defects across 50% of the total width (5 mm) of the bilateral infraspinatus tendon were excised. Models of rotator cuff injury were created at the synovium. 0.05 mL hyaluronic acid was injected into the subacromial bursa mucosa in the hyaluronic acid group,whereas 0.05 mL corticosteroids in the corticosteroids group. MAIN OUTCOME MEASURES: At three and six weeks after injury,the infraspinatus were harvested to detect EGF,PDGF,TGF-? and BFGF by reverse transcription-polymerase chain reaction (RT-PCR). 10.Genotyping of Acinetobacter calcoaceticus-Acinetobacter baumannii complex and analysis of antibiotics resistance

Objective To identify strains of Acinetobacter(A.)calcoaceticus-A.baumannii complex accurately,and investigate the species distribution of Acinetobacter calcoaceticus-A.baurnannii(ACB)complex isolated in Daping Hospital.Analysis the anti-biotics resistance of ACB complex.Methods A total of 95 clinica1 isolates of ACB complex were collected from Daping Hospital from February to April 2014,identify by sequence analysis of 1 6S rRNA gene spacer region.Use VITEK-2compact automatic iden-tification of bacteria by susceptibility meter to detect the precise identification of all ACB complex group of drug sensitivity.Results Among all the 95 strains of ACB complex identification results were A.baumanni (81,85.26%),A.pittii(10,10.53%),A.noso-comialis (4,4.21%).A.baumannii to third generation cephalosporins,aminoglycosides and carbapenem antibiotic resistance in seri-ous condition,the sensitive rate was only about 20.00%.But A.pittii and A.nosocomialis had a high sensitivity rate to commonly used antibiotics.Conclusion VITEK-2 Compact automatic identification of bacteria susceptibility meter has limitations in identifica-tion of Acinetobacter,with analysis of 1 6S rRNA gene sequencing,the ACB complex could be accurately identified.There is signifi-cant difference in the composite group-resistant characteristics of each species,clinical infection is also various.
