1.Effect of Jianwei Yuyang Granules on expression of epidermal growth factor receptor in gastric mucosa of gastric ulcer patients

OBJECTIVE: To observe the effect of Jianwei Yuyang Granules on the expression of epidermal growth factor receptor (EGFR) in the gastric mucosa of gastric ulcer patients. METHODS: Sixty gastric ulcer patients (final diagnosis by gastroscope) were randomly divided into Jianwei Yuyang Granules treated group (JWYY group, n=30) and western medicine treated group (control group, n=30). Ten patients without gastric mucosa lesion were treated as normal control group. The expression of EGFR in the mucosa was tested by immunohistochemistry and RT-PCR. RESULTS: Expression of EGFR and EGFR mRNA was not observed in patients without gastric mucosa lesion, and slight expression was observed in the edge of ulcer region of the gastric ulcer patients. The expression of EGFR and EGFR mRNA was increased in JWYY group and control group. CONCLUSION: Jianwei Yuyang Granules enhances the expression of EGFR in the mucosa of gastric ulcer patients to prevent peptic ulcer recurrence.

3.Grasp torque control based on experiential learning and haptic feedback

Objective To propose a grasp torque control based on experimental learning and haptic feedback to facilitate the manipulator in dexterous manipulation.Methods An experience database was built firstly,and then the object was recognized by tactile feedback in the grasp task.If the object had been experitentially grasped,the torque was output based on the database.In case a new object was grasped,the optimal output torque was calculated by iterative learning.Results The experiment showed that the robot hand could find the experiential output torque quickly when encountering the object in database and calculate the torque by iterative learning to achieve grasp task.Conclusion The experiential database grows up when the robot hand learns more and more experience.It can fast output torque like human in the grasp task.
4.Up-regulation of telomerase reverse transcriptase expression in guinea pig asthmaticlung tissue and human circulating lymphocytes in bronchial asthma

To investigate the expression of mRNA and protein of telomerase reverse transcriptase(TERT) in guinea pig asthmatic lung tissues and human circulating lymphocytes and to discuss the role of TERT in the pathogenesis of asthma and the clinical significance of expression of TERT in human asthmatic circulating lymphocytes in monitoring the state of asthma.Methods: In 6 guinea pig asthmatic lung tissues, 6 guinea pig normal lung tissues, 16 samples of circulating lymphocytes of asthmatic patients and 14 normal circulating lymphocytes the expression of mRNA and protein of TERT were detected with nuclei acid in situ hybridization and immunostaining. The expression of TERT in circulating lymphocyte were compared with pulmonary function and serum IgE level. Results:The expression of mRNA and protein were up-regulated in asthmatic airway epithelium and lymph nodule germinal center in guinea pig. Compared with control group,high expression were also showed in human asthmatic circulating lymphocytes.The index of TERT expressive positive lymphocytes showed negative correlation with FEV1/ predictive value and MW/predicn've value,positive correlation with serum IgE level. Conclusion: (1 )TERT expression in asthmatic airway epithelium and lymph nodule germinal centers of guinea pig and human circulating lymphocytes were up-regulated; (2)Detection of TERT expression of asthmatic circulating lymphocytes may be useful to monitor the state of asthma.

6.Prevention and treatment of in-stent restenosis after PCI by Guanxintongluo capsule combined dual an-tiplatelet drugs

Objective:To study therapeutic effect of Guanxintongluo (GXTL ) capsule combined dual antiplatelet drugs on prevention and treatment of in -stent restenosis after percutaneous coronary intervention (PCI) .Methods :A total of 82 patients with coronary heart disease undergoing stent implantation treated in our hospital were select‐ed .All patients were numbered ,and they were divided into dual antiplatelet group (n=40) and combined treatment group (n=42 ,received GXTL capsule based on dual antiplatelet group ) according to odd and even number method . Therapeutic effect was compared between two groups .Results:Compared with dual antiplatelet group after treat‐ment ,there were significant rise in total effective rates of in -stent restenosis (77.5% vs .95.2% ) ,angina pectoris (80.0% vs .95.2% ) ,ECG (77.5% vs .92.9% ) and syndrome of traditional Chinese medicine (72.5% vs .90.5% ) in combined treatment group , P< 0.05 all . There were no significant difference in liver function between two groups before and after treatment , P>0.05. Conclusion:Guanxintongluo capsule combined dual antiplatelet drugs can effectively reduce incidence rate of restenosis , improve syndrome of traditional Chinese medicine ,ECG chan‐ges ,reduce pain degree of angina pectoris in patients with coronary heart disease after PCI , which is worth extending .
7.A nonradioactive method for detecting DNA-binding activity of nuclear transcription factors.

To determine the feasibility of a nonradioactive electrophoresis mobility shift assay for detecting nuclear transcription factor, double-stranded oligonucleotides encoding the consensus target sequence of NF-kappa B were labelled with DIG by terminal transferase. After nuclear protein stimulated with phorbol 12-myristate 13-acetate (PMA) or PMA and pyrrolidine dithiocarbamate (PDTC) electrophoresed on 8% nondenaturing poliacrylamide gel together with oligeonucleotide probe, they were electro-blotted nylon membrane positively charged. Anti-DIG-AP antibody catalyzed chemiluminescent substrate CSPD to image on X-film. The results showed that nuclear proteins binded specifically to the NF-kappa B consensus sequence in the EMSA by chemiluminescent technique method and the activity of NF-kappa B in PMA group was more than that in PMA + PDTC group. It is suggested that detection of NF-kappa B by EMSA with chemiluminescent technique is feasible and simple, which can be performed in ordinary laboratories.

8.Clinical research of acute pulmonary embolism after surgery

Objective To analyze the risk factors,clinical characteristics,and missed factors of acute pulmonary embolism (APE) after surgery,so as to advise clinicians to improve understanding of the early postoperative APE and enhance prevention concepts.Methods Retrospectively analyzed the general information of 43 cases of surgical patients with APE after surgery categories,risk factors,clinical manifestations and prognosis.Results APE after surgery had obvious risk factors; often occurred within 2 weeks after surgery; commonly seen in malignant tumors (53.5%,23/43),orthopedics (27.9%,12/43) and other major surgery; clinical manifestations wasn't typical,of which chest distress and dyspnea was the most common symptom (93.0%,40/43),and other pestoperative symptoms were similar,easy to misdiagnosis.Conclusions Surgery is one of the important risk factors of APE.Preoperatively evaluating underlying diseases,postoperative strengthening preventional methods,early detection,early treatment,have great significance in reducing the occurrence of postoperative APE and improve the prognosis.

Objective:To investigate the expression and function of CD226 on NK s ubsets and its coexpression with other activation receptor and inhibition recept or on NK cells. Methods:The expression of CD226 on CD56 bright and CD56 dim NK subsets and coe xpress ion with CD16 and NKG2A in PBMC and MLC in the presence or absence of IL-2 or IL-15 were detected by double fluorescent staining and flow cytometry analysis. The level of IFN-? in the supernatants of PBMC culture and MLC treated with o r without IL-2 or IL-15 were evaluated by ELISA. 51Cr release assay w as employed to measure the specific lysis of NK cells killing target K562 cells. Results:CD226 was mainly expressed on CD56 dim NK subsets in PBMC. When s imulated by IL-2, CD226 expression was shifted to CD56 bright NK subsets, while IL-15 increased the subpopulation of NKG2A+CD226+double positive cell s. In MLC-generated NK cells, CD226 was mainly expressed on CD56 dim NK su bsets, and also shifted to CD56 bright NK subsets in the addition of IL-15 . Furt hermore, the percentage of CD16+CD226+和NKG2A+CD226+ subsets were increa sed when stimulated by IL-2 or IL-15. There was great increase in IFN-? lev el in the supernatants of PBMC culture in the presence of IL-2 or IL-15, but no difference in the supernatants of MLC treated with or without two cytokine s. Moreover, the cytotoxicity of NK cells in PBMC and MLC were greatly enhanced by IL-2 or IL-15. Conclusion:CD226 is mainly expressed on CD56 bright NK subsets in IL-2 or IL-1 5 activated NK cells, and is coexpressed with CD16 and NKG2A preferentiatly, whi ch maybe involve in the modulation of cytotoxicity of NK cells based on the balance of coexpressed activation and inhibition receptors.
