1.OBSERVATION OF ULTRASTRUCTURE AND DRUG SENSITIVITY OF IN VITRO CULTURED EXOERYTHROCYTIC FORM OF PLASMODIUM BERGHEI

The infrastructure of in vitro cultured exoerythrocytic stage (EE) of Plasmodium ber-ghei (P.b.)was observed under transmission electron microscope (TEM).The drug sensitivity of EE was also measured in vitro.The EE was cultured in monolayer host cell,fixed and embeded in situ.The ultrathin sections were prepared and examined by routine methods.The TEM pictures showed that the fine structure of in vitro cultured EE was similar with that of EE grown in rat hepatocytes in vivo,as described by Meis et al.The parasite was found within a parasitophorous vacuole,with nucleus,mitochondria,endoplasmic reticuia and Golgi apparatus were observed inside the parasite.After 24h cultivation,the EE was incubated for 48h in medium containing a serial concentrations of primaquine or chlo-roquine,then examined under light microscope.The percentage of abnormal parasites was calculated.The preliminary results showed that the sensitivity of cultured P.b.EE to primaquine and chloroquine was significantly different.At the same concentration of 1?10-5 mol/L,the percentage of abnormal parasites was 38.5?3.9% and 5.7?1.9% ,respectively These results demonstrated that the in vitro cultivation system of P.b.EE would have potential utility in antimalarial drug research.(Figs.1-6)
2.COMBINED TRANSPLANTATION OF FETAL PANCREAS AND THYMUS FOR THE TREATMENT OF DIABETES MELLITUS I: A REPORT OF ONE-YEAR OBSERVATION

Combined transplantation of fetal pancreas and thymus was performed in 10 patients suffering from severe diabetes I. They were closely followed for over one year, and it was found that among them 7 showed spectacular clinical improvement, including 2 with total discontinuation of insulin, and 5 with average reduction of insulin by 82%. In 3 patients, the dosage of insulin was reduced by an average of 56.9%. The average reduction of insulin for the 10 patients in 2 months, 6 months and one year after the transplantation was 68.3%, 74.5%, and 78.4%, respectively. The effect of warm ischemia of different durations on the donor tissues was studied with electron microscopy. The significance of combined transplantation and the relationship of the techniques of procuring the donor tissues and transplantation is discussed.
3.Clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage

Objective To explore the clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage.Methods One hundred and twenty children with rotaviral enteritis and myocardial damage were randomly divided into the control group (n =60) and observation group (n =60).Two groups were given antiviral,rehydration,nutritional support,redressing acid-base imbalance and electrolyte disorders and other conventional treatment.The patients of the control group were given 160 mg/(kg · d) 1,6Fructose Diphosphate(1 times/d) on the basis of routine treatment,while those in the observation group were given 1.0 g creatine phosphate + 150 ml 5% glucose injection(1 times/d) on the basis of routine treatment,both courses of two groups were 7 days.The levels of myocardial enzymes were measured and the clinical effect and adverse reactions were recorded.Results The total effective rate in observation group was 95.0% (57/60),obviously higher than that of control group (80.0% (48/60)) and the differences were statistically significant (x2 =6.268,P < 0.05).There were no statistical significant difference in terms of myocardial enzyme between the two groups before treatment(P >0.05).The level of lactate dehydmgenase(LDH),creatine kinase (CK),creatine kinase isoenzyme (CK-MB) and aspartate aminotransferase (AST) of observation group were (163.8 ±18.5) U/L,(152.8±26.4) U/L,(19.4±7.8) U/Land (24.4 ±6.8) U/Lafter treatment,and (193.4 ± 22.8) U/L,(168.6 ± 32.6) U/L,(27.4 ± 9.2) U/L and (35.4 ± 9.8) U/L of control group,and the differences between the two groups were significant (t =3.484,3.228,4.278 and 4.729; P < 0.05).There was no obvious adverse reaction during the treatment course.Conclusion The clinical effect of creatine phosphate on infantile with rotaviral enteritis and myocardial damage is remarkable and safe,and it is worth of clinical application.
4.Investigation on testing results of 5 blood indicators in unpaid blood donors in Handan region during 2002-2012

Objective To understand the status of the blood safety in Handan region to provide the basis for recruiting blood do-nors in our region and the data support for meeting clinical blood use.Methods The detection results of 5 legal indicators among unpaid blood donors in Handan region during 2002-2012 were investigated and analyzed.Results The total positive rate of indica-tors detection was 5.52%,in which alanine aminotransferase(ALT)was 4.06%,hepatitis B surface antigen(HBsAg),hepatitis C virus antibody(anti-HCV)was 0.54%,human immunodeficiency virus(HIV)antibody(anti-HIV)was 0.24% and treponema palli-dum antibody(anti-TP)was 0.12%.Conclusion This survey is conducive to the blood donor recruitment work and strengthening the screening of blood donors is helpful for safety of clinical blood use.

6.Determination of Four Kinds of Ingredients in Transparent Absorbent Fluid of Different Size of Huoxue Powder by Ultra Performance Liquid Chromatography Tandem Mass Spectrometry

Objective To optimize the particle size of Huoxue powder by contents comparison of emodin,phellodendrine,berberine and jatrorrhizine before and after permeabilized skin absorption of Huoxue powder in different particle size of 0.150,0.075,0.048,0.038 mm.Methods The contents of emodin,phellodendrine,berberine and jatrorrhizine in transparent absorbent fluid of Huoxue power in different particle size within 24 hours were measured by ultra performance liquid chromatography tandem mass spectrometry,and optimize its particle size by contents comparison of the effective components.Results The contents of the active components in Huoxue power with the particle size of 0.075 mm were high before and after percutaneous absorption.Conclusion Particle size of 0.075 mm is best for Huoxue powder.
7.Overview of the detection methods for equilibrium dissociation constant KD of drug-receptor interaction

Drug-receptor interaction plays an important role in a series of biological effects, such as cell pro-liferation, immune response, tumor metastasis, and drug delivery. Therefore, the research on drug-re-ceptor interaction is growing rapidly. The equilibrium dissociation constant (KD) is the basic parameter to evaluate the binding property of the drug-receptor. Thus, a variety of analytical methods have been established to determine the KD values, including radioligand binding assay, surface plasmon resonance method, fluorescence energy resonance transfer method, affinity chromatography, and isothermal ti-tration calorimetry. With the invention and innovation of new technology and analysis method, there is a deep exploration and comprehension about drug-receptor interaction. This review discusses the differ-ent methods of determining the KD values, and analyzes the applicability and the characteristic of each analytical method. Conclusively, the aim is to provide the guidance for researchers to utilize the most appropriate analytical tool to determine the KD values.
8.Establishment of a modified model of adriamycin nephrosis in rats

Objective To study the adriamycin nephrosis model in different development pathological stages. Methods The rat adriamycin nephrosis model was established by injecting adriamycin into tail-vein twice every two weeks to detect blood and urina biochemical indicators and to observe the pathological changes of the renal tissues. Results The model showed serious edema,proteinuria,hypoproteinemia,and hyperlipidemia. Podocytes were swollen and mesangial cells developed mild hyperplasia at the end of the fourth week. The nephric tubule atrophied at the end of the eighth week accompanied with adhesion between glomeruli and Bowman's capsule. Glomeruli sclerosis of mild or medium degree was observed at the end of the twelfth week with obvious lymphocyte infiltration in the renal interstitium as well as the formation of collagen fibers. Conclusion The adriamycin nephrosis model was successfully developed by injecting adriamycin 4 mg/kg into tail-vein twice every two weeks. The acute model is similar to human minimal change disease,and the chronic model is similar to human focal segmental glomerulosclerosis.
9.The optimized culture and identification of astrocytes from rat cortical tissue

Objective To obtain highly purified astrocytes and identify the cells in each stage to support further studies.Methods The cerebral cortex of a neonatal SD rat was isolated and prepared into single cell suspension.The obtained cells were purified by differential adherence and shook at a constant temperature.By inverted phase contrast microscopy and HE staining,cell morphology was observed.The immunofluorescence staining with anti-mouse GFAP was used to identify the cells.Results The primary cortical cells developed rapidly at 3 d after culture and covered the flasks at 9-12 d.At this time,the cells showed stratification and the astrocytes lay at the lower layer.GFAP positive rate was only about (67.2 ±7.1)%.After the first passage,GFAP positive rate increased obviously (84.0±6.0)%. However, oligodendrocytes and microglias could not be removed completely,and the cells also showed stratification.Through 3 times of passages,we obtained many single species of astrocytes showing satellite shape with 2 or 3 processes,big cell body and round or oval-shaped nuclei leaned to one side.Immunofluorescence staining showed that nearly all of the cells were strong positive and the positive rate reached as high as (97.6 ± 2.4 )%.Conclusion Through differential adherence and shaking at a constant temperature,more astrocytes of high purity and in good state can be obtained.
10.Effect of Muscle-region Alignment Needling plus Dermal Needle Therapy on the Long-term Therapeutic Efficacy for Post-stroke Upper-limb Spasticity

ObjectiveTo evaluate the long-term therapeutic efficacy of muscle-region alignment needling plus dermalneedle therapy in treating post-stroke upper-limb spasticity.MethodTotally 488 patients with post-stroke upper-limb spasticity were randomized into a treatment group and a control group, 244 in each group. Besides theessentialrehabilitation treatment, the treatment group was intervened by muscle-region alignment needling plus dermal needle therapy, while the control group was given regular Western medication.Thetwo groups were intervened for 3 weeks and were followed up for 6 months. The neurologic function, Functional Comprehensive Assessment (FCA), and Stroke Speciality-Quality of Life (SS-QOL) were observed for the follow-up study.ResultThe total effective rate was 93.4% in the treatment group versus 61.5% in the control group, and the difference was statistically significant (P<0.05); the neurologic function, FCA score, and SS-QOL score in the treatment group were significantly superior to that in the control group at the end of the follow-up study (P<0.05).ConclusionMuscle-region alignment needling plus dermal needle therapycan produce a content long-term therapeutic efficacy in treating post-stroke upper-limb spasticity.
