1.Observations on the Therapeutic Effect of Acupuncture on Biased Constitution-based Subhealth

Objective To investigate the clinical efficacy of acupuncture in treating subhealth.Methods One hundred and fifty-four patients with subhealth were allocated, using a random number table, to observation and control groups, 77 cases each. The control group received basic treatment by diet, exercise, rest and rest and psychological intervention and the observation group, acupuncture therapy in addition. Subhealth scale subscores and the 36-Item Short-Form Health Survey (SF-36) subscore were recorded and blood biochemical indicators were measured in the two groups before and after treatment. the clinical therapeutic effects were compared between the two groups.Results The total efficacy rate was 96.1% in the observation group and 83.1% in the control group; there was a statistically significant difference between the two groups (P<0.05). There were statistically significant pre-/post-treatment differences in subhealth scale subscores in the two groups (P<0.05). After treatment, sleep, digestion, urination, positive emotion, ability and social relation subscores were higher, and fatigue, discomfort and negative emotion subscores were lower in the observation group than in the control group (both P<0.05). There were statistically significant pre-/post-treatment differences in blood biochemical indicators in the two groups (P<0.05). After treatment, blood uric acid (UA), total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) were significantly lower and high-density lipoprotein cholesterol (HDL-C) was significantly higher in the observation group than in the control group (both P<0.05). The 36-Item Short-Form Health Survey (SF-36) subscore were higher in thetwo groups after treatment than before treatment (P<0.05). After treatment, physiological function, role-physical, role-emotional, body pain and general health subscore were significantly higher in the observation group than in the control group (P<0.05).Conclusions Acupuncture helps to improve subhealth status in population, regulate blood biochemical indicators and enhance the clinical therapeutic effect to raise the quality of life in the patients.
2.Package and Sterilization of Field Operation Materials

Objective To ensure the quality of field operation materials, and enhance rescue ability of medical corps in field condition. Methods Materials for package and sterilization were tested and selected for new way of package and sterilization instead of the traditional cotton. Results Sheet package was preserved in normal condition. Every month the microorgarism in sheet package was sampled. There were no bacteria in the operation items in the half-year sampling. So the period of validity was prolonged from one week to half year. Conclusion With the new package sterilization method, the work load of nurses is alleviated and the expenses on repeated sterilization due to short term of validity are also reduced. At the same time, operation infection rate and death rate are decreased so that the battle injury rescue level was raised.
3.Magnetic resonance imaging reflects articular cartilage degeneration caused by proteoglycan depletion in the rabbit knee

Objective To study the appearance of magnetic resonance imaging (MRI) in early stages of cartilage degeneration and to detect its values.Methods Intra articular injection of 5 units of papain causing a reversible loss of cartilage proteoglycan in the New Zealand rabbit knees.Rabbits were scanned with magnetic resonance imaging,using a 0 3 T Hitachi magnet with 16 cm coil.Gradient echo sequences and spin echo sequences were performed in the sagittal planes at 0,24,48 and 72 hours after intra injection of papain.Then signal intensity and thickness of cartilage were measured.The proteoglycan content was measured biochemically and histochemically.Results The cartilage thickness and signal intensity decreased significantly in treated knees compared with control knees at 24 and 48 hours ( P 0 05) after injection of papain.These changes on magenetic resonance (MR) images were consistent with the changes of proteoglycan concentration measured by biochemical analysis and histochemical staining of cartilage.Conclusion It is possible for MRI to detect the early stages of cartilage degeneration.
4.Effects of hydrogen-rich medium on lipopolysaccharide-induced release of inflammatory cytokines from RAW264.7 macrophages

Objective To investigate the effects of hydrogen-rich medium on lipopolysaccharide (LPS)-induced release of inflammatory cytokines from murine macrophage RAW264.7 cells.Methods The RAW264.7cells were cultured in DMEM culture medium containing 10% fetal bovine serum.The cells were seeded in 6-well plates (3 ml/hole) with a density of 2 × 106/ml and randomly divided into 4 groups (n =24 each):control group (group A),hydrogen-rich medium group (group B),LPS group (group C) and LPS + hydrogen-rich medium group (group D).Group A received no treatment.LPS 1 μg/ml was added to the medium in groups C and D,while the cells were incubated with 0.6 mmol/L hydrogen-rich medium simultaneously in groups B and D.Six wells in each group were chosen at 6,12.24 and 48 h of incubation,and the supernatant was collected to detect the concentrations of TNF-α,IL-1β and IL-10.Six wells in each group were chosen at 6 and 12 h of incubation for determination of heme oxygenase-1 (HO-1) expression in the cells by Western blot.Results Compared with group A,the concentrations of TNF-α,IL-1β and IL-10 in the supernatant were significantly increased,and the expression of HO-1 was up-regulated in group C (P < 0.05).Compared with group C,the concentrations of TNF-α and IL-1β in the supernatant were significantly decreased,the concentration of IL-10 in the supernatant was significantly increased,and the expression of HO-1 was up-regulated in group D (P < 0.05).Conclusion Hydrogen-rich medium can inhibit the release of pro-inflammatory cytokines and promote the release of anti-inflammatory cytokines from RAW264.7 cells via up-regulating HO-1 expression.
5.Effects of hydrogen gas on lipopolysaccharide-induced damage to human umbilical vein endothelial cells

Objective To evaluate the effects of hydrogen gas (H2) on lipopolysaccharide (LPS)-induced damage to human umbilical vein endothelial cells (HUVECs) in vitro.Methods HUVECs were cultured in DMEM culture medium containing 10% fetal bovine serum.The cells were seeded in 6-well plates (2 ml/hole) at a density of 2 × 106 cells/ml and randomly divided into 4 groups (n =35 each) using a random number table:control group (group C),group H2,LPS group and LPS + H2 group.The cells were cultured in the plain culture medium in C and LPS groups or in hydrogen-saturated culture medium in H2 and LPS + H2 groups.In addition,LPS 1 μg/ml was added simultaneously in LPS and LPS + H2 groups,and the equal volume of normal saline was added instead in C and H2 groups.The cells were incubated for 24 h.At 6,12 and 24 h of incubation,the cells of 5 wells were chosen and stained with Wright-Giemsa to observe the destruction of HUVEC barrier.At 6,12 and 24 h of incubation,the cells of 5 wells were chosen to detect the expression of VE-cadherin and β-catenin using Western blot.At 24 h of incubation,the cells of 5 wells were chosen to detect the expression of VE-cadherin and βcatenin using immunofluorescence.Results Pathological changes of endothelial cells were observed in LPS group.The pathological changes of the cells were lighter in LPS + H2 group than in LPS group.Compared with group C,VE-cadherin expression was significantly down-regulated (P < 0.05),while no significant change was found in β-catenin expression in LPS and LPS + H2 groups (P > 0.05).Compared with group LPS,VE-cadherin expression was significantly up-regulated (P < 0.05),while no significant change was found in ~catenin expression in group LPS + H2 (P > 0.05).Conclusion H2 can effectively reduce LPS-induced damage to HUVECs through inhibiting down-regulation of VE-cadherin expression.
6.The role of Nrf2 in the hydrogen treatment for intestinal injury caused by severe sepsis

Objective To investigate the role of Nrf2 on hydrogen treatment for intestinal injury caused by severe sepsis.Methods 152 male ICR mice were randomly divided into four groups:sham operation group,hydrogen control group,sepsis group,and hydrogen treatment group,each n=38.Sepsis model was reproduced by cecal ligation and puncture (CLP).The mice in sham operation group and hydrogen control group did not receive CLP,and the operative procedure was the same as follows.The mice in hydrogen control group and hydrogen treatment group received 1-hour inhalation of 2% hydrogen 1 hour and 6 hours after sham operation or CLP.Twenty animals in each group were selected and observed for 7-day survival rate.Eighteen animals in each group were selected and sacrificed at 6,12 and 24 hours after CLP.The intestinal tissues were obtained to determine the expression of Nrf2 and high mobility group B1 (HMGB1) protein by Western Blot,and the expression of Nrf2 mRNA by reverse transcription-polymerase chain reaction (RT-PCR).The middle portion of jejunum was obtained to evaluate the degree of septic injury by light microscope after hematoxylin and eosin (HE) staining.Results There was no statistical signifieance in variables between sham operation group and hydrogen control group.Compared with sham operation group,the 7-day survival rate was significantly decreased in sepsis group (0 vs.100%,P<0.05); compared with sepsis group,the 7-day survival rate was significantly increased in hydrogen treatment group (55% vs.0,P<0.05).Compared with sham operation group,the expression of Nrf2 protein (gray value) and Nrf2 mRNA were up-regulated in sepsis group at 6,12 and 24 hours after CLP (Nrf2 protein 6 hours:1.973 ± 0.350 vs.1.000 ± 0.000,t=4.411,P=0.002; 12 hours:2.367 ± 0.186 vs.1.000 ±0.000,t=10.210,P=0.000; 24 hours:2.517 ±0.280 vs.1.000 ±0.000,t=9.521,P=0.000; Nrf2 mRNA 6 hours:1.606 ± 0.271 vs.1.000 ± 0.000,t=3.631,P=0.002; 12 hours:1.692 ± 0.399 vs.1.000 ± 0.000,t=3.233,P=0.005; 24 hours:1.784 ± 0.341 vs.1.000 ± 0.000,t=3.894,P=0.001),and it was also the expression of HMGB1 (gray value) at 24 hours after CLP operation (1.507 ± 0.220 vs.1.000 ± 0.000,t=3.948,P=0.004).Compared with sepsis group,the expression of Nrf2 protein and Nrf2 mRNA in intestines were up-regulated at 6,12 and 24 hours after CLP in hydrogen treatment group (Nrf2 protein 6 hours:2.583 ± 0.395 vs.1.973 ± 0.350,t=2.765,P=0.024; 12 hours:2.725 ± 0.235 vs.2.367 ± 0.186,t=2.674,P=0.028; 24 hours:2.930 ± 0.212 vs.2.517 ± 0.280,t=2.595,P=0.032; Nrf2 mRNA 6 hours:2.008 ± 0.400 vs.1.606 ± 0.271,t=2.405,P=0.029; 12 hours:2.188 ± 0.475 vs.1.692 ±0.399,t=2.317,P=0.034; 24 hours:2.333 ±0.406 vs.1.784 ±0.341,t=2.728,P=0.015).Compared with sepsis group,the expression of HMGB1 was down-regulated significantly at 24 hours after CLP in hydrogen treatment group (1.147 ± 0.152 vs.1.507 ± 0.220,t=2.805,P=0.023).HE staining showed that there was significantly aggravated intestinal pathological injury in the mice of sepsis group; compared with sepsis group,the pathology was significantly less marked in hydrogen treatment group.Conclusion Through activation of Nrf2-antioxidant response element (ARE) pathway,hydrogen may increase the level of Nrf2,which is a kind of protective protein,in the intestine of mice,thus decreases the level of late pro-inflammatory factor,HMGB1,and it may protect the intestinal tissues in septic mice and increase the survival rate significantly.
7.Effects of hydrogen on nuclear factorE2-related factor 2/antioxidant response element pathway in lung tissues in septic mice

Objective To evaluate the effects of hydrogen (H2) on nuclear factorE2-related factor 2 (Nrf2)/antioxidant response element (ARE) pathway in lung tissues in septic mice.Methods Seventy-two male ICR mice,weighing 20-25 g,aged 6 weeks,were randomly divided into 4 groups (n =18 each) using a random number table:sham operation group (group SH),group H2,sepsis group (group S),and sepsis + H2 group (group S + H2).Sepsis was produced by cecal ligation and puncture (CLP).H2 and S + H2 groups inhaled 2% H2 for 1 h starting from 1 and 6 h after CLP.Six mice in each group were chosen and sacrificed at 7,12 and 24 h after CLP (T1-3).The pulmonary specimens were obtained to determine the expression of Nrf2 and HO-1 protein (by Western blot) and Nrf2 mRNA (by RT-PCR).At 24 h after CLP,the pathological changes of lungs were scored,wet/dry lung weight ratio (W/D ratio) was determined,and the expression of high mobility group box-1 (HMGB-1) in lung tissues was measured (by Western blot).Results Compared with group SH,the pathological scores and W/D ratio were significantly increased,and the expression of Nrf2 protein and mRNA,HO-1 and HMGB1 was up-regulated in S and S + H2 groups,while no significant change was found in the indexes mentioned above in group H2.Compared with group S,the pathological scores and W/D ratio were significantly decreased,the expression of Nrf2 protein and mRNA and HO-1 was up-regulated and HMGB1 expression was down-regulated in group S + H2.Conclusion The mechanism by which H2 reduces acute lung injury in septic mice is related to activation of Nrf2/ARE pathway.
8.Analyze the Effect of the Bicyclol Tablets on Platelet Based on the Data of the Hospital Information System

Objective To study the real world of bicyclol tablets in the treatment of viral hepatitis and its influence on platelets.Methods The hospital information system (HIS) was selected from 18 Triple-A hospitals in China on patients with bicyclol tablets information as exposed group (2 690 cases),patients with unused bicyclol tablet (2 690 cases) as an exposed group,an exception occurs of platelet as an index of the ending.Results The incidence of abnormal platelet difference in two groups before the treatment were not statistically significant.The incidence of abnormal platelet reduce between two groups have statistical significance,can be thought of exposure of abnormal lower rate (11.75%)lower than that of non exposed group(15.17%) (P<0.01).Conclusion Bicyclol piece have signficantly curative effect on the treatment of viral hepatitis.It shows more safety,and less impact on platelets.It is worth to be used in clinical practice.
9.Effect of Sulfureted Hydrogen Bath on Spinal Cord Injury

Objective To explore the effects of motor training in sulfureted hydrogen miniral spring on neurological function after spinal cord injury (SCI). Methods 80 SCI patients were divided into treatment (n=40) and control group (n=40). They all accepted comprehensive rehabilitation, while the treatment group accepted motor training in sulfureted hydrogen miniral spring in addition. They were assessed with ASIA scale, modified Barthel index (MBI), Ashworth scale, visual analogue scale (VAS) and limbs circumference. Results There was not significant difference between both groups after treatment in ASIA scores of motor and sense, but was in Ashworth scale (P<0.05), VAS (P<0.01), limbs circumference (P<0.01), MBI (P<0.01, not in walk), which the treatment group improved more. Conclusion Motor training in sulfureted hydrogen miniral spring can further improve the activities of daily living and neurological function
10.Role of the nuclear factor kappa B signaling pathway in the pathogenesis and development of diabetic keratopathy

In recent years, with the increasing incidence of diabetes, diabetic keratopathy has gradually attracted more attention from clinicians.Diabetic keratopathy is mainly manifested as delayed corneal epithelial healing, corneal edema, decreased corneal sensitivity, and neurotrophic corneal ulcer.There are many mechanisms of diabetic keratopathy, including the abnormal accumulation of metabolites, an inflammatory reaction, oxidative stress, the lack of neurotrophic factors, corneal limbal stem cell abnormalities, etc.At present, most of the research on diabetic keratopathy at home and abroad has mainly focused on epithelial damage repair and changes in nerve function, and the pathological mechanism is mainly inflammatory response and apoptosis.Nuclear factor kappa B(NF-κB) is an important transcription regulatory factor, and is involved in regulating the expression of inflammatory factors, chemokines, cell adhesion molecules, apoptosis-related genes, growth factors, and other target genes.Studies have reported that chronic inflammation and epithelial cell apoptosis of the diabetic corneal epithelium are involved in the regulation of the NF-κB signaling pathway, which is also involved in the regulation of diabetic peripheral neuropathy, and diabetic corneal neuropathy belongs to the category of diabetic peripheral neuropathy.The NF-κB signaling pathway is closely related to the occurrence and development of diabetic keratopathy, and this article summarizes its mechanism.
