1.PPAR-? ligand in suppressing radiation induced PAI-1 upregulation

Objective observe the post-radiation expression of rat mesangial cell plasminogen activator inhibitor-1 (PAI-1), and the effect of PPAR-? ligand on radiation induced PAI-1 expression. Methods After 10?Gy ?-ray radiation of cultured rat kidney mesangial cell, RT-PCR was used to observe PPAR-?expression. PAI-1 expression after 10?Gy radiation ?10?mmol/L pioglitazone (one of PPAR-? ligands) was measured by Northern blot and Western blot. Gel-shift method was used to study the effect of AP-1 of PPAR-? ligand on radiation induced PAI-1 expression. Methods Rat kidney mesangial cell expressed PPAR--? mRNA. PAI-1 over-expressed at 24 hour after 10?Gy radiation in a radiation dose dependent manner, but not at other time. When the cell was treated with 10?Gy + 10?mmol/L pioglitazone, PAI-1 upregulation was blunted at message and protein level. AP-1 activation was increased from the 2nd hour after 10?Gy radiation and reached the highest level at the 4th hour. When pioglitazone was added, the radiation induced increase in AP-1 activation was suppressed at the 4th hour. Conclusions Pioglitazone, mediated by AP-1 gene, can suppress radiation induced PAI-1 upregulation.

The endocrine therapy of hormone receptor -positive breast cancer has been widely recog-nized.Aromatase inhibitors are better than tamoxifen in efficacy ,tolerability and other aspects .However,AIs is on-ly applied in the menopausal status of breast cancer patients .Therefore,for premenopausal patients with tamoxifen resistance ,the presence of contraindications or the existence of risk factors ,ovarian castration undoubtedly open a gateway to the more and better option for them .Because of the disadvantages of the reversibility, controllability and side effects ,castration surgery and radiotherapy have been gradually replaced by medical castration .In this paper , the new progress are reviewed .Our article aims to make a review on the lastest research on the new progress of breast cancer medical ovariectomized on endocrine therapy ,the comparison with chemotherapy and other aspects .
3.Identification of Clostridium perfringens Causing Human Disease by Multiplex PCR

OBJECTIVE To search a method for identifying Clostridium perfringens and genotyping their toxin for gene diagnosis by multiplex PCR.METHODS The mutiplex PCR was developed with three sets of primers(designed) based on the sequences of three C.perfringens toxin genes(CP?,CP? and CPE) published in GenBank to identify C.perfringens and genotype their three toxin genes.RESULTS Three expected(sequences) were (obtained) successfully by multiplex PCR and identified by electrophoresis.CONCLUSIONS The(specific) sequences of C.perfringens could be amplified and their three genes of toxins could be identified by this multiplex PCR(system).Such method should be helpful for developing gene diagnosis well.
4.Immobilization Method on Piezoelectric Quartz Crystal DNA Sensors Based on Monolayer SAM

OBJECTIVE To discuss a highly effective method to immobilize probe on the surfaces of piezoelectric DNA sensors.METHODS Pseudomonas aeruginosa probe was immobilized on the gold surface of gene sensor(array) with routine self-assembly method(SAM)(non-reduction method) and SAM with deoxidized probe((reduction) method),respectively.The changes in frequency and time-cost were compared in reactions with(different) concentrations of probe.RESULTS Reduction method had the advantage of more probe immobilization;less time consumed in testing and higher changes in frequency during the reaction than non-reduction method.CONCLUSIONS Reduction method has a better ability to immobilize probe on the surfaces of piezoelectric DNA sensors.
5.Analysis on the monitoring of influenza in Maanshan from 2006 to 2007

Objective To explore the epidemic trend of influenza and pandemic virus variation in Maanshan, and to provide scientific basis for the prevention of influenza. Methods The ILI data of monitor hospitals and some ILI nasopharyngeal specimens were collected, then influenza virus was isolated by MDCK cells, and influenza serotypes were identified by HI. Results 15 904 cases of ILI were reported from 2006 to 2007. The detectable rate of influenza like illness in outpatients was 6.63%. 2 656 nasopharyngeal specimens of ILI were collected, and 358 strains of influenza virus were isolated, with isolation rate of 13.48%, of which 142 were H1N1 subset influenza A virus, 49 were H3N2, 72 were Victoria subset influenza B virus, and 96 were Yamagata. Conclusions During 2006 and 2007 two peaks of influenza-like illness in outpatients was in winter-spring and summer seasons of each year. More attention meeds to be paid to the monitoring of influenza.
6.Study of Stomach Using Ferric Oxide Suspension as Oral Contrast Agent in MRI

Objective To evaluate the applied value superparamagnetic Ferric Oxide oral contrast agent in the dignosis of MR forstomach.Methods Ferric Oxide Suspension was administered before MR scanning of stomach in 36 cases,ranged in age form 21 to 60 years old.MR findings were analysed.Results The fullness of stomach was 100% after administration of Ferric Oxide Suspension.Stomach canal appeared as negative contrast in all MR sequences,the relation between lesions in stomach and around tissue were clearly showed.During MRCP,it could restrict the high signal in stomach canal.Conclusion Oral Ferric Oxide Suspension as contrast agent during MR examination of stomach is easily accepted by patients,it is a safe and steady negative contrast agent for gastrointestinal MR examination.
7.Rapid detections of OprD2 gene in Pseudomoas aeruginosa using loop-mediated isothermal amplification

Objective To establish a rapid detection approach by visual interpretation directly for OprD2 resistance gene of Pseudomonas aeruginosa based on the Loop-mediated isothermal amplification (LAMP),and provide a quick and effective method for clinical monitoring of Pseudomonas aeruginosa strains.Methods Totally 47 strains of Pseudomonas aeruginosa collected from December 2011 to June 2012 in Southwest Hospital of microorganisms were prospectively studied.Four LAMP primers (two inner,two outer) were designed according to the six zones of the OprD2 gene of Pseudomonas aeruginosa.A positive reaction is indicated by the color change after adding an intercalating dye (hydroxy naphthol blue) to the reaction solution.This method was used to detect and analyze the distribution of OprD2 resistance gene in 47 strains of Pseudomonas aeruginosa and its coirelation with antibiotic resistance.Results The LAMP assays showed 100% specificity for the OprD2 gene,and the sensitivity (with the lowest detection limits of 17.414 μg/L) was 10-fold higher than that of conventional PCR assays.The OprD2 gene was negative in 23 strains by both conventional PCR and LAMP.In OprD2 negative strains,the resistance rate of cefotaxime,levofloxacin,aztreonam,piperacillin,imipenem and meropenem was 100% (23/23),57% (13/23),48% (11/23),48% (11/23),48% (11/23) and 43% (10/23).Compared with the OprD2 positive strains,statistical analysis showed that the resistance rate of imipenem,levofloxacin and meropenem in OprD2 negative strains increased significantly (chisquare value is 9.155,4.846,4.037,P value was 0.002,0.028,0.045,and so there was significant difference).Conclusions The established LAMP approach in this study enables rapid,sensitive and specific detection of OprD2 gene in Pseudomonas aeruginosa by visual interpretation.Deficiency of OprD2 gene confers Pseudomonas aeruginosa a basal level of resistance to carbapenems especially to imipenem.The identification of OprD2 gene distribution in Pseudomonas aeruginosa is helpful to the selection of antimicrobial agents in the infection treatment.
8.DNA Staining Efficiencies of Different Fluorescent Dyes in Preparative Agarose Gel Electrophoresis:A Comparative Analysis

OBJECTIVE To set-up safe alternatives to ethidium bromide(EB).METHODS Comparative analysis was performed on the DNA staining efficiencies of 4 fluorescent dyes including SYBR Gold,SYBR Green Ⅰ,(GoldView) and EB in preparative agarose gel electrophoresis.RESULTS Although both SYBR Gold and SYBR Green Ⅰ altered electrophoretic mobility and thus DNA size estimates,they were cost-effective alternatives to EB.SYBR Gold was more sensitive than SYBR Green Ⅰ at detecting short fragments,but 50 bp bands were clearly(visible) using either dye when visualized with a long integration time.CONCLUSIONS SYBR Gold or SYBR Green Ⅰ are sensitive and relatively safe alternatives to EB.
9.Association between single nucleotide polymorphism of BARD1 gene and BRCA1 gene mutation in epithelial ovarian cancer

Objective To investigate the relationship between single nucleotide polymorphism (SNP) of BARD 1 gene and BRCA1 gene in epithelial ovarian cancer (EOC).Methods Nineteen EOC patients with BRCA1 gene mutation and 50 EOC cases without BRCA1 gene mutation between January 2016 and October 2016 were collected,and all EOC were diagnosed by pathological method.BARD1 gene variants were detected by next generation sequencing (NGS).The SNP of BARD1 gene was analyzed by Pearson linear correlation.Logistic regression analysis was used to research the clinicopathologic features and BRCA1 gene mutation associated with BARD1 gene SNP.Pearson's chi-square test was used to analyze the association between BARD1 gene Val507Met,Arg378Ser and Pro24Ser with different clinicopathologic features and BRCA1 gene mutation risk.Results (1) Eight BARD1 gene variants were found in 69 ovarian cancer patients,in which Val507Met,Arg378Ser and Pro24Ser were common variants,and the rate of mutation were all 54% (37/69).(2) There was a significant linear correlation among Val507Met,Arg378Ser and Pro24Ser (all P<0.01).(3) Obvious differences were found in Val507Met,Arg378Ser and Pro24Ser of BARD1 gene between BRCA1+ and BRCA1 (all P<0.05).(4) No differences were found between BARD1 gene Val507Met,Arg378Ser and Pro24Ser and the clinicopathologic features (all P>0.05),while obvious differences were found in BRCA1 gene mutation compared to the controls group.The risk of BRCA1 mutation in Val507Met and Arg378Ser were more evident in subjects with negative family history,positive menopause history,negative tubal ligation,onset age (≤60 years old) and sensitivity to platinum-based chemotherapy in EOC (all P<0.05),while Pro24Ser was only more evident in positive menopause history of EOC (P<0.05).Conclusions BARD1 Val507Met,Arg378Ser and Pro24Ser are the common genotypes,which are associated with BRCA1 mutation in EOC.The family history,menopause history,tubal ligation,onset age and sensitivity to platinum-based chemotherapy have effects on BARD1 SNP in the risk of BRCA1 gene mutation.
10.Clinical significance and distribution of BRCA genes mutation in sporadic high grade serous ovarian cancer

Objective To investigate the mutations of BRCA genes in sporadic high grade serous ovarian cancer (HGSOC) and study its clinical significance. Methods Sixty-eight patients between January 2015 and January 2016 from the Affiliated Cancer Hospital of Zhengzhou University were collected who were based on pathological diagnosis of ovarian cancer and had no reported family history, and all patients firstly hospitalized were untreated in other hospitals before. (1)The BRCA genes were detected by next-generation sequencing (NGS) method. (2)The serum tumor markers included carcinoembryonic antigen (CEA), CA125, CA199, and human epididymis protein 4 (HE4) were detected by the chemiluminescence methods, and their correlation was analyzed by Pearson linear correlation. Descriptive statistics and comparisons were performed using two-tailed t-tests, Pearson′s chi square test, Fisher′s exact tests or logistic regression analysis as appropriate to research the clinicopathologic features associated with BRCA mutations, including age, International Federation of Gynecology and Obstetrics(FIGO)stage, platinum-based chemotherapy sensitivity, distant metastases, serum tumor markers (STM). Results (1) Fifteen cases (22%, 15/68) BRCA mutations were identified (BRCA1: 11 cases; BRCA2: 4 cases), and four novel mutations were observed. (2) The levels of CEA, CA199, and HE4 were lower in BRCA mutations compared to that in control group, while no significant differences were found (P>0.05), but the level of CA125 was much higher in BRCA mutation group than that in controls (t=-3.536,P=0.003). Further linear regression analysis found that there was a significant linear correlation between CA125 and HE4 group (r=0.494,P<0.01), and the same correlation as CEA and CA199 group (r=0.897,P<0.01). (3) Single factor analysis showed that no significant differences were observed in onset age, FIGO stage, distant metastasis, and STM between BRCA+and BRCA- group (P>0.05), while significant differences were found in CA125 and sensitivity to platinum-based chemotherapy between the patients with BRCA mutation and wild type (P<0.05). The multiple factors analysis showed that the high level of CA125 was a independent risk factor of BRCA mutations in sporadic HGSOC (P=0.007). Conclusion The combination of CA125 with BRCA have great clinical significance, the mutation of BRCA gene could guild the clinical chemotherapy regiments.
