1.Correlation between glycosylated hemoglobin level and coronary heart disease in patients with and with-out diabetes mellitus

Objective:To study the correlation between glycosylated hemoglobin (HbA1c) level and coronary heart disease (CHD) in patients with and without diabetes mellitus (DM) .Methods :A total of 100 DM patients and 100 non-DM patients were enrolled .According to complicated CHD or not ,DM patients were divided into DM +CHD group (n=44) and DM + non-CHD group (n=56) ,non-DM patients were divided into non -DM + CHD group (n=41) and non-DM + non-CHD group (n=59) .HbA1c concentration ,insulin resistance [homeostasis model-insulin resistance index (HOMA-IR) ,insulin sensitivity index (ISI)] and endothelial function [number of endothelial microparticles (EMPs) ,brachial artery flow -mediated dilation (FMD)] were measured and compared among above groups .Results:Compared with DM + non-CHD group ,there were significant rise in HbA1c con‐centration [ (7.10 ± 0.86)% vs .(9.98 ± 1.05)% ] ,HOMA-IR [ (2.28 ± 0.31) vs .(3.31 ± 0.52)] and number of EMPs [(548.29 ± 69.35)/μl vs . (861.18 ± 94.57) /μl] ,and significant reductions in ISI [(-3.23 ± 0.41) vs .(-4.62 ± 0.65)] and FMD [ (6.65 ± 0.75)% vs .(4.58 ± 0.59)% ] in DM + CHD group , P<0.05 all .Compared with non -DM + non-CHD group ,there were significant rise in HbA1c concentration ,HOMA-IR and number of EMPs ,and significant reductions in ISI and FMD in non -DM + CHD group , P<0.05 or <0.01 .Single factor regression analysis indicated that HbA1c level was positively correlated with HOMA -IR and number of EMPs (β=2.845 ,1.945 , P<0.05 both) ,and inversely correlated with ISI and FMD (β= -3.582 ,-3.293 , P<0.05 both) . Conclusion:In patients with or without diabetes mellitus ,glycosylated hemoglobin concentration significantly rising can cause insulin resistance ,endothelial function injury ,then further lead to occurrence of coronary heart disease .

Astrocyte elevated gene (AEG) -1 is cloned as a human immunodeficiency virus (HIV) -1-inducible and tumor necrosis factor-alpha (TNF-α)-inducible transcript in primary human fetal astrocytes (PHFA) by a rapid subtraction hybridization approach. AEG-1 has been reported to be up-regulated in various types of human cancers. Multivariate analyses indicat that AEG-1 correlates with the ability of growth, invasion, metastasis, angiogenesis and chemoresistance of tumors. AEG-1 over-expression activates the PI3K-Akt,nuclear factor kappa B (NF-κB) and Wnt-β-catenin signaling pathways in several crucial aspects of tumorprogression. AEG-1 represents a viable potential target for the therapy of human cancers.
3.Quality Standard of Changsheng Oral Solution

OBJECTIVE:To study the quality standard of Changsheng oral solution.METHODS:Radix Astragali,Radix Platycodi,Spica Prunellae and Radix et Rhizoma Glycyrrhizae in the formula were identified qualitatively by TLC.The content of Liquiritin in Changsheng oral solution was determined by HPLC.The chromatographic separation was performed on Nova-Pak C18 column(150 mm?4.6 mm,4 ?m)and the mobile phase consisted of acetonitrile-2.5% acetic acid(13∶87)at a flow rate of 1.0 mL?min-1.The detective wavelength was set at 276 nm.RESULTS:Clear TLC spots were noted.The linear range of Liquiritin was 0.025 04~0.626 00 ?g(r=0.999 9).The average recovery rate of Liquiritin was 98.66%(RSD=0.52%,n=6).CONCLUSION:The established standard can be used for the quality control of Changsheng oral solution.

5.Relationship between plasma D-lactate levels and prognosis of neonatal necrotizing enterocolitis

Objective To investigate the relationship between plasma D-lactate levels and prognosis in newborns with neonatal necrotizing enterocolitis (NEC).Methods One hundred and four premature infants with NEC (stase Ⅱ and Ⅲ ) and another 104 premature infants without NEC admitted into Quanzhou Children's Hospital for other diseases from January 2007 to October 2010 were selected into this case control study.The gestational age,gender and birth weight of patients in the two groups were matched.NEC patients' bloods were collected within 24 hours after NEC was confirmed and blood samples of the control group were collected at the corresponding age.Enzymelinked immunosorbent assay was used to detect the plasma D-lactate level.Receiver operating characteristic curve was used to confirm the criteria of D-lactate positive,according to which,the NEC group was divided into high D-lactate group and normal D-lactate group,and the mortality and complication rate of the two groups were compared with x2 test.And NEC group was subdivided into death group and survive group according to the prognosis of the patients,and the difference of D-lactate level between the two groups were compared with t test.Results Among the NEC group,there were 63 cases (60.6%) of stage Ⅱ and 41 (39.4%) cases of stage Ⅲ ; 88 (84.6%) survived infants and 16 (15.4%) dead infants.D-lactate level was (35.4 ± 29.1) μg /ml in stage Ⅲ NEC group,(29.5±16.2) μg/ml in stage Ⅱ NEC group and (3.7±18.4) μg/ml in control group; there were statistical differences between each other(F=5.97,P<0.05).Receiver operating characteristic curve analysis showed that if 6 μg/ml was set as the borderline,there were 87 cases(83.7%,87/104) with high D-lactate patients in NEC group,whose neonatal critical illness scoring (NCIS) was 80.9±22.6,significantly lower than that of normal D-lactate patients ( 95.8 ± 20.5) (t =2.417,P< 0.05),and higher neonatal septicemia rate (48.3%,42/87) and mortality (27.6%,24/87) compared with those [(5.9%,1/17) and (5.9%,1/17)] of normal D-lactate patients(x2 =11.539 and 7.146,P<0.05,respectively).In NEC group,compared with the survived infants,the D-lactate level [(43.2±13.5) μg/ml vs (21.9 ± 22.9) μg/ml,t =4.572,P<0.05] and the rate of septicemia [68.8% (11/16) vs 38.6% (34/88),x2 =3.445,P<0.05] were higher in dead patients,and NCIS (82.4± 29.1 vs 90.6 ± 21.3,t =2.409,P<0.05) was lower.Conctusions The level of plasma D-lactate related to the prognosis of neonatal NEC and which might be a good indicator for its prognosis and severity.

7.Cloning and sequencing of the gene encoding extended-spectrum ?-lactamases SHV-12

Objective To analyse the encoding gene sequence of extended spectrum ? lactamase produced by Klebsiella pneumoniae E95 strain in Zhejiang Province and identify its ESBLs subtype. Methods The gene of ESBLs produced by E95 strain was amplified by PCR. The purified PCR product was cloned into pGEM Teasy vector and then sequenced by Sanger's dideoxy chain termination composition method. Results The encoded gene of ESBLs was identified as SHV by PCR. Its PCR product had 812 nucleotides. It had the same gene sequence as the gene encoding SHV 12 discovered in Swiss. Conclusions The ESBLs produced by Klebsiella pneumoniae E95 strain isolated from a patient in Zhejiang Province is SHV 12.
8.Study on Quality Standard for Kechuanling Plaster

Objective To establish the quality control standard of Kechuanling Plaster. Methods Semen sinapis in Kechuanling Plaster was identified by TLC. The content of ?-asarone in Kechuanling Plaster was identified by HPLC. Results Semen sinapis in Kechuanling Plaster could be detected by TLC. ?-asarone shows a good linear relationship at range of 0.132~0.308 ?g (r=0.9999). The average recovery was 101.7% and RSD was 2.0%. Conclusion The method was simple, specific and accurate. This study provide a method for the quality control of Kechuanling Plaster.
9.Studies on the chemical constituents of Anabasis aphylla L.

The investigation on the chemical constituents of Anabasis aphylla L. was carried out by using various chromatographies, such as silica gel, Sephadex LH-20 and RP-C18 column chromatography. Further detailed investigation on the fraction of the ethanol extract of Anabasis aphylla L. yielded one new compound p-acetyl-phenol 1-O-beta-D-xylopyranosyl-(1-->2)-beta-D-glucopyranoside (1), together with five known compounds: piceine (2), isorhamnetin (3), quercetin (4), rutin (5) and isorhamnetin-3-rutinoside (6). Their structures were elucidated by spectral analysis such as NMR and MS. Among these compounds, compounds 2-6 were isolated from this plant for the first time.
10.Femoral head surface replacement for avascular necrosis of the femoral head

Prosthesis for femoral head surface replacement was designed. Between January 2002 and December 2005,28 patients underwent femoral head surface replacement in Anyang People's Hospital. The diameter of the normal femoral head was determined by CT,and the prosthesis was selected according to the measured value ±1 mm. With the muscle gap approach,the center of the femoral head was determined according to the measured distance between the femoral head center and the culminated point of the round ligament fossa based on the mark of the culminated point of the round ligament pit. Additionally,the diameter of the femoral head was measured again to ensure the precise match of the prosthesis and acetabulum. X-ray films at 24-48 months show no loosening or dislocation of the prosthesis,which was well matched with acetabulum. Furthermore,the joint space did not remarkably change. The mean score was 63 before operation and 91.5 after operation respectively according to Harris sore. The successful rate is 92.86% (excellent in 23cases,good in 3,and improved in 2). The results show that femoral head surface replacement is an effective method for young patients with avascular necrosis of the femoral head at the stage of ARCO HI.
