1.Application of diffusion weighted magnetic resonance imaging for the diagnosis of pancreatic cancer

Objective To investigate the value of diffusion weighted magnetie resonance imaging and apparent diffusion coefficient (ADC) value in the diagnosis of pancreatic cancer.MethodsThe clinical data of 36 patients with pancreatie cancer who were admitted to the Wuxi No.2 People's Hospital from March 2009 to June 2011 were retrospectively analyzed,and the clinical data of 30 healthy volunteers were collected.All candidates received diffusion weighted magnetie resonance imaging examination.The signal intensity ratios ( SIRs ) of the cancer and the pericancerous tissues in the T1 weighted-images (T1WI),T2 weighted-images (T2WI)and diffusion weighted-images (DWI) were compared by using one-way analysis of variance.The ADC values of the cancer and the paricancerous tissues were compared using the paired t test.The differences of the ADC values of the cancer and pericancerous tissues compared with those of healthy individuals were analyzed using an independent sample t test.ResultsThe accuracy rate of preoperative magnetic resonance imaging examination was 92%.The relative SIRs were 0.203 ± 0.190 in the T2 WI,0.399 ± 0.201 in the T1 WI and 0.579 ± 0.291 in the DWI,respectively,with no significant differences across the 3 kinds of images (F=5.92,6.15,6.83,P < 0.05 ),while SIRs of the T1 WI and DWI were significantly higher than that of the T2 WI ( P < 0.05 ).There was no significant difference in SIRs when comparing T1 WI and DWI ( P > 0.05 ).The mean ADC values of the cancer and pericancerous tissues of the pancreatic cancer patients and the pancreatic tissues of healthy individuals were (1.40±0.24) ×10 3 mm2/s,(1.71 ±0.10) ×10-3 mm2/s and (1.73±0.30) ×10-3 mm2/s,respectively,with significant differences across the 3 modes of images (t =10.54,12.08,P < 0.05).ConclusionDWI can show high quality images of the lesions,and ADC value is helpful in the diagnosis of pancreatic cancer.
2.Human parvovirus B19 detection in voluntary blood donors of Foshan City

Objective To investigate the infection status of human parvovirus B19(HPV B19) in voluntary blood donors of Fos‐han City .Methods The HPV B19 IgG and IgM antibodies of voluntary blood donors were detectd by using ELISA .Real‐time PCR assay was applied to detect HPV B19 DNA on HPV B19 IgG and/or IgM antibody positive specimens .Results There were 92 HPV B19 IgG positive cases in 368 specimens ,positive rate was 25 .00% ;and the HPV B19 IgM positive cases were 2 ,positive rate was 0 .54% .4 positive cases were detected in 94 antibody positive specimens ,positive rate was 4 .26% .Conclusion HPV B19 in‐fection of blood donation person in Foshan is high ,new infections and chronic infection rate are low .HPV B19 viral loads are low in people with chronic infection .

4.Iodine nutritional status before and after the adjustment of salt iodine content in Guangxi

Objective To understand the implementing situation of prevention measures for iodine deficiency disorders before (2011) and after (2013) the adjustment of salt iodine content in Guangxi,to evaluate the changes of iodine nutritional status,and to provide a basis for future prevention work.Methods Monitoring data of iodized salt,drinking water iodine and iodine nutritional status before and after the adjustment of iodine content of salt was collected.Changes in water iodine,salt iodine and urinary iodine before and after adjusting iodine content of salt were compared.Results The monitoring work of iodized salt,drinking water iodine and iodine nutritional status was carried out in all the 109 counties (cities,districts) in 2011 and 2013.①Drinking water iodine monitoring:a total of 4 968 water samples was tested in 2011,the median water iodine was 2.69 μg/L.Of which,4 210 water samples below 10.00 μg/L,the proportion was 84.74%.A total of 7 554 water samples were tested in 2013,the median water iodine was 2.11 μg/L.Of which,6 512 water samples below 10.00 μg/L,the proportion was 86.12%.②Iodized salt monitoring:a total of 30 786 salt samples were tested in 2011; the salt median iodine was 32.30 mg/kg.The iodized salt coverage rate was 98.31%,iodized salt qualification rate was 97.36%,and qualified iodized salt consumption rate was 95.98% weighted by population.A total of 32 779 salt samples were tested in 2013; the salt median iodine was 24.94 mg/kg,the iodized salt coverage rate was 98.36%,iodized salt qualification rate was 95.97%,and qualified iodized salt consumption rate was 94.49% weighted by population.The difference of salt iodine was statistically significant between 2011 and 2013 (x2 =17 830.03,P < 0.05).③Urinary iodine monitoring:a total of 8 278 urinary samples were detected in 2011; the median urinary iodine was 241.10 μg/L.Among these,889 urinary samples below 100.00 μg/L,the proportion was 10.74%; 2 174 urinary samples in 100.00 -< 200.00 μg/L,the proportion was 26.26%; 2 451 urinary samples in 200.00-< 300.00 μg/L,the proportion was 29.61%; and 2 764 urinary samples ≥300.00 μg/L,the proportion was 33.39%.A total of 10 988 urinary samples were tested in 2013; the median urinary iodine was 200.35 μg/L Among these,1 716 urinary samples below 100.00 μg/L,the proportion was 15.62%; 3 745 urinary samples in 100.00-< 200.00 μg/L,the proportion was 34.08%;2 970 urinary samples in 200.00-< 300.00 μg/L,the proportion was 27.03%; and 2 557 urinary samples ≥300.00 μg/L,the proportion was 23.27%.The difference of urinary iodine was statistically significant between 2011 and 2013 (x2 =391.98,P < 0.05).Conclusions Guangxi belongs to an area with low iodine level.The situation of iodine deficiency disorders is in accordance with the national Standard to Eliminate Iodine Deficiency Disorders.Scientific salt iodization and sustained elimination of iodine deficiency disorders should continue to ensure appropriate levels of iodine nutrition among residents in Guangxi.

Objective To study the impacts of variation of folate metabolic component including folic acid (FA), riboflavin (RF) and MTHFR C677T polymorphism on chromosome 8 and 17 segregation. Method RPMI1640 medium was modified with different combinations of low (LF) and high (HF) concentrations of folic acid (20, 200 nmol/L) and low (LR) and high (HR) concentrations of riboflavin (1 and 500 nmol/L). The lymphocytes with different MTFHR C677T genotype were cultured in the mentioned media for 9d. Cytokinesis was blocked by cytochalasin B and the slide was prepared as usual. The frequencies of aneuploidy of chromosomes 8 and 17 were determined by dual-color fluorescence in situ hybridization (FISH). BACKGROUND:As a kind of undifferentiated precursor cells,the phenotypic differentiation of bone marrow mesenchymal stem cells (BMSCs) remains immaturity,thus it presents weak rejection following transplantation.However,the in vitro directional differentiation of BMSCs into neuronal cells is easy affected by various factors.OBJECTIVE:To observe the immunomodulatory effect and the potential of BMSCs differentiate into neuronal-like cells.DESIGN,TIME AND SETTING:A contrast observation was conducted at the Department of Cytology,Third People's Hospital of Wuxi from January 2008 to March 2009.MATERIALS:Bone marrow was harvested from chips of cancellous or ilium bone dudng hip joint surgery.METHODS:Firstly,the BMSCs were separated and cultured to establish mixed lymphocyte reaction (MLR) system.Secondly,2 samples of peripheral blood mononuclear cells (1×10~5/well) were added into 96-well plate,and then BMSCs treated by mitomycin were added according to different ratios (BMSCs/peripheral blood monouclear cells).At the end,the cells were cultured as follows:Method 1:DMEM+10% fetal calf serum+1 μmol/L RA +20 μg/L basic fibroblast growth factor+20 μg/L epidermal growth factor.Method 2:DMEM+2% dimethyl sulfoxide +100 μmol/L butylated hydroxyanisole.MAIN OUTCOME MEASURES:The growth rate of lymphocyte was measured by ~3H-Thymidine,and the effect of BMSCs on lymphocyte proliferation was observed.Additionally,the differentiation potential of BMSCs into neuronel cells was determined by immunofluorescenca and immunohistochemistrical staining.RESULTS:①The BMSCs inhibited lymphocyte proliferation in MLR system and the influence on proliferation of lymphocyte was direct related to ratio of BMSCs.②Under a light microscope,cytoplasm of BMScs were shrinkd,which presented typical perikaryon morphology at hour 2 after culture with method 1.The majority of BMSCs were formed neuronal-like cells without number changes at hours 3-5,which turned to be dipolar or multipolar neuronal shapes at day 3.There were 60%-70% neuronspecific enolase,45%-50% glial fibrillary acidic protein were positive expressed.However,the positive rate of nidogen was decreased 3.4%.Cells cultured with method 2 became smaller after 2 hours,formed dipolar or multipolar body cells,and most of cells were died after 48 hours.The 40%-50% neuronspecific enolase,35%-40% glial fibrillary acidic protein was found to be positive.The positive rate of nidogen was temporary increased to 63% at hour 2 after culture;however,it was decreased to 1.6% after 48 hours.CONCLUSION:BMSCs can differentiate into neuronal-like cells,as well as inhibit lymphocyte proliferation in MLR system,which possess down regulation effect on alloimmunity-reaction.

8.Pitfall in the detection of acute lesions of transient ischemic attack with fluid-inversion prepared diffusion weighted imaging

Objective To explore the limits of fluid-inversion prepared diffusion weighted imaging (FLIPD) in detection of acute cerebral ischemic lesions.Methods From January 2012 to March 2014,forty-nine patients (33 males,16 females,age (55.6± 12.3) years) clinically diagnosed as transient ischemic attack (TIA) were included.Patients underwent brain MRI (conventional diffusion weighted imaging (DWI) and FLIPD) within 3 d after the onset of TIA.The detection ability of MRI with the two sequences was compared,and the relative signal intensity (rSI) and apparent diffusion coefficient (ADC) of acute ischemic lesions based on two sequences were compared.Kappa test and two-sample t test were used to analyze the data.Results A total of 87 acute ischemic lesions were detected in 21 patients by conventional DWI,and 54 were detected in 19 patients by FLIPD (Kappa=0.916,P<0.05).The rSI of ischemic lesions on FLIPD was significantly lower than that on conventional DWI (1.37±0.22 vs 1.57±0.26;t=6.647,P<0.001).The ADC value of ischemic lesions on FLIPD was slightly lower than that on conventional DWI:(0.54 ±0.10) ×10-3 mm2/s vs (0.57±0.13)×10-3 mm2/s (t=2.120,P<0.05).The missed lesions on FLIPD were located in the white matter (n =18),cerebellum and brainstem (n =8),and the cortex (n =7).Conclusions A slight diffuse abnormality may be missed on FLIPD,so this method is not suitable for the detection of acute ischemic lesions.FLIPD technology still needs improvement.
