Objective To detect the expression of hypoxia inducible factor-lα (HIF-1α) in both mRNA and protein and reactive oxygen species (ROS) levels in human epithelial ovarian cancer,and investigate the abnormal state of oxygen metabolism and their interaction in human epithelial ovarian cancer develoment.Methods The expression of HIF-1α in mRNA and protein in epithelial ovarian cancer were detected by real time reverse transcription-polymerase chain reaction and immunohistochemical Elivision plus two-step method,and the level of ROS was detected by fluorescence microplate reader.Results The relative content of HIF-1α mRNA in the epithelial ovarian cancer group was 4.466±0.754,which was higher than that of the benign control group (6.073±1.053) (t =-7.701,P =0.000).The expression of HIF-1α mRNA was raised gradually in tissues of the benign ovarian tumor,early and advanced stage of epithelial ovarian cancer (t =5.301,P =0.000).The expression of HIF-1α mRNA in the epithelial ovarian cancer had positive correlation with grade and lymph node metastasis (t =3.724,-3.851,P =0.000).The positive expression rate of HIF-1 α protein in epithelial ovarian cancer was 63.49 ％ (40/63),which was higher than that of the benign group (U =433,P =0.005),which had positive correlation with lymph node metastasis and stage (x2 =4.289,12.360; P =0.038,0.000).The expression of ROS in the epithelial ovarian cancer was 110.239±18.414,which was higher than that of the benign group (58.366±20.360) (t =11.068,P =0.000),which had positive correlation with grade,lymph node metastasis and stage (t =-2.055,2.816,-4.138; P =0.044,0.007,0.000).There was a moderate positive correlation between the expression of HIF-1α mRNA and ROS in epithelial ovarian cancer (r =0.572,P =0.000),and that was similar in low differentiation group and advanced group,there was a relatively high positive correlation in lymph node metastasis group (r =0.703,P =0.001),there was a relatively low correlation between HIF-1α mRNA and ROS (r =0.376,P =0.002).Conclusion Hypoxia is the basic characteristic of human epithelial ovarian carcers.This study suggested that HIF-1α is associated with human epithelial overaian cancer prognosis.ROS and HIF-1α might be involved in the genesis of human epithelial ovarian cancer,and the regulation of ROS to HIF-1α may play the important roles in the invasion and metastasis process of human epithelial ovarian cancer.The expression of HIF-1α is regulated by ROS at the transcriptional and protein levels,probably mainly on protein level.

Objective To investigate immunoadsorption in the prevention of hyperacute rejection in highly sensitized recipients of renal allografts.Methods Immunoadsorption treatment was performed on 10 patients whose panel reactive antigen (PRA) was more than 40 %. The adverse effect and hyperacute rejection were observed.Results Two cases of acute rejection of renal allografts were found in highly sensitized recipients and reversed by the therapy of immunoadsorption and adjusting immuno-suppressive drugs.Conclusion Immunoadsorption treatment could effectively and safely prevent highly sensitized recipients from hyperacute rejection post-transplantation.

Objective To study the image features and clinical significance of 3-dimensional (3-D)and multiplanar reconstruction with spiral CT(SCT) in cervical spinal fractures.MethodPlain film radiography and spiral CT scanning were taken for 26 cases of cervical spinal fractures.The type of fractures, diagnositic successful rate, parameter of scanning and late stage managing methods were investigated retrospectively to find out the effective difference between the diagnosis of plain X-ray and SCT. The imagin location of fractures,sequence of cenrum, heighth of centrum, fracture line, auxillary structure and rotation/dislocation of the fractures were emphasized to evaluate the advantages of 3-D imaging.ResultIn the 26 cases, there were 4 Jefferson fractures, (in which 1 combined with articular dislocations), 17 C2 spinous process fractures, (in which 1 had failure of plain X-ray diagnostic), 1 Hangman fracture, 2 C1～2 imbalance, (in which 1 had failure of plain X-ray diagnostic).There were 2 cases of axoid longitudinal fractures without obvious dislocation. The diagnositic successful rate were 100% in SCT group, while 65% in X-ray group in which 6 cases had diagnostic failure and 3 cases had fracture mis-diagnozing as dislocation.The anatomical structure of upper cervical spine was complicated and was significantly different from other cervical spine constructions. Spiral CT could reveal high resolution images of the upper cervical spine, morphosis of vertibral canal, dislocation information of fracture mass and its relation with neighbouring tissues. Especially in C1 burst fracture,C2 dens fracture and C1～2 dislocation, SCT could provide clear 3-D information of the size of collateral mass and the dislocation degree, furthermore, images could be rotated freely to observe the fracture in any proper section. That provides important information for the decision of operation, selection of operation types and evaluation of operation dager.ConclusionThe spiral CT scan and 3D reconstruction can reveal fracture better.The advantages of SCT are suggested that this diagnostic modality may be the standard method for the initial evaluation of the upper cervical spine injury.

Objective To elucidate the regulation of e pidermal growth factor receptor (EGFR) expression by transforming growth factor (TGF)? and epidermal growth factor (EGF) in human prostate cancer cells. Methods EGFR mRNA expression and its protein level were measur ed by means of RT-PCR and Western blot respectively in three human prostate can cer cell lines,the androgen-responsive (LNCaP) and the nonresponsive (ARCaP and PC3), all being treated with exogenous EGF or TGF?. Results TGF? enhanced EGFR mRNA two-to tenfold in all three cell lines, whereas EGF increased EGFR mRNA two-to sixfold.In all the three cell lines, TGF? induc ed total EGFR protein levels were higher than EGF. Conclusions These data indicate that TGF?/EGF-EGFR path-way serves as a key growth regulator in prostate cancer.TGF?, but not EGF, would preferentially maintain an autocrine loop in human androgen-nonresponsive prostate cancer.

OBJECTIVE:To study in vitro antiviral effect of Junduqing granules. METHODS:Human laryngeal cancer Hep2 cells were inoculated with the Nancy strain of coxsackievirus B3 (CoxB3) and respiratory syncytial virus (RSV) to establish vi-rus-infected cell models. 2 mg/ml original liquid of Junduqing granules and 2 mg/ml original liquid of chlorogenic acid which were diluted according to the ratio of 1∶2-1∶256 were used to act on the virus-infected cells. Reed-Muench method was adopted to calcu-late 50% toxic concentration (TC50) and maximal atoxic concentration (TC0). Virus-infected cells were cultured with solution of Junduqing granules of 1.25,0.625,0.312 5 and 0.156 25 mg/ml and chlorogenic acid solution 0.5,0.25,0.125,0.062 5 mg/ml, and then the degree of cytopathic effect was evaluated with a microscope. Virus-infected cells were cultured with 1.25,0.625, 0.312 5 ,0.156 25,0.078 125 mg/ml solution of Junduqing granules and 0.5,0.25,0.125,0.062 5,0.031 25 mg/ml of chlorogenic acid solution,and Reed-Muench method was adopted to calculate 50% inhibitory concentration (IC50) and therapeutic index (TI) on RSV and CoxB3 cells. RESULTS:TC50 of Junduqing granules was 1.79 mg/ml and TC0 was 1.25 mg/ml,TC50 of chlorogenic ac-id was 0.71 mg/ml and TC0 was 0.5 mg/ml. Virus-infected cells grew normally when the mass concentration of solution of Jundu-qing granules was 1.25 mg/ml. IC50 of Junduqing granules was 0.22 mg/ml and TI was 8.14;IC50 of chlorogenic acid were 0.18 and 0.36 mg/ml,TI were 3.94 and 1.97. CONCLUSIONS:Junduqing granules have in vitro anti-CoxB3 and RSV effect.

40%)were observed in HLA CREGs matching and outcome of post-transplantation.Results Patients with 0,1,2 or 3 mismatching(MM) of HLA CREGs+DR were 4(28%), 6(44%)and 4(28%)cases respectively according to the the rule of CREGs matching and no case had 3～6 MM.However the cases of 0,1,2,3 and 4 MM were 1(7%),3(21%),5(36%)and 5(36%)respectively by the standard of conventional HLA antigen matching,without 4～6 MM and only 4 cases had shared 0～1MM.Only 9 patients were developed into acute rejection, and were reversed by OKT3 treatment after transplantation.Renal function was returned to normal in all patients.Conclusions Using CREGs matching criteria would significantly increase the chance of recipients to receive well-matched kidney and provide more chance for waiting recipients.Suitable HLA matching could play an important role in reducing the incidence of acute rejection and improving graft survival in sensitized patients.

Objective To investigate the etiological diagnosis and therapy of liver function lesion after kidney transplantation.Methods By analyzing the etiological factor of 82 cases of liver function lesion,we provided a reasonable and operative therapy scheme in kidney post-transplantation.Results Of 82 cases,the number of drug toxicity and viral hepatitis B which result in liver functional lesion were 70 cases(85 5%) and 12 cases(14 5%) respectively.The former were all cured,8 cases in the latter were cured and the other 4 cases were died from acute hepatic failure.Conclusions The liver function lesion of post-transplantation should be diagnosed carefully and treated particularly.Morever,the adjustment of immuno-suppressive drugs and monitoring circlosporin A concentration should be done for the patients with liver function damaged.

Objective To observe the role of preoperative single administration of zenapax in the prevention of acute rejection after kidney transplantation, and evaluate its safety. Methods A total of 80 renal transplant recipients during the same period were divided into treatment and control groups, each group containing 40 patients. There were no difference between the two groups in treatment regime except the preoperative single administration of zenapax in the treatment group. Results Acute rejection rate was significantly lower in the treatment group(7.5%) than the control group(25%) within 6 months after kidney transplantation(P

0.05). It could protect the survival nephrons. No obvious toxic and side effects were found. It is indicated that Niaoduling capsule is safe and effective for CRF.

Objective To analysis curative effect of triple therapy and sequential therapy for eradication of Helicobacter pylori in patients with long-term aspirin use.Methods 52 patients with long-term aspirin use who were diagnosed with helicobacter pylori infection were collected.All patients were divided into sequential group and triple group according to different drugs classified, each 26 cases in each group were given corresponding drug treatment, after the end of treatment, the serum pepsinogen, Hp-IgG antibody levels and Helicobacter pylori clearance rate were detected in all patients. Results After treatment, compared with the triple Group, in the sequential group, the levels of serum PG I and PG II were lower, and PGⅠ/PGⅡwas higher, and the differences were statistically significant (P<0.05);the serum Hp-IgG antibody levels were lower in the sequential group (P<0.05);the Hp clearance rate of patients in the sequential group was higher (P<0.05).Conclusion Compared with triple therapy, sequential therapy can lower serum PG, PG II and Hp-IgG levels, and improve the Hp clearance rate and clinical efficacy, have guiding significance to clinical.
