3.Investigation on Production of Cluster Ions UsingSurface-Assisted Laser Desorption/Ionization Mass Spectrometry
Chinese Journal of Analytical Chemistry 2017;45(6):851-855
Surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) has become an indispensable tool in macromolecule and small molecule analysis for solid and liquid samples. However, there were few studies focusing on the ionization mechanisms especially for the small molecules. In this work, the compounds of pyrene, coronene and rubrene which have similar molecular structures were used to investigate the ionization mechanism via SALDI-MS.Additionally the effect of the laser power on the product ions was also investigated. It was found that strong cluster ions peaks nM+(M=pyrene, coronene) were observed for pyrene and coronene, and daughter ions of coronene by loss of C2H2 were also observed. On the contrary, no cluster ion was obtained for rubrene, only daughter ions with the losses of nC6H5 (n=1, 2, 3) were acquired. Finally, the ionization mechanisms were discussed. The formation of clusters of pyrene and coronene was attributed to the interaction of π-π bands. For rubrene, the spatial barrier weakened the interaction of π-π bands because the four phenyl groups were not on the same plane of skeleton structure, thus impeding the formation of cluster ions.

Chinese Journal of Tissue Engineering Research 2015;(39):6309-6314
BACKGROUND:Bony and structural feature often cause pulout strength decrease of pedicle screw, which induces loosening and pulout, and finaly results in fixation failure. Thus, it is very important to elevate the stability of pedicle screw.
OBJECTIVE:To detect the biomechanical stability of bone cement injectable canulated pedicle screw, and to provide reference for bone cement dosage.
METHODS: We selected T11-L4 samples of seven fresh adult corpses, containing 40 vertebral bodies. They were randomly divided into bone cement injectable canulated pedicle screw group and DTPSTM pedicle screw group (n=20). After screw implantation, 1, 2, 3 and 5 mL bone cement was injected. The diffuse distribution of bone cement was observed by imaging. The maximum axial pulout strength was measured.
RESULTS AND CONCLUSION:When the dose of bone cement was 1-3 mL, the average maximum axial pulout strength was significantly greater in the bone cement injectable canulated pedicle screw group than in the DTPSTM pedicle screw group (P < 0.05). When the bone cement dosage was 5 mL, no significant difference in the maximum axial pulout strength was detected between the two groups (P > 0.05). The regression equation was Y=25.269X+133.681 (R2=0.837) in the bone cement injectable canulated pedicle screw, and Y=32.039X+99.251 (R2=0.936) in the DTPSTM pedicle screw group. When the dosage of bone cement was 1-5 mL, the maximum axial pulout strength was highly positively correlated with bone cement dosage (|R| > 0.8). These results suggested that bone cement augmentation pedicle screw could apparently elevate the stability of the screw. The maximum axial pulout strength of the pedicle screw was positively correlated with bone cement dosage. After reaching the satisfactory fixation effects, the bone cement injectable canulated pedicle screw can reduce bone cement dosage, diminish the risk of bone cement leakage, and have more advantages than DTPSTM pedicle screw.

Objective To elucidate the regulation of e pidermal growth factor receptor (EGFR) expression by transforming growth factor (TGF)? and epidermal growth factor (EGF) in human prostate cancer cells. Methods EGFR mRNA expression and its protein level were measur ed by means of RT-PCR and Western blot respectively in three human prostate can cer cell lines, the androgen-responsive (LNCaP) and the nonresponsive (ARCaP and PC3), all being treated with exogenous EGF or TGF?. Results TGF? enhanced EGFR mRNA two-to tenfold in all three cell lines, whereas EGF increased EGFR mRNA two-to sixfold. In all the three cell lines, TGF? induc ed total EGFR protein levels were higher than EGF. Conclusions These data indicate that TGF?/EGF-EGFR path-way serves as a key growth regulator in prostate cancer.TGF?, but not EGF, would preferentially maintain an autocrine loop in human androgen-nonresponsive prostate cancer.

Medical Journal of Chinese People's Liberation Army 1983;0(05):-
Objective To investigate the early and long-term effects of enhanced ischemia/reperfusion injury on the transplanted abdominal aorta. Methods Abdominal aorta grafts from Sprague-Dawley (SD) rats were cold stored for 1 or 24 hours, and they were orthotopically transplanted to SD or Wistar recipients. The pathohistological changes and the expression of TGF-? 1 in the grafts were observed. The levels of serum lipid peroxides before and after transplantation were also measured. Results The intima was significantly thicker in aorta transplants which was cold stored, both in SD→SD and SD→Wistar groups 10 weeks and 6 weeks after transplantation, whereas grafts which were cold stored for 24 hours showed pronounced thickening as early as 2 weeks after transplation. Serum lipid peroxides levels were elevated significantly 2 hours post-transplantation in all groups, but they were lowered 4 and 24 hours post-transplantation. The expression of TGF-? 1 in 24 hours of ischemia became stronger 1 week after transplantation regardless the difference in strains of the animal. Conclusions The enhanced ischemia/reperfusion injury can aggravate the infiltration of inflammatory cells, intensify the expression of TGF-? 1, accelerate the thickening of intimal layer.
